The aim of this study was to evaluate the anti-inflammatory effects of essential oils extracted from the wood of Chamaecyparis obtusa, Pinus densiflora, Pinus koraiensis, and Larix kaempferi. Essential oils were extracted by hydrodistillation, and their chemical components were determined by GC/MS. Major chemical components of these essential oils were ${\alpha}$-cadinol (19.25%), ${\tau}$-muurolol (14.20%), and ${\alpha}$-pinene (13.74%) in C. obtusa; ${\alpha}$-pinene (47.16%), longifolene (14.31%), ${\beta}$-phellandrene (11.78%), and ${\beta}$-pinene (11.02%) in P. densiflora; ${\alpha}$-pinene (13.49%) and longifolene (10.79%) in P. koraiensis, and geranyl linalool (23.58%) and ${\alpha}$-pinene (18.57%) in L. kaempferi. To evaluate the anti-inflammatory effects of essential oils, lipopolysaccharide (LPS)-induced RBL-2H3 mast cells were treated with these essential oils; then, the changes in the mRNA expression level of the cytokines IL-4 and IL-13 were examined. Further, degranulation was evaluated by measuring ${\beta}$-hexosaminidase release. After LPS-induced RBL-2H3 mast cells were exposed to $10^{-7}%$ of all types of essential oils, the gene expression levels of IL-4 and IL-13 within the cells remarkably decreased. The relative mRNA expression level of IL-4 was 69.6% in P. densiflora, 63.2% in P. koraiensis, 55.1% in C. obtusa, and 45.8% in L. kaempferi compared with that in the group treated with LPS. The mRNA expression level of L-13 should a similar trend. The inhibitory rate of IL-13 mRNA expression of P. densiflora, P. koraiensis, C. obtusa, and L. kaempferi was 57.8%, 57.1%, 51.1%, and 34.5%, respectively. ${\beta}$-Hexosaminidase release significantly decreased following the treatment with the four types of essential oils. The rate of ${\beta}$-hexosaminidase release were 38.1% C. obtusa; 33.0% P. densiflora; 27.4% P. koraiensis; and 9.1% L. kaempferi. Among all types of essential oils, that extracted from P. densiflora wood showed the highest anti-inflammatory activity. These results show that the tested essential oils exert an anti-inflammatory effect through the inhibition of degranulation and expression of cytokines.
Jeungaektang (JAT) is the herbal formula, has the effect of moistening the dryness by activating lung Qi and by nourishing Yin, has being used for dryness syndromes. Generally the herbal formulae for moistening dryness are used for exogenous or endogenous dryness syndromes. JAT has been clinically used for the treatment of endogenous dryness syndromes. It is composed of Scrophulariae Radix. Rehmanniae Radix and Liriopis Tuber. Recent studies showed that JAT has a protective effect against $CCl_{4}-induced$ hepatotoxicity and anti-inflammatory effects against ear swelling of mouse induced by Crotonis Fructus. However, the effect of JAT on the immunological activity was rarely studied. Therefore, this study evaluated the effects of JAT the regulatory mechanism of nitric oxide (NO) and cytokines in the lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. After the treatment of JAT water extract, cell viability was measured by MTT assay, NO production was monitored by measuring the nitrite content in culture medium. Cyclooxygenase-2 (COX -2) and inducible nitric oxide synthase (iNOS) were determined by immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results provided evidence that JAT inhibited the production of nitrite and nitrate ($0.1{\sim}1.0$ mg/ml), iNOS ($0.1{\sim}1.0$ mg/ml), $interleukin-1{\beta}$ ( $0.1{\sim}1.0$ mg/ml) and tumor necrosis $factor-{\alpha}$ ($0.1{\sim}1.0$ mg/ml) in RAW 264.7 cells activated with LPS. Furthermore, JAT inhibited the expression of COX-2 expression and production of prostagladin E2 ($0.1{\sim}1.0$ mg/ml). These findings suggest that JAT can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections.

This study was conducted to investigate the effects of hydrolyzed Lactobacillus supplementation with digestive enzymes treatment on the macrophage activation, the induction of nitric oxide(NO), interleukin (IL)-6 and tumor necrosis factor(TNF)-$\alpha$. The RAW 264.7 murine macrophage was exposed to porcine Lactobacillus strains which were digested with both pepsin and pancreatin. The production of NO, TNF-$\alpha$ and IL-6 in the macrophage were strain and dose-dependent, respectively. The induction of NO and cytokines were higher in both 3149 and 3146 strains compared with other Lactobacillus strains. Overall, the level of NO was observed at the lower range between 10 and 150 μg hydrolysates per ml, whereas IL-6 and TNF-$\alpha$ were observed at relatively higher concentration between 50 and 300 μg hydrolysates per ml. Lactobacillus strains which produced a high level of NO also showed a high induction of TNF-$\alpha$ and IL-6. Therefore, the present results suggest that hydrolysates of Lactobacillus strains are related to induction of several macrophage mediators, and then it could be beneficially used to modulate gastrointestinal immune function of the host. Also, the methodogly employed in this study might be useful to investigate the effects of lactic acid bacteria on gastrointestinal immunity.

