A New Report of Biscogniauxia petrensis Isolated from Mosquitoes in Korea

  • A fungal strain designated KNU-WDM2A2 was isolated from mosquitoes in Gimcheon, Korea. The pure culture was transferred to potato dextrose agar (PDA) and synthetic nutrient agar (SNA) media and attained a diameter of 90 mm after 10 days of incubation at 25℃. The colonies were whitish to light pink and cottony to wooly, with an abundant production of aerial mycelia. The strain produced hyaline to slightly yellowish conidiophores that were rough-walled and branched, with conidiogenous cells arising terminally or laterally. Conidia were unicellular, hyaline to light brown, smooth, and oval or ovoid to clavate, with a size of 4.1-6.9×2.5-3.3 ㎛ (n=65). A phylogenetic analysis was conducted using the internal transcribed spacer (ITS) regions and 28S rDNA of large subunit (LSU) sequences, to support the cultural and morphological characteristics. The KNU-WDM2A2 strain was identified here as Biscogniauxia petrensis, new to Korea.

Genetic Clarification of Auricularia heimuer Strains Bred and Cultivated in Korea Using the ITS and IGS1 rDNA Region Sequences

  • Auricularia is one of the broadly cultivated edible mushrooms in Korea. Most of the Korean Auricularia strains used for cultivation and breeding are known as A. auricula-judae. Recently, this species has been reported to belong to a species complex. Therefore, this study was carried out to genetically clarify the bred and cultivated Korean A. auricula-judae strains. The internal transcribed spacer (ITS) and IGS1 rDNA region sequences were determined from 10 A. auricula-judae strains by PCR and sequencing. Variation in the nucleotide sequence and sequence length of the two rDNA regions were found among the seven A. auricula-judae strains. A maximum-likelihood (ML) phylogenetic tree based on the ITS sequences clearly placed all the 10 Korean A. auricula-judae strains in the A. heimuer clade of the A. auriculajudae complex. A. heimuer is diverged from A. auricula-judae. An ML phylogenetic tree based on the IGS1 sequences revealed the close relationship between Korean A. heimuer strains to Chinese A. heimuer strains. But each strain could be distinguishable by the IGS1 sequence. Furthermore, progeny strains in the seven Korean strains could be differentiated from their parental strains by the IGS1 sequence based phylogenetic tree. Our results are expected to be used to complement the distinction of domestic Auricularia cultivars.

First record of a marine microalgal species, Jaagichlorella roystonensis (Trebouxiophyceae) isolated from Jungmun Saekdal Beach, Jeju Island, Korea

  • A eukaryotic marine microalga was isolated from Jungmun Saekdal Beach, Jeju Island, Korea and an integrated approach, including molecular phylogeny and morphology, was used to determine its taxonomical status. Molecular phylogenetic evidence inferred from the small subunit (SSU) 18S rRNA sequence and internal transcribed spacer (ITS) secondary structure analysis clearly showed that the isolate belonged to the recently described species, Jaagichlorella roystonensis. Distinctive morphological keys of the species were also observed by light microscopy and scanning/transmission electron microscopy(S/TEM). In this study, a Korean marine J. roystonensis species was described for the first time and was subsequently added to the national culture collections in Korea.

Biodegradation of Organochlorine Insecticide Endosulfan by the Fungus Eutypella sp. KEF-1

  • BACKGROUD: ${\alpha}$- and ${\beta}$- Endosulfan isomers of endosulfan, an endocrine disrupting chemical, are widely used cyclodiene organochlorine pesticide in worldwide, and it has widespread application in agriculture and can contaminate river-system as runoff from soil or aerial deposition METHOD AND RESULTS: In this study, an attempt was made to isolate an endosulfan degrading fungus from endosulfan-polluted agricultural soil. Through repetitive enrichment and successive subculture in media containing endosulfan and its metabolites as the sole carbon source, a fungus designated KEF-1 was isolated. Based on phylogenetic analysis, strain KEF-1 was assigned to the genus Eutypella. Also, the ITS (internal transcribed spacer) sequences of KEF-1 were submitted to GenBank under accession number EF581006. In potato dextrose broth containing 8 ${\mu}g$/mL endosulfan, strain KEF-1 completely degraded the endosulfanin 12 days. CONCLUSION: These results suggested that Eutypella sp. KEF-1 has potential as a biocatalyst for endosulfan bioremediation

