Discovery of Eurytrema Eggs in Sediment from a Colonial Period Latrine in Taiwan

  • In this study we take a closer look at the diseases that afflicted Japanese police officers who were stationed in a remote mountainous region of Taiwan from 1921 to 1944. Samples were taken from the latrine at the Huabanuo police outpost, and analyzed for the eggs of intestinal parasites, using microscopy and ELISA. The eggs of Eurytrema sp., (possibly E. pancreaticum), whipworm and roundworm were shown to be present. True infection with Eurytrema would indicate that the policemen ate uncooked grasshoppers and crickets infected with the parasite. However, false parasitism might also occur if the policemen ate the uncooked intestines of infected cattle, and the Eurytrema eggs passed through the human intestines. These findings provide an insight into the diet and health of the Japanese colonists in Taiwan nearly a century ago.

Effect of Dietary Fat Sources and Levels on Plasma and Tissue Cholesterol (식이내 지방의 종류와 수준이 혈장 콜레스테롤 및 조직내 콜레스테롤 함량에 미치는 영향)

  • The present studies were designed to compare the effects of dietary fat sources and levels on plasma and tissue cholesterol. Changes in plasma total cholesterol and HDL-cholesterol, cholesterol concentration in aortas, liver and intestines were determined in young rats fed diets providing dietary fat as corn-oil or butter and levels as 10%, 20% or 30% of calories respectively 1) Plasma total cholesterol and HDL-cholesterol levels were little sensitive to the modification of dietary fat sources and levels. 2) Dietary cholesterol increased the levels of plasma total cholesterol and this effect was accentuated by feeding butter. But dietary cholesterol did not increase the levels of HDL-cholesterol in the butter-fed groups, but decreased in the corn-oil-fed groups. 3) Liver cholesterol concentrations were significantly higher in rats fed diets of corn-oil than those fed diets of butter. On the contrary, cholesterol concentrations of intestines were significantly higher in the butter-fed groups than the corn-oil-fed groups. However, in aortas, no significant differences were found.

Isolation of Bacteriocin-producing Lactic Acid Bacteria from Human Intestines and the Characteristics of their Bacteriocins (Bacteriocin을 생산하는 장내 유산균의 분리 및 Bacteriocin 특성조사)

  • Lactobacillus strains were isolated from volunteer's feces, including from newly-born infants and adults in their 20's, by using differential MRS-BPB plates. Total 56 presumptive Lactobacillus strains were isolated and the bacteriocin productions by the isolates were examined by agar diffusion method. Six bacteriocin-producing strains were confirmed. Among them, two isolates, HU-1 and H22-3, showed the most outstanding antimicrobial activities, which were not affected by pH adjustments or catalase treatments of culture. HU-1 was originated from a two-years old boy and H22-3 was originated from a newly-born infant. HU-1 and H22-3 had the same morphology(short rod) when examined by scanning electron microscope, and the same biochemical traits including growth temperature range, salt tolerance and sugar-fermenting abilities. But the growth-inhibition spectrum and plasmid profiles of HU-1 and H22-3 were different. Both strains inhibited the growth of various Gram (+) microorganisms including Listeria monocytogenes. Micrococcus luteus, and Staphylococcus aureus in addition to many species of lactic acid bacteria, indicating the production of broad-spectrum bacteriocins. Bacteriocins produced by HU-1 and H22-3 were stable up to 90℃, 15 min heat treatments. Their activities were not affected by pepsin or trypsin treatments but destroyed by proteinaseK or pronase treatments.

