Detection of Gamma-Irradiated Spices by ESR Spectroscopy (ESR Spectroscopy에 의한 감마선 조사된 향신료의 확인)

  • Electron spin resonance(ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity of irradiated spices and to identify the stability of radicals after storage. Red, white and black peppers, and garlic powders were irradiated with doses of 0, 1, 5, 10, 20 and 30 kGy at room temperature using a Co-60 irradiator. Triplet ESR signals were observed in irradiated pepper powders, while singlet ESR signals were observed in irradiated garlic powders. Those characteristic signals were not detected in non-irradiated samples. The strength of ESR signals linearly increased with the applied doses(1~3 kGy). Highly positive correlation coefficients ($R^2$=0.9757~0.9933) were obtained between the irradiation doses and the corresponding ESR signal intensities. The signal intensities of irradiated samples were stable even after 97 days of storage at room temperature.

  • Electron spin resonance (ESR) spectroscopy was used to detect irradiated meat containing bones (chicken, pork and beef), to investigate the effect of irradiation dose on the ESR signal intensity and to identify the stability of radicals under 9 weeks of storage. Chicken, pork and beef were irradiated with doses 0, 1, 3, 5 and 7 kGy at room temperature using a Co-60 irradiator. Bones were pieced and dried, which were placed in a quartz tube within an Electron paramagnetic resonance (EPR) spectrometer resonator cavity. The irradiated bone presented an asymmetric absorption in shape, different from that of a non-irradiated one. The signal intensity of smaller animals are lower than larger species. Variation was observed between samples of the same species depending on the calcification status of the bone. Moreover different irradiation doses produced different signal areas that make possible to estimate the absorbed dose of treated meat. The ESR signal stability after irradiation was stable in even after a 9 week storage at room temperature.

Utilization of Brabender Visco-Amylograph to Detect Irradiated Starches

  • A study was carried out to establish the detection method of irradiated corn, potato, and sweet potato starches. The samples were packed in polyethylene bags and irradiated with 1, 3, 5, 7, 10, and 15 kGy using a Co-60 irradiator. The maximum viscosity of irradiated and unirradiated corn, potato, and sweet potato starches reduced by increase of irradiation dose levels and showed significant differences which clearly showed the effect of irradia-tion dose levels (p<0.05). Regression expressions and coefficients (p<0.000) or corn, potato, and sweet potato starches were y=-38.538x+718.23(r2=0.9761), y=669.97e-0.1372x (r2=0.9820) and y=-42.544x+730.26(r2=0.9939), respectively. Nor-malized parameter A,B and C values showed a dose dependent relationship and were a better parameter for detecting the irradiated starches than that of the maximum viscosity itself.

  • To determine the radiosensitivity and dose-survival characteristics of jejunal crypt cells, experimental study was done using total 40 mice. Single irradiation of 1,000 rad to 1,600rad was delivered to whole bodies of mice, using a cesium 137 animal irradiator. The number of regenerating crypts per jejunal circumference was counted, by using a jejunal crypt cell assay technique, and dose response curve was measured. The average number of jejunal crypt Per circumference in control group was $140\pm10$. Mean lethal dose$(D_0)$ of moose jejunal crypt cell was 135rad.

Study on the Degradation of Pigskin Collagen Using Irradiation Technique (방사선조사를 이용한 돈피 콜라겐의 저분자화 연구)

  • This study was intended to observe possibility of which radiation technique can be used for oligopeptide production from pigskin collagen to reduce environmental pollution in processing and simplify the processing steps. Raw pigskin was ground using chopper, and then defatted in acetone cooled at $-20^{\circ}C$ freezer. Defatted dried pigskin was irradiated at 20, 40, 60, 100, 150, 200, 250, and 300 kGy using Co-60 gamma rays irradiator. With irradiation doses, the amount of soluble proteins increased, and the viscosity and turbidity of soluble proteins decreased, which could be clue of that irradiation degrade high molecular proteins directly. pH of soluble proteins from defatted pigskin increased in the sample above 150 kGy, and low molecular weight components (below 24 kDa) in SDS-PAGE increased. From gel permeation chromatography of the hydrolysates of pigskin irradiated at 300 kGy showed the major peak of 9,000, 8,200, 860, and 170 Da.

Production of Food and Fodder Yeasts from Seaweed (해조(海藻)를 이용한 효모제조(酵母製造)에 관한 연구)

  • 1) For the purpose of preparation of food and fodder yeasts from nonedible seaweed, two suitable Candida yeasts have been isolated from seaweed compost. 2) They had the ability of fermenting galactose, sucrose and glucose, and could not ferment maltose and mannit, but could assimilate mannit. 3) NaCl concentration from 1 to 2% had no remarkable effect on growth of yeast and the optimum pH was $4{\sim}5$. 4) In the acid hydrolyzate of brown seaweed (Ecklonia cava Kjellman, Sargassum fulvellum AGARDH) an amorphous deposit was produced during storage after neutralization of media and its removal always delayed yeast growth, but addition of $(NH_4)_2SO_4$ and $NaH_2PO_4$ to media could increase the assimilation of reducing sugar and yeast yield. 5) $Co^{60}$ gamma ray irradiation (dose rate : 1 Mrad/hr, BNL shipboard irradiator) of seaweed had not so much effect on the hydrolysis of carbohydrates and nitrogen compounds in seaweed but could increase the yeast production from seaweed hydrolyzate. 6) The yeast yield was $7{\sim}8$ g of dry yeast per 100 g of seaweed by cultivation with jar fermentor.