Screening of Antioxidative Activity of Bifidobacterium Species Isolated from Korean Infant Feces and Their Identification

  • Among 59 Korean isolated, 20 were confirmed as members of the genus Bifidobaferium species based on gram staining, microscopic examination of cell morphology and the TLC method, The oxygen tolerance and antioxidative activities of these 20 Bifidobacterium strains and 5 standard Bifidobaferium strains were tested. All the strains demonstrated antioxidative activities as regards inhibiting linoleic acid peroxidation. The antioxidative activities of isolated and standard strains were found to range from 10.7-46.4% and from 10.7-22.2%, respectively. In addition, all tested strains exhibited a Scavenging ability on DPPH free radicals, range from 15-41% for the isolated strains and 8.3-22% for the standard strain. Accordingly. the isolated Bifidobarterium strains demonstrated higher antioxidative artivities than the 5 standa rd Bifidobarterium strains. On the base of grades for each test, HJL 7511 was identified 35 the best strain, followed by HJL 7501. 2 strains were identified with Polymerase Chain Reaction (PCR) assay using group-specific primers designed from the nucleotide Sequences of the 16S rRNA and internal transcribed spacer (ITS) regions of the Bifidobacteria. Based on the Sequencing results, HJL 7511 and HJL 7501 were identified as Bifidobacterium infantis.

A New Species of Thelonectria and a New Record of Cephalotrichum hinnuleum from Gunwi and Ulleungdo in Korea

  • Three fungal strains belonging to the class Sordariomycetes were isolated from soils collected from Gyeongsangbuk-do in Korea. They were identified as Cephalotrichum hinnuleum (UD CT 1-3-3 and KNU-19GWF1) and Thelonectria chlamydospora sp. nov. (UD ST 1-2-1). T. chlamydospora sp. nov. was morphologically identical to T. truncata, but its specific macroconidial dimensions, lower number of septations, and chlamydospore diameter render it distinct from the strains of the genus Thelonectria. The strains UD CT 1-3-3 and KNU-19GWF1 were developed flat, velvety to felty, and golden gray to brown-gray after 14 days of incubation at 25 ℃ on PDA. These strains were produced polyblastic conidiogenous cells and conidia were pale brown to brown, smooth, thin-walled, subglobose to ellipsoidal, arranged in chains, and the diameters of 6.7-9.0 × 3.7-5.1 ㎛. The strains were also confirmed by using the multi-locus genes using internal transcribed spacer (ITS) regions, partial large subunit (LSU), translation elongation factor 1α (TEF1-α), β-tubulin (TUB2), and actin (ACT) genes. This is the discovery of T. chlamydospora sp. nov. and Cephalotrichum hinnuleum, a new record from Korea.

Novel Fungal Species Belonging to the Genus Acaulium Isolated from Riptortus clavatus (Heteroptera: Alydidae) in Korea

  • A survey of insect-associated fungi in Korea revealed a novel fungal strain isolated from the bean bug Riptortus clavatus (Heteroptera: Alydidae). Culturally and morphologically, the fungal strain designated KNUF-20-INY03, shares features with members of the genus Acaulium. Phylogenetic analyses based on the concatenated nucleotide sequences of the internal transcribed spacer regions (ITS) regions and partial sequences of the translation elongation factor 1-alpha (TEF1-α), and β-tubulin (β-TUB), and large subunit of the nuclear ribosomal RNA (LSU) genes showed that the isolate is part of a clade that includes other Acaulium species, but it occupies a distinct phylogenetic position. Based on the shape, size, and color of its conidia and conidiogenous cells, strain KNUF-20-INY03 is readily distinguishable from the closely related A. acremonium, A. albonigrescens, A. caviariformis, A. pannemaniae, and A. retardatum. The conidial length-to-width ratio (1.6) of the novel isolate is significantly lower than that of A. acremonium (1.9), A. albonigrescens (2.4), and A. pannemaniae (2.4), and KNUF-20-INY03 produces hyaline conidia and elliptical conidiogenous cells while A. caviariformis forms brown conidia and A. retardatum produces flask-shaped conidiogenous cells. Thus, both phylogenetic and morphological analyses indicate that this strain is a novel species in the genus Acaulium, and we propose the name Acaulium microspora sp. nov.

