Application and Development of 'Chestnut Management Standard Diagnostic Table' (밤나무 경영 표준진단표의 개발 및 적용)

  • The chestnut management standard diagnostic table is developed and would be utilized in order that a manager raising chestnuts checks where the own level of management is and grasps current state of own for the purpose of planning aims and advancing toward a higher level. The developed 'Chestnut management standard diagnostic table' consisted of 3 first classified items, 19 second classified items and 2 product indicator items by the chestnut experts consultative meeting. A survey of 212 farmhouses in 4 major producing area was conducted. Except invalid survey of 53 farmers, 159 farmhouses interviewed were used in analysis. Total score was calculated with sum of each item's score. According to the survey results, average score is 68.0 and Buyeo received the highest score of 69.7 and Suncheon received the lowest score of 61.8 by regional groups. The higher the group in score, the better it is in output per hectare. But the property of 'the ratio of the best products in total products' does not show a statistical correlation. Generally the score of 'management-based evaluation indicator part' and 'management and sale capacity indicator part' in Suncheon was low because of many elderly people. In part of 'manufacturing technology indicator' as Environmentally-Friendly production is progressed in over 70% of four regions, when comes to a disease and insect pest control there are rarely farmhouses having a way of crop dusting.

Flight of Matsucoccus thunbergianae Males in Response to Synthetic Pheromone Placed at Various Heights above Ground and the Wind Speed (합성(合成)페로몬의 지상(地上)높이별(別) 위치(位置) 및 풍속(風速)에 따른 솔껍질깍지벌레 수컷의 비행(飛行))

  • Matsucoccus thunbergianae is a major insect pest of Pinus thunbergiana in southern Korean peninsula. To study the flight behavior of M. thunbergianae males responding to the synthetic pheromone, five sticky traps were placed on a bamboo pole at various heights, between 0.1m and 2.0m above ground. A bait impregnated with the synthetic pheromone was placed at 0.1m, 1.0m or 2.0m above ground and the number of male catches on each trap was counted. In an open area, numbers of males caught per trap were not different between heights when the bait was placed at 2m or 1m above ground ; when the bait was placed at 0.1m height, male flight was aggregated near the ground. In a forest with low crown closure, trap catches on five traps on the same bamboo pole were not different one another when the bait was placed at 2m height, but most males were flying near the bait when it was placed at 1m height. In a dense pine forest, most males were flying around the bait regardless of the bait position. In all three places, most males were caught on the trap near the ground when the baits were placed 0.1m above ground. Thus, for monitoring the frontal zone of infestation of the scale, placing the pheromone trap near the ground was considered the most efficient. When the males perceived pheromone, they tended to fly in the air with low wind speed.

Culture Method of Spore for Entomopathogenic Fungus Using Natural Zeolite Ceramic Ball (천연제오라이트 세라믹볼을 이용한 곤충병원성 곰팡이 포자 생산 방법)

  • BACKGROUND: Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. This investigated to determine the optimal culture conditions in ceramic balls for maximal sporulation of entomopathogenic fungi Beauveria bassiana M130 by use rice bran extract.METHODS AND RESULTS: METHODS AND RESULTS: A culture of entomopathogenic fungi for 12day on rice bran extract(1:8, w/v) incubated in ceramic matrix at 28℃. Natural zeolite ceramic ball was high production of 4.2×108 conidial/mL. The culture condition optimized initial pH, temperature, rice bran extract concentration, adhesives substance and concentration of NaCl, respectively. The high production of spore optimal conditions were temperature 28℃, initial pH 3, rice bran extract 3 mL, starch 33 g, 5 % NaCl and sopre suspension 7 mL, respectively.CONCLUSION: This study was carried out for the mass production of entomopathogenic fungi conidia recover rate 65% in matrix of natural zeolite ceramic ball, and to develop ingredient-used formulation of Beauveria bassiana M130 conidia for biological control agents.

Application and utilization of marker assisted selection for biotic stress resistance in hybrid rice (Oryza sativa L.)

