Hydrolysis of Olive Oil by Lipase, Immobilized on Hydrophobic Support

  • Two commercially available lipases, Lipase OF (non-specific lipase from Candida rugosa) and Lipolase 100T (1, 3-specific lipase from Aspergillus niger), were immobilized on insoluble hydrophobic support HDPE (high density polyethylene) by the physical adsorption method. Hydrolysis performance was enhanced by mixing a non-specific Lipase OF and a 1, 3-specific Lipolase 100T at a 2 : 1 ratio. The results also showed that the immobilized lipase maintained its activity at broader temperature ($25~55^{\circ}C$) and pH (4-8) ranges than soluble lipases. In the presence of organic solvent (isooctane), the immobilized lipase retained most of its activity in upto 12 runs of hydrolysis experiment. However, without organic solvent in the reaction mixture, the immobilized lipase maintained most of its activity even after 20 runs of hydrolysis experiment.

The Optimum Effect of Long Chain Fatty Monoglyceride from Microemulsion by Lipase Catalyst (마이크로에멀젼에서 리파아제 촉매에 의한 고급지방산 모노글리세리드의 생성에 있어 최적효과)

  • Mono alkyl glycerides have bean obtained in good yield by enzyme catalyst from soybean oil. The reaction was carried out in an oil rich microemulsion formula. Best results were obtained with sodium bis(2-ethyl hexyl) sulfo succinate(AOT), isooctane as hydrocarbon component and buffer of pH 7. The enzyme used was a 1,3-specific lipase which leaves the 2-position intact. However, the 2-monoglyceride formed slowly undergoes long chain acyl migration to 1-mono-glyceride. Optimal reaction time at $35^{\circ}C$ reaction temperature was found to be three hour.

Separation of Lipases Using Reversed Micelles (역미셀을 이용한 리파아제의 분리)

  • The liquid-liquid extraction of lipase A from Candide cylindracea and lipase B from porcine pancrease was carried out using reversed micellar organic solvents. Effects of various factors such as ionic strength, pH, and species and concentration of surfactant, on lipase solubilization were studied. A cationic surfactant cetyl-trimethyl ammonium bromide (CTAB) in isooctane/nhexanol(1:1) was found to be an effective solvent and its optimum concentration was 50 mM. KCl among various salts tested was the most effective and the efficiency of solubilization of lipase increased with decreasing the ionic strength of salts. The maximum activity and solubiliz ation of protein were obtained at pH 8. The stripping efficiency has a maximum value at pH 4 and increases with KCl concentration in the range of 0.2∼1.0 M. After the solubilization and stripping, the overall recovery efficiency of mass and specific activity of lipases was 62% and 66%, respectively.

  • This study reports the extraction behavior of $\alpha$-lactalbumin using bis(2-ethylhexyl) sulfosuccinate sodium (AOT) reverse micelles. Forward extraction of $\alpha$-lactalbumin in the reverse micellar organic phase from aqueous feed solutions was strongly dependent on the AOT concentration and the complete forward extraction of 0.03 mM $\alpha$-lactalbumin was successfully achieved at an AOT concentration of ca. 100 mM. A similar dependency of the forward extraction on the AOT concentration was obtained in isooctane, n-hexane, and n-octane systems. In the backward extraction from the micellar organic phase, the recovery of the protein as high as ca. 90% was obtained with pH control and/or salt addition.

  • Bovine serum albumin(BSA) was solubilized into the reverse micellar phase consisting of sodium bis(2-ethylhexyl) sulfosuccinate(AOT) and isooctane using the phase transfer method. Of particular interest in this study were the effects of pH and the added salt type and concentration on the solubilization efficiency. When univalent or divalent salts such as KCl, NaCl, $MgCl_2$, or $CaCl_2$ were added to the aqueous phase at a concentration of 0.1 M, maximum solubilization efficiency was attained at a pH ranging from 5 to 7, depending on the added salt type. Increased salt concentration up to 1 M resulted in an increased solubilization efficiency for $CaCl_2$ and NaCl, while the addition of $MgCl_2$ beyond 0.1 M showed an anomalous trend. Further, it was noteworthy that too a large extent the protein precipitated in the interface between the organic and aqueous phases at lower pHs and lower salt concentrations. The size of the reverse micelle water pool was estimated by measuring the molar ratio of the surfactant to the water, $W_0$. Irrespective of pH in the aqueous phase, the resulting value of $W_0$ was almost constant, eg., 20 for $MgCl_2$ . However, the value of $W_0$ decreased with increased salt concentration in the cases of KCl and $CaCl_2$.

  • Effect of additives such as NP series nonionic surfactant and cosurfactant on AgCl nanoparticles was investigated where nanoparticles were prepared using two different types of water-in-oil (W/O) microemulsions containing silver nitrate and sodium chloride, respectively. Phase behavior experiments showed that the region of one phase W/O microemulsion was found to be broadened with an increase in the ethylene oxide length of a nonionic surfactant mainly due to an increase in hydrophilic nature of a surfactant. Photomicrographs obtained by transmission electron microscopy indicated that an increase in ethylene oxide length of a nonionic surfactant results in both increases in particle size and size distribution. Phase behavior experiments for the systems containing AOT surfactant, isooctane and aqueous solution of an inorganic salt showed that addition of a cosurfactant caused a shrinkage in phase region of one phase W/O microemulsion, especially water contents contained in W/O microemulsion with an increase in the chain length or the concentration of a cosurfactant used. Photomicrographs obtained by transmission electron microscopy indicated that characteristics of AgCl nanoparticles produced were dependent both on the radius of spontaneous curvature and film rigidity of a microemulsion.

