Effect of Ginseng Saponin on the Integrity of Lysosomes (인삼사포닌이 Lysosome의 안정성에 미치는 영향)

  • The effect of ginseng saponin on acid phosphatase (AP) activity in liver Iysosomes was investigated and the mechanism by which ginseng saponin may function on the integrity of Iysosomes was discussed. The experimental results obtained are summarized as follows; 1, A very marked increase in the AP activity was observed in the supernatant of hypotonic medium, as compared with that of isotonic medium, indicating that the hypoosmotic shock per so results in activation through osmotic Iysis of particles. 2. Ginseng saponin had no effect on the activity of AP if once released from Iysosomes when Iysed in the hypotonic medium, suggesting that ginseng saponin has no effect on the enzyme molecules per se. 3. The AP activity in isotonic medium suspensions was decreased at the concentrations of 10-6, 10-5 and 10-4% of ginseng saponin, but increased at 10-2 and 10-1%. It's suggested that ginseng saponin enhances the integrity of Iysosomes at 10-6, 10-5 and 10-4%, but decreases it at 10-2 and 10-1%. 4. Suspending particles in distilled water resulted in no correlation of AP activity with treatment with ginseng saponin. 5, The AP activity was decreased in the presence of ATP, showing the possible significance of ATP as a Iysosomal stabilizer and the possibility that ginseng saponin may affect a membrane bound ATPase system by which Iysosomal AP release may be controlled.

폐염균 다당류 정제 방법의 개발 및 한국형 폐염균의 수집

  • 한국에서 유행하는 페염균 type을 결정하기 위한 기초 실험을 수행하여 서울대병원과 서울중앙병원의 환자로부터 분리한 88개의 폐염균주를 담즙산에 의한 용해도 및 Optochin에 대한 민감성 등을 측정하고 폐염균의 type을 결정하기 위해서 Double diffusion, Countercurrent immunoelectrophoresis, Rocket immunoelectrophoresis 방법 등으로 폐염균 항혈청과 반응하였다. 또한 ATCC로부터 구입한 폐염균주를 이용하여 배지조성의 변화에 따른 폐염균 생장을 측정하기 위해 CAT broth, Brain Heart Infusion broth, Defined Media에서 3가지 type의 균주(Type I, III, IV)를 접종하고 37$^{\circ}C$에서 배양하면서 1시간마다 550nm에서 흡광도를 측정하였다. 3가지 type (Type I, III, IV)의 폐염균주로부터 협막다당류를 phenol Iysis 추출법으로 폐염균 Capsule 다당류의 분리하여 Total Sugar 함량, Uronic acid 함량, 0-Acetyl 함량, 단백질 함량, 핵산 함량, 분자량분포 Countr-current immunoelectrophoresis 등을 실시하여 폐염균 백신 규격에 부합되는지 확인하였다. 수집한 88개의 폐염균주가 모두 담즙산에 의해 용해되고 6mm Optochin disc를 사용하였을때 성장 저지원이 모두 14mm 이상으로 나타나 감수성이 있어서 폐염균임을 확인하였으며 폐염균의 type 결정에 필요한 방법을 설정하였다. 배지조성에 따른 폐염균의 생장을 측정한 결과 CAT broth 에서는 Type I만이 잘 배양되었고 Type III, IV는 늦게 배양되었으나, Brain Heart Infusion broth에서는 3가지 Type이 모두 잘 배양되었고, Defined Media 에서는 Type I만이 성장되었을 뿐 나머지 두 Type은 배양되지 않았다. 본 실험방법으로 분리된 폐염균 다당류는 폐염균 백신 규격중의 6가지 항목에서 적합하였으며 Statens(Denmark)로부터 구입한 항체와 뚜렷한 침강선을 나타내었다.

