한우 PPARγ 유전자의 동정과 mRNA의 발현 (Molecular Cloning and mRNA Expression of the Bovine Peroxisome Proliperator Receptor Gamma(PPARγ))

  • 지방세포분화 과정에서 중추적인 역할을 한다고 보고되어지고 있는 PPAR$\gamma$ 유전자를 cloning 하기 위하여 한우 12개월 지방조직을 이용하여 Total RNA를 추출하고 RT-PCR을 수행하여 1515bp, 505 아미노산 서열을 가진 PPAR$\gamma$ 유전자를 cloning 하였다. bovine을 제외한 4종의 다른 동물과의 아미노산 서열을 비교한 결과 모두 90% 이상의 상동성을 나타내었다. 특히 NCBl에 보고된 Norwegian cattle PPAR$\gamma$ (NCBI Accession No. O18971)의 아미노산 서열과 비교한 결과 99.2%의 상동성을 나타내었으며 아미노산 서열중 81번째 이소루신, 140번째 아르기닌, 157번째 글루탐산, 486번째 리신이 각각 트레오닌, 트립토판, 글리신, 이소루신으로 치환된 상태의 아미노산 서열을 나타내었다. 특히 nuclear hormone receptor의 DNA binding domain 영역에 포함 되는 140번째, 157번째 아미노산의 치환은 앞으로 한우PPAR$\gamma$ 유전자가 조절하는 다른 유전자와의 관계를 밝히는데 매우 중요한 점이 될 것으로 사료되어진다. 한우 PPAR$\gamma$ 의 발현은 지방조직에서 매우 높은 발현을 보였으며, 한우 12개월령과 30개월령 지방조직에서의 PPAR$\gamma$ 발현을 비교 분석하였을 때 12개월령에서보다 30개월령 지방조직에서 PPAR$\gamma$ 발현이 약 6배정도 높았다. 이와 같은 결과는 한우 PPAR$\gamma$ 유전자도 사람과 생쥐에서와 같이 지방분화에 관여하고 있다는 보고와 일치하는 결과라고 사료된다.

한국의 장흡충에 관한 연구 IX. 실험동물내에서의 Fibricola Seoulens양 충체위수률 및 발육 (Studies on Intestinal Trematodes in Korea IX. Recovery Rate and development of Firbricola seoulensis in Experimental Animals)

  • 몇가지 실험동물의 Firbricola seoulensis 실험감염에 대한 감수성을 알아보고 숙주로서의 적합도를 관찰하고자 중간숙주로 알려진 뱀으로부터 피예유충을 분리하여 50∼2,000개군으로 나누고 횐쥐, 마우스, 개, 고양이, 토끼 및 닭등 총 127마리에 각각 실험감염시킨 다음 3일부터 8주까지 각 숙주내에서의 충체회수률 및 충체성숙도를 관찰하고 비교분석 하였다. 그 결과는 다음과 같다. 1. 총 충체회수률은 횐쥐에서 가장 높아 40.0%이었으며, 그 다음이 마우스로 33.9%이었고, 고양이 20.9%, 개 11.4%, 토끼 0.05% 및 닭 O%의 순으로 나타났다. 충체회수율은 감염기계이 길어짐에 따라 차차 저하되는 경향을 보였으며 감염피낭유충수에 따라서도 다양하게 변화하는 것으로 나타났다. 2. 횐쥐에는 피예유충 1,000개를, 마우스에는 200개를 감염시켰을때 충체회수률이 가장 높았으며 이 정도의 파낭유충수가 이들 동물을 감염시키는데 적당한 감염량으로 생각되었다. 3. 각 실험동물에서 주 기생부위는 소장이었고 고 중에서도 특히 십이지장이었다. 4. 각 동물에서 회수한 충체의 성숙도(성숙충체수/관찰충체수)는 횐쥐와 마우스에서 모두 100%이었고 개에서는 22.7%이었으며 고양이에서는 0%로 전혀 성숙된 충체가 없었다. 이상의 결과를 종합해볼 때 6종의 실험동물중 충체회수율 및 총체성숙도가 높은 횐쥐와 마우스가 Fibricola seoulensis의 가장 좋은 호적숙주로 생각되었다.

