  • In Japan, Wagyu cattle include four Japanese breeds; Black, Brown, Shorthorn, and Polled. Today, the renowned brand name Wagyu includes not only cattle produced in Japan, but also cattle produced in countries such as Australia and the United States. In recent years, the intramuscular fat percentage in beef (longissimus muscle) from Japanese Black cattle has increased to be greater than 30%. The Japanese Black breed is genetically predisposed to producing carcass lipids containing higher concentrations of monounsaturated fatty acids than other breeds. However, there are numerous problems with the management of this breed including high production costs, disposal of untreated excrement, the requirement for imported feed, and food security risks resulting from various viral diseases introduced by imported feed. The feeding system needs to shift to one that is more efficient, and improves management for farmers, food security for consumers, and the health environment for residents of Japan. Currently, we are developing a metabolic programming and an information and communications technology (ICT, or Interne of Things) management system for Wagyu beef production as future systems. If successful, we will produce safe, high-quality Wagyu beef using domestic pasture resources while solving the problems of how to utilize increasing areas of abandoned agricultural land and to make use of the plant-based feed resources in Japan's mountainous areas.

Metabolomic profiling of postmortem aged muscle in Japanese Brown beef cattle revealed an interbreed difference from Japanese Black beef

  • Objective: Japanese Brown (JBR) cattle, especially the Kochi (Tosa) pedigree (JBRT), is a local breed of moderately marbled beef. Despite the increasing demand, the interbreed differences in muscle metabolites from the highly marbled Japanese Black (JBL) beef remain poorly understood. We aimed to determine flavor-related metabolites and postmortem metabolisms characteristic to JBRT beef in comparison with JBL beef. Methods: Lean portions of the longissimus thoracis (loin) muscle from four JBRT cattle were collected at 0, 1, and 14 d postmortem. The muscle metabolomic profiles were analyzed using capillary electrophoresis time-of-flight mass spectrometry. The difference in post-mortem metabolisms and aged muscle metabolites were analyzed by statistical and bioinformatic analyses between JBRT (n = 12) and JBL cattle (n = 6). Results: A total of 240 metabolite annotations were obtained from the detected signals of the JBRT muscle samples. Principal component analysis separated the beef samples into three different aging point groups. According to metabolite set enrichment analysis, post-mortem metabolic changes were associated with the metabolism of pyrimidine, nicotinate and nicotinamide, purine, pyruvate, thiamine, amino sugar, and fatty acid; citric acid cycle; and pentose phosphate pathway as well as various amino acids and mitochondrial fatty acid metabolism. The aged JBRT beef showed higher ultimate pH and lower lactate content than aged JBL beef, suggesting the lower glycolytic activity in postmortem JBRT muscle. JBRT beef was distinguished from JBL beef by significantly different compounds, including choline, amino acids, uridine monophosphate, inosine 5'-monophosphate, fructose 1,6-diphosphate, and betaine, suggesting interbreed differences in the accumulation of nucleotide monophosphate, glutathione metabolism, and phospholipid metabolism. Conclusion: Glycolysis, purine metabolism, fatty acid catabolism, and protein degradation were the most common pathways in beef during postmortem aging. The differentially expressed metabolites and the relevant metabolisms in JBRT beef may contribute to the development of a characteristic flavor.

축종별 일본뇌염바이러스에 대한 항체보유율 조사 (Serological survey of Japanese encephalitis virus in domestic animals)

  • A total of 1,024 sera were collected from cattle(227), pigs(465), chickens(257) and dogs(75) raised or slaughtered in Daejeon metropolitan city from April to September 2002. Japanses encephalitis virus(JEV) antibodies in sera were detected by the haemagglutination inhibition test. The prevalence rates of JEV antibodies were 99.1 %, 54.0 %, 63.0 % and 98.7 % in cattle, pigs, chickens and dogs, respectively. In case of cattle and dogs, the monthly antibody-positive rates were as high as 85.7∼100.0 % and there were no differences among six months. In case of pigs, the monthly antibody-positive rate showed the lowest in April(6.4 %) and the highest in July(100.0 %) and it remained above 50 % during the summer-time. In case of chickens, the monthly antibody-positive rate was 100.0 % in July & August, 80.5 % in June, 40.0 % in May, 7.5 % in September and 5.0 % in April in order and there were distinct differences in seasons.

