Hydrodynamic Characteristics of Self-expandable Graft Stents in Steady Flow (정상유동에서 자가팽창성 그래프트 스텐트의 수력학적 특성)

  • This experimental study is aimed at evaluating the hydrodynamic performance of newly designed self-expandable graft stents under steady flow condition. Two graft stents with different coating materials and a bare TiNi metallic stent for comparison test were used in the experiment. Pressure variation and velocity distribution at the upstream and downstream of the stents were measured at flow rates of 5, 10, and 15 l/min, respectively. Pressure loss due to insertion of the stent increased with increasing flow rate exponentially as expected. At a flow rate of 15 l/min, pressure loss of Polyure-thane(PU)-coated graft stent was 6 times higher than that of TiNi metallic stent, while the pressure loss of a porous Polytetrafluoroethylene(PTFE)-coated graft stent was comparable to a bare TiNi metallic stent. Velocity profiles of the porous PTFE-coated graft stent were similar to those of a bare TiNi metallic stent regardless of flow rate. Furthermore, the velocity profile of PU-coated graft stent revealed an asymmetrical and relatively low central velocity at a higher flow rate than 10 1/min, expecially, where the effects resulted in increases of wall shear stress and normal stress. The worse hydrodynamic behavior of PU-coated graft stent than the other two stents might be attributed to formation of folds due to poor flexibility of coated material when inserting the graft stent into the pipe with a more smaller size, which later gave rise non-symmetry of flow area, increase of surface roughness and jet flow via the crevice between the stent and cylinder wall.

Detrimental Effect of Bovine Serum Albumin in a Maturation Medium on Embryonic Development after Somatic Cell Nuclear Transfer in Pigs

  • This study was designed to evaluate the effect of bovine serum albumin (BSA) in a maturation medium on oocyte maturation and embryonic development in pigs. Immature pig oocytes were matured for 44 h in a medium supplemented with 0.4% (w/v) BSA, 0.1% (w/v) polyvinyl alcohol (PVA), or 10% (v/v) pig follicular fluid (PFF). After IVM, oocytes reached metaphase II stage were activated for parthenogenesis (PA) or used as cytoplasts for somatic cell nuclear transfer (SCNT). Nuclear maturation (89.5%, 90.7% and 91.3% for BSA, PVA and PFF, respectively) and intraoocyte glutathione contents (1.20, 1.16 and 1.00 pixels/oocyte for BSA, PVA and PFF, respectively) were not altered by the macromolecules added to maturation medium. IVM of oocytes in a medium containing BSA (21.4%) and PVA (20.7%) showed significantly lower blastocyst formation after PA than culture in medium with PFF (39.2%). After SCNT, oocytes matured in medium with BSA showed decreased embryonic development to the blastocyst stage (9.2%) compared to those matured in medium with PFF (28.9%), while 23.6% of SCNT oocytes matured in medium with PVA developed to the blastocyst stage. When the effect of BSA in a maturation medium during the first 22 h and the second 22 h of IVM in combination with PFF or PVA was examined, PVA-BSA showed a higher nuclear maturation (94.1%) than BSA-PFF (84.5%). However, there was no significant difference in the blastocyst formation among tested combinations (47.3, 52.2, 50.0, 44.4 and 49.0% for PFF-PFF, PFF-BSA, PVA-BSA, BSA-PVA and BSA-PFF, respectively). Our results demonstrate that BSA and PVA added to maturation medium can support oocyte maturation comparable to PFF-supplemented medium. However, maturation of oocytes in a BSA-containing medium decreases embryonic development after PA and SCNT when compared with the medium supplemented with PFF.

