효모의 배양시기에 따른 인산화합물의 합성 및 효흡능에 미치는 탄수원의 영향 (Effect of the Carbon sources on the Synthesis of phosphate compounds and Respiratory activity of Yeast (saccharomyces uvarm) during growth phases)

  • The growth rate of yeast population (Saccharomyces uvarum) cultivated in the Knopp's modified medium (plus various carbon sources) appeared the highest value when the Knopp's minimal medium was treated to 1.5% with disaccharide such as maltose and sucrose. Also the treatment of lactose and raffinose resulted in polulation growth as to the population size in case of maltose and sucrose. However, the gorwth of yeast was not occurred at all when a polysaccharide, such as inulin, was added as carbon source. The growth from of yeast population in Knopp's modified medium are characterized by the fact that log phase continued 100hrs after inoculation and that stationary state phase appeared in general 250hrs after inoculation. Applying the various carbon sources to respiration substrate for yeast cell, the respiration rate of yeast showed the highest value in treatment of maltose and followed in order of raffinose, lactose, glucose, and sucrose. Determined the amount of poly-phosphate and turn over pathway of poly-phosphate according to culture phase of yeast, it is revealed that the yeast synthesized 3 types of poly phosphate (poly-P A,B, and C) and postulated that turn over pathway of poly-phosphate as follows ; Inorganic phosphate is converted into each kind of polyphosphates, and then one part of poly-P-C is converted into poly-P-B, the rest poly-p-C and poly-P-B are converted into poly-P-A. The synthesized poly-phosphate is considered to have a role as energy pool utilizing to synthesis of cellular organic materials. Of the 13 carbon sources used in this experiment, the useful carbon sources for biosynthesis of poly-phosphate and cellular organic materials are confirmed as disaccharide (maltose and sucrose) as well as glucose. Protein synthesis in yeast cell showed the two peaks on 6th and 8th day after inoculation ; nucleic acid on 2nd day (48hrs), carbohydrates on 2nd day (48hrs), and phospholipid on 2nd and 8th day after inoculation, respectively.

의왕시내 BTEX 오염 부지에서의 자연 정화법 이용 적합성 고찰 (Assessment of Monitored Natural Attenuation as Remediation Approach for a BTEX Contaminated Site in Uiwang City)

  • In the United States (U.S.), the monitored natural attenuation (MNA) approach has been used as an alternative remedial option for organic and inorganic compounds retained in soil and dissolved in groundwater. The U.S. Environmental Protection Agency (EPA) defines the MNA as“in-situ naturally-occurring processes include biodegradation, diffusion, dilution, sorption, volatilization, and/or chemical and biochemical stabilization of contaminants and reduce contaminant toxicity, mobility or volume to the levels that are protective of human health and the environment”. The Department of Soil Environment. National Institute Environmental Research (NIER) is in the process for demonstrating the MNA approach as a potential remedial option for the BTEX contaminated site in Uiwang City. The project is charactering the research site in terms of the nature and extend of contamination, biological degradation rate, and geochemical and hydrological properties. The microbial-degradation rate and effectiveness of nutrient and redox supplements will be determined through laboratory batch and column tests. The geochemical process will be monitored for determining the concentration changes of chemical species involved in the electron transfer processes that include methanogenesis, sulfate and iron reduction, denitrification, and aerobic respiration. Through field works, critical soil and hydrogeologic parameters will be acquired to simulate the effects of dispersion, advection, sorption, and biodegradation on the fate and transport of the dissolved-phase BTEX plume using Bioplume III model. The objectives of this multi-years research project are (1) to evaluate the MNA approach using the BTEX contaminated site in Uiwang City, (2) to establish a standard protocol for future application of the approach, (3) to investigate applicability of the passive approach as a secondary treatment remedy after active treatments. In this presentation, the overall picture and philosophy behind the MNA approach will be reviewed. Detailed discussions of the site characterization/monitoring plans and risk-based decision-making processes for the demonstration site will be included.