The importance of NIR spectroscopy in the estimation of nutritional quality of grains for ruminants

  • The production of grain for export and domestic use is one of Australia's most important agricultural industries, and the NIR technique has been used extensively over many years for the routine monitoring of grain quality, particularly moisture and protein content. Because most Australian grain is intended for human food production, the determinants of grain quality for livestock feed, apart from protein, have been largely ignored. However the increasing use of grain for feeding to pigs, poultry, beef cattle and dairy cows has led to an important national research project entitled “Premium Grains for Livestock”. Two of the objectives of this project are to determine the compositional and functional characteristics of grains which influence their nutritional quality for the various classes of livestock, and to adopt rapid and objective analytical tests for these quality criteria. NIR has been used in this project firstly to identify a set of grain samples from a large population of breeders' lines which showed a wide spectral variation, and hence a potentially wide variation in nutritional value. The selected samples were not only subjected to an extensive array of chemical, physical and in vitro analyses, but also were grown out to produce sufficient quantities of grain to feed to animals in vivo studies. Additional grains were also strategically selected from farms in order to include the effect of weather damage, such as rain, drought and frost. In this study to date, NIR calibrations have been derived or attempted, on both ground and whole grains, for in vivo dry matter digestibility (DMD), pepsin-cellulase dry matter disappearance, protein, fat, acid detergent fibre, neutral detergent fibre, starch, in sacco DMD and in vitro assays to simulate starch digestion in the lumen and small intestine. Results so far indicate high calibration accuracy for chemical components (SECV 0.3 to 2.6%) and very promising statistics for in vivo DMD (SECV 1.8, $R^2$ 0.93, SD 7.0, range 61.9 to 92.3, n=60). There appears to be some potential for NIR to estimate some in vitro properties, depending upon the accuracy of reference methods and appropriate sample populations. Current work is in progress to extend the range of grains with in vivo DMD values (a very laborious and expensive process) and to increase the robustness of the various NIR calibrations, with the aim of implementing uniform testing procedures for nutritional value of grains throughout Australia.

The Promotive Effects of Antioxidative Apigenin on the Bioavailability of Paclitaxel for Oral Delivery in Rats

  • This study was to investigate the effect of apigenin on the bioavailability of paclitaxel after oral and intravenous administration in rats. The effect of apigenin on P-glycoprotein (P-gp), cytochrome P450 (CYP)3A4 activity was evaluated. The pharmacokinetic parameters of paclitaxel were determined in rats after oral (40 mg/kg) or intravenous (5 mg/kg) administration of paclitaxel with apigenin (0.4, 2 and 8 mg/kg) to rats. Apigenin inhibited CYP3A4 activity with 50% inhibition concentration ($IC_{50}$) of 1.8 ${\mu}M$. In addition, apigenin significantly inhibited P-gp activity. Compared to the control group, apigenin significantly increased the area under the plasma concentration-time curve (AUC, p<0.05 by 2 mg/kg, 59.0% higher; p<0.01 by 8 mg/kg, 87% higher) of oral paclitaxel. Apigenin also significantly (p<0.05 by 2 mg/kg, 37.2% higher; p<0.01 by 8 mg/kg, 59.3% higher) increased the peak plasma concentration ($C_{max}$) of oral paclitaxel. Apigenin significantly increased the terminal half-life ($t_{1/2}$, p<0.05 by 8 mg/kg, 34.5%) of oral paclitaxel. Consequently, the absolute bioavailability (A.B.) of paclitaxel was significantly (p<0.05 by 2 mg/kg, p<0.01 by 8 mg/kg) increased by apigenin compared to that in the control group, and the relative bioavailability (R.B.) of oral paclitaxel was increased by 1.14- to 1.87-fold. The pharmacokinetics of intravenous paclitaxel were not affected by the concurrent use of apigenin in contrast to the oral administration of paclitaxel. Accordingly, the enhanced oral bioavailability by apigenin may be mainly due to increased intestinal absorption caused via P-gp inhibition by apigenin rather than to reduced renal and hepatic elimination of paclitaxel. The increase in the oral bioavailability might be mainly attributed to enhanced absorption in the gastrointestinal tract via the inhibition of P-gp and reduced first-pass metabolism of paclitaxel via the inhibition of the CYP3A subfamily in the small intestine and/or in the liver by apigenin. It appears that the development of oral paclitaxel preparations as a combination therapy is possible, which will be more convenient than the i.v. dosage form.

