Production of Glucoamylase from Hybrid Constructed by Intergenic Nuclear Transfer between Saccharomycopsis sp. and Saccharomyces sp. (핵전이법에 의해 형성된 Saccharomycopsis 속과 Saccharomyces 속의 잡종에서 glucoamylase 생산에 관한 연구)

  • The glucoamylase was purified from the induced culture filtrate of hybrid between Saccharomycopsis sp. and Saccharomycopsis sp. made by nuclear transfer and characterized for some enzyme properties. The enzymewas purified 76-fold in an overall yield of 16% from the culture medium by ammonium sulfate fractionation,Sephadex G-150 gel permeation chromatography and DEAE-Sephadex A-50 ion exchage chromatography.The molecular weight of the purified glucoamylase was estimated to be 57.5 KDa on SDS-polyacrylamidegel electrophoresis and Sephadex G-150 gel permeation chromatography. The purified enzyme was active atpH-5.0 and $40^{\circ}C$. The Km value for soluble starch was 2.6 mg/ml. The enzymatic activity was stimulated inthe presence of TEX>$Ca^{2+}$, EDTA, $Co^{2+}$, $Mg^{2+}$, and $Mn^{2+}$

Biochemical Properties and Antibacterial Activity of Lactoferrin from Korean Native Cow (한우 Lactoferrin의 생화학적 특성 및 항균 활성)

  • The purpose of this study was to demonstrate biochemical properties and antibacterial activity of lactoferrin(Lf) obtained from the colostrum of Korean native cow. Lactoferrin was isolated from the colostrum of Korean native cow by purification steps using batch extraction, ion exchange chromatography, gel filtration chromatography, affinity chromatography. Other whey protein components that is similar molecular weight and affinity to lactoferrin were gradually removed from crude Korean Native cow's lactoferrin during the purification steps. The molecular weight of the purified Korean native cow's Lf(K-Lf) was 81 kDa, the isoelectric point was 9, and the content of iron was 0.56mg/g, which is indicated that iron saturation of the K-Lf was 40.6%. Amino acid composition and a-helix content were different K-Lf from bovine Lf(B-Lf). Antibacterial activity of E. coli O111 by K-Lf was lower than that of B-Lf. A minimal inhibitory concentration(MIC) of K-Lf and B-Lf was 2.75mg/ml and 1.5mg/ml respectively.

How to Design Membrane Chromatography for Bioseparations: A Short Review (바이오분야 분리용 막크로마토그래피 설계 방안)

  • While there are increasing demands on biomolecules separation, resin chromatography lacks in terms of throughput and membrane chromatography is an alternative with high binding capacity and enhanced mass transfer properties. Unlike typical membrane processing, where the performance can only be empirically assessed, understanding how mechanisms work in membrane chromatography is decisive to design biospecific processing. This short review covers three separation mechanisms, including affinity interaction modes for selectively capturing bulk molecules using biospecific sites, ion exchange modes for binding biomolecules using net charges and hydrophobic interaction modes for binding targeted, hydrophobic species. The parameters in designing membrane chromatography that should be considered operation-wise or material-wise, are also further detailed in this paper.

Determination of Trace Metals in Atmospheric Particulates by Ion Chromatography

  • A simple and fast ion chromatographic method is developed for the determination of transition metals such as Fe, Cu, Ni, Zn and Co in atmospheric particulates. The method involves acid digestion, on-column preconcentration, and subsequent ion chromatogaphic detection. The precision of the method is less than 3${\%}$ RSD at parts per billion level for the metals studied. No significant interferences are observed. The results obtained with this method agree well with those by ICP-AES.

  • Optimal operation in chromatography is needed to save operation time and the solvent used in multiple chromatographic runs. To this end, many simulation studies of chromatography process have been performed. The relationship between the distribution coefficient and the ionic strength is important in gradient elution ion chromatography. Experimental runs and computer simulations were carried out under linear gradient elution condition in order to compare the experiments and the simulation. Experiments were performed with formic acid under isocratic conditions to determine the simulation equation parameters. Computer simulation was based on three equations which related distribution with ionic strength as follows; K=${\alpha}$I(sup)-${\beta}$, K=A+BI+Cl$^2$and K=y(sub)0+A$_1$$.$e(sup)(-I/m$_1$). The effects of gradient slope on the chromatograms are discussed, and good agreement between the experimental and the simulated results is shown.

