Cellular Protective Effect and Liposome Formulation for Enhanced Transdermal Delivery of Isoquercitrin (Isoquercitrin의 세포 보호 작용과 피부 흡수 증진을 위한 리포좀 제형 연구)

  • In this study, the cellular protective effect of isoquercitrin against $H_2O_2$ and rose bengal-indued HaCaT cell damage was investigated. The ethosome and elastic liposome for enhanced transdermal delivery were prepared. Particle size, loading efficiency and cumulative permeated amounts of them were evaluated. Isoquercitrin didn't show any characteristic cytotoxicity at 50 ${\mu}M$. When HaCaT cells were treated with 5 mM $H_2O_2$ and 25 ${\mu}M$ rose bengal, isoquercitrin protected the cells against the oxidative damage in a concentration dependent manner (6.25 ~ 50 ${\mu}M$). The size of 0.03 % isoquercitrin loaded ethosome was 222.85 nm and the loading efficiency was 82.26 %. The ethosome loaded with 0.03 % isoquercitrin was stable and maintained the constant particle size for 2 weeks after being prepared. The ethosome exhibited more enhanced skin permeability than general liposome and ethanol solution. The optimal ratio of lipid to surfactant of 0.1 % isoquercitrin loaded elastic liposomes was observed to be 89 : 5 through evaluating particle size (341.2 nm), deformability index (59.89), loading efficiency (54.3 %), and skin permeability (54.4 %).

Effect of Isoquercitrin-containing Nelumbo nucifera Leaves Extract on Skin Wrinkle Improvement (Isoquercitrin 함유 연꽃잎 추출물의 피부주름개선 효능 연구)

  • In this study, we conducted research to find anti-wrinkle skin care ingredients from natural sources via the measurement of procollagen type 1 levels in the medium of normal human dermal fibroblast (NHDF) and found that a Nelumbo nucifera leave extract (NLE) was as the best effective ingredient. By the high performance liquid chromatography analysis, we confirmed isoquercitrin was one of the main compounds of NLE. Moreover, it also increased the procollagen type 1 production without cytotoxicity of NHDF. NLE and isoquercitrin exerted free radical scavenging activity. Especially, isoquercitrin exhibited strong intracellular antioxidant capacity in human skin-derived cells, HaCaT and NHDF. Finally, we performed clinical test for the inhibitory effect of NLE on skin wrinkle formation. A randomized study was conducted in 22 healthy female volunteers, aged between 30 and 65 yrs, with moderate to moderately severe facial wrinkles. Volunteer applied a 1.7% NLE cream (isoquercitrin 51 ppm) and a control cream at each facial side (left/right) twice daily for 8 weeks. The 1.7% NLE cream improved skin roughness through reducing the wrinkle of the craw's feet significantly without any skin side effect. Our results demonstrate that NLE and isoquercitrin can increase the collagen production and exert antioxidant activity. Therefore, we expect that the new non-toxic herbal extract, isoquercitrin-containing NLE will be interesting natural ingredient of cosmetics with anti-wrinkle efficacy.

Optimization of Extraction Conditions and Quantitative Analysis of Isoquercitrin and Caffeic Acid from Aster scaber

  • To determine the optimum extraction conditions that give the highest yield of isoquercitrin and caffeic acid from Aster scaber, the effects of four extraction variables (solvent concentrations, extraction time, number of repeated extraction, and solvent volumes) on isoquercitrin and caffeic acid yield was examined via HPLC-UV. Our results showed that the highest extract and isoquercitrin yield were observed when A. scaber was extracted with 450 mL distilled water for 8 hr repeatedly for three times. In case of caffeic acid, the content was higher in the two repeated extracts. Also, content analysis of isoquercitrin in Aster species was performed in which A. fastigiatus, A. ageratoides, and A. scaber exhibited the highest isoquercitrin content at 6.39, 5.68, and 2.79 mg/g extract, respectively. In case of caffeic acid, the highest content of A. scaber and A. glehni was 0.64 and 0.56 mg/g extract, respectively. This study reports an optimized method for extraction of isoquercitrin and caffeic acid from A. scaber and evaluates potential sources of the compounds.

Improvement and Validation of an Analytical Method for Quercetin-3-𝑜-gentiobioside and Isoquercitrin in Abelmoschus esculentus L. Moench (오크라 분말의 Quercetin-3-𝑜-Gentiobioside 및 Isoquercitrin의 분석법 개선 및 검증)

  • This study aimed to investigate the validation and modify the analytical method to determine quercetin-3-𝑜-gentiobioside and isoquercitrin in Abelmoschus esculentus L. Moench for the standardization of ingredients in development of functional health products. The analytical method was validated based on the ICH (International Conference for Harmonization) guidelines to verify the reliability and validity there of on the specificity, linearity, accuracy, precision, detection limit and quantification limit. For the HPLC analysis method, the peak retention time of the index component of the standard solution and the peak retention time of the index component of A. esculentus L. Moench powder sample were consistent with the spectra thereof, confirming the specificity. The calibration curves of quercetin-3-𝑜-gentiobioside and isoquercitrin showed a linearity with a near-one correlation coefficient (0.9999 and 0.9999), indicating the high suitability thereof for the analysis. A. esculentus L. Moench powder sample of a known concentration were prepared with low, medium, and high concentrations of standard substances and were calculated for the precision and accuracy. The precision of quercetin-3-𝑜-gentiobioside and isoquercitrin was confirmed for intra-day and daily. As a result, the intra-day precision was found to be 0.50-1.48% and 0.77-2.87%, and the daily precision to be 0.07-3.37% and 0.58-1.37%, implying an excellent precision at level below 5%. As a result of accuracy measurement, the intra-day accuracy of quercetin-3-𝑜-gentiobioside and isoquercitrin was found to be 104.87-109.64% and the daily accuracy thereof was found to be 106.85-109.06%, reflecting high level of accuracy. The detection limits of quercetin-3-𝑜-gentiobioside and isoquercitrin were 0.24 ㎍/mL and 0.16 ㎍/mL, respectively, whereas the quantitation limits were 0.71 ㎍/mL and 0.49 ㎍/mL, confirming that detection was valid at the low concentrations as well. From the analysis, the established analytical method was proven to be excellent with high level of results from the verification on the specificity, linearity, precision, accuracy, detection limit and quantitation limit thereof. In addition, as a result of analyzing the content of A. esculentus L. Moench powder samples using a validated analytical method, quercetin-3-𝑜-gentiobioside was analyzed to contain 1.49±0.01 mg/dry weight g, while isoquercitrin contained 1.39±0.01 mg/dry weight g. The study was conducted to verify that the simultaneous analysis on quercetin-3-𝑜-gentiobioside and isoquercitrin, the indicators of A. esculentus L. Moench, is a scientifically reliable and suitable analytical method.

