Enhanced Expression of TREK-1 Is Related with Chronic Constriction Injury of Neuropathic Pain Mouse Model in Dorsal Root Ganglion

  • Neuropathic pain is a complex state showing increased pain response with dysfunctional inhibitory neurotransmission. The TREK family, one of the two pore domain $K^+$ (K2P) channel subgroups were focused among various mechanisms of neuropathic pain. These channels influence neuronal excitability and are thought to be related in mechano/thermosensation. However, only a little is known about the expression and role of TREK-1 and TREK-2, in neuropathic pain. It is performed to know whether TREK-1 and/or 2 are positively related in dorsal root ganglion (DRG) of a mouse neuropathic pain model, the chronic constriction injury (CCI) model. Following this purpose, Reverse Transcription Polymerase Chain Reaction (RT-PCR) and western blot analyses were performed using mouse DRG of CCI model and compared to the sham surgery group. Immunofluorescence staining of isolectin-B4 (IB4) and TREK were performed. Electrophysiological recordings of single channel currents were analyzed to obtain the information about the channel. Interactions with known TREK activators were tested to confirm the expression. While both TREK-1 and TREK-2 mRNA were significantly overexpressed in DRG of CCI mice, only TREK-1 showed significant increase (~9 fold) in western blot analysis. The TREK-1-like channel recorded in DRG neurons of the CCI mouse showed similar current-voltage relationship and conductance to TREK-1. It was easily activated by low pH solution (pH 6.3), negative pressure, and riluzole. Immunofluorescence images showed the expression of TREK-1 was stronger compared to TREK-2 on IB4 positive neurons. These results suggest that modulation of the TREK-1 channel may have beneficial analgesic effects in neuropathic pain patients.

Physicochemical Characteristics of Medicinal Herbs Ganjang (약용식물 추출물을 첨가한 간장의 이화학적 특성)

  • We investigated the physicochemical properties of Ganjang added to a medicinal herb extract (functional Ganjang). 1be proximate compositions of conventional Ganjang (control) and functional Ganjang (supplemented with 3% or 5% of an hem extract) were not significantly different Analysis of total and free amino acids indicated 17 different amino acids in both types of Ganjang. Glutamic acid was the most common, followed by valine, leucine, isoleucine, lysine, methionine, phenylalanine, and threonine. Sensory tests of conventional and functional Ganjang found no significant differences. An analysis of flavoring components indicated that flavor-active compounds (paeonol, senkyunolide A, and cnidilide) were present in both forms of Ganjang. Our results suggest that the extract could be used to manufacture fermented soybean products.

  • Two-dimensional materials offer unique characteristics for membrane applications to water technology. With its atomic thickness, availability and stackability, graphene in particular is attracting attention in the research and industrial communities. Here, we present a brief overview of the recent research activities in this rising topic with bringing two membrane architecture into focus. Pristine graphene in single- and polycrystallinity poses a unique diffusion barrier property for most of chemical species at broad ambient conditions. If well designed and controlled, physical and chemical perforation can turn this barrier layer to a thinnest feasible membrane that permits ultimate permeation at given pore sizes. For subcontinuum pores, both molecular dynamics simulations and experiments predict potential salt rejection to envisage a seawater desalination application. Another novel membrane architecture is a stack of individual layers of 2D materials. When graphene-based platelets are chemically modified and stacked, the interplanar spacing forms a narrow transport pathway capable of separation of solvated ions from pure water. Bearing unbeknownst permeance and selectivity, both membrane architecture - ultrathin porous graphene and stacked platelets - offer a promising prospect for new extraordinary membranes for water technology applications.