Effect of Hh-Ag1.5 Treatment on the In Vitro Development and Apoptosis of In Vitro Fertilized Embryos in Pigs (Hh-Ag1.5 처리가 돼지 체외수정란의 발육 및 세포사멸에 미치는 영향)

  • The present study was performed to investigate the effect of Hh-Ag1.5, a small-molecule chemical agonist of SMOothened receptor, on the in vitro maturation and development of in vitro fertilized (IVF) embryos in pigs. Oocytes or fertilized embryos were cultured in a maturation or embryo culture medium supplemented with 0 (control), 25, 50 or 100 nM of Hh-Ag1.5, respectively. Although the maturation rate were not different among treatment groups, the blastocyst formation rate in the group treated with 25 nM Hh-Ag1.5 was significantly increased compared to other groups (P<0.05). While the highest dose of Hh-Ag1.5 (100 nM) did negatively affect to the embryo development and cell number in blastocysts compared to other groups (P<0.05), the apoptotic cell index in blastocysts was significantly lower in 25 and 50 nM groups than in control and 100 nM groups (P<0.05). The mRNA expression of the proapoptotic gene Bax and the ratio of Bax/Bcl-XL decreased in among treatment groups compared to control (P<0.05). The embryo quality related genes, Tert and Zfp42, were significantly decreased in 50 and 100 nM groups compared with control and 25 nM groups (P<0.05). In conclusion, the addition of 25 nM Hh-Ag1.5 to in vitro maturation and culture medium can enhance the developmental potential as well as quality of IVF embryos in pig.

Effect of OVOIL oil on B6D2F1 Mice Oogenesis

  • This study was performed to investigate the effect of types of oil (OVOIL vs. OIL) on B6D2F1 mice oogenesis. In this study, B6D2F1 F1 mice were used in order to maximize oogenesis. The expansion rate of cumulus cells ($82.0%{\pm}0.2$ vs. $78.0%{\pm}0.1$), in vitro fertilization rate ($92.0%{\pm}0.1$ vs. $88.0%{\pm}0.1$), developmental rate ($91.0%{\pm}0.1$ vs. $87.0%{\pm}0.2$), blastocysts formation rate ($56.0%{\pm}0.1$ vs. $57.0%{\pm}0.1$), and zona hatched rate($41.4%{\pm}0.2$ vs. $24.0%{\pm}0.1$) were not different between groups (NS; P>0.05). However, there was a significant difference in maturation rate; the OVOIL group showed higher maturation rate compared to that of the OIL group ($96.0%{\pm}0.1$ vs. $87.0%{\pm}0.1$; P<0.05). In the blastocysts cell numbers, the total cell numbers ($83.9{\pm}26.1$ vs. $56.9{\pm}23.9$), ICM cell numbers ($15.7{\pm}8.8$ vs. $6.3{\pm}3.5$), TE cell numbers ($68.3{\pm}25.7$ vs. $50.7{\pm}24.1$), % ICM ($21.6%{\pm}0.1$ vs. $12.7%{\pm}0.1$), and the ratio of ICM:TE ($1:6.2{\pm}6.5$ vs. $1:10.3{\pm}7.0$) were significantly higher in the OVOIL group than the OIL group (P<0.05). These results suggested that it is expected to achieve the more developmental ability of B6D2F1 mice depending on the type of oil (OVOIL vs. OIL). In addition, the results can provide essential information for culture condition on B6D2F1 mice. Henceforth, thus, it is expected that these results herein might be used for in vitro culture of human embryos.

Endoplasmic Reticulum (ER) Stress and Apoptosis in Parthenogenetic Porcine Embryos following Different Combination of Activation Methods

  • This study was conducted to investigate the effect of activation method on the endoplasmic reticulum (ER) stress induction, apoptosis and in vitro development of porcine parthenogenetic embryos. Porcine in vitro matured oocytes were activated by four activation methods; 1) electric stimulus (ES) (E), 2) $ES+10{\mu}M$ Ca-ionophore (A23187) treatment (EC), 3) ES+2 mM 6-dimethylaminopurine (6-DMAP) treatment (ED), or 4) ES+A23187 and 6-DMAP treatments (ECD). Parthenogenetic embryos were sampled to analyze x-box binding protein 1 (Xbp1) mRNA, ER stress-associated genes and apoptosis genes at 3 h after ES and the 1-cell and blastocyst stages. In the EC group, the band intensity of spliced Xbp1 (Xbp1s) mRNA was higher than those of the other groups at the 3 h and 1-cell stage, and higher than that of the E group at the blastocyst stage. Four ER stress-associated genes were expressed at the highest level in the EC group and weakly expressed in the ED group at 3 h after activation. However, most of the genes were highly expressed at the 1-cell and blastocyst stages with some variation in the EC and ECD groups. Expression of Bcl-2-associated X protein (Bax) and caspase-3 mRNA was significantly higher in the EC group than in the other groups at all development stages. The developmental rates to the blastocyst stage were higher in the ED and ECD groups than in the E and EC groups. These results suggest that the intracellular ER stress of parthenogenetic porcine embryos is affected by the activation method and subsequently lead to the apoptosis of embryos.