A Study on the Performance Evaluation of Standard Gamma Irradiation System Using Monte Carlo Code (몬테카를로 코드를 활용한 표준 감마선 조사장치의 성능평가에 관한 연구)

  • In this study, we compared the measured values of the effective beam size of standard gamma irradiator with the simulation results to provide a useful means to the effective beam area determination. Results of the simulation and measured using ion chamber was distributed in a relative error of 4.5 ~ 7.3% of the case of air kerma rate. The size of the effective beam area is when the simulation was implemented in the horizontal direction 27cm, 21.6cm vertical direction, the measured result using a film was obtained similar results with the horizontal direction 26.5cm, 21.9cm vertical direction. The relative error in the horizontal direction is 1.85% and 1.38% vertical effective beam area was also similarly distributed around the field gamma rays. As a result of the study, it was confirmed that the effectiveness of the simulation was sufficient for the gamma irradiation system. In particular, it is small relative errors in the effective beam size than the air kerma rate is considered to be due to the size of the beam is determined by geometric factors rather than the capacity of the standard source. A further study is needed to improve the reliability of the photon energy distribution diagram using simulation.

AN EXPERIMENTAL STUDY ON THE LOW-DOSE RADIOSENSITIVITY OF TUMOR CELL LINES (수종의 암세포주의 저선량 방사선감수성에 관한 실험적 연구)

  • The purpose of this study was to aid in the radiation therapy of head and neck cancer patients. For this study, radiation survival curves were generated for Bl6, MG-63 and YAC-l cell lines using semiautomated MTT assay and Dye Exclusion Assay. Irradiation of 2, 4, 6, 8, 10Gy were delivered at room temperature at a dose rate of 210.2cGy/min using / sup 60/Co γ-ray Irradiator ALOORAOO 8. The viable cells were determined for each radiation dose and compared to control values. The obtained results were as follows: 1. There was significantly different absorbance at 10Gy on B16 cell line in MTT assay(P<0.05). 2. There was significantly different absorbance at 4, 6, 8, 10Gy on MG-63 cell line in MTT assay(P<0.05). 3. YAC-l cell line was more sensitive than B16 or MG-63 cell line to all doses of radiation(P<0.05). 4. There was significantly different absorbance among all tumor cell lines except between B16 and MG-63 cell line at ZGy in MTT assay(P<0.05). 5. Good correlation was obtained between MTT assay and DEA(P<0.05). The efficient of correlation of B16, MG-63 and YAC-l cell line was 0.845, 0.824 and 0.906, respectively.

돈피 콜라겐 유래 올리고펩타이드 제조를 위한 방사선조사의 이용

  • 환경친화적 기술로 알려진 방사선조사기술(RT: Radiation Technology)을 이용하여 돈피 유래 올리고펩타이드를 제조하고자 하였다. 생 박 돈피를 hammer mill과 chopper를 이용하여 조분쇄한 후 $-20^{\circ}C$ 아세톤으로 탈지하였고, ${\gamma}$-ray irradiator를 이용하여 0, 20, 40, 60, 100, 150, 200, 250, 300 kGy의 총 흡수량을 얻도록 탈지돈피에 방사선조사를 실시하였다. 방사선조사에 의한 탈지돈피의 pH 변화는 0${\sim}$100 kGy 조사선량에서는 미비했으나, 150 kGy 이상에서는 소폭 증가하였다. 탈지돈피의 단백질 함량 중 콜라겐 함량은 93% 이었으며 방사선조사된 돈피콜라겐을 효소처리하면 효소반응 시간이 길어질수록 약 24 kDa 범위에서 밴드가 확인되었고, 100 kGy 이상의 고선량에서는 효소반응 2시간 이후 10% polyacrylamide 전기영동 겔의 최 하단에 머무는 분자량의 펩타이드가 다량 관찰되었다. 용해도 변화는 20${\sim}$60 kGy의 선량에서는 효소반응 시간이 길어질수록(1시간${\sim}$4시간) 최대 65${\sim}$80%의 용해도 증가를 보였고, 반면에 100 kGy 이상에서는 효소반응 시간에 관계없이 80% 이상, 300 kGy에서는 90% 이상의 용해도를 보여주려다. 점도와 탁도는 100 kGy 이상의 고선량에서 짧은 효소반응 시간(1시간)에 급격히 감소하였다. 가수 분해물(300 kGy)을 gel permeation chromatography한 결과 분자량 9,000 Da의 주 피크가 검출되었다.

Effects of low dose irradiation on the calcific nodule formation in MC3T3-E1 osteoblastic cell line (저선량 방사선이 MC3T3-E1 골모세포주의 석회화결절 형성에 미치는 영향)

  • Purpose: To investigate the effects of low dose irradiation on the calcium content and calcific nodule formation of the MC3T3-El osteoblastic cell line. Materials and Methods: Cells were irradiated with a single dose of 0.2, 0.4 and 0.6 Gy at a dose rate of 5.38 Gy/min using Cs-137 irradiator. After irradiation, the calcium content and calcific nodule formation were examined on the 1st, 2nd, 3rd and 4th week. Results: We did not find any significant difference of total calcium content after irradiation of 0.2, 0.4 and 0.6 Gy when compared with the unirradiated control group. There was no significant difference of total calcium content between 0.2, 0.4 and 0.6 Gy irradiated groups. We found an increased tendency of the calcific nodule formation after irradiation of 0.2, 0.4 and 0.6 Gy when compared with the unirradiated control group without significant difference of calcific nodule formation between 0.2, 0.4 and 0.6 Gy irradiated groups. Conclusion : The results showed an increased tendency of the calcific nodule formation after low dose irradiation. However, this tendency did not increase with the increase of irradiation dose.