Characterization of Lactobacillus plantarum strains isolated from black raspberry and their effect on BALB/c mice gut microbiota

  • The objective of this study was to evaluate probiotic effects of two Lactobacillus plantarum strains (GBL16 and 17) isolated from black raspberry. Results revealed that the number of GBL16 was gradually decreased as bile salt concentration was increased from 0.3 to 1%. However, GBL17 did not show any difference when GBL17 was applied to 1% bile salt, and it indicates that GBL17 is more tolerant to bile salt than GBL16. GBL17 exhibited higher heat resistance and adhesion ability to Caco-2 cells than GBL16. Regarding gut microbiome, no significant change in the number of total bacteria in intestines of mice after treatment with GBLs was determined. However, the combination of GBL16 and GBL17 significantly increased the number of total bacteria in intestines of mice after they were orally administered. Therefore, the results suggest that both GBL16 and 17 strains could be one of major probiotics that can improve human gut health.

Paralytic Ileus Secondary to Electrolyte Imbalance: A Case Study in a 16 Year Old Female

  • Paralytic ileus is a metabolic state in which the intestines fail to transmit peristalsis due to failure of the neuromuscular mechanism in the small intestines and colon. It is a major cause of morbidity in hospitalized patients especially during late presentations and points of mismanagement. The causes include infections, electrolyte imbalance (hypokalemia, hyponatremia), surgeries and medications. When the exact cause of the disease condition is identified and corrected, paralytic ileus is usually resolved. This case report is that of a 16 year old female who was admitted and managed as a case of paralytic ileus. The patient presented with symptoms of fever, abdominal pain, abdominal distension, vomiting and inability to pass stool or flatus. There was associated body weakness, reduced urine output and weight loss. She was properly examined clinically and sent for various investigations. Investigations such plain abdominal X-Ray, serum electrolyte estimation, chest X-Ray and full blood count were carried out. The results of the investigations done were in keeping with the diagnosis of paralytic ileus, electrolyte imbalance and ongoing sepsis. She was subsequently managed through nil per oral, adequate fluid rehydration, antibiotics and correction of electrolyte imbalance. Following stable clinical state and investigation results, she was discharged and advised on follow-up.

Serotonin and Energy Metabolism (세로토닌과 에너지 대사)

  • Serotonin, a biogenic amine widely found in many organisms, functions as both a neurotransmitter and hormone. Although serotonin is involved in various physiological processes, this study aimed to review its role in energy metabolism. Given that serotonin cannot cross the blood-brain barrier and is synthesized by two different isoforms of tryptophan hydroxylase in the central nervous system (CNS) and peripheral tissues, it is reasonable to assume that serotonin in the CNS and peripheral tissues functions independently. Recent studies have demonstrated how serotonin influences energy metabolism in metabolic target organs such as the intestines, liver, pancreas, and adipose tissue. In summary, serotonin in the CNS induces satiety and appetite suppression, stimulates thermogenesis, and reduces body weight. Conversely, serotonin in the periphery increases intestinal motility, stimulates gluconeogenesis in the liver, suppresses glucose uptake by hepatocytes, promotes fat uptake by liver cells, stimulates insulin secretion while suppressing glucagon secretion in the pancreatic islets, promotes lipogenesis in white adipose tissue, inhibits lipolysis and browning of white adipose tissue, and suppresses thermogenesis in brown adipose tissue, thereby storing energy and increasing body weight. However, considering that most experimental results were obtained using mice and conducted under specific nutritional conditions, such as high-fat diets, whether serotonin acts in the same way in humans, whether it will act similarly in individuals with normal versus obese weights, and whether its effects vary depending on the type of food consumed, remain unknown.

Histochemical Study of the Intestinal Mucosubstances in Sebastes schlegeli, Halichoeres poecilopterus, Bryzoichthys lysimus and Takifugu pardalis (조피볼락, 용치놀래기, 송곳니베도라치 및 졸복 장관 점액질에 대한 조직화학적 연구)

