수도의 세포간극 물질 추출 (Extraction of Intercellular Material from Rice Leaf Tissue)

  • 본 연구는 세포간극물질을 추출코자 실시하였다. 2. Vacuum flask에 물을 넣은 후 볏잎을 물에 잠기게 하고, 280mb로 기압을 줄여 벼조직의 세포간극내의 공기를 뺀 후 Vacuum을 끊고 증류수로 세포간극을 채운다. 볏잎에 묻은 증류수를 제거하고 본 연구에서 사용한 leaf holder에 끼워 3,000rpm으로 5분간 원심분리하면 세포간극물질만 채취할 수 있다. 3. 세포내물질과 세포간극물질의 성분분석 및 Peroxidase와 Easterase의 isozyme pattern을 비교한 결과 내용성분의 함량이 서로 다르고 효소의 활성과 isozyme pattern이 다르게 나타나 세포간극물질 추출액 속에는 세포내물질이 혼입되어 있지 않음을 증명하였다.

무지개송어와 은연어간 잡종3배체의 부화자어에 대한 동위효소 분석 (Isozyme Analysis on the Allotriploid between Rainbow Trout (Oncorhynchus mykiss) and Coho Salmon (O. kisutch))

  • 본 연구는 우리나라의 대표적인 연어과 어류인 은연어, 무지개송어 및 그 잡종3배체의 부화자어에 있어서의 동위효소를 분석하여 부화후 초기의 유전적 특징과 유전현상을 규명하고 이를 토대로 이들 종 및 잡종의 식별을 위한 genetic marker를 찾아보고자 5개의 동위효소에 대한 분석을 실시하였다. 이들 중 PGI에서는 종간 차이를 나타내지 않은 반면에 LDH, MDH, IDH 및 PGM 등은 은연어와 무지개송어간에 종 특이적 pattern을 나타내었는데 이는 이들 어종간의 발생 초기에 종의 식별을 위한 유전적 marker로써 유용한 것으로 판단되며, 또한 잡종 및 잡종3배체의 genetic marker로서도 상기 네 가지의 동위효소는 유용한 것으로 나타났다.

Protoplast fusion between Lentinula edodes and Coriolus versicolor

  • Protoplast fusion between isoleucine-, argihine- and thymidine-requiring auxotroph $(Ile^{-}, Arg^{-}, Thy^{-})$ of Lentinula edodes and arginine-requiring auxotroph $(Arg^-)$ of Coriolus versicolor has been achieved using 30% polyethylene glycol (M.W.4000) in 10 mM $CaCl_2$-glycine solution (pH 8.0). Fusion hybrids were selected in the 0.6 M sucrose supplemented minimal media on the basis of nutritional complementation with fusion frequency of $7.4{\times}10{-6}$ The hybrids included both parental and non-parental types in colony morphology, growth rate and isozyme patterns. We succeeded inter-order protoplast fusion between the auxotrophs of Lentinula edodes and Coriolus versicolor overcoming the natural barriers of incompatibility. We examined the characteristics of the hybrids and clarified the fusion rocess using electron microscopy.

유기린계 살충제가 흰쥐의 Cholinesterase활성에 미치는 영향 (Effects of Organophosphorous Insecticides on Mice Cholinesterase Activity)

  • Changes in cholinesterase (ChE) activity, electrophoretic pattern of ChE and histopathologic state on the mice serum, brain and liver by administration of organophos-phorous insecticides were studied. The mice ChE activities on serum, brain and liver decreased by increasing of concentration and time both administration of malathion and DDVP, whereas on serum and brain the activities of the 7 days after administration decreased, and then presented the gradually slight recovery in course of time. The ChE on serum and liver showed many isozyme bands by polyacrylamide gel electrophoresis but several on brain. And isozyme bands disappeared and diffused by administration of organophosphorous insecticides and development of time. The mice liver with administration group of malathion on histopathologic test showed midzonal necrosis between central vein and portal area, and with administration group of DDVP mainly presented portal necrosis on location of potal area.