  • Development of disease resistant plant is one of the important objectives in rice breeding programs because biotic stresses can adversely affect rice growth and yield losses. This study was conducted to identify lines with multiple-resistance genes to biotic stress among 173 hybrid rice breeding lines and germplasms using DNA-based markers. Our results showed that one hybrid rice line [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66)] possessed 5 bacterial blight resistance genes (Xa4, xa5, Xa7, Xa13 and Xa21) while two hybrid rice lines [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66) and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessed 3 bacterial blight resistance genes (Xa4, Xa7 and Xa21, and Xa3, Xa4 and xa5). Molecular survey on rice blast disease revealed that most of these lines had two different resistant genes. Only 11 lines possessed Pib, Pi-5, and Pi-ta. In addition, we further surveyed the distribution of insect resistant genes, such as Bph1, Bph18(t), and Wbph. Three hybrid breeding lines [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66), IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), and 7292s (IR75589-31-27-8-33S(S1) /IR102758B)] contained all three resistance genes. Finally, we obtained four hybrid rice breeding lines and germplasms [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), Damm-Noeub Khmau, 7290s, and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessing six-gene combination. They are expected to provide higher level of multiple resistance to biotic stress. This study is important for genotyping hybrid rice with resistance to diverse diseases and pests. Results obtained in this study suggest that identification of pyramided resistance genes is very important for screening hybrid rice breeding lines and germplasms accurately for disease and pest resistance. We will expand their cultivation safely through bioassays against diseases, pests, and disaster in its main export countries.

  • Greenhouse whitefly, $Trialeurodes$ $Vaporariorum$, is the most important insect pest in environmental friendly tomato production. Natural enemies, $Nesidiocoris$ $tenuis$ and environmental friendly agricultural materials (EFAM) are major control agents in Korea. The aim of the present work is to evaluate release time of $N.$ $tenuis$ in controlling of $T.$ $vaporariorum$ and to select environmental friendly agricultural materials (EFAM) to suppress both $T.$ $vaporariorum$ and $N.$ $tenuis$ which if the density were high causing necrotic rings. Six EFAM selected to control of $Trialeurodes$ $Vaporariorum$ in three times spray with seven days intervals. Three EFAM showed high mortality 67.7-70.5% in spray after release plot but low in release after spray plot. It should be careful to spray with $N.$ $tenuis$. When $N.$ $tenuis$ were early release at 7 days after transplanting which before occurring and establishment of greenhouse whitefly, it could suppress the density. Extracts of bead tree + matrin, matrin 1, Chrysanthemum showed high mortality on $T.$ $vaporariorum$ adults and $N.$ $tenuis$. The results suggested that this three EFAM could be control agents of the damage of necrotic rings from $N.$ $tenuis$.

Screening of Resistance Genes Linked to Brown Planthopper Using STS Marker in Aromatic Rice Germplasm

  • Brown planthopper (BPH) is a serious insect pest of rice crop throughout rice growing countries, and yield loss due to its infection can be up to 60%. This study aimed to evaluate efficiency of molecular markers for screening BPH resistance accessions among 86 aromatic rice germplasm Eighty-six accessions of aromatic rice germplasm included two accessions of Tongil type (bred in Korea), 28 accessions of japonica type and 56 accessions of indica type. We applied eight STS markers (pBPH9, pBPH19, pBPH20, pBPH21, AJ09-b, RG457L, RG457B, and 7312.T4A) which were linked to four of BPH resistance genes, Bph1, Bph13(t), Bph10, and Bph18(t) respectively. One japonica type accession, 415XIr352, and six indica type accessions possessed one or four positive bands when tested with four STS markers linked to Bph1 gene. One indica type aromatic rice, Basmati9-93, showed the target bands linked to the Bph10 gene. The other accessions did not show same fragments as the respective resistant lines. Bph13(t) is the most widely introduced resistance gene and only one accession showed positive bands implying that this accession might harbor Bph10 and Bph18(t) genes. Three aromatic accessions, Domsiah, Khao Dawk Mali 105 and 415XIr352 showed gene pyramiding of Bph1 and Bph13(t). Two indica aromatic rice, Ds 20 and Basmati 9-93, possessed at least two BPH resistance genes, Bph1, Bph18(t) and Bph13(t), Bph18(t), respectively. These results indicates that aromatic rice germplasm have narrow diversities of BPR resistance genes.

An Empirical Model for the Prediction of the Onset of Upward-Movement of Overwintered Caccopsylla pyricola (Homoptera: Psyllidae) in Pear Orchards (배과원에서 꼬마배나무이 월동성충의 수상 이동시기 예측 모형)

  • Pear psylla, Caccopsylla pyricola (Homoptera: Psyllidae), is a serious insect pest in pear orchards. C. pyricola overwinters as adults under rough bark scales of pear trees. When the weather warms up in the spring, the overwintered adults become active, climb up to the tree branches, and inhabit on fruit twigs to lay eggs. This study was conducted to develop a forecasting model for the onset of upward-movement of overwintered C. pyricola adults to control them by timely spraying of petroleum oil. The adult population densities were observed under rough barks (B) and on fruit twigs (T) of pear trees. Relative upward-movement rates (R) were calculated as T/(B+T). Low threshold temperatures for the activation of overwintered C. pyricola adults were selected arbitrarily from 5 to $9^{\circ}C$ at a $1^{\circ}C$ interval. Then, the days (D) when daily maximum air temperatures were above each low threshold temperature were counted from 1 February until to the dates with R $\geq$ 0.8. The same methods were applied for the prediction of the first observation of eggs. The variation of coefficients (CV) for the mean Des were lowest with the low threshold temperature of $6^{\circ}C$. At this selected threshold temperature, the upward movement of C. pyricola adults occurred with 12 D and they started laying eggs with 25 D. In the field validation, the model outputs with the $6^{\circ}C$ threshold temperature reasonably well explained the observed data in Suwon and Cheonan in 2002. Practical usages of the model were also discussed.

