본질적 난연 및 내열성 헤테로환식 폴리머의 특성에 관한 연구 (A Study on the Characterization of Intrinsic Flame Retardant and Heat Resistant Sulfur-Bridged Heterocyclic Polymers)

  • 본질적인 난연 및 내열성의 PQXS, PIQS, PQS 3종류의 폴리머를 2,3-dichloroquinoxaline, 1,3-dichloroisoquinoline, 4,7-dichloroquinoline을 각각 출발 물질로 하여 합성하였다. 이들을 FT-IR, UV/Vis 분광분석, DTA, 원소분석 등에 따라 특성분석을 행하였다. 이들 폴리머소재는 내열 및 난연특성 시험에서 녹는점 기준 $350^{\circ}C$ 이상의 높은 내열성과 LOI값 기준으로 41~42의 높은 수치의 난연특성을 나타내었으며 또한 수직연소시험에서 UL 94 V-0 등급의 높은 난연특성을 나타내었다.

Protoberberine Alkaloids and their Reversal Activity of P-gp Expressed Multidrug Resistance (MDR) from the Rhizome of Coptis japonica Makino

  • Six protoberberine alkaloids were isolated from the chloroform layer of the rhizome of Coptis japonica Makino (Ranunculaceae). The structures of the isolated compounds were determined to be 6-([1,3]dioxolo[4,5-g]isoquinoline-5-carbonyl)-2,3-dimethoxy-benzoic acid methyl ester (1), oxyberberine (2), 8-oxo-epiberberine (3), 8-oxocoptisine (4), berberine (5) and palmatine (6) by physicochemical and spectroscopic methods. The compound 3 (8-oxo-epiberberine) was first isolated from natural sources. The compounds were tested for cytotoxicity against five tumor cell lines in vitro by SRB method, and also tested for the MDR reversal activities. Compound 4 was of significant P-gp MDR inhibition activity with ED50 value $0.018\;{\mu}g/mL$ in MES-SA/DX5 cell and $0.0005\;{\mu}g/mL$ in HCT15 cell, respectively.

Neuroprotective Effects of Berberine in Neurodegeneration Model Rats Induced by Ibotenic Acid

  • Berberine, an isoquinoline alkaloid found in Coptidis Rhizoma(goldenthread) extract, has multiple pharmacological effects such as anti-inflammatory, antimicrobial and anti-ischemic effects. In the present study, we examined the effects of berberine on neuronal survival and differentiation in a hippocampal precursor cell line and in the memory deficient rat model. Berberine increased in a dose dependent manner the survival of hippocampal precursor cells as well as differentiated cells. In addition, berberine promoted neuronal differentiation of hippocampal precursor cells. In the memory deficient rat model induced by stereotaxic injection of ibotenic acid into entorhinal cortex(Ibo model), hippocampal cells were increased about 2.7 fold in the pyramidal layer of CA1 region and about 2 fold in the dentate gyrus by administration of berberine after 2 weeks of ibotenic acid injection. Furthermore, neuronal cells immunoreactive to calbindin were increased in the hippocampus and entorhinal cortex area by administration of berberine. Taken together, these results suggest that berberine has neuroprotective effect in the Ibo model rat brain by promoting the neuronal survival and differentiation.

Activating transcription factor-3 induction is involved in the anti-inflammatory action of berberine in RAW264.7 murine macrophages

  • Berberine is an isoquinoline alkaloid found in Rhizoma coptidis, and elicits anti-inflammatory effects through diverse mechanisms. Based on previous reports that activating transcription factor-3 (ATF-3) acts as a negative regulator of LPS signaling, the authors investigated the possible involvement of ATF-3 in the anti-inflammatory effects of berberine. It was found berberine concentration-dependently induced the expressions of ATF-3 at the mRNA and protein levels and concomitantly suppressed the LPS-induced productions of proinflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). In addition, ATF-3 knockdown abolished the inhibitory effects of berberine on LPS-induced proinflammatory cytokine production, and prevented the berberine-induced suppression of MAPK phosphorylation, but had little effect on AMPK phosphorylation. On the other hand, the effects of berberine, that is, ATF-3 induction, proinflammatory cytokine inhibition, and MAPK inactivation, were prevented by AMPK knockdown, suggesting ATF-3 induction occurs downstream of AMPK activation. The in vivo administration of berberine to mice with LPS-induced endotoxemia increased ATF-3 expression and AMPK phosphorylation in spleen and lung tissues, and concomitantly reduced the plasma and tissue levels of proinflammatory cytokines. These results suggest berberine has an anti-inflammatory effect on macrophages and that this effect is attributable, at least in part, to pathways involving AMPK activation and ATF-3 induction.