Effect of IOA and Media on Production of Somatic Hybrid by the Asymmetric Protoplast Fusion Between Three Solanum Species

  • In this study, we surveyed the influence of IOA (iodoacetamide) and media upon protoplast fusion for the efficient production of the somatic hybrid among S. sisymbriifolium and other Solanum species (S. intergrifolium and S. toxicarium). Regardless of a breed, as the IOA concentration increases, the cell division tends to decrease. As the influence of the media on the colony formation, we could get 5 colonies from the fusion of S. sisymbriifolium and IOA-treated S. integrifolium protoplast, but none was observed in other fusions. As a result of analyzing their IDH isozyme, we found a somatic hybrid in 2 objects.

  • In order to evaluate the applicability of enzyme electrophoresis for the identification of intra/interspecific hybride obtained by the protoplast fusion in Trichoderma, soluble proteins, intracellular soluble enzymes and extracellular $\beta$-glucosidase were analyzed by polyacrylamide gel electrophorsis. As the results, patterns of soluble protein, and isozyme patterns of peroxidase, malate dehydrogenase, and $\beta$-glucosidase in hydrids were defferent from those in parental and wild type strains. Therefore, it was established that the analysis of protein pattern by electrophoresis could be applied to isolate and identify the hybrids from the protoplast fusion.

Breeding of Bivoltine Breeds of Bombyx mori L Suitable for Variable Climatic Conditions of the Tropics

  • The success of rearing with presently available conventional bivoltine is unpredictable in some seasons of the tropical regions due to highly fluctuating adverse climatic conditions. Thus, in order to popularize bivoltine breeds in tropical parts of India, it is very much essential to have a bivoltine breed(s), which can give stable cocoon crop under variable environments. With this objective a breeding programme was undertaken to improve the survival trait in bivoltine silkworm by introducing multivoltine genes into bivoltine through back crossing. Resultant bivoltine lines showed significantly higher survival in compared to the receptor (Bivoltine) parent and control bivoltine breed. Esterase isozyme analysis revealed similar banding pattern in the developed bivoltine and in the donor multivoltine, which predicts the introgression of multivoltine character into evolved bivoltine.

  • 사과의 수령(樹齡), 지역(地域) 및 부위(部位)에 따른, 그리고 주요(主要) 6개(個) 품종(品種)(국광(國光), 후지, 홍옥(紅玉), 스타킹, 골덴 델리셔스, 쓰가루) 및 그들의 교잡개체(交雜個體)의 peroxidase isoenzyme pattern의 차이(差異)를 조사(調査)하였던 바 다음과 같은 결과(結果)를 얻었다. 1. 후지품종(品種)의 수령(樹齡)은 zymogram에 영향(影響)을 미치지 않았다. 2. 후지와 쓰가루 두 품종(品種)의 zymogram은 재식지(栽植地)에 따라 차이(差異)가 없었다. 3. 후지와 쓰가루 두 품종(品種)의 가지와 잎의 zymogram은 뚜렷한 차이(差異)가 인정(認定)되었으나 채취부위별(採取部位別) 재식지(栽植地)에 따른 차이(差異)는 인정(認定)되지 않았다. 4. 국광(國光), 후지, 홍옥(紅玉), 스타킹, 골덴 델리셔스, 쓰가루 등(等) 여섯 품종(品種)에서 그 품종(品種) 고유(固有)의 zymogram이 관찰(觀察)되었다. 5. 교잡개체(交雜個體)의 zymogram은 1) 양친(兩親)의 band를 모두 갖고 있는 형(形), 2) 양친(兩親)의 band 중(中) 일부(一部)를 소실(消失)한 형(形), 3) 양친(兩親)에는 없는 새로운 band를 갖고 있는 형(形)으로 분류(分類)되었다.

The Protein and Isozyme Patterns During in vitro Plant Regeneration of Yooja (Citrus junos Sieb.) and Trifoliate Orange (Poncirus trifoliata Rafin.)

