  • Fracture characteristics of plate shape using shock tube for glass filled ceramics was carried out. Glass filled ceramics have been considered as a promising candidate material for the dome port cover of air breathing engine. This part of the air breathing engine has an important role separating solid and liquid fuel, and needs the frangible characteristics that the fracture of a part should not affect the internal components of combustion. The objectives of this study are to evaluate the fracture pressures for various thicknesses and diameters of shock impact area. Also fracture phenomena of separated membrane using a shock tube are observed. The experimental apparatus of shock tube consists of a driver, a driven section and a dump tank. The used material is glass filled ceramic made from Corning company. Specimens are used 3, 4.5 and 6mm thickness. Also diameters of shock wave area are chosen 70, 60 and 50 mm. It is expected that the results obtained from this study can be used in the basic data for the dome port cover design of an air breathing engine.

Static and dynamic analysis of circular beams using explicit stiffness matrix

  • Two new elements with six degrees of freedom are proposed by applying the equilibrium conditions and strain-displacement equations. The first element is formulated for the infinite ratio of beam radius to thickness. In the second one, theory of the thick beam is used. Advantage of these elements is that by utilizing only one element, the exact solution will be obtained. Due to incorporating equilibrium conditions in the presented formulations, both proposed elements gave the precise internal forces. By solving some numerical tests, the high performance of the recommended formulations and also, interaction effects of the bending and axial forces will be demonstrated. While the second element has less error than the first one in thick regimes, the first element can be used for all regimes due to simplicity and good convergence. Based on static responses, it can be deduced that the first element is efficient for all the range of structural characteristics. The free vibration analysis will be performed using the first element. The results of static and dynamic tests show no deficiency, such as, shear and membrane locking and excessive stiff structural behavior.

  • 사하중과 단조 활하중을 받고 있는 철근콘크리트판에 대한 일반적인 유한요소 해석방법을 개발하였다. 이 방법을 통하여 탄성, 비탄성 및 극한범위에서 하중-변형 응답과 균열전파를 추적할 수 있었고 또한 응답 경로를 통하여 콘크리트와 철근의 내부응력을 결정할 수 있었다. 면내응력과 휨응력간의 상호작용을 고려하는 층분할된 8 절점 등매개변수 요소를 개발하였다. 수치해를 구하는 방법은 접선증분 강성도법을 이용하였다. 본 해법에 대한 유효성을 검토하기 위하여 다른 해석결과들과 비교하였다.

단백질의 구조 전환과 기능 조절 (Conformational Switch and Functional Regulation of Proteins)

  • In common globular proteins, the native form is in its most stable state. However, the native form of inhibitory serpins (serine protease inhibitors) and some viral membrane fusion proteins is in a metastable state. Metastability in these proteins is critical to their biological functions. Our previous studies revealed that unusual interactions, such as side-chain overpacking, buried polar groups, surface hydrophobic pockets, and internal cavities are the structural basis of the native metastability. To understand the mechanism by which these structural defects regulate protein functions, cavity-filling mutations of a 1-antitrypsin, a prototype serpin, were characterized. Increasing conformational stability is correlated with decreasing inhibitory activity. Moreover, the activity loss appears to correlate with the decrease in the rate of the conformational switch during complex formation with a target protease. We also increased the stability of a 1-antitrypsin greatly via combining various stabilizing single amino acid substitutions that were distributed throughout the molecule. The results showed that a substantial increase of stability, over 13 kcal/mol, affected the inhibitory activity with a correlation of 11% activity loss per kcal/mol. The results strongly suggest that the native metastability of proteins is indeed a structural design that regulates protein functions and that the native strain of a 1-antitrypsin distributed throughout the molecule regulates the inhibitory function in a concerted manner.

  • 공기막구조물의 공간형성방법은 크게 단일공기막과 이중공기막 방식으로 나뉜다. 내압으로 인해 평형곡면이 형성되는 공기막구조물의 형상은 형태의 변화가 외력에 의해 크게 변형되거나 응력이 집중되지 않는 등장력 곡면으로 많이 이용된다. 따라서 본 연구에서는 공기막 구조물의 초기 형성된 형상에 대해서 설계하중조합에 대한 구조해석을 비선형 전용해석프로그램인 NASS를 이용하여 수행한 후 변형형상을 살펴보고, 최대응력을 이용하여 안전성을 검토한다. 해석 모델은 사면에서 지지되는 장방형 단일공기막 구조물을 대상으로 하며, 직교이방성을 고려하여 비선형 증분해석을 수행한다.