  • Recently, in Korea, demand for establishment of systematic risk assessment techniques for construction projects has increased, especially after the large construction failures occurred during construction such as New Haengju Bridge construction projects, subway construction projects, gas explosion accidents etc. Most of existing risk analysis modeling techniques such as Event Tree Analysis and Fault Tree Analysis may not be available for realistic risk assessment of construction projects because it is very complex and difficult to estimate occurrence frequency and failure probability precisely due to a lack of data related to the various risks inherent in construction projects like natural disasters, financial and economic risks, political risks, environmental risks as well as design and construction-related risks. Therefor the main objective of this paper is to suggest systematic probabilistic risk assessment model and demonstrate an approach for probabilistic risk assessment using advanced Event Tree Analysis introducing Fuzzy set theory concepts. It may be stated that the Fuzzy Event Tree AnaIysis may be very usefu1 for the systematic and rational risk assessment for real constructions problems because the approach is able to effectively deal with all the related construction risks in terms of the linguistic variables that incorporate systematically expert's experiences and subjective judgement.

Cholesterol Uptake by Lactobacillus acidophilus: Its Fate and Factors Influencing the Uptake (Lactobacillus acidophilus에 의한 콜레스테롤의 흡착)

  • Cholesterol assimilated by Lactobacillus acidophilus ATCC 43121 was not metabolically degraded in that most of it was recovered with the cells. Cells grown in the presence of cholesterol micelles and bile salts were more resistant to Iysis by sonication than those grown in their absence, suggesting a possible alteration of cellular membranes. Cholesterol assimilation occurred during growth at pH 6.0, the amount of which was more than that by cells grown without pH control. Cholesterol assimilated by cells was recovered in the membrane fractions of cells both grown at pH 6.0 and without pH control. The effect of unsaturated fatty acids on cholesterol assimilation was not clear, since there was no significant (P> 0.05) difference in the amount taken up from micelles prepared using L-${\alpha}$-phosphatidylcholine, dioleoyl or L-${\alpha}$-phosphatidylcholine, distearoyl. Without Tween 80, little, if any, cell growth or cholesterol uptake was observed. In the presence of 0.05% Tween 80, cholesterol uptake increased dramatically as did growth. However, as the amount of Tween 80 increased beyond 0.05%, cholesterol uptake decreased while the amount of growth remained the same.

  • This study was conducted in order to find out biological control of sesame wilt and blight caused by Fusarium of oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica by using Streptomyces spp. Two sesame pathogens, Fusarium oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica were purely isolated from diseased sesame plants of the field. Streptomyces species were isolated from 72 soil samples collected from red pepper and sesame uplands in Chungbuk and selected as antagonists according to the results of dual culture. The selected Streptomyces isolates such as St-11 and St-20 were confirmed their antagonistic effect through mycelial inhibition zone and inhibitory effects on the mycelial growth of the pathogens by culture filterate of the antagonists. Inhibitory effects on the conidial germination of Fusarium oxysporum vasinfectum and Phytophthora nicotianae parasitica by the antagonists were also tested in addition to mycelial Iysis. The antagonists St-11 and St-20 showed inhibitory effect on growth of sesame seedlings after seeds soaked in the suspension. Effect of soil inoculation with antagonist St-11 showed 40 to 78 percent of control effect for two diseases in comparison with control under greenhouse.

Bacteriological Control of Cyanobacterial Bloom. (시안세균 수화의 세균학적 조절)

  • A Gram (-), rod-shaped bacterium in size of 1.3∼$1.8{\times}0.35{\mu}m$ inhibiting the growth of cyanobacterium (Ana-baena cylindrica) was isolated and designated NG-2 in this manuscript. This isolate showed positive reactions for catalase and oxidase, and optimal growth conditions of 35∼TEX>$40<^{\circ}C$ and pH 9.0. In a mixed-culture of A. cylindrica and the isolate, each microorganism grew inverse-proportionally, and the cyanobacterial vegetative cells almost completely disappeared within 24 hours. NG-2 lysed A. cylindrica only under light, which means that lytic activity of NG-2 was dependent on the photosynthetic activity of host. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas het-erocysts have not been lysed. When cyanobacterial cell walls have been lysed partly, NG-2 attatched around A. cylindrica filament and formed colony, then encouraged complete lysis of cyanobacterial cells. The isolate showed similar lytic activity in natural water as in an artificial medium. And lytic activity of NG-2 was enhanced when attached on expandable polystyrene bead.