동일비료 24년 연용이 콩의 수량(收量) 및 품질(品質)에 미치는 영향(影響) (The Effect of Long-Term Fertilization on the Yield and Quality of Soybean)

  • 본 시험은 석회 및 규산의 시용이 콩의 수량 및 종실에 미치는 영향을 조사하기 위하여 수행하였다. 콩의 수량은 NPK+석회 $1.5Mg\;ha^{-1}$ 처리가 가장 높았다. 탄수화물 함량은 무비에서 가장 높았고 단백질 함량은 NPK+석회 $1.5Mg\;ha^{-1}$ 및 NPK+규산 $1.5Mg\;ha^{-1}$ 처리구에서 높았으며 지방 및 회분함량은 NPK+규산 $1.5Mg\;ha^{-1}$ 처리구에서 높았다. 무기성분 함량은 처리간에 차이가 없었다. $P_2O_5$/T-N과 탄수화물 사이에는 $-0.99^{**}$, $P_2O_5$/T-N과 지방 사이에는 $-0.94^{**}$의 상관이 있었다. 그리고 Mn과 탄수화물사이에는 $0.95^*$의 상관이 있었다. 석회의 시용은 콩의 수량에, 규산의 시용은 단백질 함량증가에 효과적이었다.

  • 토양(土壤)에 축적(蓄積)되어 있는 난용성(難溶性) 인산염(燐酸鹽)을 이용(利用)하고자 인산염(燐酸鹽) 가용화능(可溶化能)을 가지고 있는 Pseudomonas putida, Penicillium속(屬) 및 Aspergillus niger를 우리 나라 주요(主要) 경작지(耕作地) 토양(土壤)에서 분리(分離)하여 이들 균(菌)의 용해능(溶解能)을 측정(測定)하였다. 인산염(燐酸鹽) 용해량(溶解量)은 배지내(培地內)의 pH 저하(低下)와 밀접(密接)한 관계(關係)를 가지고 있었다. Pseudomonas putida, Penicillium속(屬)에 비(比)해 Aspergillus niger의 인산염(燐酸鹽) 가용화능(可溶化能)이 높았다. Aspergillus niger에 의한 인산염(燐酸鹽) 종류별(種類別) 최대(最大) 가용화량(可溶化量)은 질소원(窒素源)이 $KNO_3$일 때는 aluminium phosphate 776ppm, hydroxyapatite 665ppm, tai-calcium phosphate 593ppm이었으며 인광석(燐鑛石)은 질소원(窒素源)이 $(NH_4)_2SO_4$일 때 411ppm이었다.

Monoculture and Mixture Effects of Green Manure Crops on Soil Quality, Weed Suppression and Organic Red-pepper Production

  • Organic farming is rapidly expanding worldwide. Crop growth in organic systems greatly depends on the functions performed by soil microbes, and nutrient supply weed suppression by green manure crops input. Four red-pepper production systems were compared: 1) bare ground (conventional system); 2) hairy vetch monoculture; 3) rye monoculture; and 4) hairy vetch-rye mixture. Soil inorganic N reached the peak at 30 DAI and hairy vetch monoculture was the highest ($192mg\;kg^{-1}$) and soil total carbon was fluctuated sporadically during the experiment. Carbohydrate and phenolic compounds in soil kept significantly higher in green manure crops systems from 10 DBI to 30 DAI, however the level was the maximum at 10 DBI (carbohydrate) and 30 DAI (phenolic comounds). Incorporation of green manure crops residue enhanced soil microbial biomass C and N throughout the growing season except that MBN in rye was reduced after incorporation. Green manure crops systems suppressed weed occurrence and, in particular, it was prominent in rye monoculture. Mineral elements composition and production in red-pepper fruits were markedly decreased in green manure crops systems although hairy vetch monoculture has come close to bare ground (NPK-applied). Therefore, it was suggested that higher biomass production should be performed not only to improve soil quality and suppress weeds but to yield suitable red-pepper fruits in green manure crops-based organic farming.

