Arabidopsis thaliana (L.) Heynh. is a small plant species that serves as a model organism of plant biology and genetics. Here, we present the first complete mitochondrial genome of Korean A. thaliana natural isolate (named as 180404IB4), which is 368,875 bp long and contains 58 genes (33 protein-coding genes, 22 tRNAs, and three rRNAs), with a GC ratio of 44.8%. Sixty-four single-nucleotide polymorphisms and 11 insertion and deletion regions (1,089 bp in length) are identified against the Col-0 ecotype, showing one large insertion of 1,069 bp without structural variation. Phylogenetic trees constructed from 30 conserved genes indicate that the 180404IB4 mitochondrial genome is clustered with Col-0 and three East Asian ecotypes.
We have determined the complete chloroplast genome of Erigeron Canadensis isolated in Korea. The circular chloroplast genome of E. canadensis is 152,767 bp long and has four subregions: 84,317 bp of large single-copy and 18,446 bp of small single-copy regions are separated by 25,004 bp of inverted repeat regions including 133 genes (88 protein-coding genes, eight rRNAs, and 37 tRNAs). The chloroplast genome isolated in Korea differs from the Chinese isolate by 103 single-nucleotide polymorphisms (SNPs) and 47 insertions and deletion (INDEL) regions, suggesting different invasion sources of E. canadensis in Korea and China. A nucleotide diversity analysis revealed that the trend of the nucleotide diversity of E. canadensis followed that of 11 Erigeron chloroplasts, except for three peaks. The phylogenetic tree showed that our E. canadensis chloroplast is clustered with E. canadensis reported from China. Erigeron canadensis can be a good target when attempting to understand genetic diversity of invasive species.
The intraspecific susceptibility of twelve Echinochloa colona(L.) Link ecotypes to herbicides was studied at three different stages of growth. At germination and post-germination stage, the Batangas ecotype was most susceptible to both butachlor (N-butoxymethyl-2-chloro-2', 6'-diethylacetanilide) and thiobencarb (S-4-dichlorobenzyldiethylthiocarbamate), whereas the Bukidnon ecotype was the most tolerant. The susceptibility of the ecotypes used was not associated with the seed weight and maturity. When the preemergence herbicides were applied at the 2-leaf stage of the ecotypes, the Nueva Ecija ecotype was least susceptible to butachlor at 1 ppm, but it was most susceptible to thiobencarb. The reverse was exhibited by the Cagayan ecotype. Differential susceptibility of the ecotypes to propanil (3', 4'-dichloropropionanilide) as determined by the necrotic length of the third leaf of 20-day-old seedlings was pronounced in the Camarines Sur, Iloilo, and IRRI (red) ecotypes. However, no necrotic tip appeared in the Cagayan and Batangas ecotypes. The differential responses varied with the type and the concentration of the herbicide studied and the growth stage of the ecotypes.
The genetic variation and intraspecific relationships between 10 individuals of seven cultivars and one Ulleungdo native of Wasabia japonica were investigated using RAPD (Randomly Amplified Polymorphic DNA) analysis. The 21 primers out of 50 random primers were amplified for all tested plants. The 68 (47.2%) among 144 bands derived from 21 primers showed polymorphism, and 3.2 bands per primer were observed. Number of bands per primer was ranged from 2 to 13, and average numbers were 6.8. The phenograms for 11 analyzed individuals by RAPD markers were not matched well with those of the result by morphological characters since they were clustered monophyletic at the similarity coefficient value ranged from 0.81 to 0.96. The Ulleungdo native individual was clustered sister to Daruma, Simanesairal, Sawa, and Hujidaruma cultivars. The RAPD markers were not useful to evaluate the intraspecific variations in Wasabia japonica cultivars, therefore need to more specific molecular phylogenetic characters such as AFLP technology and gene sequence of nuclear and chloroplast DNA.
Taxonomic studies on morphological, principal component analysis (PCA), palynological, RAPD and PCR-RELP analysis were conducted to intraspecific relationships of Rubus oldhami. Three types of Rubus oldhami based on the flower characters such as petal length and number were used in this study. Among the 14 morphological characters, perianth length, calyx lobe length, apical leaflet shape and leaflet length were used to distinguish for each type. The pollen characters such as shape, aperture number, surface sculpture were showed very similar among three types. Eight primers out of 20 arbitrary primers were screened for three types, and were revealed 33 ($60\%$) polymorphic bands. The phonogram by RAPD data showed incongruent with morphological analysis. Even though ten restriction endonucleases produced 20 restriction sites, polymorphic bands were not observed. Based on the results, three types of Rubus oldhami divided well by morphological characters, but pollen and DNA data were not supported. Therefore, type 1 and 2 which different from type 3 by flower characters considered as a temporary hybrid or ecotype because of their similar habitats.