ITS 영역의 HRM 분석을 통한 참당귀(Angelica gigas Nakai)의 특이적 SNP 분자표지 개발 (Development of specific single nucleotide polymorphism molecular markers for Angelica gigas Nakai)

  • 당귀는 약용으로 널리 사용되는 대표적인 다년생 식물이다. 다양한 당귀 계통의 유전적 다양성에 대한 정보는 당귀의 유전자원의 발굴 및 보존의 차원에서 매우 중요하다. 당귀는 현재 한국에 등록 된 중요한 약용 식물 종이지만, 다른 나라의 다른 유사한 종과 구별 할 수 있는 분자표지가 없는 실정이다. 그러므로, 본 연구에서는 HRM 분석을 통해 국내 토종 당귀계통인 참당귀를 식별하기 위해 유전체 염기서열의 핵리보솜 DNA 내부(ITS)영역에서 단일염기다형성(SNP) 분자 표지를 개발하였다. 본 연구에서는 또한 5가지 당귀계통들의 혼합된 염색체 DNA 시료를 이용하여 HRM 분석을 수행하였으며, 최종적으로는 혼합된 염색체 DNA의 다양한 비율을 사용하여 혼용되는 대표적인 당귀계통인 참당귀와 중국 당귀를 재료로 HRM 분석을 진행하였다. 그러므로, 본 연구에서 개발된 SNP 분자표지는 다양한 지역 또는 국가에서 서식하는 당귀 계통들의 신속한 확인을 위해 매우 유용하게 이용될 것으로 생각된다.

Fasciola hepatica: Infection Status of Freshwater Snails Collected from Gangwon-do (Province), Korea

  • Fasciola hepatica is a trematode that causes zoonosis, mainly in cattle and sheep, and occasionally in humans. Few recent studies have determined the infection status of this fluke in Korea. In August 2015, we collected 402 samples of freshwater snails at Hoenggye-ri (upper stream) and Suha-ri (lower stream) of Song-cheon (stream) in Daegwalnyeong-myeon, Pyeongchang-gun in Gangwon-do (Province) near many large cattle or sheep farms. F. hepatica infection was determined using PCR on the nuclear ribosomal internal transcribed spacer 2 (ITS-2). Among the 402 samples, F. hepatica 1TS-2 marker was detected in 6 freshwater snails; thus, the overall prevalence in freshwater snails was 1.5%. The prevalence varied between collection areas, ranging from 0.0% at Hoenggye-ri to 2.9% at Suha-ri. However, F. gigantica ITS-2 was not detected in the 6 F. hepatica-positive samples by PCR. The nucleotide sequences of the 6 F. hepatica ITS-2 PCR-positive samples were 99.4% identical to the F. hepatica ITS-2 sequences in GenBank, whereas they were 98.4% similar to F. gigantica ITS-2 sequences. These results indicated that the prevalence of F. hepatica in snail intermediate hosts was 1.5% in Gangwon-do, Korea; however the prevalence varied between collection areas. These results may help us to understand F. hepatica infection status in natural environments.

ITS2 부위의 염기서열 및 RAPC-PCR에 의한 Pseudo-nitzschia 4종의 유연관계 (Phylogenetic Relationships Using ITS2 Sequence and RAPD-PCR Data from Four Species of Korean Pseudo-nitzschia (Bacillariophyceae))

  • ITS2 부위를 시퀀싱하여 Pseudo-nitzschia delcatissima, P. multiseries, P. pungens, P. subfraudulenta 상호간의 유전자 다양도를 조사함과 아울러 RAPD-PCR pattern을 이용하여 유사도를 구하였다. 유전자 거리를 근거로 했을 때 P. delicatissima 종은 P. multiseries와 P. pungens와는 유전적 거리가 상당히 요원하였고, 심지어 P. subfraudenlta와도 거리를 보였다. 유사도의 경 P. multiseries와 P. pungens는 0.31로 보인 반면에, P delicatissima는 다른 세종과 0.81를 나타내었다. 따라서 P. delicatissima 종은 P. multiseries, P. pungens, P. subfraudulenta와는 유전적으로 밀접하지 않는 관계로 보였다. ITS2부위는 Pseudo-nitzschia 동정에 사용될 수 있는 유용한 도구로 보이며 형태적으로 구분할 수 없는 P. multiseries와 P. pungens을 구분할 수 있다. 또한 RAPD-PCR 방법도 단시간에 Pseudo-nitzchia을 분리시키는데 사용될 것으로 보인다.
