상엽 물추출물이 다이옥신 유도 독성에 대한 흰쥐의 혈액 생화학지표에 미치는 영향 (The Effect of Mulberry Leaf Extract on Blood biochemical parameters in White Rats Exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD))

  • TCDD 독성이 유발된 흰쥐 혈액의 생화학적 지표에 미치는 상엽 물추출물 (MLE)의 효과를 조사하였고 통계자료는 일원분산분석과 Scheffe의 사후검증을 하였다. TCDD 투여 시 감소했던 백혈구와 혈소판의 수가 MLE투여로 급격하게 회복(p<0.01)되어 면역기능 개선효과가 있었다. TT에 의해 감소했던 혈중 칼슘 및 마그네슘의 농도는 MLE 투여 시 유의하게 증가(p<0.01)하였으나 인의 농도는 TT군에 비하여 MLE군이 유의하게 감소(p<0.01)하였다. 신장 기능과 관련된 creatinine 및 blood urea nitrogen은 TT군과 MLE군 사이에 유의성은 없었으며, 요산은 TT군과 MLE군 모두에서 감소하였다. 한편 TT에 의해 증가했던 glucose 농도, amylase 및 lipase의 효소 활성도는 MLE 투여시 유의하게 감소(p<0.01)하여 혈당상승 억제효과가 있었다. Total protein, ${\gamma}$-GTP 활성은 TT와 MLE의 영향을 거의 받지 않았으나 TT에 의해 증가했던 AST, ALT 및 ALP의 활성도는 MLE 투여 시 모두 유의하게 감소(p<0.01)하여 간기능 개선의 효과가 있었다. 한편 Total cholesterol, triglyceride와 LDL-cholesterol의 경우 TT에 의해 유발된 고지혈 증상이 MLE 투여 시 유의하게 감소(p<0.01)하여 항고지혈 효과가 있었으며 HDL-cholesterol은 TT군과 MLE군 모두에서 증가하였다. 이와 같이 MLE는 TCDD독성이 유발된 흰쥐에서 독성을 완화시켜 주는 작용이 있음을 알 수 있었다.

Construction of a Thermotolerant Saccharomyces cerevisiae Strain for Bioethanol Production with Reduced Fermentation Time and Saccharifying Enzyme Dose

  • A thermotolerant Saccharomyces cerevisiae mutant strain, TT6, was constructed after multi-parental hybridization of five mutant strains obtained by UV or NTG treatment of the original strain, S. cerevisiae KV1. When incubated at $40^{\circ}C$ in YPD broth, TT6 began to grow exponentially in 10 h, but KV1 did not show any noticeable growth even after 22 h. The thermotolerant growth of TT6 was confirmed by serial dilution assay at $42^{\circ}C$; TT6 grew at a cell concentration ($10^{-5}$) 10,000 times lower than that of KV1 ($10^{-1}$). Whereas ethanol production from YP containing 23% (w/v) glucose by KV1 decreased with increasing temperature from $30^{\circ}C$ to $36^{\circ}C$, ethanol production by TT6 did not decrease at temperatures up to $37^{\circ}C$. When TT6 was tested for ethanol production at $36^{\circ}C$ by simultaneous saccharification and fermentation (SSF) from 23% corn, 24 h of fermentation time or 50% of the glucoamylase dose was saved when compared with KV1 at $30^{\circ}C$. The ethanol yield from corn by SSF with TT6 at $36^{\circ}C$ was 91.7% of the theoretical yield, whereas that of KV1 at $30^{\circ}C$ was 90.6%.

Carboxy-Terminal Region of a Thermostable CITase from Thermoanaerobacter thermocopriae Has the Ability to Produce Long Isomaltooligosaccharides

  • Isomaltooligosaccharides (IMOs) have good prebiotic effects, and long IMOs (LIMOs) with a degree of polymerization (DP) of 7 or above show improved effects. However, they are not yet commercially available, and require costly enzymes and processes for production. The N-terminal region of the thermostable Thermoanaerobacter thermocopriae cycloisomaltooligosaccharide glucanotransferase (TtCITase) shows cyclic isomaltooligosaccharide (CI)-producing activity owing to a catalytic domain of glycoside hydrolase (GH) family 66 and carbohydrate-binding module (CBM) 35. In the present study, we elucidated the activity of the C-terminal region of TtCITase (TtCITase-C; Met740-Phe1,559), including a CBM35-like region and the GH family 15 domain. The domain was successfully cloned, expressed, and purified as a single protein with a molecular mass of 115 kDa. TtCITase-C exhibited optimal activity at 40℃ and pH 5.5, and retained 100% activity at pH 5.5 after 18-h incubation. TtCITase-C synthesized α-1,6 glucosyl products with over seven degrees of polymerization (DP) by an α-1,6 glucosyl transfer reaction from maltopentaose, isomaltopentaose, or commercialized maltodextrins as substrates. These results indicate that TtCITase-C could be used for the production of α-1,6 glucosyl oligosaccharides with over DP7 (LIMOs) in a more cost-effective manner, without requiring cyclodextran.