Exogenous Nitric Oxide Donation During In Vitro Maturation Improves Embryonic Development after Parthenogenesis and Somatic Cell Nuclear Transfer in Pigs

  • Nitric oxide (NO) has an important role in oocyte maturation and embryonic development in mammals. This study examined the effect of exogenous NO donor S-nitroso-N-acetylpenicillamine (SNAP) in a maturation medium on meiotic progression and embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) in pigs. When oocytes were exposed to $0.1{\mu}M$ SNAP for first 22 h of in vitro maturation (IVM) in Experiment 1, SNAP significantly improved blastocyst development in both defined and standard follicular fluid-supplemented media compared to untreated control (48.4 vs. 31.7-42.5%). SNAP treatment significantly arrested meiotic progression of oocytes at the germinal vesicle stage at 11 h of IVM (61.2 vs. 38.7%). However, there was no effect on meiotic progression at 22 h of IVM (Experiment 2). In Experiment 3, when oocytes were treated with SNAP at 0.001, 0.1 and $10{\mu}M$ during the first 22 h of IVM to determine a suitable concentration, $0.1{\mu}M$ SNAP (54.2%) exhibited a higher blastocyst formation than 0 and $10{\mu}M$ SNAP (36.6 and 36.6%, respectively). Time-dependent effect of SNAP treatment was evaluated in Experiment 4. It was observed that SNAP treatment for the first 22 h of IVM significantly increased blastocyst formation compared to no treatment (57.1% vs. 46.2%). Antioxidant effect of SNAP was compared with that of cysteine. SNAP treatment significantly improved embryonic development to the blastocyst stage (49.1-51.5% vs. 34.4-37.5%) irrespective of the presence or absence of cysteine (Experiment 5). Moreover, SNAP significantly increased glutathione (GSH) content and inversely decreased the reactive oxygen species (ROS) level and mitochondrial oxidative activity in IVM oocytes. SNAP treatment during IVM showed a stimulating effect on in vitro development of SCNT embryos (Experiment 7). These results demonstrates that SNAP improves developmental competence of PA and SCNT embryos probably by maintaining the redox homeostasis through increasing GSH content and mitochondrial quality and decreasing ROS in IVM oocytes.

Surface Ozone Episode Due to Stratosphere-Troposphere Exchange and Free Troposphere-Boundary Layer Exchange in Busan During Asian Dust Events

  • The current paper reports on the enhancement of O$_3$, CO, NO$_2$, and aerosols during the Asian dust event that occurred over Korea on 1 May 1999. To confirm the origin and net flux of the O$_3$, CO, NO$_2$, and aerosols, the meteorological parameters of the weather conditions were investigated using Mesoscale Meteorological Model 5(MM5) and the TOMS total ozone and aerosol index, the back trajectory was identified using the Hybrid Single-Particle Lagrangian Integrated Trajectory Model(HYSPLIT), and the ozone and ozone precursor concentrations were determined using the Urban Ashed Model(UAM). In the presence of sufficiently large concentrations of NO$\sub$x/, the oxidation of CO led to O$_3$ formation with OH, HO$_2$, NO, and NO$_2$ acting as catalysts. The sudden enhancement of O$_3$, CO, NO$_2$ and aerosols was also found to be associated with a deepening cut-off low connected with a surface cyclone and surface anticyclone located to the south of Korea during the Asian dust event. The wave pattern of the upper trough/cut-off low and total ozone level remained stationary when they came into contact with a surface cyclone during the Asian dust event. A typical example of a stratosphere-troposphere exchange(STE) of ozone was demonstrated by tropopause folding due to the jet stream. As such, the secondary maxima of ozone above 80 ppbv that occurred at night in Busan, Korea on 1 May 2001 were considered to result from vertical mixing and advection from a free troposphere-boundary layer exchange in connection with an STE in the upper troposphere. Whereas the sudden enhancement of ozone above 100 ppbv during the day was explained by the catalytic reaction of ozone precursors and transport of ozone from a slow-moving anticyclone area that included a high level of ozone and its precursors coming from China to the south of Korea. The aerosols identified in the free troposphere over Busan, Korea on 1 May 1999 originated from the Taklamakan and Gobi deserts across the Yellow River. In particular, the 1000m profile indicated that the source of the air parcels was from an anticyclone located to the south of Korea. The net flux due to the first invasion of ozone between 0000 LST and 0600 LST on 1 May 1999 agreed with the observed ground-based background concentration of ozone. From 0600 LST to 1200 LST, the net flux of the second invasion of ozone was twice as much as the day before. In this case, a change in the horizontal wind direction may have been responsible for the ozone increase.