한국의 간흡충에 관한 연구 X. Fibricola seoulensis 표피의 전자현미경적 관찰 (Studios on Intestinal Trematodes in Korea X. Scanning Electron Microscopic Observation on the Tegument of Fibricola seoulensis)

  • 유혈목이에서 얻은 피낭유충을 생쥐(mouse)에 실험 감연시킨후 소장에서 수집한 Fibricole seoulensis 성충의 표피 메세구조를 주사전자현미경을 사용하여 관찰하였다. 1. 충체전반부는 자갈모양 원형질돌기(cobblestone-like cytoplasmic processes)로, 충체후반부는 손가락모양 원형질돌기(finger-like cytoplasmic processes)로 덮혀 있었다. 2. 전단 배면에는 상모양가시(spade shape spine)가 60∼70개 있었으며, 충체전반부에는 끝이 3∼4분지된 가시가 분포하고 있었는데, 특히 배면 전방 중앙부분에 밀집되어 있었다. 3. 구흠반 후면과 닥흡반에는 끝이 2∼3분지된 가시가 배열되어 있었다. Tribocytic organ 외면은 고사상으로 주름져 있었으며 stout recurved spine으로 무장되어 있었다. 충체후반부의 앞 1/3까지 끝이 분지되지 않은 가시가 발견되 었다. 4. 제 I형 감각유두(ciliated knob-like papilla)는 충체전반부 배면과 복면에 거의 과우대칭적으로 분포하였으며, 특히 구흡반, 복홈반, tribocytic organ 주위와 전반부의 외연에 밀집되어 있었다. 제 II형 감각유두(non-ciliated round swellings)는 구흡반과 복흡반의 외연(lip)에 각각 24개씩 배열되어 있었다. 제 III형 감각유두(plate-like elevation)는 충체후반부 전면에 걸쳐 분포하였다. 이 3가지 감각유두는 그 구조와 분포로 보아 tangoreceptor 혹은 rheoreceptor로 생각되었다. 비만 억제를 위한 췌장 리파아제 도메인에 대한 특이 난황항체의 개발 (Development of Egg Yolk Antibody Specific to the Pancreatic Lipase Domain for Anti-Obesity)

  • 사람 췌장 리파아제는 췌장에서 합성되어 소장으로 분비된 후, 음식물속의 지방을 가수분해하는 소화효소이다. 췌장 리파아제 단백질은 촉매활성도메인과 코리파아제 결합도메인의 2개의 도메인으로 구성되어 있다 본 연구에서는 리파아제 전체단백질과 촉매활성도메인 및 코리파아제 결함도메인 유전자를 PCR 방법으로 증폭하여 발현벡터에 넣은 후, 대장균에서 발현시켜 얻은 3가지 재조합 단백질을 산란계에 3차례 주사하여 면역반응을 유도하였다. 난황단백질을 분리하여 항체가를 측정한 결과, 코리파아제 결합도메인의 항원성이 가장 높은 것으로 밝혀졌다. 또한, 돼지 췌장 리파아제의 활성저해 실험에서도 코리파아제 결합도메인에 의해 만들어진 난황항체가 지방분해활성 저해작용이 가장 큰 것으로 밝혀졌다. 코리파아제 결합도메인 난황항체가 첨가된 지방을 실험동물에게 급여하고 혈중 주요 성분을 분석한 결과, 코리파아제 결합도메인에 의한 난황항체를 섭취한 경우에 혈중 중성지방의 수치가 대조군에 비해서 유의적으로 감소했음을 화인하였다. 결론적으로 코리파아제 결합도메인을 항원단백질로 사용하여 얻은 난황항체가 생체 내에서 췌장 리파아제의 활성을 억제할 수 있으며 비만억제제로의 개발 가능성이 있음을 보여주었다.