Anticoagulant 1,2,3,4,6-Pentagalloyl-$\beta$-D-Glucopyranose Isolated from Geranium (Pelargonium inquinans Ait)

  • Geranium (Pelargonium inquinans Ait) leaves were extracted with $80\%$ MeOH, and partitioned into n-hexane, ethyl acetate, BuOH and $H_2O$ to isolate the anticoagulant principles. The EtOAc fraction was found to be the most active, and was further purified using silica and octadecylisilane column chromatography employing a bioassay-guided fractionation method. The active compound was isolated and identified as $1,2,3,4,6-pentagalloyl-\beta-D-glucopyranose$(PGG) (compound I). The isolated anticoagulant significantly prolonged the activated partial thrombin time (APTT) and thrombin time (TT) using normal human plasma. One microgram of $1,2,3,4,6-pentagalloyl-\beta-D-glucopyranose$ showed 0.063 heparin units in the APTT and 2.73 heparin units in the TT for anti-thrombosis. This is the first report of the isolation of PGG from geranium plants.

증기발생기 전열관 Alloy 690TT의 소성변형이 표면특성 및 미세조직에 미치는 영향 (Effects of Plastic Deformation on Surface Properties and Microstructure of Alloy 690TT Steam Generator Tube)

  • Denting of steam generator (SG) tube is defined as the reduction in tube diameter due to the stresses exerted by the corrosion products formed on the outer diameter surface. This phenomenon is mostly observed in the crevices between SG tube and the top-of tubesheet or tube support plate. Despite the replacement of SG tube with Alloy 690, which has better corrosion resistance than Alloy 600, the denting of SG tube still remains a potential problem that could decrease the SG integrity. Deformation of SG tube by denting phenomenon can affect the surface properties and microstructure of SG tube. In this study, the effects of plastic deformation on surface properties and microstructure of Alloy 690 thermally treated (TT) tube was investigated by using the various analysis techniques. The plastic deformation of Alloy 690 increased the surface roughness and area. Many surface defects such as ripped surface and micro-cracks were observed on the deformed Alloy 690TT specimen. Based on the electron backscatter diffraction analysis, the dislocation density of deformed SG tube increased compared to non-deformed SG tube. In addition, the effects of changes in surface properties and microstructure of SG tube on general corrosion behavior were discussed.

황금찰수수의 혈액응고저해 및 혈전용해 효과 (Anticoagulant and Fibrinolytic Activities of Hwanggeumchal Sorghum In Vitro)

  • 잡곡류의 항혈전작용을 조사하기 위해 국내산 11종의 잡곡으로부터 80% 에탄올 추출물을 얻어 혈액응고저해활성을 thrombin time (TT)법으로 측정한 결과, 황금찰수수의 에탄올 추출물이 가장 높은 혈액응고저해활성을 보였으며, 뒤이어 청차조 유래의 에탄올 추출물도 혈액응고저해활성을 보였다. 황금찰수수의 에탄올 추출물을 n-hexane, methylene chloride, ethyl acetate 및 n-butanol로 분획하였을 때, 대부분의 TT-저해활성은 hexane과 methylene chloride 분획에 분포하였으며, 이들 분획의 활성은 동일 농도의 aspirin (최종 농도 480 ${\mu}g/ml$)보다 더 높은 것으로 나타났다. 또한 황금찰수수의 에탄올 추출물의 혈액응고저해활성은 activated partial thromboplastin time (APTT)법 및 prothrombin time (PT)법으로도 확인되었다. 이때 APTT-저해활성은 hexane 및 methylene chloride분획에 주로 분포하였으나 PT-저해활성은 hexane 분획에 주로 분포하였다. 이들 분획들의 APTT-저해활성 및 PT-저해활성은 warfarin (최종농도 3.13 mg/ml) 보다 더 높은 것으로 확인되었다. 황금찰수수 유래 에탄올 추출물의 TT-저해활성은 열 안정성 및 pH 안정성이 매우 우수하였다. 한편, 이와 같은 황금찰수수 유래 에탄올추출물, hexane 분획 및 methylene chloride 분획의 경우는 피브린 응괴를 가용화할 수 있는 피브린용해활성을 지닌 것으로 나타났다. 이러한 결과들은 황금찰수수가 혈액응고저해활성과 혈전용해활성을 지니고 있음을 보여주며, 아울러 항혈전 식이요법의 소재가 될 수 있음을 시사한다.