Amino acid, fatty acid, and carbohydrate metabolomic profiles with ginsenoside-induced insecticidal efficacy against Ostrinia furnacalis (Guenee)

  • Background: Previous studies have shown the insecticidal efficacy of ginsenosides. In the present study, we aimed to investigate the metabolic mechanism related to the inhibitory effect of panaxadiol saponins (PDSs) against the Asian corn borer Ostrinia furnacalis (Guenee). Methods: Third instar larvae of O. furnacalis were fed normal diets with different concentrations of PDSs for 4 days. The consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food were recorded. A targeted gas chromatographye-mass spectrometry assay was performed to detect the profiles of amino acids, fatty acids, and carbohydrates in larvae of O. furnacalis. In addition, the activity of detoxification-related enzymes was determined. Results and Conclusions: PDSs decreased the consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food in the 3rd instar larvae of O. furnacalis in a dose-dependent manner. PDSs decreased 15 free amino acids, 16 free fatty acids, and 5 carbohydrates and increased the levels of palmitoleic acid, palmitic acid, and 9-octadecenoic acid in the 3rd instar larvae. The activity of detoxification-related enzymes, such as acetylcholinesterase, glutathione S-transferase, cytochrome P450, carboxylesterase, trehalase, acid phosphatase, and alkaline phosphatase, was reduced in a dose-dependent manner in the 3rd instar larvae exposed to PDSs. These data confirmed the inhibitory effect of PDSs against growth, food utilization, and detoxification in the 3rd instar larvae of O. furnacalis and the potential for using PDSs as an efficient tool for insect pest management for O. furnacalis larvae.

Seasonal Earthworm Casting Activity on Korean Golf Courses (골프장에서 지렁이 분변토 발생의 계절적 변화)

  • Earthworm cast occurred in surface of turfgrass in golf courses which affect turfgrass maintenance and golf play. In this study several survey of seasonal fluctuation of earthworm cast in different golf courses (Anseong, Gapyeong, Gunpo and Yongin in Gyeonggi province, and Geumjeong in Busan) was done. A number of earthworm, soil temperature and moisture of detection site of earthworm cast in each golf course were also done. Cast occurred in different golf courses are found mostly from April to November on turf surface and the cast number varied in different month as well as in golf courses. In the same golf courses, a number of detected cast was difference from individual sites. A number of cast and earthworm was correlated. Also, in higher soil moisture showed the higher cast density in different golf courses. Soil temperature and moisture of detecting cast ranging from $2.1^{\circ}C$ to $33.1^{\circ}C$ and 4.9 to 44.1%, respectively. In case of cast, soil temperature level lies in between $10-15^{\circ}C$ where the highest soil moisture was 25%.

Control System of Whitefly, Trialeuodes vaporariorum, in Cucumber by the Alternate Application of Insecticides within Each Conventional Group (오이에서 살충제 계열내 교호처리에 의한 온실가루이 방제 체계)

  • In order to establish a whitefly control system using conventional groups of insecticide(carbamate, organophosphorus and pyrethroid insecticides), three alternative application methods were designed on the medium growth stage of cucumber. To discriminate the effectiveness of these sequences observed were the residual activity of insecticides, frequency of insecticide applications, residue of insecticides in cucumber leaves, development of insecticide resistance in whitefly, and yield of fruits. Spraying furathiocarb, a carbamate insecticide, was very effective in reducing the frequency of application for the control of white flies. The effectiveness of furathiocarb was enhanced by the potentiation process to carbofuran, the long residual activity, and the lower development rate of insecticide resistance. Methion, an organophosphorus insecticide, did not show resistance development after successive use but resulted in short residual activity. However, other organophosphates, profenofos and phenthoate, lost their activity by the resistance development. Decreasing activity was common to pyrethroids, deltamethrin and zetacypermethrin due to resistance. From these results it could be drawn a conclusion that furathiocarb, a carbamate insecticide, was the most desirable among conventional insecticide groups for the management of greenhouse whitefly population on the cucumber. To prevent an outbreak of the insect pest by various cause, it was recommended to choose acetamiprid, a nicotinoid, which showed very good control efficacy to the resistance insects to conventional insecticides.