간외 담도암 고선량률 관내근접방사선치료 시 몬테카를로 시뮬레이션을 통한 주변장기의 선량평가 연구 (Study of Radiation dose Evaluation using Monte Carlo Simulation while Treating Extrahepatic Bile Duct Cancer with High Dose Rate Intraluminal Brachytherapy)

  • MCNPX를 통하여 계산한 상대선량과 고체팬텀과 전리함을 이용하여 측정한 상대선량을 비교하여 몬테카를로 시뮬레이션의 정확성을 평가하였다. 그리고 간외 담도암 관내근접방사선치료를 몬테카를로 시뮬레이션에 적용하기 위해 192Ir 밀봉방사성선원을 모사하였고, 한국 성인남성 표준인을 기초로 하는 KMIRD형 팬텀을 이용하여 담도 및 주변 장기를 제작하였다. 간외 담도암 관내근접방사선치료를 MCNPX를 이용하여 담도 주변 정상장기의 비유효에너지와 초기방사능을 1 Ci로 설정하여 흡수선량을 산정하였다. 몬테카를로 시뮬레이션의 정확성 평가에서 상대선량 차이가 가장 많은 지점이 1.96%로 MCNPX에서 제시한 상대오차 2%를 만족하는 것으로 나타났다. 또한, 담도 주변 정상장기의 비유효에너지 및 흡수선량은 담도와비교적 인접한 위치에 있는 우측신장, 간, 췌장, 횡행결장, 척수, 위장, 소장이 높았고, 담도와의 거리가 떨어져 있는 장기들인 좌측신장, 비장, 상행결장, 하행결장, S상결장이 낮게 나타났다.

Effects of Dietary Supplementation with the Combination of Zeolite and Attapulgite on Growth Performance, Nutrient Digestibility, Secretion of Digestive Enzymes and Intestinal Health in Broiler Chickens

  • This study was designed to investigate the effects of basal diets supplemented with a clay product consisting of zeolite and attapulgite (ZA) at 1:1 ratio on growth performance, digestibility of feed nutrients, activities of digestive enzymes in small intestine and intestinal health in broiler chickens. In experiment 1, 112 one-day-old male chickens were randomly divided into 2 groups with 8 replicates of 7 chickens each. In experiment 2, 84 one-day-old male chickens were randomly allocated into 2 groups consisting 6 replicates of 7 chickens each. The experimental diets both consisted of a maize-soybean basal control diet supplemented with 0% or 2% ZA. The diets were fed from 1 to 42 days of age. The results showed that ZA supplementation could increase body weight gain (BWG) and feed intake (FI), but had no significant effect on feed conversion ratio. The apparent digestibility values of crude protein and gross energy were significantly increased (p<0.05) by ZA from 14 to 16 d and 35 to 37 d. Dietary ZA treatment significantly increased (p<0.05) the activities of amylase, lipase and trypsin in jejunal digesta and the activities of maltase and sucrase in jejunal mucosa on days 21 and 42. The ZA supplementation also significantly increased (p<0.05) the catalase activity, reduced (p<0.05) the malondialdehyde concentration in the jejunal mucosa. In addition, a decrease of serum diamine oxidase activity and an increase (p<0.05) in concentration of secretory immunoglobulin A in jejunal mucosa were observed in birds treated with ZA on 21 and 42 days. It is concluded that ZA supplementation (2%) could partially improve the growth performance by increasing BWG and FI. This improvement was achieved through increasing the secretion of digestive enzymes, enhancing the digestibilites of nutrients, promoting intestinal health of broiler chickens.

Apparent and standardized ileal nutrient digestibility of broiler diets containing varying levels of raw full-fat soybean and microbial protease