XPG is Predictive Gene of Clinical Outcome in Advanced Non-small-cell Lung Cancer with Platinum Drug Therapy

  • Polymorphisms in XPG are considered to contribute to the clinical outcome of patients receiving platinum drug chemotherapy. We aimed to investigate the role of five potential SNPs of XPG gene on the response to platinum-based chemotherapy in advanced Chinese NSCLC patients. A total of 451 patients with newly diagnosed and histopathologically confirmed primary NSCLC were consecutively collected. XPG rs2296147, rs4150261, rs17655, rs1047768 and rs2094258 were genotyped by the Taqman real-time polymerase chain reaction (PCR). In our study, we found patients carrying rs1057768 TT genotype had a significantly lower treatment response when compared with the CC genotype (OR=0.38, 95% CI=0.18-0.78). Patients carrying rs1047768 TT genotype showed a significantly short median PFS (11.2 months) and OS (13.6 months) than CC genotype, and the hazard ratios (HR) for PFS and OS were 2.06 (1.01-4.50) and 2.29 (1.21-2.49), respectively. Moreover, we found a significant decreased risk of death from NSCLC among patients carrying the rs2296147 TT genotype when compared with the CC genotype, the HR (95% CI) for OS being 0.50 (0.27-0.95). In conclusion, our study found that polymorphisms in rs1047768 C/T and rs2296147 C/T are associated with response to platinum-based chemotherapy in advanced NSCLC, and XPG polymorphisms could be predictive of prognosis.

  • 영어 교육용 소프트웨어는 다양한 기법을 적용하여 개발, 활용되고 있으며 영어 능력을 중요시하는 현재의 시대상을 반영하듯 학교, 학원, 직장 외에 개인 PC를 이용해 시간과 장소의 제약 없이 쉽고, 재미있게 영어 공부를 돕는데 목적이 두고 지속적으로 개발되고 있다. 따라서 본 논문에서는 영어 교육의 중요성을 높게 판단하여 영어 단어를 중점적으로 학습하는데 목표를 둔 시스템을 구현하였다. 즉, TTS를 활용하여 음성적 교육 방식을 적용하였으며 영어 단어 학습 과정과 문제 풀이 과정으로 분류하여 영어 공부에 대한 흥미를 유발시키는 시스템을 설계하였으며 개인 계정을 통한 학습 과정 진도를 저장 및 관리하는 시스템 환경을 구현하였다.

A Novel Anticoagulant Protein with High Affinity to Blood Coagulation Factor Va from Tegillarca granosa

  • A novel inhibitory protein against blood coagulation factor Va (FVa) was purified from muscle protein of granulated ark (Tegillarca granosa, order Arcoida, marine bivalvia) by consecutive FPLC method using anion exchange and gel permeation chromatography. In the results of ESI-QTOF tandem mass analysis and database research, it was revealed that the purified T. granosa anticoagulant protein (TGAP) has 7.7 kDa of molecular mass and its partial sequence, HTHLQRAPHPNALGYHGK, has a high identity (64%) with serine/threonine kinase derived from Rhodopirellula baltica (order Planctomycetales, marine bacteria). TGAP could potently prolong thrombin time (TT), corresponding to inhibition of thrombin (FIIa) formation. Specific factor inhibitory assay showed that TGAP inhibits FVa among the major components of prothrombinase complex. In vitro assay for direct-binding affinity using surface plasmon resonance (SPR) spectrometer indicated that TGAP could be directly bound with FVa. In addition, the binding affinity of FVa to FII was decreased by addition of TGAP in dose-dependant manner ($IC_{50}$ value = 77.9 nM). These results illustrated that TGAP might interact with a heavy chain of FVa ($FVa_H$) bound to FII in prothrombin complex. The present study elucidated that non-cytotoxic T. granosa anticoagulant protein (TGAP) bound to FVa can prolong blood coagulation time by inhibiting conversion of FII to FIIa in blood coagulation cascade. In addition, TGAP did not significantly (P < 0.05) show fibrinolytic activity and cytotoxicity on venous endothelial cell line (ECV 304).
