Molecular Cloning and Characterization of a Novel Metallothionein Isoform Expressed in Tiger Shark(Scyliorhinus torazame) (두툽상어(Scyliorhinus torazame)Metallothionein cDNA의 cloning 및 이의 분자적 특성)

  • A complementary DNA encoding metallothionein(MT), a heavy metal-responsive protein was cloned from a cartilaginous shark species. Scyliorhinus torazame. An expressed sequence tag(EST)from the shark liver, which showed high similarity with a MT gene, was isolated and its full-length sequence(390bp)was determined. The putative shark MT cDNA sequence contained an open reading frame consisting 68 amino acids and 182bp of 3-untranslated region including the poly (A+) signal. The deduced amino acid sequence was 41-54% identical to those of other animals including mammals and fish species. Tiger shark MT cDNA showed high conservation in the Cys regions. however, peculiarly contained not only additional five amino acids just prior to the conserved beta-domain but also a Ser residue at C terminal, which has not been seen in other MT sequences.

Constructions of a Transfer Vector Containing the gX Signal Sequence of Pseudorabies Virus and a Recombinant Baculovirus

  • Constructions of a transfer vector and a recombinant baculovirus using the thymidine kinase gene of the Herpes simplex virus type 1 strain F (HSV -1) were carried out. Newly cloned transfer vector, pHcgXIIIB, was constructed by insertion of the glycoprotein gX gene signal peptide sequence of Pseudorabies virus into the baculovirus vector pHcEV-IV. The gX sequence was inserted just downstream from the promoter for the polyhedrin gene of the Hyphantria cunea nuclear polyhedrosis virus (HcNPV). HSV-1 thymidine kinase(tk) gene (1.131 kb) was used as a candidate gene for transferring into the baculovirus expression system. The tk gene was inserted into a BamHI site downstream from the gX sequence-promoter for the polyhedrin gene in the pHcgXIIIB transfer vector and was transferred into the infectious lacZ-HcNPV expression vector. Recombinant virus was isolated and was named gX-TK-HcNPV. The recombinant virus produced a 45 kDa gX-TK fusion protein in Spodoptera frugiperda cells, which was confirmed by Western blot analysis. Microscopic examination of gX-TK-HcNPV-infected cells revealed normal multiplication. Fluorescent antibody staining indicated that the gX-TK fusion protein was present in the cytoplasm. These results indicated that the transfer vector successfully transferred the gX-tk gene into the baculovirus expression system.

Jo Jeongsan in Context: "Second Founders" in New Religious Movements

  • Scholars of new religious movements have emphasized the role of "second founders," such as Judge J.F. Rutherford for the Jehovah's Witnesses, Brigham Young for the Mormons, or Deguchi Onisaburo for Oomoto. They systematize and structure movements often created by the "first founders" with a minimal organization only. The paper argues that the model for the sequence first founder/second founder described by these scholars is the relationship between Jesus and Paul of Tarsus at the origins of Christianity. It proposes a comparison between Jesus of Nazareth and Kang Jeungsan, who established the tradition leading to present-day Daesoon Jinrihoe. It then summarizes the biography of Jo Jeongsan, recognized by Daesoon Jinrihoe as its "second founder" within the same tradition, and discusses the analogies between his connection to the "first founder," Kang Jeungsan, and the connection Paul of Tarsus established with Jesus Christ. The paper considers recent scholarship about Paul, often described as the "New Perspective on Pauline Scholarship." Paul never personally met Jesus Christ, except after the latter's death through a spiritual revelation, just as Jo Jeongsan never met Kang Jeungsan, except after his death, when he manifested himself to him in spirit. Nonetheless, Paul was able to decisively shape the largest branch among the followers of Jesus Christ, just as Jo Jeongsan originated the lineage leading to Daesoon Jinrihoe, currently the largest religious order among those recognizing Kang Jeungsan as the incarnated Supreme God.

Analysis of Reproduction and CO I Gene Sequence between Two Earthworm Populations Used in Vermicomposting Organic Wastes in Different Localities (서로 다른 지역에서 유기성 폐기물 처리에 이용되는 두 지렁이 집단의 생식 및 CO I 유전자 분석)

  • Two domestic earthworm populations used in vermicomposting in different lacalities were collected. The one was from Hongcheon (Kangwon province) and the other was from Youngdong (Chungcheong province). Reproductive capacities and the degree of reproductive isolation of two population were investigated. CO I gene sequences were also compared. There was no difference in their reproductive capacities. And there was no reproductive isolation between two populations. Two populations were identified as Eisenia andrei or Eisenia fetida by CO I gene biomarkers. Phylogenetic tree formulated by CO I gene sequence strongly suggested that two populations were just the same species.

First Description of the Adult Scolecenchelys aoki (Ophichthidae, Anguilliformes) Collected from the South Sea of Korea, with Comments on the Synonym Scolecenchelys borealis (한국 남해에서 채집된 동해물뱀(Scolecenchelys aoki) 성어의 첫기재 및 Scolecenchelys borealis의 동종이명에 관한 검토)

  • Scolecenchelys borealis was first added to the Korean fish fauna based on one leptocephalus specimen (total length (TL) 59.2 mm) collected from Dokdo, in the East Sea. Recently, however, Scolecenchelys borealis has been treated as a junior synonym of Scolecenchelys aoki, necessitating review of the scientific name S. borealis. This study describes the morphological characteristics of S. aoki based on an adult specimen (TL 230.0 mm) collected from the sea south of Korea and compares its mitochondrial DNA 12S rRNA sequence with that of the S. borealis leptocephalus. The adult S. aoki is characterized by several morphological features: vertebral formula 59-55-135; preanal lateral pores 55; infraorbital pores two; teeth on upper jaw visible externally when the mouth is closed; the dorsal fin originates just behind the anus; both jaws and vomerine teeth are conical, arranged in two rows. An analysis of an 886-base paire sequence of the 12S rRNA showed that adult S. aoki was concordant with the S. borealis leptocephalus (sensu Ji et al., 2012), indicating that both are the same species.