Ultrastructural Changes in Cryptosporidium parvum Oocysts by Gamma Irradiation

  • Cryptosporidium parvum is known as one of the most highly resistant parasites to gamma irradiation. To morphologically have an insight on the radioresistance of this parasite, ultrastructural changes in C. parvum sporozoites were observed after gamma irradiation using various doses (1, 5, 10, and 25 kGy) following a range of post-irradiation incubation times (10 kGy for 6, 12, 24, 48, 72, and 96 hr). The ultrastructures of C. parvum oocysts changed remarkably after a 10-kGy irradiation. Nuclear membrane changes and degranulation of dense granules were observed with high doses over 10 kGy, and morphological changes in micronemes and rhoptries were observed with very high doses over 25 kGy. Oocyst walls were not affected by irradiation, whereas the internal structures of sporozoites degenerated completely 96 hr post-irradiation using a dose of 10 kGy. From this study, morphological evidence of radioresistance of C. parvum has been supplemented.

Cloning and characterization of a gene encoding ABP57, a soluble auxin-binding protein

  • Auxin-binding protein 57 ($ABP_{57}$), a soluble auxin-binding protein, acts as a receptor to activate plasma membrane (PM) $H^+-ATPase$. Here, we report the cloning of abp57 and the biochemical characterization of its protein expressed in E. coli. The analysis of internal amino acid sequences of $ABP_{57}$ purified from rice shoots enabled us to search for the corresponding gene in protein DB of NCBI. Further BLAST analysis showed that rice has four abp57-like genes and maize has at least one homolog. Interestingly, Arabidopsis seems to have no homolog. Recombinant $ABP_{57}$ expressed in E. coli caused the activation of PM $H^+-ATPase$ regardless of the existence of IAA. Scatchard analysis showed that the recombinant protein has relatively low affinity to IAA as compared to natural $ABP_{57}$. These results collectively support the notion that the cloned gene is responsible for $ABP_{57}$.

Hydrogen Peroxide Induces Apoptosis of BJAB Cells Due to Formation of Hydroxyl Radicals Via Intracellular Iron-mediated Fenton Chemistry in Glucose Oxidase-mediated Oxidative Stress

  • The aim of this study was to determine if hydrogen peroxide ($H_2O_2$) generated by glucose oxidase (GO) induces apoptosis or necrosis of BJAB cells and which radical is the direct mediator of cell death. We found that GO produced $H_2O_2$ continuously in low concentrations, similar to in vivo conditions, and decreased proliferation and cell viability in a dose-dependent manner. The GO-mediated cytotoxicity resulted from apoptosis, and was confirmed by monitoring the cells after H33342/Annexin V/propidium iodide staining. Decreases of mitochondrial membrane potential and intracellular glutathione level were found to be critical events in the $H_2O_2$-mediated apoptosis. Additional experiments revealed that $H_2O_2$ exerted its apoptotic action through the formation of hydroxyl radicals via the Fenton rather than the Haber-Weiss reaction. Moreover, intracellular redox-active iron, but not copper, participated in the $H_2O_2$-mediated apoptosis.

콘크리트용 무기계 방수제의 침투깊이 측정방법에 관한 연구 (A Study on the Penetration Depth Measurement Methods of Inorganic Waterproofing Agent for Concrete)

  • 콘크리트용 무기계 방수제는 내부 막을 형성하지 않기 때문에 무기계 방수제의 침투깊이 정량적으로 평가하기 어렵다. 그러나 무기계 방수제의 침투깊이 평가는 일관된 침투효율을 확보하는데 있어서 매우 중요한 평가지표 중 하나이다. 따라서 본 논문에서는 콘크리트용 무기계 방수제의 침투깊이 측정방법을 고안하였으며, 고안된 침투깊이 측정방법을 통하여 기존 무기계 방수제의 침투깊이를 평가하였다. 실험결과 고안된 무기계 방수제의 침투깊이 측정방법을 통하여 평가된 침투깊이는 90% 이상의 신뢰성이 있는 것으로 나타났다.