양식장 배출물 발효물의 어류 사료 첨가에 따른 넙치, Paralichthys olivaceus의 혈액학적 변동에 미치는 영향 (Effects of oral administration with fermented product from sewage in land-based seawater fish farm on haematological factors of olive flounder, Paralichthys olivaceus)

  • 본 연구는 우리나라 주요 양식어류인 넙치, Paralichthys olivaceus를 대상으로 육상수조식 양식장 배출물 발효물의 어류 사료 첨가에 따른 혈액학적 변동에 미치는 영향을 검토하였다. 젖산균 및 효모를 사용하여 발효한 양식장 배출물의 이화학적 분석을 실시하였고, 발효물을 0.1, 0.5, 1.0 및 5.0% 농도로 8주간 투여하였다. 투여 4주째 및 8주째에 각 투여 구간별로 혈액학적 항목을 중심으로 그 차이를 확인하였다. 배출물 발효농도별 유의한 차이는 나타나지 않았지만, 분석시기에 따른 적혈구수, 혈색소 농도 및 혈색소 지수가 증가하는 경향을 나타내었으나, 주요 혈청 무기 및 유기성분은 배출 발효물 농도별로 유의한 차이는 나타나지 않았다. 또한 혈청 효소학적 조사에서는 조사시기별로 변동은 나타났지만, 동일 조사시기에서 각 구간별 유의한 차이는 나타나지 않았다. 따라서 이러한 육상 양식장 유래의 배출물 발효산물에 대한 다양한 방면의 연구가 진행되어야겠다.

Dielectric properties and microstructures of (CaxSr1-x)ZrO3 ceramics

  • The effects of Ca/Sr ratio and the sintering temperature on the properties of $(Ca_xSr_{(1-x)})ZrO_3$ (CSZ) ceramics were investigated in this study. CSZ ceramics were prepared using solid-state reaction process, which were sintered in air at temperatures ranging from $1350^{\circ}C$ to $1450^{\circ}C$. Their structures were characterized by X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), and Transmission Electron Microscopy (TEM). The change in Ca/Sr ratio significantly affected the crystalline phase and the dielectric properties of the $(Ca_xSr_{(1-x)})ZrO_3$ ceramics. The secondary phase, $Ca_{0.15}Zr_{0.85}O_{1.85}$, was observed and increased correspondingly with the rising of sintering temperatures. In order to understand the effects of secondary phase on the dielectric properties of CSZ ceramics, the $Ca_{0.15}Zr_{0.85}O_{1.85}$ phase was prepared individually using solidstate method. The $Ca_{0.15}Zr_{0.85}O_{1.85}$ ceramics sintered at $1500^{\circ}C$ for 2 hours possessed a dielectric constant (${\varepsilon}_r$) of 21.7, a dielectric loss ($tan{\delta}$) of $49.510^{-4}$ and an Insulation Resistance (IR) of $2.1{\times}10^{10}{\Omega}$. The ($Ca_{0.7}Sr_{0.3})ZrO_3$ ceramics exhibited the best dielectric properties, with a permittivity of 29, a dielectric loss ($tan{\delta}$) of $2.7{\times}10^{-4}$, and an Insulation Resistance (IR) of $2.6{\times}10^{12}{\Omega}$.

  • The purpose of this review is to summarize studies that investigate blood and the main components of fetal bovine serum (FBS) in vertebrates, including major livestock, and review the current research on commercializing cultured meat. Detailed research on FBS is still lacking; however, some studies have shown that FBS consists of proteins, carbohydrates, growth factors, cytokines, fats, vitamins, minerals, hormones, non-protein nitrogen, and inorganic compounds. However, there are few studies on how the composition of FBS differs from blood or serum composition in adult animals, which is probably one of the main reasons for not successfully replacing FBS. Moreover, recent studies on the development of FBS replacers and serum-free media have shown that it is difficult to conclude whether FBS has been completely replaced or serum-free media have been developed successfully. Our review of the industrialization of cultured meat reveals that many basic studies on the development of cultured meat have been conducted, but it is assumed that the study to reduce or replace ingredients derived from fetuses such as FBS has not yet been actively developed. Therefore, developing inexpensive and edible media is necessary for the successful industrialization of cultured meat.