This study aimed to evaluate seaweed polysaccharide extracts as a cosmetic material. To assess anti-microbial efficacy, Staphylococcus aureus (S. aureus) was treated with seaweed polysaccharide extracts and zones of inhibition were measured. In addition, the anti-inflammatory effect was confirmed in RAW 264.7cells, and seaweed polysaccharide extracts was applied to the dorsal skin of Sprague-Dawley (SD) rats to evaluate single-dose toxicity. As a results, seaweed polysaccharide extracts did not exhibit cytotoxicity at concentrations up to $1,000{\mu}g/mL$ in skin fibroblasts. Furthermore, when S. aureus was treated with 1% seaweed polysaccharide extracts, clear zones of $1.52{\pm}0.34cm$ formed, confirming sufficient anti-microbial activity. When RAW 264.7 cells were treated with seaweed polysaccharide extracts extract, nitric oxide (NO) production decreased in a concentration-dependent manner and the production of inflammation-related cytokines, such as interleukin 1 beta ($IL1{\beta}$), tumor necrosis factor alpha ($TNF{\alpha}$), and prostaglandin E2(PGE2), decreased. When seaweed polysaccharide extracts extract was applied at various concentrations to rats, symptoms did not change for more than 14 d, and there was no change in body or organ weights. In conclusion we found that seaweed polysaccharide extracts is not cytotoxic and has anti-microbial and anti-inflammatory effects. Therefore, it is suitable for use as a cosmetic material.

Algae is a potential resource with various biological activities. In this study, the anti-inflammatory effect of Grateloupia imbricata Holmes ethanol extract (GIHEE) from red algae was investigated in LPS-induced RAW 264.7 cells. As a result, reduced secretion of pro-inflammatory cytokines [tumor necrosis factors-${\alpha}$, interleukin (IL)-$1{\beta}$, and IL-6] and nitric oxide (NO) was observed in a dose-dependent manner. Expression of nuclear factor-kappaB (NF-${\kappa}B$) as well as inducible NO synthase and cyclooxygenase-2 proteins was reduced by GIHEE, suggesting that the anti-inflammatory activity of GIHEE is related to suppression of NF-${\kappa}B$ signaling pathways. In addition, GIHEE reduced phosphorylation of mitogen-activated protein kinases. These results suggest that GIHEE can be used as a potential anti-inflammatory therapeutic.
Saussurea lappa is known for a variety of physiological activities as a component but has not known to show the effect of the cellular signaling pathway. We investigated the anti-inflammatory effects by Saussurea lappa ethanol extract on the LPS(lipopolysaccharide) induced nitric oxide (NO) production by RAW 264.7 cell line. It shows the expressions of iNOS and COX-2 at the transcriptional level (RT-PCR). The Saussurea lappa ethanol extract showed transcriptional expression levels of pro-inflammatory cytokine TNF- and IL-$1{\beta}$ induced by LPS(lipolysaccharide) in RAW264.7 cell line. Saussurea lappa ethanol extract reduced the LPS-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the RNA levels in a concentration-dependent manner. The finding that ethanol extract of Saussurea lappa has an influence on NO (nitric oxide) homeostasis in the study of the action mechanism on the macrophage-mediated inflammatory reaction was considered in terms of convergence. And it is to provide an important basis for the prevention and treatment of inflammatory diseases in the future.
Objectives: This study was performed to identify the anti-inflammatory effect of Pulsatilla koreana (PK) methanol extract on lipopolysaccharide (LPS) induced inflammatory. Methods: There were five groups. They were control group, LPS-exposed PK methanol extract group ($0{\mu}g/ml$, $10{\mu}g/ml$, $30{\mu}g/ml$, $100{\mu}g/ml$). To measure out cytotoxicity of PK, we performed the MTT assay. To evaluate the anti-inflammatory effect of PK, we examined the inflammatory mediators, such as nitric oxide (NO), prostaglandin E2 (PGE2), and pro-inflammatory cytokines (interleukin $[IL]-1{\beta}$, IL-6, tumor necrosis $factor-{\alpha}$ [$TNF-{\alpha}$], IL-10). Results: 1. The extract of PK (${\sim}100{\mu}g/ml$) itself did not have any cytotoxic effect in RAW 264.7 cells. 2. The concentration of plasma NO and PGE2 in PK methanol extract group showed a lower values than those of control group. 3. The concentration of plasma $IL-1{\beta}$, plasma IL-6, and plasma $TNF-{\alpha}$ in PK methanol extract group showed a lower values than those of control group. 4. The concentration of Plasma IL-10 in PK methanol extract groups showed higher than control group; however, these values showed no significantly different. Conclusions: According to this study, the extract of PK could be used as a protective agent against inflammation.