  • Bacillus thuringiensis var. thuringiensis produces an extracellular insecticidal thermostable .betha.-exotoxin, which was purified through microfiltering, barium precipitation, charcoal absorption chromatography, ion exchange column chromatography and gel filtration. The exotoxin in each purification step was detedted by thin layer chromatography, high pressure liquid chromatography and paper electrophoresis with efficient results. The exotoxin productivity on time course was checked by spectrophotometric absorbance at 258nm with the result that the exotoxin was initially produced in 6 hour culture and reached maximum value in 36 hour culture. Anti-bacterial effect test on Micrococcus flava was applied as toxicity test. The results showed that frowth inhibition of M. flava could be shown in plate assay of cell free filtered supernatant, alkaline eluant from charcoal and purified exotosin obtained from gel filtration column chromatography on Sephadex G-10 appeared to be 740. Heat stability of the exotoxin was confirmed through autoclaving twice.

Environmental and biological monitoring of workers exposed to methyl bromide through quarantine fumigation (방역작업자의 브롬화메틸(methyl bromide) 노출수준과 생물학적 모니터링)

  • This study was performed to estimate environmental and biological monitoring of workers exposed to methyl bromide through quarantine fumigation. Airborne methyl bromide and it's metabolites were analyzed by gas chromatography and ion chromatography, respectively. The results are as followings; airborne concentration of methyl bromide(TWA) was $2.08{\pm}1.56ppm$(N=8). Dispersion and setting/degas groups were $0.67{\pm}0.12ppm$(N=2) and $2.54{\pm}1.53ppm$(N=6), respectively. Bromide ion concentration in serum was $23.40{\pm}14.91mg/{\ell}$(N=10) in the exposed workers and $4.74{\pm}0.82mg/{\ell}$(N=21) in the non-exposed workers. Bromide ion concentration in urine was $35.56{\pm}26.89mg/{\ell}$(N=11) in exposed group and $6.62{\pm}2.31mg/{\ell}$(N=21) in non-exposed group. Good correlation was observed between concentration of serum and urine ($r^2$=0.890 p<0.01). No significant correlations of other determinants were observed. Calculated from a regressive curve, the biological half lifes of serum and urine were 10.7 and 5.9 days. In these results, biological monitoring of bromide ion of serum and urine provided useful information for evaluating exposure of workers to methyl bromide, so that an availability of bromide ion of biological samples was showed as biological monitoring indices for methyl bromide.

Research for the Analytical Method of Various Pesticides in Raw Milk by Gas Chromatography-Mass Spectrometry (GC/MSD를 사용한 원유 내 잔류농약의 분석법 연구)

  • The aim of this study was to optimize a simple, fast, and economical analysis procedure for the determination of 16 different pesticides in raw milk via GC/MSD. Analyses were performed via gas chromatography with electron impact mass spectrometric detection in the selected ion monitoring mode (GC/MSD-SIM) using Pentachloronitrobenzene as the internal standard. The modified sample preparation methodology was based on the Pesticide Analytical Manual (PAM) of the FDA concerning fat extraction, ACN-ether partitioning, and clean-up of the Sep-Pak florisil cartridge. The modified methodology for the determination of the 16 pesticides was validated. The range of LOQs of the 16 pesticides was likely three times lower than their Maximum Residence Levels (MRLs). The recoveries of most of the pesticides were acceptable at the fortification levels of 0.5 and 1.0 ${\mu}g/mL$ and their RSD (%) level was less than 20%. None of the 16 pesticides were detected in the selected raw milk samples.

  • Phenylalanine ammonia-lyase (PAL; EC 4.3.1.5), the first enzyme in the phenylpropanoid biosynthesis, catalyzes the elimination reaction of ammonium ion from L-phenylalanine. PAL was purified from the cytosolic fraction of Chinese cabbage (Brassica campestris ssp. napus var. pekinensis) through ammonium sulfate fractionation, DEAE-cellulose chromatography, Sephadex G-200 chromatography, and Q-Sepharose chromatography. It consists of four identical subunits, the molecular mass of which was estimated to be about 38,000 daltons on SDS-PAGE. The optimal pH and temperature of the purified enzyme are 8~9 and $45^{\circ}C$, respectively. Its activity is greatly inhibited by $Zn^{2+}$ ion, and strongly activated by caffeic acid. The purified PAL has some different characteristics compared to those obtained with other PALs.

  • Changes in esterase activity and zymogram pattern during development in Pieirs rapae L. were investigated and three esterases (E2, E6 and E11) from the late 5th instar larvae were purified. Esterase activity in whole body increased rapidly during 5th instar larval stages and reached a peak at the late 5th instar larval stage. The number and intensity of esterase band from whole body and midgut also showed a peak at the late 5th instar larval stage. Purification of esterase was performed using gel filtration on Sephadex G-lOO, ion-exchange chromatography on DEAE-trisacryl and preparative electrophoresis. The final purities of these enzymes were about 30 to 60-fold.
