Isolations of Flavonoids and a Higher Alcohol from the Aerial Parts of Brassica juncea (갓 지상부에서 플라보노이드와 고급 알콜 화합물의 분리)

  • The aerial parts of Brassica juncea (Cruciferae) called by brown or oriental mustard have been widely used as a spice in food and also traditional folk medicine as stimulant, diuretic and expectorant agent. And Gatkimchi made of the aerial parts of this plant are very popular in Korea. The aerial parts of this plant was refluxed with MeOH and then fractionationed with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. One higher alcohol compound and three flavonoids were isolated from the EtOAc fraction through silica gel and Sephadex LH-20 column chromatographies. Their structures were elucidated as n-hexacosanol(1), kaempferol(2), isorhamnetin $3-O-{\beta}-D-glucopyranoside(3)$ and isorhamnetin $3-O-(6'-O-acetyl)-{\beta}-D-glucopyranoside(4)$ by comparison of spectral data with those in references. And compounds 1 and 4 were firstly isolated from this plant.

In vitro-growth and Gene Expression of Porcine Preantral Follicles Retrieved by Different Protocols

  • This study was conducted to determine how the isolation method of the porcine preantral follicles influenced the following follicular growth in vitro. Mechanical and enzymatical isolations were used for retrieving the follicles from prepubertal porcine ovaries, and in vitro-growth of the follicles and the expression of folliculogenesis-related genes were subsequently monitored. The enzymatic retrieval with collagenase treatment returned more follicles than the mechanical retrieval, while the percentage of morphologically normal follicles was higher with mechanical retrieval than with enzymatic retrieval. After 4 days of culture, mechanically retrieved, preantral follicles yielded more follicles with normal morphology than enzymatically retrieved follicles, which resulted in improved follicular growth. The mRNA expression of FSHR, LHR Cx43, DNMT1 and FGFR2 genes was significantly higher after culture of the follicles retrieved mechanically. These results suggest that mechanical isolation is a better method of isolating porcine preantral follicles that will develop into competent oocytes in in vitro culture.

  • A single degree of freedom system and transmissibility are key concepts in many problems of vibration isolation. In order to apply this approach, however, several assumptions must be satisfied, which are often not realistic. For examples, in practical systems, vibration transmissions at multi-point with multi-degree of freedom(translational and rotational DOF) take place and mobilities or impedances of receiver structures cannot be ignored any more especially over high frequency range. Therefore, a multi-dimensional treatment is required for accurate estimation of dynamic behavior of the system. In this paper, an approach using vibrational power flow is introduced to deal with analysis of multi-dimensional vibration isolation system in a more practical way and in aspects of vibration isolations and vibration path analysis. Procedures of this approach and some results of research for vibrational power path analysis with rotational terms included are presented. Difficulties in this method are also discussed.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

Studies on Nosocomial Pathogens Isolated from Veterinary Hospitals in Gwangju (광주지역 동물병원에서 분리한 의원성 병원체에 관한 연구)

  • We investigated the contamination of animal hospital floor, beauty table, computer keyboard, exam table, operation table and forcep handle by isolations of aerobic bacteria in small animal hospitals in Gwangju. The total number of aerobic bacteria was 52 isolates and Staphylococcus spp. (38 isolates) were the predominant isolates (69.71 %) of them. The prevalent contaminated areas were floor (17 isolates), beauty table (13 isolates) and computer keyboard (9 isolates). The detection of methicillin-resistant (mecA) gene, determined by PCR, showed that 3 of the 17 coagulase-negative Staphylococcus spp. (CNS) isolates possessed the mecA gene. For evaluating the antibiotic susceptibility patterns of the isolates, disk diffusion method was used. The majority of isolates showed high susceptibility to amoxicillin (92.1 %), ceftiofur (84.2%) and polymixin B (73.7%). Also they showed the high resistant to ampicilline (66.7%), penicillin (65%) and kanamycin (56.5%). These results suggest extensive contamination of aerobic bacteria in animal hospital environment.