  • The conventional histochemical staining were used to study mucosubstances properties of intestinal striated border and goblet cells in four teleostean species, i. e., Sebastes schlegeli, Halichoeres poecilopterus, Bryzoichthys lysimus, and Takifugu pardalis. The following methods were used; PAS, AB pH 2.5, AB pH 1.0, AB pH 2.5-PAS, AF pH 1.7-AB pH 2.5 and HID-AB pH 2.5 stains. The mucosubstances of striated border in the proximal intestine and rectum of Sebastes schlegeli contained with neutral mucin, middle and distal intestine contained with neutral mucin and acid mucin. The striated border of all the intestines of Halichoeres poecilopterus contained with neutral mucin and acid mucin, and those of Bryzoichthys lysimus and Tnkifugu pardalis contained with neutral mucin only. The amounts of neutral mucin were moderate to considerable in Sebastes schlegeli and Halichoeres poecilopterus, minimal to small in Bryzoichthys lysimus and Tnkifugu pardalis. The amounts and properties in mucosubstances of intestinal goblet cells showed differences in species and regions. The intestinal goblet cells of Bryzoichthys lysimus, and Tnkifugu pardalis contained neutral mucin only while Sehastes schlegeli and Halichoeres poecilopterus contained mixture of neutral mucin, sulfomucin and sialomucin. The amounts of neutral mucin were considerable to large in distal intestine and rectum of Tnkifugu pardalis, while moderate to considerable in all intestines of Sehastes schlegeli, all the intestines except for middle intestine of Bryzoichthys lysimus, and proximal and middle intestine of Tnkifugu pardalis. Also it was minimal to small in middle intestine of Halichoeres poecilopterus. The intestinal goblet cells of Sehastes schlegeli contained mixture of minimal amounts of strong sulfmucin, weak sulfomucin and minimal to small amounts of sialomucin, and those of Halichoeres poecilopterus except for rectum contained mixture of minimal to small amounts of strong sulfomucin and sialomucin.

Effects of the Gamihyangsayukgunjatang on Radioprotection and Apoptosis in Small Intestines of Mice (가미향사육군자탕(加味香砂六君子湯)이 생쥐 소장(小腸)에서 방사선조사(放射線照射) 후(後) 보호효과(保護效果)와 Apoptosis에 미치는 영향(影響))

  • The present study was performed in order to evaluate the effects of the gamihyangsayukgunjatang on radioprotection and apoptosis in small intestines of mice after whole body irradiation. Two hundred forty mice were divided into 40 groups according to the radiation dose and the gamihyangsayukgunjatang treatment. The extracts of the herbal medicines were orally administered to each group differently before and/or after irradiation. The gamihyangsayukgunjatang treated groups were divided into 3 groups. Sample Ⅰ was the group treated with the gamihyangsayukgunjatang for 3 days before the radiation, sample Ⅱ was the group treated with the gamihyangsayukgunjatang for 3 days after the radiation. Sample Ⅲ was the group treated with the gamihyangsayukgunjatang for both 3 days before and after the radiation. To analyze the crypt survival, the micradony survival assay was used according to the Withers and Elind's method. To analyze the apptosis, the TUNEL assay was done. The results obtained are a follows : 1. From the microcolony survival assay, the gamihyangsayukgunjatang treated groups showed the radioprotective effect with a statistical significance(p<0.05), as compared to the control group. Comparing the radioprotective effect among the 3 groups, sample III was statistically more significant than sample I and II (p<0.05). Sample I showed no effect. In accordance with the research mentioned above, it is suggested that the radioprotective effect of the gamihyangsayukgunjatang is more useful for the treatment of the radiation injury rather that the prevention. 2. The results of the TUNEL assay showed that the apoptotic index in the gamihyangsayukgunjatang treated group was slightly decreased with no effectiveness, as compared to the control group. According to the above results, it could be suggested that the gamihyangsayukgujatang has a prominent protective effect in mice intestines against the radiation damage. However, the radioprotective effect does not seem to be related to inhibition of the apoptosis.