Effect of Cosurfactant on Preparation of Silica Nanoparticles using Water in Oil Microemulsion of Nonionic Surfactant (보조계면활성제가 비이온 계면활성제의 Water in Oil 마이크로에멀젼을 이용한 실리카 나노입자 제조에 미치는 영향)

  • The effects of cosurfactant on silica nanoparticles were investigated in systems containing surfactant, oil and aqueous ammonia solution where nanoparticles were prepared using a single phase water-in-oil (W/O) microemulsion. For the same oil phase, a single phase region was dependent on the interaction between surfactant and oil. For the cyclohexane system, NP-5 surfactant showed a wider single phase region than NP-4. The addition of n-propanol as a cosurfactant resulted in an increase or a decrease of a single phase W/O microemulsion region depending on the continuous oil phase. For both cyclohexane and isooctane systems, the addition of n-propanol resulted in a decrease in the single phase region. On the other hand, for n-heptane system, the addition of n-propanol expanded a single phase W/O microemulsion region. Silica nanoparticles prepared within a single phase region showed that relatively large number of particles of irregular shape were obtained with the addition of n-propanol to NP surfactant system. The addition of n-propanol to LA-5 surfactant and n-heptane system produced a decrease in average particle size and an increase in the number of particles formed due to a decrease in the intermicellar exchange rate among microemulsion droplets.

Solid Bases as Racemization Catalyst for Lipase-catalyzed Dynamic Kinetic Resolution of Naproxen 2,2,2-Trifluoroethyl Thioester (리파아제에 의한 나프록센 2,2,2-트리플로로에틸 씨오에스터의 Dynamic Kinetic Resolution을 위한 라세미화 촉매로서의 고체 염기)

  • A variety of solid bases such as inorganic bases, basic anion exchange resins, and resin-bound bases were tested as a catalyst for racemization of (S)-naproxen 2,2,2-trifluoroethyl thioester in isooctane at 45$^{\circ}C$. Among the various bases, DIAIOM WA30, which is a weakly basic anion exchange resin with a tertiary amine based on a highly porous type styrene-divinylbenzene copolymer, showed the highest catalytic activity. The second-order interconversion constant of DIAION WA30 was 8.6${\times}$10$\^$-4/ mM$\^$-1/h$\^$-1/ and about 3 times higher than that of trioctylamine under the same conditions. The rate of DIAION WA30-catalyzed racemization decreased with increasing an amount of water added to the reaction medium. Lipase-catalyzed kinetic resolution of racemic naproxen 2,2,2-trifluoroethyl thioester was successfully carried out under in situ racemization of substrate with DIAION WA30 in isooctane at 45$^{\circ}C$. More than 60% conversion and 99% enantiomeric excess for the desired (S)-naproxen product were obtained. Furthermore, such a solid base catalyst could be easily separated and reused in contrast to trioctylamine.

Ketoprofen Resolution by Enzymatic Esterification and Hydrolysis of the Ester Product

  • Immobilized Candida antarctica lipase was used to catalyze the separation of ketoprofen into its components by means of esterification followed by the enzymatic hydrolysis of the ester product. In this study, ketoprofen underwent esterification to ethanol in the presence of isooctane. When the reaction was complete, 58.3% of the ketoprofen had been transformed into an ester. The ketoprofen remaining in solution after the reaction was complete consisted primarily of its S-enantiomer (83.0%), while the 59.4% of the ketoprofen component of the ester consisted of its R-enantiomer. We then subjected the ester product to enzymatic hydrolysis in the presence of the same enzyme and produced a ketoprofen product rich in the R-enantiomer; 77% of this product consisted of the R-enantiomer when 50% of the ester had been hydrolyzed, and 90% of it consisted of the R-enantiomer when 30% of the ester had been hydrolyzed. By contrast, the R-enantiomer levels only reached approximately 42 and 65%, respectively, when 50 and 30% of the racemic ester was hydrolyzed under the same conditions.

Fabrication of metal structure using AI sacrificial layer (알루미늄 희생층을 이용한 금속 구조물의 제작)

  • In this paper, novel release technique using wet etch is proposed. The results of this technique and the results of SAMs (Self-Assembled monolayers) coated after release using this technique are compared. Fabricated structure have 100 um in width and experimental length is from 100 um to 1 mm. Thickness of aluminum sacrificial layer is 2 um and structure thickness is 2.5 um. Cantilevers and bridges are fabricated with electroplated gold and silicon nitride deposited on substrate. An aluminium sacrificial layer was evaporated thermally and removed in various wet etching solutions. Detachment length of cantilever is 200 um and detachment length of bridge is 1 mm after isooctane rinsing. And the SAMs coating condition which is appropriate for gold and nitride are studied respectively.