Isolations of the Orchid mycorrhizal Fungi from the Roots of the Korean native Orchids and Inoculations of the Isolates to Four different Orchids (한국 자생란에 난 균근균의 분리와 유묘난에 접종)

  • Ten isolates of the orchid mycorrhizal fungi were isolated from the roots of Korean native orchid plants (Cymbidium goeringii) which inhabitate mainly in southern and western areas of Korea. The growth rates and color of the isolates in potato dextrose agar (PDA) were various. Microscopic observations of the hyphae isolated were identified as Rhizoctonia repens and R. endophytica var endophytica or their related species. R. repens was isolated from the roots of the Korean native orchids, but R. endophytica var endophyica was only isolated from the roots of the commercial orchids introduced from foreign countries. Also, the polymorephic patterns of genomic DNA extracted from selected isolates were compared with those of DNA extracted from the orchid mycorrhizal fungi isolated previously and similar band patterns were observed among those isolates. Five isolates of R. repens were selected and cultured at the oatmeal agar for investigating their symbiosis with orchid plants. The symbiotic specificity between orchid plants and isolated orchid mycorrhizal fungi was observe by growing orchids about six months in the greenhouse. The symbiotic responses of the commercial orchid plants with selected isolates were quite different form different isolates due to the genetic variations.

Low Profile Dual Feeding Microstrip Antenna with High Isolation (높은 격리도를 갖는 Low Profile 이중급전 마이크로스트립 안테나)

  • In this paper, a novel low profile dual feeding microstrip antennas with microstrip and H-shaped aperture feeding are proposed to achieve high isolation and easy feeding, and the validity of the proposed antennas is demonstrated experimentally. In the proposed single and 1$\times$4 array antenna, the measured isolations are less than -49 dB and -48 dB, impedance bandwidths are 4 % at 12.5 GHz and 2.1 % at 14.25 GHz for the VSWR<1.5 in both antennas and maximum gains are 8.25 dBi and 14.2 dBi.

Design of A Compact Single-Balanced Mixer for UWB Applications

  • The design and implementation aspects of a new single-balanced mixer for ultra-wideband (UWB) applications are presented in this study. The proposed mixer utilizes a miniaturized UWB ring coupler as a balun, consisting of a pair of in-phase and inverted-phase transitional structures. The well-balanced UWB performance of the ring coupler, aside from the optimized diode matching, results in improved conversion loss and inter-port isolations for a wide bandwidth. The size of the implemented single-balanced diode mixer is reduced to about 60% of the area of the conventional single-balanced ring diode mixer. The measured results of the proposed mixer exhibit an average conversion loss of 7.5 dB (minimum 6.7 dB) and a port-to-port isolation of greater than 18 dB over a UWB frequency range of 3.1-10.6 GHz. The measured results agree well with the simulated results.

  • The seismic performance evaluation and retrofit process are very important in old existing bridges. If the result is not appropriate. then a retrofit process are required. Among various retrofit methods, the seismic isolation is a very useful method. because it can be applied by replacing old bridge bearings. In this study, the effectiveness of seismic isolation is rationally verified. For this purpose, two seismic isolations used widely are selected and non-linear static and dynamic analyses are performed. The responses of existing bridges are compared with those of retrofited bridges by seismic isolation bridge for earthquake of target level. and seismic performances are evaluated.

  • Human have eaten various traditional fermented foods for a numbers of million years for health benefit as well as survival. The beneficial effects of fermented foods have been resulted from complex microbial communications within the fermented foods. Therefore, the holistic approaches for individual identification and complete microbial profiling involved in their communications have been of interest to food microbiology fields. Microbiome is the ecological community of microorganisms that literally share our environments including foods as well as human body. However, due to the limitation of culture-dependent methods such as simple isolations of just culturable microorganisms, the culture-independent methods have been consistently developed, resulting in new light on the diverse non-culturable and hitherto unknown microorganisms, and even microbial communities in the fermented foods. For the culture-independent approaches, the food microbiome has been deciphered by employing various molecular analysis tools such as fluorescence in situ hybridization, quantitative PCR, and denaturing gradient gel-electrophoresis. More recently, next-generation-sequencing (NGS) platform-based microbiome analysis has been of interest, because NGS is a powerful analytical tool capable of resolving the microbiome in respect to community structures, dynamics, and activities. In this overview, the development status of analysis tools for the fermented food microbiome is covered and research trend for NGS-based food microbiome analysis is also discussed.
