Gut-residing Microbes Alter the Host Susceptibility to Autoantibody-mediated Arthritis

  • K/BxN serum can transfer arthritis to normal mice owing to the abundant autoantibodies it contains, which trigger innate inflammatory cascades in joints. Little is known about whether gut-residing microbes affect host susceptibility to autoantibody-mediated arthritis. To address this, we fed C57BL/6 mice with water containing a mixture of antibiotics (ampicillin, vancomycin, neomycin, and metronidazol) for 2 weeks and then injected them with K/BxN serum. Antibiotic treatment significantly reduced the amount of bacterial genomic DNA isolated from fecal samples, in particular a gene encoding 16S ribosomal RNA derived from segmented filamentous bacteria. Arthritic signs, as indicated by the arthritic index and ankle thickness, were significantly attenuated in antibiotic-treated mice compared with untreated controls. Peyer's patches and mesenteric lymph nodes from antibiotic-treated mice contained fewer IL-17-expressing cells than those from untreated mice. Antibiotic treatment reduced serum C3 deposition in vitro via the alternative complement pathway. IL-$17^{-/-}$ congenic C57BL/6 mice were less susceptible to K/BxN serum-transferred arthritis than their wild-type littermates, but were still responsive to treatment with antibiotics. These results suggest that gut-residing microbes, including segmented filamentous bacteria, induce IL-17 production in GALT and complement activation via the alternative complement pathway, which cause the host to be more susceptible to autoantibody-mediated arthritis.

Porcine Circovirus Infection in Weaned Pigs with Postweaning Multisystemic Wasting Syndrome in Korea (국내 이유자돈의 써코바이러스 감염에 의한 이유후전신소모성 증후군)

  • Eight nursery to grower pigs exhibiting weight loss and sudden death were diagnosed as postweaning multisystemic wasting syndrome (PMWS) based on the results of gross findings, histopathology, immunohistochemistry, fluorescent antibody test, virus isolation, PCR, serology, and electron microscopy. Groosly, the pigs had a rough hair coats and were severely emaciated. And moot lymph nodes were pale and enlarged. Lungs were not fully collapsed and exhibited 10 to 40% pale red cranioventral consolidation. Histopathologically, typical lymphohistiocytic interstitial to bronchointerstitial pneumonia, chronic lymphadenitis, severe lymphoid depletion, and basophilic intracytoplasmic inclusions were noted in the most lymphoid tissues. Porcine circovirus panicles were observed in the inguinal lymph node of the pigs by electron microscopy. Porcine circovirus type 2 (PCV2) antigens or viral DNAs were detected in the lesions of all pigs using immunohistochemistry or PCR. Two PCV2 were isolated from a homogenate of pooled lung and lymph node in 2 of the 5 pigs. Additionally, antigens of porcine reproductive and respiratory syndrome virus (PRRSV) and Hemophilus (H.) parasuis were also detected by immunofluorescent antibody test. Serologically, 55% of randomly selected sows and fattening pigs was serum antibody positive to PCV2 by an indirect fluorescent antibody (IFA) test and approximately 18 % of animals in the herd were serologically pooitive by the ELISA kit for PRRSV. To our knowledge, this is the first report of PMWS co-infected with PCV-2, PRRS, and H. parasuis in Korea.

Selective Attentive Learning for Fast Speaker Adaptation in Multilayer Perceptron (다층 퍼셉트론에서의 빠른 화자 적응을 위한 선택적 주의 학습)

  • In this paper, selectively attentive learning method has been proposed to improve the learning speed of multilayer Perceptron based on the error backpropagation algorithm. Three attention criterions are introduced to effectively determine which set of input patterns is or which portion of network is attended to for effective learning. Such criterions are based on the mean square error function of the output layer and class-selective relevance of the hidden nodes. The acceleration of learning time is achieved by lowering the computational cost per iteration. Effectiveness of the proposed method is demonstrated in a speaker adaptation task of isolated word recognition system. The experimental results show that the proposed selective attention technique can reduce the learning time more than 60% in an average sense.

Preferential Expression of IgA Isotype Switching-associated Transcripts in Mouse Intestinal Lymphoid Tissues (마우스 내장 림프조직에서 우세하게 발현되는 IgA Isotype Switching 관련 전사체의 분석)

  • Background: Transforming growth factor-${\beta}$ (TGF-${\beta}1$) directs class switch recombination (CSR) to IgA isotype, which is a predominant antibody in mucosal surfaces. Although IgA is preferentially committed in mucosal lymphoid tissues, it is not definitely established whether hallmarks of IgA CSR such as IgA germ-line transcripts (GLT ${\alpha}$), post-switch transcripts (PST ${\alpha}$) and circle transcripts (CT ${\alpha}$) are readily expressed in such tissues. Therefore, we compared the expression of these transcripts among mouse Peyer's patches (PP), mesenteric lymph nodes (MLN), and spleen. Methods: Levels of GLTs, PSTs and CTs were measured by RT-PCR in isolated PPs, MLNs and spleen cells. Results: GLT ${\alpha}$ and PST ${\alpha}$ were well expressed in PP and MLN cells but in spleen cells. Similar patterns were observed in the expression of GL ${\gamma}$2b and PST ${\gamma}$2b. On the other hand, these transcripts were only inducible in spleen cells upon stimulated with LPS and TGF-${\beta}1$. In addition, CT${\alpha}$ and CT${\gamma}$2b were detected in PP cells. Conclusion: PP B cells readily express IgA GLT, PST, and CT. Overall expression patterns of these transcripts were similar in MLN cells. Thus, these results suggest that microenvironment of PP and MLN influences spontaneous IgA CSR, which lacks in systemic lymphoid tissues such as spleen.