A lot of clones of the genus Dioscorea have been introduced from some tropical and subtropical regions since 1997. In 33 clones of water yams (Dioscorea alata L.), some morphological characteristics were investigated at the field. Variation ranges of the total weight and tuber number per stump were within the ranges from 90 to 2,147 g with an average of 610 g ; and 1.3-4.7 with an average of 2.8, respectively. The color tones observed on the tuber-flesh were sorted into 3 color-categories, i.e., white, pale brown and pale purple, and those on leaves were sorted into 3 color-categories, i.e., green, heavy green and purplish green. Intraspecific genetic relationship of 19 variation types of the Yam classified by their external morphological characteristics such as leaf and tuber shape was assessed by DNA using random and specific primers. Twenty two out of 113 primers (100 random[10-mer] primers, two 15 mer [M13 core sequence, and (GGAT)4 sequence]) had been used in PCR-amplification. Only 12 primers, however, were successful in DNA amplification in all of the analyzed plants, resulting in 93 randomly and specifically amplified DNA fragments. The analyzed taxa showed very high polymorphisms(69 bands, 71.0%), allowing individual taxon to be identified based on DNA fingerprinting. Monomorphic bands among total amplified DNA bands of each primer was low under the 50%. Similarity indices between accessions were computed from PCR(polymerase chain reaction) data, and genetic relationships among intraspecific variations were closely related at the levels ranging from 0.66 to 0.90.
We performed RAPD analysis to investigate the inter-/intraspecific relationships and regional variations of Phragmites japonica and P. communis according to the environmental change. One hundred and fourty nine genetic effective polymorphic bands between 300 bp and 1,900 bp were marked from RAPD PCR with nine oligoprimers. From the RAPD analysis by Nei-Li's genetic distance, the dissimilarity indices among the populations of Phragmites japonica were relatively low from 0.012 to 0.061, and Phragmites communis were also low from 0.033 to 0.095. It showed the close genetic relationships among the same species populations, and both species were distinctly independent with relatively high level of dissimilarity indices (0.043 - 0.132). The obvious genetic markers to distinguish two species were confirmed and those profiles were suggested. From the UPGMA phenogram by RAPD analysis, both species showed the water environment related cluster patterns by distributional regions. RAPD analysis was useful to delimit two species taxonomically and to investigate the genetic relationships among inter-/intraspecific populations.

This study was conducted to provide the genetic diversity on Safflower collections and to identify the variations which could be utilized in Safflower breeding. The RAPDs analysis was used to clarify the genetic relationships among 32 Safflower collections. Among 37 primers applied in RAPD analysis, 25 primers that generated appropriate PCR products for identification of the genetic characters in safflower collections were used. Amplified PCR showed the highly reproducible bands at $0.1{\sim}4.0kb$. The number of bands amplified in each primer showed the variations ranging from 1 to 9, with the average of 5.6. A total of 25 bands were identified among twenty-five selected primers and 23 bands (19.2%) showed polymorphism. Based on the similarity value of 0.042 in dendrogram derived from the cluster analysis, the 32 Safflower collections were classified into 6 groups. The two main groups, II and III included 12 collections (38%) and 12 collections (38%), respectively.
Toxoplasma gondii is a eukaryotic parasite of the phylum Apicomplexa, which infects all warm-blood animals, including humans. In the present study, we examined sequence variation in dense granule 20 (GRA20) genes among T. gondii isolates collected from different hosts and geographical regions worldwide. The complete GRA20 genes were amplified from 16 T. gondii isolates using PCR, sequence were analyzed, and phylogenetic reconstruction was analyzed by maximum parsimony (MP) and maximum likelihood (ML) methods. The results showed that the complete GRA20 gene sequence was 1,586 bp in length among all the isolates used in this study, and the sequence variations in nucleotides were 0-7.9% among all strains. However, removing the type III strains (CTG, VEG), the sequence variations became very low, only 0-0.7%. These results indicated that the GRA20 sequence in type III was more divergence. Phylogenetic analysis of GRA20 sequences using MP and ML methods can differentiate 2 major clonal lineage types (type I and type III) into their respective clusters, indicating the GRA20 gene may represent a novel genetic marker for intraspecific phylogenetic analyses of T. gondii.

Background and objective: Native to the limestone zones of the Korean Peninsula, Zabelia tyaihyonii is a popular plant for landscaping. As it is now classified as a rare species, the conservation of its genetic resources is necessary. Methods: In this study, which aimed to understand the morphological variation of Z. tyaihyonii, 18 characteristics of Z. tyaihyonii from five habitats were examined. Results: Of these 18 characteristics, 16 characteristics showed significant differences among sites, and the coefficient of variation ranged from 5.4% (for corolla lobe number) to 31.3% (for flower number). Notable variations were observed in the size of flower and calyx lobe. When the corolla length and calyx lobe length were used as the classification key of Z. tyaihyonii, the sites were divided into those with small, intermediate, and large values. Hair was observed on the filament of all samples, a finding which conflicts with an earlier report. Rather than classifying Z. tyaihyonii into different species on the basis of corolla length (COL) and calyx lobe length (CALL) values, we recommend modifying the species description to incorporate the variation in these characteristics of interest. Principal component analysis results showed that the first main component was highly correlated with the traits related to the size of the calyx lobe (length: 0.819, width: 0.758), and the second main component was highly correlated with the traits related with the size of the inflorescence (length: 0.790, width: 0.626). Conclusion: Several notable variations were identified among the characteristics related to inflorescence and calyx lobe. There is little genetic exchange among groups, or each group is influenced by micro environmental factors, because sites that are located nearby. In addition, the difference between COL and CALL, which is used as the classification key for Z. tyaihyonii, was divided into small group, large group, and intermediate group, regardless of the sites' geographical distance.