Study on the Synthesis of Isoquercitrin by Enzymatic Partial Hydrolysis (효소를 이용한 부분 가수분해에 의한 isoquercitrin의 합성에 관한 연구)

  • Isoquercitrin (IQ), quercetin monoglycoside, is classified as a polyphenol, and a minute quantity of IQ is known to be present in several plants. Recently, it was reported that IQ can be prepared by the partial enzymatic hydrolysis of quercetin diglycoside (rutin, RU). In this paper, the effects of enzyme types, enzyme amounts, and substrate concentrations on the reactivity were investigated using a series of $Pecinex^{TM}$ multi-enzymes. The reaction, when a 8 ml of Ultra Clear to 1 g of RU was applied with the substrate concentration of 1% at $50^{\circ}C$, was found to be optimum, based on the reaction rate and the selectivity to IQ.

Chemical Constituents of Domestic Quercus spp. Leaves (국내산 참나무속 수종 잎의 추출성분)

  • This study was carried out to investigate chemotaxonomical correlation an d the chemical constituents of domestic Quercus sp. leaves. The leaves of Q. mongolica, Q. aliena, Q. serrata, Q. acutissima, Q. dentata and Q. variabilis were collected in the experimental forest of Kangwon National University. The combined extracts were successively fractionated with n -hexane, methylene chloride and ethyl acetate using a separation funnel. A portion of the ethyl acetate and $H_2O$ soluble materials of each species were chromatographed on a Sephadex LH-20 column using various aqueous MeOH and EtOH-hexane as washing solvents. Spectrometric analysis such as NMR and MS, including TLC, were performed to characterize the structures of the isolated compounds. Gallic acid, (+)-catechin, (-)-epicatechin, (+)-gallocatechin, kaempferol, astragalin, astragalin-6"-O-gallate, isoquercitrin, isoquercitrin-6"-O-gallate and myricetin were isolated from Q. mongolic a leaves. Gallic acid, kaempferol and quercetin were characterized from Q. acutissima leaves. Gallic acid, (+)-catechin, (-)-epicatechin, (+)-gallocatechin, (-)-epigallocatechin, kaempferol, quercetin, guajaverin and tamarixin were identified from Q. dentata leaves. Gallic acid, (+)-catechin, (-)-epicatechin, kaempferol, quercitrin, isoquercitrin and myricetin were purified from Q. serrata leaves. Gallic acid, (+)-catechin, astragalin, astragalin-6"-O-gallate and isoquercitrin were isolated from Q. variabilis leaves. Gallic acid was isolated from all the leaves and could be a taxonomic index on Quercus spp..

Biological Activities of Flavonoid Glycosides Isolated from Angelica keiskei (신선초에서 분리된 flavonoid glycosides의 생리활성)

  • Recently, much attention has been focused on plant antioxidants, because they are expected to protect against oxidative damage, possibly preserving biological functions of cells. Antioxidant compounds were isolated from Angelica keiskei through extraction with 80% EtOH, and fractionations were carried out sequentially with n-hexane, chloroform, ethyl acetate, n-butanol, and water. Two active compounds were isolated from ethyl acetate fraction by silica gel column chromatography, and were identified as isoquercitrin ($quercetin-3-O-{\beta}-D-glucose$) and hyperoside ($quercetin-3-O-{\beta}-D-glucose$). Isoquercitrin and hyperoside showed strong antioxidative potency, as revealed by evaluation of their ABTS, DPPH, OH, and $H_{2}O_{2}$ radical-scavenging activities, and ex vivo DNA damage-protecting effects.

Quantitative analysis of hyperoside and isoquercitrin in methanolic extract of Stewartia koreana leaves using HPLC-DAD

  • Since Stewartia koreana leaves are registered with the Food and Drug Administration as edible herbal materials, they are used in the development of functional foods, cosmetics, and medicines. In this study, we established an analysis method that can simultaneously analyze two indicators, hyperoside (quercetin 3-O-galactoside) and isoquercitrin (quercetin 3-O-glucoside) contained in the leaves of S. koreana using HPLC-DAD. In accordance with the Ministry of Food and Drug Safety's health functional food guidelines, the analysis method was verified for specificity, accuracy, precision, limit of quantification, and linearity. The analysis method established in this study showed more than 0.9989 of the correlation coefficient values (r2) for the calibration. The total recovery rates of isoquercitrin and hyperoside were 100.55 and 98.87% with 0.14-0.78 and 0.47-0.67% of the relative standard deviation, respectively. Therefore, it was suggested that the new analytical method would be applied to standardize raw materials and high value-added products originated from the leaves of the S. koreana in the future.