A Proteomics Based Approach Reveals Differential Regulation of Visceral Adipose Tissue Proteins between Metabolically Healthy and Unhealthy Obese Patients

  • Obesity and the metabolic disorders that constitute metabolic syndrome are a primary cause of morbidity and mortality in the world. Nonetheless, the changes in the proteins and the underlying molecular pathways involved in the relevant pathogenesis are poorly understood. In this study a proteomic analysis of the visceral adipose tissue isolated from metabolically healthy and unhealthy obese patients was used to identify presence of altered pathway(s) leading to metabolic dysfunction. Samples were obtained from 18 obese patients undergoing bariatric surgery and were subdivided into two groups based on the presence or absence of comorbidities as defined by the International Diabetes Federation. Two dimensional difference in-gel electrophoresis coupled with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was carried out. A total of 28 proteins were identified with a statistically significant difference in abundance and a 1.5-fold change (ANOVA, $p{\leq}0.05$) between the groups. 11 proteins showed increased abundance while 17 proteins were decreased in the metabolically unhealthy obese compared to the healthy obese. The differentially expressed proteins belonged broadly to three functional categories: (i) protein and lipid metabolism (ii) cytoskeleton and (iii) regulation of other metabolic processes. Network analysis by Ingenuity pathway analysis identified the $NF{\kappa}B$, IRK/MAPK and PKC as the nodes with the highest connections within the connectivity map. The top network pathway identified in our protein data set related to cellular movement, hematological system development and function, and immune cell trafficking. The VAT proteome between the two groups differed substantially between the groups which could potentially be the reason for metabolic dysfunction.

A Study on the Recognition of Korean Numerals Using Recurrent Neural Predictive HMM (회귀신경망 예측 HMM을 이용한 숫자음 인식에 관한 연구)

  • In this paper, we propose the Recurrent Neural Predictive HMM (RNPHMM). The RNPHMM is the hybrid network of the recurrent neural network and HMM. The predictive recurrent neural network trained to predict the future vector based on several last feature vectors, and defined every state of HMM. This method uses the prediction value from the predictive recurrent neural network, which is dynamically changing due to the effects of the previous feature vectors instead of the stable average vectors. The models of the RNPHMM are Elman network prediction HMM and Jordan network prediction HMM. In the experiment, we compared the recognition abilities of the RNPHMM as we increased the state number, prediction order, and number of hidden nodes for the isolated digits. As a result of the experiments, Elman network prediction HMM and Jordan network prediction HMM have good recognition ability as 98.5% for test data, respectively.

Effusion Cytology of Ki-1 Positive Anaplastic Large Cell Lymphoma - A Case Report - (Ki-1 양성 역형성 대세포 림프종의 체액 세포학적 소견 - 1예 보고 -)

  • Ki-1 positive anaplastic large cell lymphoma is a newly described high-grade lymphoma and is defined by histopathological and immunologic criteria. We experienced a case of systemically involving Ki-1 positive anaplastic large cell lymphoma in a 44 year-old female which initially manifested as pleural effusion. Abdominopelvic CT scan showed the evidence of marked lymphadenopathy in retroperitoneal and both external and inguinal lymph nodes. On cytologic examination of pleural fluid, tumor cells revealed pleomorphic large isolated cells with prominent nucleoli and abundant cytoplasms. The nuclei were large with irregular profiles including some deep invaginations. Also, occasional multilobed/multinucleated and binucleated nuclei were seen. Immunohistochemical examination was performed to differentiate from the undifferentiated adenocarcinoma, Hodgkin's disease, non-Hodgkin's lymphoma and malignant histiocytosis. The neoplastic cells were positive for leukocyte common antigen, CD3, CD30(Ki-1) but negative for cytokeratin, epithelial membrane antigen, and CD15. A histologic diagnosis of Ki-1 positive anaplastic lymphoma was made by biopsies of the inguinal lymph node, polypoid lesions of the stomach and cecum.

