Nucleotide Analysis of Phaffia rhodozyma DNA Fragment That Functions as ARS in Saccharomyces cerevisiae

  • The chromosomal DNA fragment from Phaffia rhodozyma CBS 6938 which is able to autonomously replicate in the yeast Saccharomyces cerevisiae was cloned on an integrative URA3 plasmid. Its minimal fragment exhibiting autonomously replicating activiy in the S. cerevisiae gave a higher frequency transformation efficiency than that found for centromere-based plasmid, and enabled extrachromosoma1ly stable transmission of the plasmids in one copy per yeast cell under non-selective culture condition. The 836-bp DNA element lacked an ORF and did not contain any acceptable match to an ARS core consensus. Sequence analysis, however, displayed a cluster of three hairpin-Ioop-sequences with individual $\triangle {G_{25}}^{\circ}C$ free energy value of -10.0, -17.5, and -17.0 kcal. $mor^{-l}$as well as a 9-bp sequence with two base pair mismatches to the S. cerevisiae/E. coli gyrase-binding site. This 836-bp sequence also included one 7-bp sequence analogous to the core consensus of centromeric DNA element III (CDEIII) of S. cerevisiae, but CDEIII-like 7 bp sequence alone did not give a replicative function in this yeast.

  • Sigma delta modulation has been the preferred technique for oversampling conversion. In this paper we present the basic principles of oversampled sigma-delta Converters. Basic operation and theory behind sigma-delta modulation is reviewed. The different structures of the sigma-delta converters are described and the concepts of designing modulators and digital filters are discussed. The latest designs are also reviewed.

  • In this paper, we introduce the concepts of fuzzy r-pre-semiopen and r-pre-semiclosed sets. With them we define fuzzy r-pre-semiinterior and r-pre-semiclosure. We also introduce and investigate the properties of a fuzzy r-pre-semicontinuous map, a fuzzy r-pre-semiopen map and a fuzzy r-pre-semiclosed map. These concepts are generalizations of the Bai Shi-Zhongs fuzzy pre-semicontinuity.

  • In this paper, an analysis has been done on the performance of SCTP header compression by using Robust Reader Compression (ROHC)[1] method. And it is assumed that the operating mode for ROHC is unidirectional mode (U-Mode) and the possible states are IR and SO states. The throughput of SCTP packets in wireless link and the impact of size of W-LSB encoding window on throughput are discussed.

  • This paper presents a transmission cost allocation through application of a new reliability index in transmission pricing in competitive electric industry. The proposed method allocates a fair use of transmission system charge given separately to individual generator by capacity usage, based on the contribution of individual generator considering N-1 contingency in reliability margin of transmission capacity, and offers more alternatives of pricing in using transmission capacity and transmission margin.

Cloning of Autonomously Replicating Sequence from Phaffia rhodzyma

  • A Phaffia rhodozyma chromosomal fragment (approximately 3.8 kb) capable of functioning as an origin for the replication of a kanamycin resistance ($Km^r$) plasmid in S. cerevisiae was isolated by the use of origin search plasmid, pHN134. In S. cerevisiae, transformation frequencies using the plasmid pHN134 containing an autonomously replicating sequence of P. rhodozyma was 450-580 CFU/$\mu g$ DNA. The stability of the recombinant plasmid were 16-19$\%$.

The Study of the Dyeability of Brassica Campestris on the Cotton Fabric (유채꽃잎을 이용한 면직물에서의 염색성)

  • The dyeability of the cotton fabric with Brassica campestris extract was investigated. The colorant was extracted with methanol. Cotton fabrics were dyed at various conditions such as temperatures, concentrations, dyed times, and mordanting methods. The maximum wavelength of extract was 421nm. The highest K/S value was showed at 200% dye concentration at $60^{\circ}C$, 45 minutes. As the effect of dyed temperature and mordanting on dyeability was not great, the Brassica campestris was one-color dyestuff.