소 Y 염색체 특이 Microsatellite를 이용한 품종별 대립유전자 빈도 분석 (Allele Frequency of the Bovine Y-chromosomal Microsatellite Locus in the Cattle Breeds)

  • Bovine microsatellite 마커인 INRA124는 소에 있어 Y 염색체 특이 마커로서 130 및 132 bp의 두 개 대립유전자를 가지고 있는데, Bos taurus 특이 대립유전자는 132 bp이고, Bos indicus 특이대립유전자는 130 bp이다. 이번 연구는 이 유전좌위를 이용하여 한우집단에 대한 Bos taurus 및 Bos indicus 유래의 대립유전자 분석을 통하여 한우의 유전적 특성을 이해하고자 하였다. 동북아시아, 중국내륙, 유럽, 인도 및 아프리카 유래의 20개 소 품종 822두의 수컷을 공시하여 분석해 본 결과, 유럽계통, 일본 화우 및 한우에서는 Bos indicus 유래의 대립유전자는 전혀 검출되지 않았으며, 인도 및 아프리카 유래의 Bos indicus 품종에서는 132 bp의 대립유전자가 전혀 검출되지 않았다. 한우, 브라만 및 샤롤레의 교잡우 집단(CBK)에서는 Bos indicus 특이 대립유전자인 130 bp가 0.19의 빈도로 검출되었고, 중국내륙 품종인 노서우 및 난양우는 각각 0.46 및 0.29의 빈도로 검출되었다. 이로서 한우의 기원은 기존 혼합설과 달리 유럽계통의 Bos taurus만으로 기원하였을 가능성을 제기한다.

Genealogical Relationship between Pedigree and Microsatellite Information and Analysis of Genetic Structure of a Highly Inbred Japanese Black Cattle Strain

  • Japanese Black cattle of Hyogo prefecture (Tajima strain) are famous for its ability to produce high-quality meat and have been maintained as a closed system for more than 80 years. In order to assess the usefulness of microsatellite markers in closed cattle populations, and evaluate the genetic structure of the Tajima strain, we analyzed representative dams of the Tajima strain comprised of the substrains Nakadoi and Kinosaki. Genetic variability analyses indicated low genetic diversity in the Tajima strain. In addition, a recent genetic bottleneck, which could be accounted for by the high level of inbreeding, was detected in both substrains. In phylogenetic analyses, relationship coefficients and genetic distances between individuals were calculated using pedigree and microsatellite information. Two phylogenetic trees were constructed from microsatellite and pedigree information using the UPGMA method. Both trees illustrated that most individuals were distinguished clearly on the basis of the two substrains, although in the microsatellite tree some individuals appeared in clusters of different substrains. Comparing the two phylogenetic trees revealed good consistency between the microsatellite analysis tree and the pedigree information. The correlation coefficient between genetic distances derived from microsatellite and pedigree information was 0.686 with a high significance level (p<0.001). These results indicated that microsatellite information may provide data substantially equivalent to pedigree information even in unusually inbred herds of cattle, and suggested that microsatellite markers may be useful in revealing genetic structure without accurate or complete pedigree nformation. Breed Discrimination Using DNA Markers Derived from AFLP in Japanese Beef Cattle

  • In the meat industry, correct breed information in food labeling is required to assure meat quality. Genetic markers provide corroborating evidence to identify breed. This paper describes the development of DNA markers to discriminate between Japanese Black and F1 (Japanese Black${\times}$Holstein) breeds. The amplified fragment length polymorphism method was employed to detect candidate markers absent in Japanese Black but present in Holstein. The 1,754 primer combinations yielded eleven markers that were converted into single nucleotide polymorphism markers for high-throughput genotyping. The allele frequencies in both breeds were investigated for discrimination ability using PCR-RFLP. The probability of identifying F1 was 0.9168 and probability of misjudgment was 0.0066 using four selected markers. The markers could be useful for discriminating between Japanese Black and F1 and would contribute to the prevention of falsified breed labeling of meat.

일본중부(日本中部)와 남부(南部)의 우(牛)에서 일본뇌염감염(日本腦炎感染)에 관한 역학적(疫學的) 연구(硏究) (Japanese encephalitis virus infection in cattle: comparison of antibody distribution in the central and southern regions of Japan)