Evaluation of Recent Magma Activity of Sierra Negra Volcano, Galapagos Using SAR Remote Sensing (SAR 원격탐사를 활용한 Galapagos Sierra Negra 화산의 최근 마그마 활동 추정)

  • Detection of subtle ground deformation of volcanoes plays an important role in evaluating the risk and possibility of volcanic eruptions. Ground-fixed observation equipment is difficult to maintain and cost-inefficient. In contrast, satellite remote sensing can regularly monitor at low cost. In this paper, following the study of Chadwick et al. (2006), which applied the interferometric SAR (InSAR) technique to the Sierra Negra volcano, Galapagos. In order to investigate the deformation of the volcano before 2005 eruption, the recent activities of this volcano were analyzed using Sentinel-1, the latest SAR satellite. We obtained the descending mode Sentinel-1A SAR data from January 2017 to January 2018, applied the Persistent Scatter InSAR, and estimated the depth and expansion quantity of magma in recent years through the Mogi model. As a result, it was confirmed that the activity pattern of volcano prior to the eruption in June 2018 was similar to the pattern before the eruption in 2005 and was successful in estimating the depth and expansion amount. The results of this study suggest that satellite SAR can characterize the activity patterns of volcano and can be possibly used for early monitoring of volcanic eruption.

Reproductive performance of Korean native cattle (Hanwoo) focusing on calving interval and parity (분만간격과 산차를 중심으로 한국 재래종인 한우의 번식능력 분석)

  • The Korean native cattle, Hanwoo, is the most popular breed of beef cattle in Korea. However, the reproductive performance data are limited although reproduction is one of the most economically and biologically important in beef production. Therefore, this study was undertaken to investigate reproductive performance parameters including calving interval, parity for life time production. Data collected from 206,827 calvings were analyzed. There were no significant differences in calving interval and gestation days as parity increased from 2nd and 13rd parity cow, from spring to winter. However, we found a dramatic increase in calving interval after year 2000. About 1 month were increased per year ( y = 30.578x + 344.45 $R^2=0.9157$). Interestingly, we observed that parities for life time can be affected by birth weight. Calves with 23 kg at birth showed highest parities, $3.4{\pm}2.0$ times. In summary, this study provides valuable data on reproductive performance of Hanwoo and the data presented here can be used as a standard target for optimising and enhancing reproductive performance.

Behavior of Overtopping Flow of Caisson Breakwater with Dissipating Block: Regular Wave Conditions (소파블록피복 케이슨 방파제에서 월파의 거동분석: 규칙파 조건)

  • The present study investigates the behaviour of overtopping flows falling on the leeside of a caisson breakwater with dissipating blocks through laboratory measurements. The falling overtopping flows in the leeside are expected to directly affect the leeside stability of the breakwater. This study focuses on not the resultant stability but the characteristic pattern of the overtopping flows depending on wave conditions through examining front velocity and plunging distance in the leeside. Regular waves were used to investigate the dependence of the overtopping flow pattern on wave conditions and a modified image velocimetry combining the shadowgraphy and cross-correlation method was employed for measurements of image and velocity. From the measurements, it is shown that the plunging distance and front velocity of the overtopping flow in the breakwater leeside increase as the wave period or height increases. From non-dimensional relationships between the variables, empirical formula for the velocity and overtopping distance are suggested.