Carcinoembryonic Antigen (CEA) in the Gallbladder Bile (b-CEA) of Gastric Carcinoma Patients with Long-term Follow up (위암 환자의 담즙 CEA 농도와 장기 생존율 및 간전이와의 연관성)

  • Purpose: Despite numorous reports on the relationship between the level of carcinoembryonic antigen (CEA) in gall bladder bile and liver metastasis in colorectal cancer, no similar studies have been carried out for gastric carcinomas. We, therefore, undertook the present study to establish the relationship between the gall bladder bile CEA and liver metastasis as well as the post-operative survival rate in gastric carcinoma patients with curative resections. Materials and Methods: In 373 gastric cancer patients (252 males, 121 females, age $21\∼76$ years) operated on at Kosin University Hospital between 1989 1996, the CEA concentration in the gall bladder bile was determined during the operation and the value was related to the rates of post-operative survival and liver metastasis during follow-up period. Results: The overall rate of patient survival decreased gradually with increase in TNM stage. The 13-year postoperative survival rates for stages Ia, Ib, II, IIIa, and IIIb were $95.7\%,\;92.5\%,\;79.9\%,\;50.9\%,\;and\;43.3\$, respectively, and the 10-year survival rate for stage IV was $22.6\%$. The patients with a high ($\geq$10 ng/ml) biliary CEA showed a significantly lower rate of survival than those with a low (<10 ng/ml) biliary CEA. The 13-year cumulative survival rate was $55.4\%$ for the high CEA group and $76.5\%$ for the low CEA group (P<0.01). Also, the patients with a high biliary CEA showed a significantly higher rate ($11.5\%$) of liver metastasis than those with a low biliary CEA ($1.9\%$) (P<0.000). In patients with TNM stages (I and II), the CEA level did not affect the post-operative survival rates ($95.4\%\;and87.7\%$ in the high and low CEA groups, P>0.10), but in those with high TNM stages (III and IV), the survival rate was significantly lower in the high CEA group ($25.9\%$) than in the low CEA group ($57.8\%$) (P<0.05). Conclusion: These result suggest that the gall bladder bile CEA level obtained in an advanced-staged gastric cancer operation may be used in predicting the post-operational survival rate and in sorting out patients with a high risk for cancer recurrence, especially in the liver area.

Family history of chronic renal failure is associated with malnutrition in Korean hemodialysis patients

  • The present study was to investigate the nutritional status and factors related to malnutrition in end-stage renal disease (ESRD) patients requiring hemodialysis (HD) in South Korea. Subjects were ESRD outpatients from general hospitals or HD centers in Seoul referred to the dialysis clinic for maintenance HD care. A total of 110 patients (46 men and 64 women; mean ages $58.6{\pm}1.0y$) were eligible for this study. The family history of chronic renal failure (CRF) was considered positive if a patient reported having either a first-degree or second-degree relative with CRF. Malnutrition was defined as a triceps skinfold thickness or mid-ann muscle circumference below the fifth percentile for age and sex and forty-seven of the 110 patients were malnourished. Almost all (94%) patients had anemia (hemoglobin: <13 g/dL for men and <12 g/dL for women). Energy intake was below the recommended intake levels of energy [30-35 kcal/kg ideal body weight (IBW)] and protein (1.2 g/kg IBW) in 60% of patients. The duration of HD was longer in malnourished HD patients (P=0.0095). Malnutrition was more prevalent in women (P=0.0014), those who never smoked (P=0.0007), nondiabetic patients (P=0.0113), and patients with bone diseases (P=0.0427), adequate HD (spKt/$V{\geq}1.2$) (P=0.0178), and those with a family history of CRF (P=0.0255). Multiple logistic regression was used to examine the relationship between malnutrition and potential risk factors. After adjusting for age, sex, and other putative risk factors for malnutrition, the OR for malnutrition was greater in HD patients with a family history of CRF (OR, 3.290; 95% CI, $1.003{sim}10.793$). Active nutrition monitoring is needed to improve the nutritional status of HD patients. A family history of CRF may be an independent risk factor for malnutrition in Korean HD patients. A follow-up study is needed to investigate whether there is a causal relationship between a family history of CRF and malnutrition in Korean ESRD patients.