Incidence of Plasmids in Marine Bacteria Isolated from the Bunker-C Oil Enriched Culture (Bunker-C유 집식배양으로부터 해양세균 Plasmid의 분포)

  • Samples used for the enrichment culture were collected from the sea water of suspected chronic petroleum contamination in the vicinity of Pusan, Chungmu and Ulsan ports, Korea. Alkaline Iysis and agarose gel electrophoresis techniques were employed to screen these isolates for the presence of plasmid DNA. There were n little differences in the percentage of isolates containing plasmids between sampling sites of unpolluted sen water (22%) and polluted son water (25%). Bacterial isolates taken from the Bunker-C oil enriched culture showed significantly more plasmid incidence (29%). About two thirds of strains grown on a variety of hydrocarbons were Gram negative strains of which 33% contained one or more plasmids. Multiple plasmids were observed in 23% of the plasmid-carrying strains. Forty one percent of the plasmids detected were estimated to have a mass of 20 kb or more.

Effect of irradiation on the Streptococcus mutans (방사선조사가 Streptococcus mutans에 미치는 영향)

  • Purpose : To observe direct effect of irradiation on cariogenic Streptooccus mutans. Materials and Methods : S. mutans GS5 was exposed to irradiation with a single absorbed dose of 10, 20, 30, and 40Gy. Viability and changes in antibiotic sensitivity, morphology, transcription of virulence factors, and protein profile of bacterium after irradiation were examined by pour plate, disc diffusion method, transmission electron microscopy, RT-PCR, and SDS-PAGE, respectively. Results : After irradiation with 10 and 20Gy, viability of S. mutans was reduced. Further increase in irradiation dose, however, did not affect the viability of the remaining cells of S. mutans. Irradiated 5. mutans was found to have become sensitive to antibiotics. In particular, the bacterium irradiated with 40Gy increased its susceptibility to cefotaxime, penicillin, and tetracycline. Under the transmission electron microscope, number of morphologically abnormal cells was increased as the irradiation dose was increased. S. mutans irradiated with 10 Gy revealed a change in the cell wall and cell membrane. As irradiation dose was increased, a higher number of cells showed thickened cell wall and cell membrane and Iysis, and appearance of ghost cells was noticeable. In RT-PCR, no difference was detected in expression of gtfB and spap between cells with and without irradiation of 40Gy. In SDS-PAGE, proteins with higher molecular masses were gradually diminished as irradiation dose was increased. Conclusion : These results suggest that irradiation affects the cell Integrity of S. mutans, as observed by SDS-PAGE, and as manifested by the change in cell morphology, antibiotic sensitivity, and eventually viability of the bacterium.

Purification of a 68-kDa cysteine proteinase from crude extract of Pneumocystis carinii

  • The present study intended to verify activities of cysteine proteinase of Pneumocystis carinii from rats and to purify the enzyme. In order to exclude the contamination of host-derived enzymes, concentrates of P. carinii was primarily treated with a mixture of proteinase inhibitors before Iysis of P carinii. A 68-kDa cysteine proteinase was finally purified from the crude extract of P. carinii by 4 sequential chromatographic methods. The enzyme showed an optimal activity at pH 5.5 in 0.1 M sodium acetate, and its activity was specifically inhibited by L-trans-epoxy-succinylleucylamido (4-guanidino) butane (E-64) and iodoacetic acid, suggesting that the enzyme is a cysteine proteinase. The 68-kDa proteinase weakly digested rnacrornolecules such as collagen, hemoglobin and fibronectin. The present study demonstrated the activity of cysteine proteinase at the 68-kDa band of P. carinii, and purified and characterized the molecule.

Isolation and Characterization of Algicidal Bacteria KY1 (살조세균 KY1의 분리와 특성조사)

  • Algicidal bacteria were isolated from the mud of south coastal sea of Korean peninsula and screened to evaluate algicidal activity on Cochlodinium polykrikoides. The optimum condition for the development of highest algicidal activity was determined, The optimum sodium chloride concentration for algicidal activity of isolated algicidal bacteria was $3\%$. The optimum temperature, pH and culture time for the highest algicidal activity were $30^{\circ}C$, pH 7 and above 24 hr, respectively. The algicidal activity were significantly increased at the stationary phase of the cell growth and continuously increased up to maximum at the death phase, probably due to the release of algicidal substance by cell Iysis. The effect of zinc ion addition on algicidal activity, was observed and indicated that the substance requires zinc for its activity. The candidate for the algicidal substance may be $\alpha$-mannosidase.