Investigation of Microbial Communities in Sulculus diversicolor supertexta Through 16S rRNA Sequencing and Antibacterial Monitoring of Harmful Strains (16S rRNA 염기서열 분석을 통한 오분자기(Sulculus diversicolor supertexta)내 미생물 군집 조사 및 인체유해 질병세균에 대한 항균활성 모니터링)

  • This study investigated the muscles, intestines, and gonads of Sulculus diversicolor supertexta to examine the diversity of microbial communities within examples collected from the Jeju Coast. Using different media, initial pure isolation in MA, 1% BHIA, and 1% TSA indicated that the muscles, intestines, and gonads supported more communities, respectively. In analysis of relative similarity with 16s rRNA sequencing, 190 pure colonies were isolated, and further analysis with NBLAST identified 71 species, 39 genera, 25 families, and five phyla. Homogeny with the reference strain was 91-100%. Microbial communities in S. supertexta consisted of gamma and alpha Proteobacteria (48%), Actinobacteria (32.5%), Firmicutes (16.9%), Deinococcus-Thermus (1.3%), and Bacteroides (1.3%). In all tissue, Psychrobacter cibarius in Moraxellaceae was dominant. Alteromonadaceae, Enterobacteriaceae, Pasturellaceae, Moraxellaceae, Rhodobacteraceae, Geminicoccaceae, Dietziaceae, Intrasporangiaceae, Microbacteriaceae, Micrococcaceae, Micromonosporaceae, Streptomycetaceae, Aerococcaceae, Bacillaceae, Paenibacillaceae, Planococcaceae, and Staphylcoccaceae were commonly isolated across all tissues, and Flavobacteriaceae, Corynebacteriaceae, Yesiniaceae, Vibrionaceae, Hahellaceae, Pseudomonadaceae were also identified from the intestines. In microbial monitoring of four harmful bacteria, Streptomyces albus (96%) showed antibacterial activity against all four strains, and Agrococcus baldri (99%) and Psychrobacter nivimaris (99%) presented against E. Coli and E. aerogens. In addition, some strains with low homogeny were isolated and further experiments are therefore required, for example to refine the antimicrobial substances including new strain investigations. These additional experiments would aim to establish generic resources for the microbial communities in S. Supertexta and provide basic data for applied microbiological research.

Paralytic shellfish toxins (PSTs) and tetrodotoxin (TTX) of Korean pufferfish

  • Paralytic shellfish toxins (PSTs) and tetrodotoxin (TTX) are neurotoxins that display pharmacological activity that is similar to that of specific sodium channel blockers; they are the principle toxins involved in shellfish and puffer fish poisoning. In Korea, puffer fish is a very popular seafood, and several cases of accidental poisoning by TTX have been reported. Therefore, it is necessary to determine whether puffer fish poisoning incidents are caused by PSTs or by TTX. In this study, we used mouse bioassay (MBA) and liquid chromatograph-tandem mass spectrometry (LC-MS/MS) to determine the presence of PSTs and TTX in puffer fish from an area near Mireuk-do, Tong-Yeong on the southern coast of Korea from January through March, 2014. The toxicity of PSTs and TTX extracts prepared from three organs of each specimen was analyzed by MBA. Most of the extracts killed mice with typical signs of TTX and PSTs. The LC-MS/MS analysis of seven specimens of Takifugu pardalis and Takifugu niphobles, each divided into muscles, intestines, and liver, were examined for TTX. In T. pardalis, the TTX levels were within the range of 1.3-1.6 ㎍/g in the muscles, 18.8-49.8 ㎍/g in the intestines, and 23.3-96.8 ㎍/g in the liver. In T. niphobles, the TTX levels were within the range of 2.0-4.5 ㎍/g in the muscles, 23.9-71.5 ㎍/g in the intestines, and 28.1-114.8 ㎍/g in the liver. Additionally, the toxicity profile of the detected PSTs revealed that dcGTX3 was the major component in T. pardalis and T. niphobles. When PSTs were calculated as saxitoxin equivalents the levels were all less than 0.5 ㎍/g, which is below the permitted maximum standard of 0.8 ㎍/g. These findings indicate that the toxicity of T. pardalis and T. niphobles from the southern coast of Korea is due mainly to TTX and that PSTs do not exert an effect.
