2-deoxy-D-glucose가 첨가된(25 pg/ml) 최소액체 배지에 8.5시간 전 배양하여 swelling 시킨 conidiospore에 2% driselase와 2% ${\beta}-glucuronidase$를 동량 혼합한 효소용액을 처리하여 반응시킨 결과 protoplast형성 시간 2시간이내로 단축 시킬 수 있었다. 5% Ficoll(m.w. 400,000)용액을 사용하여 conidial protoplast를 보다 순수하게 분리 벙제할 수 있었으며, protoplast를 Giemsa로 핵 염색한 결과, 대부분의 protoplast는 하나의 핵을 갖고 있었다. 또한 전자현미경으로 관찰한 결과 conidiospore에서 유래된 protoplast는 세포벽 성분이 남아 있지 아니한 완전한 protoplast인 것으로 판명되었다. 영양요구성 돌연변이주에서 유래된 conidial protoplast의 융합률은 $3.4{\times}10^{-1}$에서 $4.9{\times}10^{-1}$수준으로 이는 mycelial protoplast의 경우보다 5-28배 가량 높은 수준의 것이었다.
본 실험에서는 에탄올로 추출한 Psidium guajava 잎과 컬럼크로마토그래피를 이용한 메탄올 분획물의 항산화 능력을 측정하기 위해 DPPH, ABTS 라디컬소거능법, 총 페놀, 플라보노이드법, SOD assay법을 활용하였다. 항산화 활성 결과에서 40, 60% 메탄올 분획물이 의미있는 측정값을 나타냈고, 두 분획물을 선정하여 HPLC와 핵자기공명(NMR)을 활용한 화학적 분석을 통해 주요 성분의 분리 및 물질탐색을 진행하였다. HPLC 정제를 통해 quercitrin (1)와 isoquercetin (2) 두 종류의 quercetin 유도체를 분리 및 구조 동정하였다. SOD assay 결과에서 컬럼크로마토그래피법을 활용하여 얻은 몇몇의 분획물과 단일물질로 분리한 화합물들은 추출물보다 향상된 항산화 활성을 보였다.
국내산 갈조류로부터 생리 기능성이 우수한 수용성 식이섬유 소재를 개발하기 위한 기초자료를 얻기위해 다시마와 미역(엽상체와 포자엽 각각)으로부터 fucoidan을 추출, 정제하여 그 특성을 조사하였다 Fucoidan의 추출 조건은 pH 2.0의 산성 추출액으로 $65^{\circ}C$I에서 1시간 씩 3회 추출을 하였고 cetylpyridinium chloride를 사용하여 부분 정제하였다. 부분 정제 fucoidan의 수율은 다시마 $2.71\%$, 미역포자엽 $6.65\%$, 미역 엽상체$0.40\%$였다. 특히 미역포자엽의 수율이 가장 높은 반면에 미역 엽상체는 가장 낮아 부위에 따른 차이가 매우 큼을 알 수 있었다. DEAE-Sephadex A-25 이온 교환 칼럼으로 부분 정제 fucoidan을 분획한 결과 다시마, 미역포자엽 모두 3종의 fraction으로 분획되었고 각 fraction은 전기 영동상에서 2종 이상의 band가 검출되어 불균일함을 알 수 있었다 이온 교환 칼럼으로 분획한 주fraction을 다시 알콜 분별 침전법으로 재분획한 결과 다시마와 미역포자엽은 에탄올농도 $60-70\%$에서 침전된 fraction이 cellulose acetate 전기 영동 및 gel filteration chromatography상에서 균일성을 나타내었다. 균일하게 정제된 fucoidan의 fucose : galactose : 황산기의 mole 조성비는 다시마, 미역포자엽이 각각 1 : 0.31 : 2.43,1 : 0.87 : 1.99였다. 분자량은 다시마가 31,000, 미역포자엽이 38,000으로 비교적 적었다.