전기자동차의 난방용 면상 후막히터의 제조방법과 성능에 관한 실험적 연구 (An Experimental Study on the Manufacturing Method and Performance of Planar Thick Film Heaters for Electric Vehicle Heating)

  • Currently used heating elements are metal and non-metal heating elements, including various types of heaters, and resistance line heating elements have a problem of decreasing thermal efficiency over time, so to solve this problem, a planar heating element using high-purity carbon materials and oxidation-resistant inorganic compounds was applied. Through the manufacture of planar heating elements using CNT, ruthenium composite materials, and ruthenium oxide, physicochemical performance and capacity were increased, and instantaneous responsiveness was increased. Through thick film technology applicable to various base bodies, fine patterns were formed by the screening method in consideration of the fact that the performance of the heat source depends on the viscosity and pattern shape. The heating element was manufactured by thick film printing technology by mixing ruthenium oxide, CNT, Ag, etc. The characteristics of each paste were analyzed through viscosity measurement, and STS 430 was used as a base. Surface temperature and efficiency were measured by testing heaters manufactured for small wind tunnels and real-vehicle experiments. The surface temperature decreased as the air volume increased, and the optimal system boundary was found to be about 200 mm. Among the currently used heating elements, this paper manufactured a planar heating element using thick film technology to find out the relationship between air volume and temperature, and to study the surface temperature.

시화호 산화-환원 환경하의 용존 유, 무기 화합물의 생지화학적 연구 (Biogeochemical Study of Dissolved Organic and Inorganic Compounds under Oxic/Anoxic Environment in Lake Shihwa)

  • 인공적으로 조성된 시화호는 현재 부분적인 해수혼입을 제외하고는 해수의 순환이 차단된 폐쇄 환경을 이루고 있으며 끝막이 공사 후 6 m 이심에 갇혀있는 잔존 저층해수로 인하여 수직적으로 강력한 염분 의존형 밀도 성층에 의한 2층 구조(two-layered system)를 보이고있다. 이러한 물리적 조건에서 시화호 주변의 6개 주요 하천으로부터 유입되는 막대한 양의 유기물질 및 암모니아염은 시화호의 저산소 및 무산소 환경을 가속화시키고 있으며 이에 따라 시화호의 생지화학적 환경은 흑해(Black Sea)와 같이 전 계절을 통하여 6 m 부근 심도의 밀도경사면을 상하로 뚜렷한 산화와 환원 환경으로 나뉘어지고 있다. 본 연구는 이러한 인위적 환경에서 나타나는 여러 유, 무기 화합물과 원소의 생지화학적 분포특성과 종분화 그리고 과정에 대하여 산화-환원 경계면을 중심으로 다루고자 하였다. 연구 결과 용존산소는 유기물이 축적되어 있는 밀도 경계면 이심의 저층 잔존 해수층에서 고갈되어 있으며 이에 띠라 약 1억톤에 달하는 저층 해수수괴가 환원환경을 이루고있다. 질산염과 아질산염은 산화 환경인 저염의 표층에서 높게 나타났고 환원 환경인 고염의 저층에서는 급격히 감소하여 나타났다. 반면 암모니아염은 저층수괴에서 75에서 360 ${\mu}M$에 이르는 매우 높은 농도를 보이는데 이는 저층으로 침강 유입된 유지불의 혐기성 분해에 따른 ammonification이 주된 원인으로 보인다. 1996년 4월부터 8월까지 약 3억톤의 시화호 내 표층수가 연안으로 방류되고 상당량의 외해수 혼입에도 불구하고 시화호 내 저층수의 환원환경이 유지 또는 가속화되는 주된 원인은 저층의 잔존 해수수괴에 trap 된 침강 유기물의 산소 소비 속도와 제한적으로 공급되는 산소공급이 불균형을 이루고 있기 때문이다. 시화호의 수질 환경면에서 볼 때 현재와 같이 2층구조의 염분도의존형의 밀도 성층이 유지되는 한 사화호 수질의 개선 전망은 밝지 못한 것으로 판단된다.