Berberine suppresses in vitro migration of human aortic smooth muscle cells through the inhibitions of MMP-2/9, u-PA, AP-1, and NF-κB

  • Berberine, a type of isoquinoline alkaloid isolated from Chinese medicinal herbs, has been reported to have various pharmacological activities. Studies have demonstrated that berberine has beneficial effects on vascular remodeling and alleviates restenosis after vascular injury. However, its mechanism of action on vascular smooth muscle cell migration is not fully understood. We therefore investigated the effect of berberine on human aortic smooth muscle cell (HASMC) migration. Boyden chamber assay was performed to show that berberine inhibited HASMC migration dose-dependently. Real-time PCR and Western blotting analyses showed that levels of matrix metalloproteinase (MMP)-2, MMP-9, and urokinase-type plasminogen activator (u-PA) were reduced by berberine at both the mRNA and protein levels. Western blotting assay further confirmed that activities of c-Fos, c-Jun, and NF-${\kappa}B$ were significantly attenuated. These results suggest that berberine effectively inhibited HASMC migration, possibly by down-regulating MMP-2, MMP-9, and u-PA; and interrupting AP-1 and NF-${\kappa}B$ mediated signaling pathways.

Aldose Reductase Inhibitory Alkaloids from Corydalis ternata

  • A methanolic extract of Corydalis ternata having aldose reductase inhibitory activity was examined as a possible aldose reductase (ALR2) inhibitor, a key enzyme involved in diabetic complications. Seven alkaloids, tetrahydrocoptisine (1), corydaline (2), tetrahydropalmatine (3), isocorybulbine (4), corybulbine (5), dehydrocorydaline (6), and N-methyltetrahydroberbinium (7) were isolated from $CHCl_3$ fraction of C. ternata methanol extract. Among them, compounds 1, 5, and 7 exhibited $5.04{\pm}1.97%$, $5.00{\pm}1.26%$, and $1.80{\pm}2.33%$ inhibitions, respectively at $40{\mu}M$. The activities of the single compounds were not comparable to that of the whole extract, suggesting that the whole combination of each single compound was responsible for the activity of the extract as shown in many cases of natural medicines. Even though this is the second report on aldose reductase inhibition activity of C. ternata, recombinant human aldose reductase was employed in this study unlike in the previous report. Furthermore, the aldose reductase inhibitory activities of isocorybulbine, corybulbine, and N-methyltetrahydroberbinium, to the best of our knowledge, were evaluated for the first time in this study. These results suggest a use of the extract of C. ternata for ameliorating diabetic complications.

  • 3-Arylisoquinolin-1(2H)-ones (2) are possible bioisosteres of the $5-[4^{l}-(piperidinomethyl)phenyl]-2<$,3-dihydroimidazo[2,1-$\alpha$]isoquinoline (1) which is in clinical evaluation for the treatment of cancer. Structure-activity relationship studies of 3-arylisoquinolin-1(2H)-ones. (2) led to the synthesis of 3-arylquinolin-2(1H)-ones (3). A number of 3-phenyl substituted quinolin-2(1H)-ones were synthesized and tested for their in vitro antitumor activity against four different human tumor cell lines and 3-phenyl-N-benzyl-3,4-dihydroquinolin-2(1H)-one (12) showed the most potent activity.