토양으로부터 tyrosinase 저해제를 생산하는 방선균 F-97을 분리하여, 이 균이 생산하는 tyrosinase 저해제를 정제하였다. 먼저 배양액을 원심분리하고 이 여액을 pH 4.0으로 조절한 후 IRC-120($NH_4^+$ type column chromatography, Silica gel column chromatography, C18 column chromatography, Sephadex LH-20 column chromatography를 사용하여 정제하였다. 정제도 확인은 ODS HPLC를 이용하여 확인하였으며, 최종 정제 수율은 5.24%이었다. 물리 화학적 특성으로 tyrosinase 저해제는 water, methanol, ethanol 등에는 잘 녹았으나, acetone, butanol, ethylacetate, chloroform 등에는 녹지 않는 수용성 물질이었다. 물을 용매로 UV 흡광도를 측정한 결과 194nm에서 최대 흡광도를 나타내었다. 본 tyrosinase 저해제는 Iodine, Ninhydrin, Millon, Sakaguchi, Xanthoproteic, Emerson 시험에서는 음성이었고, Molish, Benedict, conc. $H_2SO_4$, $KMnO_4$ 시험에서는 양성이었다. 저해제의 열 안정성은 $100^{\circ}C$ 50분까지 안정하였고, pH $4{\sim}9$에서 안정하였다. Tyrosinase 저해제의 mushroom tyrosinase에 대한 $IC_{50}$ 값은 $19.2{\mu}g/ml$이었고, Streptomyces bikiniensis NRRL B-1049에 대한 저해활성은 $1,000{\mu}g/ml$ 일 때 27mm이었다. 그리고 본 저해제의 저해 양상은 경쟁적 저해로 확인되었다.
Ovarian follicular fluid contains both stimulatory and inhibitory agents that influence the growth and maturation of oocyte. In the present study, an attempt was made to isolate and study the biological properties of ovarian follicular fluid peptide(s) in buffaloes. Bubaline ovarian follicular was made steroid- and cell-free. A protein fraction was obtained by saturation (30-35% level) of the follicular fluid with ammonium sulfate. The protein fraction was purified with Sephadex-G 50 gel filtration chromatography and a single peak was obtained in the eluant volume, which was lyophilized. SDS-PAGE of the lyophilized fraction revealed a single band and the molecular weight of the peptide was 26.6 kDa. The peptide stimulated the cumulus cell expansion and in vitro maturation rate of oocytes in buffaloes in a dose dependent manner when it was incorporated at different dose levels (0, 10, 25, 50, 100 and 1,000 ng $ml^{-1}$ of maturation medium). The basic culture medium consisted of TCM 199 with Bovine serum albumin (0.3%). The in vitro maturation rates were comparable to those obtained with a positive control medium (TCM 199+20 ng EGF $ml^{-1}$+steer serum (20%)). Further purification and biological assays may throw more light on the nature and functions of this peptide.
We previously reported the isolation of a sialic acid-specific lectin eluted from the bark of Maackia fauriei using alkaline buffer on a fetuin-affinity column. Application of a borate-based elution buffer in the present study increased the specific activity of purified lectin from crude protein extract by 2.6-fold, whilst only slightly decreasing the recovery by 1.13%. The biological properties of the lectin eluted with borate buffer were the same as those of the lectin eluted with alkaline buffer such as in terms of the hemagglutination activity, hemagglutination inhibition activity, molecular mass, purity, and cytotoxicity to human breast cancer cells. A prepared biotin-labeled lectin conjugate was used to investigate the binding to various glycoproteins. Our results indicate that eluting with borate buffer is more efficient than using alkaline buffer to isolate the lectin adsorbed in a fetuin-affinity column.

본 실험에서는 토마토의 종자를 기내 배양하여 얻어진 1g 이하의 무균 잎 조직을 이용하여 미토콘드리아를 분리 정제하여 MitoTracker를 이용하여 세포생물학적으로 확인하였고, 이들의 mt를 이용하여 미토콘드리아 DNA와 RNA를 추출과 검정을 하였다. 또한 고농도의 이온성을 이용하여 미토콘드리아와 mtDNA 및 mtRNA을 추출할 수 있었으며, 식물의 여러 종류에도 사용되어질 수 있을 것이다. mtDNA는 PCR 분석에 의하여 plastid DNA와 혼재되어 있지 않음을 확인하였다. mtRNA는 RT-PCR 분석을 통하여 plastid RNA와 흔재되어 있지 않음을 확인할 수 있었다.
In the study presented here, identification, purification, and partial characterization of a hemin-regulated protein in Prevotella nigrescens were carried out. The results of this study confirm that the availability of hemin influences the expression of a selected membrane protein as well as the growth rate of P. nigrescens ATCC 33563. The 50 kDa cell envelope associated protein, whose expression is hemin regulated, is considered to be a putative hemin-binding protein from P. nigrescens. Disulfide bonds were not present in this protein, and N'-terminal amino acid sequence analysis revealed that this protein belongs to a new, so far undescribed protein. The 50 kDa protein was found to be rich in hydrophilic amino acids, with glycine comprising approximately 60% of the total amino acids. The study described here is the first to identify, purify, and biochemically characterize a putative hemin-binding protein from P. nigrescens. Work is in progress to further characterize the molecular structure of this protein.
