Notes on Endophytic Fungi Isolated from Roots of Oreorchis patens in Korea (감자난초 뿌리에서 분리한 내생균의 동정)

  • Endophytic fungi were isolated from surface-sterilized roots of a species of orchid, Oreorchis patens, in Korea. The isolated strains were identified based on morphological characteristics and sequence analyses of the internal transcribed spacer (ITS) and large subunit (LSU) of rDNA regions. Two species, Thelonectria veuillotiana and Phialocephala humicola, are first reported in Korea with their descriptions.

  • We isolated endophytic fungi from the leaves of the conifer Juniperus chinensis var. sargentii inhabiting Mt. Hallasan. The fungal isolates were identified based on phylogenetic analysis using nucleotide sequences of the internal transcribed spacer region, large subunit region of ribosomal DNA, translation elongation factor, and ${\beta}$-tubulin region. In this study, we confirmed four species of unreported endophytic fungi in Korea, Allantophomopsis lycopodina, Epicoleosporium ramularioides, Kabatina juniperi, and Seiridium podocarpi. We describe their morphological characteristics and the results of the phylogenetic analysis.

  • Three endophytic fungal strains were isolated from aquatic plants collected in Jeju. The fungi were identified by sequencing and phylogenetically analyzing their internal transcribed spacer (ITS) regions. Consequently, the three strains NNIBRFG2982, NNIBRFG2984 and NNIBRFG 3040 were confirmed as the fungal species Acremonium tubakii, Colletotrichum cliviae and Cylindrocarpon pauciseptatum, respectively, all of which are new to Korea.

  • We isolated endophytic fungal strains from the roots of three medicinal crops, Ligusticum chuanxiong, Angelica gigas, and Cnidium officinale, cultivated at Yeongju, Korea. The fungal strains were identified based on their morphological characteristics and molecular analyses of their internal transcribed spacer, large subunit rDNA, and beta-tubulin regions. Thereby, we identified three previously unrecorded endophytic fungal species, Dactylonectria pauciseptata, Rhizopycnis vagum, and Sistotrema sernanderi, in Korea. In this report, we describe the morphological characteristics and phylogenetic analyses of these three fungal species.

  • We isolated endophytic fungi from the leaves of woody plants inhabiting the Ulleungdo Island and Yeosu in South Korea. The fungal strains were identified based on their morphological characteristics in conjunction with phylogenetic analysis, which was performed using sequences of the internal transcribed spacer, large subunit rDNA, and β-tubulin DNA. We confirmed the presence of Neofusicoccum mangiferae and Thyronectria cucurbitula, two endophytic fungal species that had not been previously recorded in Korea. In this report, we describe the morphological characteristics of these fungal strains and results of the DNA-based phylogenetic analysis.

Analysis and evaluation of morphological and molecular polymorphism in the hybridization of Elaeagnus ×maritima and E. ×submacrophylla (잡종 기원 녹보리똥나무와 큰보리장나무의 형태학적 및 분자적 다양성 분석 및 평가)

  • The taxonomic identity of Elaeagnus ×maritima and E. ×submacrophylla (Elaeagnaceae) in Korea is unclear, yet they are presumed to be hybrid taxa based on their morphology. To determine their hybrid origins, a morphological analysis (field surveys and specimen examinations) and a molecular analysis involving two nuclear ribosomal DNA (nrDNA) regions (internal transcribed spacer and 5S non-transcribed spacer) and one chloroplast DNA (cpDNA) region (matK) were conducted. The morphological analysis revealed that E. ×maritima showed certain morphological similarities to E. glabra, whereas E. ×submacrophylla showed certain morphological similarities to E. pungens. However, the molecular analysis indicated that E. ×maritima exhibited additive species-specific sites of E. glabra and E. macrophylla in the nrDNA regions. Notably, E. ×submacrophylla showed various aspects, with some individuals exhibiting additive species-specific sites of E. pungens and E. macrophylla in the nrDNA and E. macrophylla sequences in the cpDNA regions, some individuals exhibiting E. macrophylla sequences in the nrDNA and E. pungens sequences in the cpDNA regions, and some individuals displaying E. macrophylla sequences in both the nrDNA and cpDNA regions, despite an intermediate morphology between E. pungens and E. macrophylla. These results indicate that these two species are of hybrid origin and frequently cross between parental and hybrid individuals.

PCR-DGGE and PCR-RFLP Analyses of the Internal Trascribed Spacer(ITS) of Ribosomal DNA in the Genus Rhizopus

  • To estimate genetic relationships within the genus Rhizopus, genetic variations in 20 strains were investigated by DGGE and PCR-RFLP of rDNA ITS region (ITSI, ITS2,5.8S). The size of the amplified products showed the interspecific polymorphisms, 650 bp,700 bp, and 900 bp. The DGGE approach allowed the separation of PCR amplicons of the same length according to their sequence variations. When the rDNA ITS region was digested with six restriction enzymes, 20 strains were classified into five RFLP haplotypes. The range of similarity between the 20 strains by PCR-RFLP was 42.3-100%. Based on the results of DGGE aud PCR-RFLP, the 20 strains were divided into four groups, R. oryzae, R. stolonifer, R. microsporus and R. homothallicus. Further study of R. homothallicus is required.

  • The length and G/C concent of regions of an aphid rDNA unit that spans 13,061bo with 59% G/C content. flolowing belowing below are the those results, 5’ETS is 843bp in length with 69% G/C content, 18S is 2,469bp in length with 59% G/C content, ITS I is 229bp in length with 70% G/C content, 5.8S is 160bp in length with 63% G/C content, ITS II is 325bp in length with 70% G/C content, 28S is 4, 147bp in length with 60% G/C content, IGS is 4,888bp in length with 55% G/C content.

  • The family Ulvaceae species are difficult to distinguish from one another on the basis of morphological and cytological criteria alone. ITS2 sequences are hewn to evolve quickly and have been reported to be useful for the study of intraspecific and interspecific variation and biogeography in algae (Bakker et al., 1992). We will here describe the basic characteristics of the ITS2 sequences in Ulva and Effteromorpha to compare our result with the above previous studies. (omitted)

First Report of Rhizopus oryzae as a Postharvest Pathogen of Apple in Korea

  • Soft rot in apple caused by Rhizopus oryzae was found for the first time in Korea. A detailed description of the specimen is given along with its internal transcribed spacer rDNA sequence. The fungus was identified as Rhizopus oryzae based on the mycological characteristics, molecular data, and pathogenicity testing.