Berberine이 구속 스트레스 부가후 Mouse의 혈중 Catecholamine 함량에 미치는 영향 (Effects of Berberine on Serum Levels of Catecholamines after Immobilization Stress in Mice)

  • Berberine, a protoberberine isoquinoline alkaloid, showed inhibitory effects on dopamine content in PC12 cells $(53.8\%\;inhibition\;at\;20\;{\mu}M)$. Tyrosine hydroxylase, the rate-limiting enzyme in the catecholamine biosynthetic pathway, was inhibited at $20\;{\mu}M)$ of berberine by $21.8\%$ relative to control. Thus, we hypothesized that the inhibition of tyrosine hydroxylase by berberine might be partially contributed to the decrease in dopamine content in PC12 cells. Furthermore, we investigated the effects of berberine on catecholamine content of serum after immobilization and cold stress in mice. Adult male mice were either subjected to 30 min of restraint or to 2 hr of cold chamber at $4-6^{\circ}C$. Serum norepinephrine, 16.8 pmol/ml, in control mice was increased to 28.8 pmol/ml by immobilization and the stress-induced rise in serum norepinephrine was partially blocked by the treatment of berberine (10 mg/kg, i.p.) once a day for 6 days. Berberine (10 mg/kg/day for 3 days, i.p.) also inhibited the increase in serum norepinephrine by cold stress in mice. These results suggest that berberine may be developed as the promising antistress agent.

Liriodenine이 PC12 세포중의 Dopamine 생합성에 미치는 영향 (Effects of Liriodenine on Dopamine Biosynthesis in PC12 Cells)

  • The effects of liriodenine, an aporphine isoquinoline alkaloid, on dopamine content in PCl2 cells were investigated. Treatment of PC12 cells with liriodenine decreased dopamine content in a dose-dependent manner (33.6% inhibition at $10\;{\mu}M$ for 12 h). The $IC_{50}$ in value of liriodenine was $8.4\;{\mu}M$. Dopamine content decreased at 3 h and reached a minimal level at 12 h after the exposure to liriodenine. Under these conditions, the activities of tyrosine hydroxylase and aromatic L-amino acid decarboxylase were also inhibited at $10\;{\mu}M$ of liriodenine by 10.1% and 20.2% relative to control, respectively. In addition, liriodenine inhibited the increase in dopamine content induced by L-DOPA Treatments $(50-100\;{\mu}M)$ in PC12 cells. These results suggest that liriodenine inhibited dopamine biosynthesis and L-DOPA-induced increase in dopamine content by reducing the activities of tyrosine hydroxylase and aromatic L- amino acid decarboxylase in PC12 cells.

Effects of Protein Kinase C Modulation on Hepatic Hemodynamics and Glucoregulation

  • This study evaluated the effects of PKC activation using phorbol 12-myristate 13-acetate (PMA) and PKC inhibition using the isoquinoline sulfomide derivative H-7 on hemodynamics and glucoregulation in the isolated perfused rat liver. Livers were isolated from fed male Holtzman rats and perfused with Krebs Ringer bicarbonate solution under a constant flow of 50 ml/min at $35^{\circ}C.$ Portal vein pressure, glucose and lactate concentrations in the medium and oxygen consumption rates were continuously monitored by a Grass polygraph, YSI glucose and lactate monitors, and a YSI oxygen monitor, respectively. PMA at concentration of 2 to 200 nM increased the portal vein pressure, glucose and lactate production, but decreased oxygen consumption rate in a dose-dependent fashion. H-7 $(200\;{\mu}M)$ attenuated PMA (50 nM)-induced vasoconstriction $(15.1{\pm}1.36\;vs\;10.56{\pm}1.17\;mmHg),$ glucose production rate $(91.3{\pm}6.15\;vs\;71.8{\pm}2.50\;{\mu}moles/g/hr),$ lactate production rate $(72.4{\pm}6.82\;vs\;53.6{\pm}4.82\;{\mu}moles/g/hr)$ and oxygen consumption rate $(33.7{\pm}1.41\;vs\;27.9{\pm}1.75\;{\mu}l/g/min).$ The effects of PMA were blocked either by addition of verapamil $(9\;{\mu}M)$ or perfusion with $Ca^{2+}-free$ KRB. These results suggest that the hemodynamic and glucoregulatory changes in the perfused rat liver are mediated by protein kinase C activation and require $Ca^{2+}$ influx from the extracellular fluid.