  • 1982년(年)부터 1984년도(年度)까지 3년간(年間)에 걸처, 일본내(日本內) 중부(中部) Saitama현(縣)의 우(牛) 1,306두(頭)와 남부(南部)의 Kagoshima현(縣)의 우(牛) 536두(頭)를 대상(對象)으로 하여 일본뇌염(日本腦炎)바이라스 (JEV)의 적혈구응집억제(赤血球凝集抑制) (HI) 항체양성율(抗體陽性率)을 검사(檢査)한 바, Kagoshima현(縣)에선 68.8% 그리고 Saitama 현(縣)에서는 65.5%가 양성(陽性)이었다. 계절별(季節別)로는 양지역(兩地域)이 공(供)히 하절(夏節)에 항체양성율(抗體陽性率)이 높았고, 연령별(年齡別) 양성율(陽性率)은 Saitama 현(縣)의 경우 64.0%부터 82.8%까지 분포(分布)하고, Kagoshima현(縣)의 우(牛)는 1세군(歲群)에서 29.4%, 2세군(歲群)에선 50.0%, 3세군(歲群)에선 47.4% 그리고 4세군(歲群)에선 74.5%의 양성율(陽性率)을 나타내었다. 그리고 양지역(兩地域)의 연령(年齡)과 항체력가간(抗體力價間)에는 상관성(相關性)이 없었고, Saitama현(縣) 우(牛)의 역가(力價)는 연령(年齡)에 따라 15.3~22.5, Kagoshima 현우(縣牛)은 20.0~32.3이었다.

Mitochondrial DNA Polymorphism, Maternal Lineage and Correlations with Postnatal Growth of Japanese Black Beef Cattle to Yearling Age

  • Mitochondrial DNA haplotypes from the displacement-loop (D-loop) region (436 bp) were genotyped and sequenced in Japanese Black beef cattle raised in the same herd. Correlation coefficients between mitochondrial DNA haplotypes, maternal lineage, birth weight, preweaning average daily gain, weaning weight, post weaning average daily gain and yearling weight were computed. The objective was to study the relationship between maternal and postnatal growth traits and to investigate if postnatal growth of calves to yearling age could be accurately predicted from mitochondrial DNA haplotypes. Results of the phylogenetic analysis revealed 17 maternal lineages and four mitochondrial DNA haplotypes. There were strong, positive and highly significant (p<0.001) correlations among maternal traits ranging from 0.52 to 0.98. Similarly, among postnatal growth traits, most of the correlations were also strong, positive and highly significant (p<0.001); the highest correlation of 0.94 was between preweaning average daily gain and weaning weight. However, correlations between mitochondrial DNA haplotypes and postnatal growth traits were very low, mostly negative and non-significant (p>0.05) ranging from -0.05 to 0.1. Prediction of postnatal growth from mitochondrial DNA yielded very low $R^{2}$ values ranging from 0.002 to 0.019. It was concluded that mitochondrial DNA polymorphism has no significant association with postnatal growth from birth to yearling age, and by implication, nuclear rather than cytoplasmic DNA, accounts for most of the genetic variation observed in postnatal growth of Japanese Black cattle. Therefore, mitochondrial DNA genotyping at an early age has no bearing on the accurate prediction of the future growth performance of calves.

Vitamins A and E in Carcass Fat from Japanese Black and F1 Cross Cattle

  • This study was carried out to determine the contents of vitamins A and E in carcass fat from Japanese Black and F1 cross cattle, because the method of controlling the amount of vitamin A in feed has been used in the production of expensive beef with high marbling in Japan. We determined the contents of tocopherol and retinol in subcutaneous and perirenal fats for 40 carcasses of Japanese Black and F1 (Holstein${\times}$Japanese Black crossed) cattle that had been sent to market from 10 prefectures. In bovine fat, free ${\alpha}$-tocopherol was present in the greatest amount, the level of ${\gamma}$-tocopherol was about 10% and there was only a trace of ${\delta}$-tocopherol. The means${\pm}$standard deviation of ${\alpha}$-tocopherol content in subcutaneous and perirenal fats were $0.68{\pm}0.33mg/100g$ and $0.54{\pm}0.23mg/100g$, respectively. The means${\pm}$standard deviation of retinol content in subcutaneous and perirenal fats were $2.9{\pm}4.5{\mu}g/100g$ and $2.9{\pm}5.1{\mu}g/100g$, respectively. Both vitamins differed widely between animals and were present at a relatively low level compared to other reports (${\alpha}$-tocopherol 2.4-5.4 mg/100 g fat tissue and $30-220{\mu}g/100g$ muscle tissue). The correlation coefficients between subcutaneous and perirenal fats for retinol and ${\alpha}$-tocopherol content were 0.97 (p<0.01) and 0.82 (p<0.01), respectively, which showed high correlation. However, the correlation coefficients between retinol and ${\alpha}$-tocopherol contents for subcutaneous and perirenal fats were 0.11 and 0.08, respectively, which showed low correlation. Breed effect was observed in both vitamins. These results indicated that vitamins A and E in fat from Japanese Black and F1 cross cattle under the vitamin A controlling method are present at relatively low levels and feeds need to be supplemented with vitamin E.