Seroprevalence of Dogs with Dirofilaria immitis, Anaplasma phagocytophilum, Borrelia burgdorferi and Ehrlichia canis Infection in the Daejeon City and Kangwon Province (대전시와 강원도에서 심장사상충증, 아나플라즈마증, 보렐리아증 및 얼리키아증의 혈청학적 발생율 조사)

  • This study was conducted to survey Dirofilaria (D.) immitis antigen, and Anaplasma (A.) phagocytophilum, Borrelia (B.) burgdorferi, and Ehrlichia (E.) canis antibodies among clinically healthy dogs in the Daejeon city and Kangwon province. Whole blood samples were collected from two hundred and fifty two dogs (127 females and 125 males, 147 outdoors and 105 indoors), and they were tested by using ELISA kit (SNAP 4Dx, IDEXX Laboratories, USA). Chi-squared analysis showed that A. phagocytophilum of female dogs in outdoors group was significantly higher prevalence than that of female dogs in indoors group (p < 0.05). In addition, infection rate of A. phagocytophilum of over 7 year-old-dogs in outdoors group revealed significantly higher prevalence than that of indoors group (p < 0.01). In E. canis, the under 4 year-old-dogs (p < 0.05), 4~7 year-old-dogs (p < 0.05) and over 7 year-old-dogs (p < 0.01) in outdoor group showed significantly higher prevalence than that of indoors group, respectively. In conclusion, this study is the large scale survey of canine D. immitis antigen, A. phagocytophilum, B. burgdorferi, and E. canis antibodies in the Daejeon city and Kangwon province, and it provides an useful reference for clinicians.

The Performance of Anaerobic Co-digester of Swine Slurry and Food Waste

  • In order to assess the performance of co-digester using pig slurry and food waste at the farm scale biogas production facility, the anaerobic facility that adopts the one-stage CSTR of 5 $m^3\;day^{-1}$ input scale was designed and installed under the conditions of the OLR of 2.33 kg $m^3\;day^{-1}$ and HRT of 30 days in an pig farmhouse. Several operation parameters were monitored for assessment of the process performance. The anaerobic facility was operated in three stages to compare the performance of the anaerobic co-digester. In the Stage I, that was fed with a mix of pig slurry to food waste ratio of 7:3 in the input volume, where input TS content was 4.7 (${\pm}0.8$) %, and OLR was 0.837-1.668 kg-VS $m^3\;day^{-1}$. An average biogas yield observed was 252 $Nm^3\;day^{-1}$ with methane content 67.9%. This facility was capable of producing an electricity of 626 kWh $day^{-1}$ and a heat recovery of 689 Mcal day-1. In Stage II, that was fed with a mixture of pig slurry and food waste at the ratio of 6:4 in the input volume, where input TS content was 6.9 (${\pm}1.9$) %, and OLR was 1.220-3.524 kg-VS $m^3\;day^{-1}$. The TS content of digestate was increased to 3.0 (${\pm}0.3$) %. In Stage III, that was fed with only pig slurry, input TS content was 3.6 (${\pm}2.0$) %, and OLR was 0.182-2.187 kg-VS $m^3\;day^{-1}$. In stage III, TS and volatile solid contents in the input pig slurry were highly variable, and input VFAs and alkalinity values that affect the performance of anaerobic digester were also more variable and sensitive to the variation of input organic loading during the digester operation. The biogas produced in the stage III, ranged from 11.3 to 170.0 $m^3\;day^{-1}$, which was lower than 222.5-330.2 $m^3\;day^{-1}$ produced in the stage II.

Evaluation of energy and amino acids of brown rice and its effects on laying performance and egg quality of layers

  • Two experiments were conducted to determine apparent metabolizable energy (AME), nitrogen-corrected AME (AMEn), and ileal digestible amino acid (AA) content of brown rice (BR) and to investigate the effect of dietary supplementation of BR on laying performance and egg quality of laying hens. In Exp. 1, 72 Hy-line Brown layers (49-week-old) were allocated to two treatments using a completely randomized block design, and each treatment included six cages per treatment and six hens per cage. A semi-purified diet was formulated to include BR as the sole source of AA and energy and an N-free diet was used to determine basal endogenous loss of AA. The hens were fed a commercial layer diet for adaptation to the experimental environment and diet for 7 days from d 0, and then fed experimental diets for 5 days from d 7. Excreta were collected from d 10 to 11 and ileal digesta were collected on d 12. On a dry matter (DM) basis, the AME and AMEn of BR was determined at 3,773 and 3,729 kcal/kg, respectively. The apparent ileal digestibility (AID) of BR ranged from 32.7% for Thr to 73.7% for Arg. The range of the standardized ileal digestibility (SID) value was between 79.4% for Met and 96.6% for Lys. In Exp. 2, 252 Hy-line Brown layers (44-week-old) were divided into four groups, comprising seven replicates of nine birds each and assigned to four experimental diets containing 0 (Control), 5%, 10%, or 15% BR for 5 weeks. The BR-containing diets were formulated to be equal in the content of AMEn and digestible AA to those of the diet without BR. No significant differences were observed in laying performances. Egg quality and blood profiles were not linearly or quadratically affected by dietary treatments. These results suggest that up to 15% BR can be included into layer feed without any adverse effects on laying performance and egg quality, if its energy and digestible AA values are well evaluated.