  • When human beings acquire and develop knowledge through education, their prior knowledge influences the next learning process. As this is a fact that should be considered in machine learning, we need to examine the effects of controlling the order of training sequence on machine learning. In this research, the role of the supervisor is extended to control the order of training samples, in addition to just instructing the target values for classification problems. The supervisor sequences the training examples categorized by SOM to the learning model which in this case is MLP. The proposed method is distinguished in that it selects the most instructive example from categories formed by SOM to assist the learning progress, while others use SOM only as a preprocessing method for training samples. The result shows that the method is effective in terms of the number of samples used and time taken in training.

Strengthening sequence based on relative weightage of members in global damage for gravity load designed buildings

  • Damage caused by an earthquake depends on not just the intensity of an earthquake but also the region-specific construction practices. Past earthquakes in Asian countries have highlighted inadequate construction practices, which caused huge life and property losses, indicating the severe need to strengthen existing structures. Strengthening activities shall be proposed as per the proposed weighting factors, first at the higher weighted members to increase the capacity of the building immediately and thereafter, the other members. Through this study on gravity load-designed (GLD) buildings, relative weights are assigned to each storey and exterior and interior columns within a storey based on their contribution to the energy dissipation capacity of the building. The numerical study is conducted on mid-rise archetype GLD buildings, i.e., 4, 6, 8, and 10 stories with variable storey heights, in the high seismic zones. Non-linear static analysis is performed to compute weights based on energy dissipation capacities. The results obtained are verified with the non-linear time history analysis of 4 GLD buildings. It was observed that exterior columns have higher weightage in the energy dissipation capacity of the building than interior columns up to a certain building height. The damage in stories is distributed in a convex to concave parabolic shape from bottom to top as building height increases, and the maxima location of the parabola shifts from bottom to middle stories. Relative weighting factors are assigned as per the damage contribution. And the sequence for strengthening activities is proposed as per the computed weighting factors in descending order for regular RCC buildings. Therefore, proposals made in the study would increase the efficacy of strengthening activities.

  • Recently e-commerce trade on the web has grown rapidly in scale and complexity, just as web site designs and web servers have become more complicated. In view of these complexities, it is obviously difficult to analyse web user's data since they web users employ so many different web paths. The existing association rule investigation algorithms identify all items with a high correlation. However even though users often only want to find items in which they have interest, it is still difficult to find the rules they want out of all of the many association rules found by existing algorithms. In this paper, we propose a system linking each node with the sequence association rule, linking all routes after finding a path corresponding to a user with the association rule-one of the data mining techniques which identify user patterns in web user paths. The suggested system helps us construct individualized or customer-subdivided sites using the sequence association rule in order to harmonize the paths of web users with user characters.

Effect of Post-Activation Potentiation according to Sequence of Velocity Using Isokinetic Device on Short-Term Performance of Lower Extremity: Taekwondo Athletes and Healthy Adults

  • Purpose: The purpose of this study was to figure out how PAP (Post-Activation Potentiation) phenomenon affects short-term performance efficiently. Methods: This study was conducted with 18 Taekwondo athletes and 16 healthy adults. By using isokinetic dynamometer, two different intervention, TDP (Top-down program) and BUP (Bottom-up program), were performed to measure isokinetic parameter; (peak torque: PT, total work: TW, average power: AP, and average torque: AT) of knee extensor for intragroup, intergroup comparison and two-way ANOVA. Results: The Taekwondo athletes group showed statistically significant differences in all isokinetic parameters PT, TW, AP, and AT after TDP (p<0.05). However, in the healthy adult group, the difference in isokinetic parameters according to the exercise sequence was not statistically significant. (p>0.05). PT and TW at TDP were statistically significant (p<0.05) when the rate of change in TDP and BUP was compared and analyzed considering the difference in physical ability between the Taekwondo athlete group and the healthy adult group. However, AP and AT were not statistically significant. Finally, when examining the interaction between the two groups and two exercise sequence according to isokinetic parameters, only TW (p<0.05) showed a statistically significant interaction, while PT (P=0.099), AP (P=0.103), and AT (P=0.096) did not. This study suggests that short-term performance can be improved through the PAP phenomenon when TDP is applied to the Taekwondo group. Conclusion: According to our result, for Taekwondo athletes, if the goal is to improve short-term performance just before the main game, we suggest a training program through TDP.

  • One of the important roles of a DNA chip is the capability of detecting genetic diseases and mutations by analyzing DNA sequence. For a successful electrochemical genotyping, several aspects should be considered including the chemical treatment of electrode surface, DNA immobilization on electrode, hybridization, choice of an intercalator to be selectively bound to double standee DNA, and an equipment for detecting and analyzing the output signal. Au was used as the electrode material, 2-mercaptoethanol was used for linking DNA to Au electrode, and methylene blue was used as an indicator that can be bound to a double stranded DNA selectively. From the analysis of reductive current of this indicator that was bound to a double stranded DNA on an electrode, a normal double stranded DNA was able to be distinguished from a single stranded DNA in just a few seconds. Also, it was found that the peak reduction current of indicator is proportional to the concentration of target DNA to be hybridized with probe DNA. Therefore, it is possible to realize a sim71e and cheats DNA sensor using the electrochemical measurement for genotyping.