  • In this study, plant regeneration through in vitro culture from plantlet stems of Yooja (C. junos Sieb.) and trifoliate orange (P. trifoliata Rafin.) was attempted to make mass-production system of virus-free plants having the same genotype with mother plant. In order to investigate physiological change depending on the developmental stage of plant regeneration, the changes of total protein, peroxidase and esterase activity and their isozyme patterns as well were examined in 1/2 MS medium. The results are as follows : 1. The MS medium for the optimal callus induction and shoot formation was utilized. The medium was supplemented either with 2,4-D and Kinetin or with BA and NAA. The optimal concentrations were the combination of 1.0mg/ 2,4-D +0.3mg/ Kinetin and 1.0mg BA +0.3mg NAA in callus induction and shoot formation, respectively. 2. For the plant regeneration from somatic embryos, 1/2 MS medium was used with supplements of growth regulators (free, 1.0mg/ IBA +1.0mg/ BA ,0.5mg/ IBA +0.5mg/ BA). Shooting and rooting were the best in the treatment of 0.5mg/ IBA and 0.5mg/ BA combination. 3. The total protein content has a tendency of increase with the developmental stage of embryo, but it was decreased at the plantlet. Also it was the highest at 8 and 6 weeks stage in C. junos Sieb. and P. trioliata Rafin, respectively. In the SDS-PAGE pattern of protein, C. junos Sieb. showed bands of 29.0 and 40kDa at 10 weeks. The 45,66 and 97.4 kDa bands at 10 weeks of culture were shown in P. trifoliata Rafin. 4. The highest esterase activity was shown at the 6 and 8 weeks of culture in C.junos Sieb. and P. trifoliata Rafin.., respectively. 5. Esterase isozyme patterns were shown difference according to the developmental stage. In C. junos Sieb. a new band was observed at pl 7.7 following 4 weeks culture. On the other hand, new bands in P. trifoliata Rafin. were observed at pl 7.5~6.5 following 4 and 6 weeks culture, respectively.

스트레스 물질에 의한 벼 glutathione peroxidase 활성패턴 변화 (Alterations of Glutathione Peroxidase Patterns by Stressor Treatment in Rice Seedling Roots)

  • 동진벼 유묘 뿌리에 환원제인 ascorbic acid, 신호전달물질인 salicylic acid와 methyl jasmonic acid, 중금속인 $NiCl_2$$CuSO_4$ 및 NaCl를 다양한 농도로 처리한 후 항산화효소인 glutathione peroxidase(GPX)의 활성과 동위효소의 패턴 변화를 살펴보았다. Ascorbic acid 처리에 의한 GPX의 총활성은 ascorbic acid 농도 증가에 의존적으로 증가하였으며 이러한 GPX활성 증가는 GPX1 동위효소의 현저한 활성증가에 기인하였다. GPX는 신호전달물질인 salicylic acid와 methyl jasmonic acid에 대하여 서로 다른 반응성을 보였다. GPX의 활성은 0.1 mM salicylic acid에 의해 증가하였다가 이후 감소하였다. 이에 비해 GPX는 methyl jasmonic acid의 농도증가에 의존하여 점진적으로 증가하여 1 mM methyl jasmonic acid에 의하여 약 3배의 활성증가를 보였다. 뿐만 아니라 GPX1 동위효소는 salicylic acid 농도가 증가할수록 활성이 감소한 반면 methyl jasmonic acid 농도가 증가할수록 현저하게 증가하였다. GPX의 총활성은 $NiCl_2$ 농도 증가에 따라 점진적으로 증가되었으나, $CuSO_4$ 처리군의 경우 GPX의 총비활성도는 0.5 mM $CuSO_4$에 의하여 약 2배 증가한 이후 점차 감소하였다. $NiCl_2$$CuSO_4$ 처리에 의한 GPX 활성증가도 주로 GPX1 동위효소의 활성증가에 기인하였다. NaCl 처리에 의한 GPX 총활성은 300 mM NaCl 처리군에서 약 1.7배 증가되었다가 이후 감소하였다. 특이하게도 NaCl 농도가 증가함에 따라 GPX2 동위효소 활성이 점차 증가하였다.