Cutting Propagation and Seedling Growth Effect According to Fertilizer Application of Elsholtzia minima Nakai (좀향유의 삽목 증식 및 시비에 따른 유묘의 생장 효과)

  • This study was performed to establish a production system for in situ and ex situ conservation of Elsholtzia minima Nakai, an endemic plant grown in Jeju Island. Moreover, this study aimed to identify root-growth characteristics according to the use of pre-treatment agents and seedling growth effect according to fertilizer application. The mean temperature was similar in greenhouse and vinyl-moist chamber, but air humidity was higher in vinyl-moist chamber than in greenhouse. After stem planting of Elsholtzia minima Nakai, initial root growth was observed after 10 days in greenhouse and after 7 days in vinyl-moist chamber. Root growth rate was more rapid in vinyl-moist chamber. Moreover, survival rate, root growth rate and root number was slightly higher in vinyl-moist chamber than in greenhouse, indicating that vinyl-moist chamber is more effective in plant growth. When pre-treatment agents were used to remove root growth-inhibiting substances, a higher root growth rate of more than 95% was found in pre-treatment groups, excluding the group treated with AgNO3 at 77.5%. Thus, Elsholtzia minima Nakai is thought to have less root growth inhibitors. In the analysis of nitrogen application rate and Osmocote application by seedling container, a difference was found in survival rate and growth according to application rate and container conditions. When Osmocote, a slow release fertilizer, was applied to the soil surface around each culture container, survival rate and the growth of aerial and root parts were most favorable. Thus, Osmocote fertilizer is thought to be desirable for seedling propagation of Elsholtzia minima Nakai.

Derivation of Neural Precursor Cells from Human Embryonic Stem Cells

  • Human embryonic stem (ES) cells are derived from the inner cell mass of the preimplantation embryo. Human ES cells have the capacity to differentiate into various types of cells in the body. Human ES cells are indefinite source of cells for cell therapy in various degenerative disorders including neuronal disorders. Directed differentiation of human ES cells is a prerequisite for their clinical application. The objective of this study is to develop the culture condition for the derivation of neural precursor cells from human ES cells. Neural precursor cells were derived from human ES cells in a stepwise culture condition. Neural precursor cells in the form of neural rosette structures developed into neurospheres when cultured in suspension. Suspension culture of neurospheres has been maintained over 4 months. Expressions of nestin, soxl, sox2, pax3 and pax6 transcripts were upregulated during differentiation into neural precursor cells by RT-PCR analysis. In contrast, expression of oct4 was dramatically downregulated in neural precursor cells. Immunocytochemical analyses of neural precursor cells demonstrated expression of nestin and SOX1. When induced to differentiate on an adhesive substrate, neuro-spheres were able to differentiate into three lineages of neural systems, including neurons, astrocytes and oligo-dendrocytes. Transcripts of sox1 and pax6 were downregulated during differentiation of neural precursor cells into neurons. In contrast, expression of map2ab was elevated in the differentiated cells, relative to those in neural precursor cells. Neurons derived from neural precursor cells expressed NCAM, Tuj1, MAP2ab, NeuN and NF200 in immunocytochemical analyses. Presence of astrocytes was confirmed by expression of GFAP immuno-cytochemically. Oligodendrocytes were also observed by positive immuno-reactivities against oligodendrocyte marker O1. Results of this study demonstrate that a stepwise culture condition is developed for the derivation of neural precursor cells from human ES cells.