Salicornia herbacea L. (Chenopodiaceae: S. herbacea) is a salt marsh plant, which has long been prescribed in traditional medicines for the treatment of intestinal ailments, nephropathy, and hepatitis in Oriental countries. In order to elucidate the mechanisms of this herb, we conducted an anti-oxidative activity, the inhibition of nitric oxide (NO) production, and the suppression of the pro-inflammatory cytokine genes, with the solvent-extracts of S. herbacea. We found that both ethyl acetate and n-butanol tractions showed potent anti-oxidative effects in comparison to other fractions using xanthine oxidase assay with $IC_{50}$ values of $66.0{\pm}0.5\;{\mu}g/ml$ and $82.5{\pm}3.8\;{\mu}g/ml$, respectively. In addition, both ethyl acetate and n-butanol fractions showed more electron donating activity (EDA) than other tractions, according to DPPH (2, 2-Diphenyl-lpicrylhydrazyl radical) assay. The EDA of ethyl acetate fraction ($IC_{50}$ values of $117.5{\pm}3.8\;{\mu}g/ml$) is more significant than that of n-butanol fraction ($IC_{50}$ values of $375.0{\pm}12.5\;{\mu}g/ml$). Among potential anti-oxidative tractions, ethyl acetate traction dose-dependently suppressed lipopolysaccharide (LPS, $0.1\;{\mu}g/ml$)-induced nitric oxide (NO) production in RAW264.7 cell, while n-butanol did not. As expected, ethyl acetate fraction suppressed the expression of inducible NO synthase (iNOS) in RAW264.7 cell stimulated by $0.1\;{\mu}g/ml$ of LPS. Moreover, the ethyl acetate traction suppressed the expression of interleukin-1 $(IL)-1{\beta}$ and granulocyte/macrophage colony-stimulating factor (GM-CSF) mRNA in LPS-stimulated RAW264.7 cells. Therefore, these results suggest that S. herbacea may have anti-oxidative and anti-inflammatory activities by modulating radical-induced toxicity and various pro-inflammatory responses.

The purpose of the present study was to evaluate the effect of Squalene, Alkoxy Glycerol, Batyl and Chimyl solutions on the production of nitric oxide (NO), tumor necrosis factor-alpha (TNF- $\alpha$ ) and interleukin-6 (IL-6) in RAW 264.7 macrophage cells after treatment of C.albicans. The cytotoxic effects was evaluated by the lactate dehydrogenase (LDH). All solutions did not affect on the LDH and NO production by itself. At 6th hour, the TNF- $\alpha$ and IL-6 production was not affected by each solutions. However, at 24th hour, the TNF- $\alpha$ and IL-6 production was affected by itself (p < 0.05). Each solution in the presence of C. albicans decreased the C. albicans-induced TNF- $\alpha$ and IL-6 production compared with C. albicans only (p < 0.05, p < 0.01). These results suggest that Squalene, Alkoxy Glycerol, Batyl and Chimyl solutions will increase the immune response on the C. albicans-induced damage.
Traditionally, the larvae of Allomyrina dichotoma (AD), a species of the rhinoceros beetle, have been widely used for their antidiabetic, antihepatofibrotic, antineoplastic, and antiobesity effects. The United Nations' Food and Agriculture Organization has reported on the possibility of using edible insects in human dietary supplements in the future. However, despite the growing interest in insect-based bio-active products, the biological activities of these products have rarely been studied. Previously, we reported that AD larvae inhibit the in vitro differentiation of adipocytes via transcription factor downregulation. In this study, our objective was to evaluate the effects of a hot-water extract of AD larvae on allergy and inflammation. To investigate the inhibitory effect of the extract on allergic reactions, we measured the levels of ${\beta}-hexosaminidase$, tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-4 (IL-4), and cyclooxygenase-2 (COX-2) after activation of RBL-2H3 cells using Compound 48/80. In addition, the inhibitory effect of the extract on inflammation was determined using Raw 264.7 cells after lipopolysaccharide (LPS) stimulation. The extract significantly inhibited the ${\beta}-hexosaminidase$, $TNF-{\alpha}$, IL-4, and COX-2 levels in RBL-2H3 cells. Furthermore, it effectively inhibited the inflammatory cytokine IL-6, nitric oxide, and inducible nitric oxide synthase expression in LPS-stimulated Raw 264.7 cells. These results suggest that AD larval extract can be potentially developed as an antiallergic and anti-inflammatory therapeutic agent.