Clinical and Histopathogical Studies of Experimentally Induced Listeriosis in Korean Native Goats (한국재래산양의 Listeria병에 관한 임상 및 병리조직학적 연구)

  • These studies were taken experimentally to clarify the clinical and histopathological findings of listeriosis in Korean native goat(KNC). Four KNGs of 4 to 5 months of age were inoculated orally or in-travenously (IV) with Listeria monocytogenes isolated from a field case of KNG. 1. On the clinical findings, depression, anorexia and fever were observed in all inoculated goats, and nasal discharge, keratoconjunctivitis and diarrhea in 3 of 4 goats. Highest rectal temperature after in-oculation was 2.5$^{\circ}C$ higher in IV inoculated goats and 1.9$^{\circ}C$ higher in orally inoculated than normal rectal temperature observed before inoculation. Durations of clinical course after inoculation in IV and orally inoculated goats were 5 days and 8 days, respectively. 2. On the gross lesions, swelling of the lymph nodes, hemorrhage and .inflammation of the small intestine and rigor mortis were observed in 4 of 4 goats, and keratoconjunctivitis, hemorrhage and inflammation of the large intestine, swelling of the spleen, pneumonia and hydropericardium in 3 of 4 goats. Congestion of the visceral organs and ecchymosis of the sin in a fetus were observed. Keratoconjunctivitis, hemorrhage of the abomasum, swelling of the lymph node, hemorrhage and inflammation of the small intestine, swelling of the spleen, necrosis of the liver and pneumonia were observed as severe lesions. These lesions were more severe in IV inoculated goats than those in orally inoculated goats. 3. On histopathological findings, focal necrosis found throughout the livers occurred mainly on peripheral areas of hepatic lobules. These necrotic foci consisted of neutrophils, lymphocytes, macrophages, short rod bacteria and necretic hepatic cells. Suppurative pneumonia of the lung, hyperemia, congestion, epithelial necrosis and sloughing of the small and large intestine, swelling in periventricular regions of the central nervous system, swelling of the kidney, spleen and lymph node were observed as listerial lesions.

Outbreaks of Strangles due to Capnophilic Streptococcus equi subsp equi in South Korea (호이산화탄소성 Streptococcus equi subspecies equi에 의한 선역의 발생)

  • We reported an outbreak of clinical strangles in thoroughbred horses due to capnophilic Streptococcus equi subsp equi in South Korea. On three different farms, we isolated 17 S equi subsp equi isolates from 29 horses with or without abscesses in their lymph nodes. Of the 17 isolates, two isolates from clinical cases grew well in aerobic conditions, whereas 7/7 isolates from clinical cases and 8/22 isolates from the nasal discharges of horses did not. The latter 15 isolates were capnophilic, oxygen-sensitive, and $CO_2$-requiring S equi subsp equi, which could not grow in aerobic conditions, but which grew well in a $CO_2$ incubator with 5% $CO_2$, in anaerobic conditions using a GasPak, and with reduced oxygen tension in a candle jar. This study is the first report of a strangles outbreak caused by capnophilic S equi subsp equi in South Korea.

Pathological studies on exudative epidermitis in experimentally infected pigs I. Macroscopical and histopathological observations (실험적 돼지 삼출성 표피염에 관한 병리학적 연구 I. 육안적 및 병리조직학적 관찰)

  • To elucidate morphologic lesion of porcine exudative epidermitis which is occurred sporadically in Korea, Staphylococcus hyicus subsp. hyicus isolated from the naturally affected pigs was inoculated to suckling pigs. The infected piglets were observed grossly and histopathologically. Although affected piglets were taking acute, subacute, or chronic course, some piglets suffered from chronic disease showed poor prognosis and marked growth depression. Affected peglets had erythematous skin on the face, ear, and abdomen and these localized lesions appear as brownish spots of exudative epidermitis and fromed crust in the early stage. But, after this stage, the skin were covered by viscous greasy exudate and formed blackish brown crust and appeared fissures and hypertrophy. Grossly, there has been hemorrhage with the removal of crust-like materials of epidermis and edematous subcutis. The superficial lymph nodes were edematous and swollen or congested and hemorrhagic. Some piglets had swollen ureters, cysts in the renal cortex, or polyarthritis. A few cases had mild edematous swelling of kidney, intestinal catarrh and congestion of brain. Microscopically, skin lesions had detachment of keralinized layer and parakeratosis of epidermis, hydropic degeneration of epidermal cell, and retrogressive degeneration of hair root sheath. Dermis had edema, and infiltration of neutrophils and mononuclear cells. As the disease was proceeded, there was marked perivasculitis with lots of mononuclear inflammatory cells. More chronic lesions formed granuloma-like bodies(nodules) due to more mononuclear, perivascular inflammatory cell infiltration and proliferation of fibroblast. Lots of plasma cells and eosinophils were also present in dermis. Epidermis was hyperplastic by proliferation of basal cells stratum germinativum and epidermal pegs often extended into the dermis. In secondary infection, lots of neutrophils could be seen in epidermis and derms. Kidney had neutrophilic infiltration, necrotic and cystic glomeruli, and dilation of renal tubules and ureters. Purulent arthritis was sometimes observed in joints. Three days old mice administrated Staphylococcus hyicus subsp hyicus subcutaneously before had focal congestion and hemorrhage, necrosis, and subcutaneous edema of the skin. This observation was also seen in the study of mice administrated exfoliatin toxin of Staphylococcus which evoked human staphylococcal scalded skin syndrome.