  • Background: Although soybean meal (SBM) is excellent source of protein in diets for poultry, it is sometimes inaccessible, costly and fluctuates in supply. The SBM can partially be replaced by full-fat SBM, but the meals prepared from raw full-fat soybean contain antinutritional factors. To avoid the risk of antinutritional factors, heat treatment is always advisable, but either excessive or under heating the soybean could negatively affect the quality. However, the potential for further improvement of SBM by supplementing with microbial enzymes has been suggested by many researchers. The objective of this study was to evaluate the performance and ileal nutrient digestibility of birds fed on diets containing raw soybeans and supplemented with microbial protease. Methods: A $3{\times}2$ factorial, involving 3 levels of raw full-fat soybean (RFFS; 0, 45 or 75 g/kg of diet) and 2 levels of protease (0 or 15,000 PROT/kg) was used. The birds were raised in a climate-controlled room. A nitrogen-free diet was also offered to a reference group from day 19 to 24 to determine protein and amino acid flow at the terminal ileum and calculate the standardized ileal digestibility of nutrients. On days 10, 24 and 35, body weight and feed leftover were recorded to calculate the body weight gain (BWG), feed intake (FI) and feed conversion ratio (FCR). On day 24, samples of ileal digesta were collected at least from two birds per replicate. Results: When RFFS was increased from 0 to 75 g/kg of diet, the content of trypsin inhibitors was increased from 1747 to 10,193 trypsin inhibitors unit (TIU)/g of diets, and feed consumption of birds was also reduced (P < 0.05). Increasing RFFS level reduced the BWG from hatch 0 to 10 d (P < 0.01) and hatch to 24 d (P < 0.05). The BWG of birds from hatch to 35 was not significantly (P = 0.07) affected. Feed intake was also reduced (P < 0.05) during 0 to 35 d. However, protease supplementation improved (P < 0.05) the BWG and FCR during 0 to 24 d. Rising levels of RFFS increased the weight of pancreas (P < 0.001) and small intestine (P < 0.001) at day 24. Except for methionine, apparent and the corresponding standardized ileal digestibility of CP and AA were reduced (P < 0.01) by increasing levels of RFFS in diets. Conclusion: This study showed that some commercial SBM could be replaced by RFFS in broiler diets, without markedly compromising productivity. The AID and SID of CP and lysine were slightly improved by dietary supplementation of microbial protease.

Effects of Moist Extruded Full-fat Soybeans on Gut Morphology and Mucosal Cell Turnover Time of Weanling Pigs

  • Ten barrows, weaned at 28 days (7.2$\pm$0.1 kg BW), were used to evaluate the effects of feeding extruded full-fat soybeans on intestinal morphology and mucosal cell turnover time. All pigs were fed corn-based diets with half of the pigs receiving diets supplemented with 15.5% soybean meal and 3% soybean oil and the remaining pigs fed a diet in which the soybean meal and oil were replaced by 18.5% extruded full-fat soybeans. The pigs were individually placed in $80{\times}150cm$ metabolic cages and fed twice daily an amount approximately equal to their ad libitum intake for a period of 14 days. On day 14, pigs were weighed and then injected intraperitoneally with $^3$H]thymidine ($100{\mu}Ci/kg$ of BW, specific activity 20 Ci/mmol) 6 h after the morning meal. A pig from each treatment was killed 1, 4, 8, 16, or 24 h postinjection and intestinal tissues were collected. Daily gains for pigs fed the soybean diet and extruded full-fat soybean diet were 0.24 and 0.31 kg/day (p=0.05) with feed conversions of 1.58 and 1.39 (p=0.05), respectively. In comparison with pigs fed soybean meal, pigs fed moist extruded full-fat soybeans had a decreased crypt depth in their duodenum and cecum (p<0.1), while the villus height in the mid jejunum and ileum and the total height (villus height plus crypt depth) of the ileum and mid jejunum increased (p<0.05). The villus width in the duodenum and mid jejunum decreased (p<0.05). The number of crypt epithelial cells in the upper jejunum increased but decreased in the ileum, colon and cecum (p<0.05). The number of villus epithelial cells in the ileum and the upper and mid jejunum increased (p<0.05). The time for migration of epithelial cells in the crypt-villus column decreased (p<0.05) in all sites except the upper jejunum, ileum and cecum. The mucosal turnover rate for all intestinal sites except the upper jejunum, colon and cecum decreased (p<0.05). From these data, we conclude that inclusion of moist extruded full-fat soybeans in weanling pig diets can improve the intestinal morphology and slow the migration rate and turnover time of epithelial cells of the small intestine, especially in the mid jejunum compared with soybean meal.
