Phylogenetic Relationships between the Genus Inonotus and its Related Genera Based on the Nucleotide Sequences of Internal Transcribed Spacers (ITS 염기서열에 기초한 차가버섯과 근연속간 유연관계분석)

  • In this study the ITS1, ITS2 and 5.8S ribosomal DNA sequences from 29 strains of the Genus Inonotus and its related genera were compared with 31 strains obtained from GenBank database. Using the neighbor-joining (NJ) method and most parsimonious analysis the phylogenetic tree was constructed. The hymenochaetales formed no monophyletic group and several non-hymenochaetales appeared as intermingled with the Hymenochaetales. Strains 6, 46, 49, 50, 53, 55 showed no certain affinities within the Hymenochaetales, whereas Inonotus sp. (51) was closely related to Phellinus baumii, and Inonotus sp. (52), and Inonotus glomeratus (10) was related to Phellinus linteus, and Fomes fomentarius (30) was related to Ganoderma lucidum. Inonotus sp. and Phellinus sp. formed no monophyletic groups and a subdivision in the following genera is accepted: Inonotus sp. Phellinus baumii, Phellinus linteus, Phellinus igniarius, Phellinus pini, Hericium erinaceum, Ganoderma lucidum and Sparassis sp. were confirmed and separated genera. The taxonomic status of Inonotus remained uncertain. Eight new combinations are proposed.

Antioxidant and Anticancer Effects of Fermentation Vinegars with Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus (상황버섯, 차가버섯, 느타리버섯 발효식초의 항산화와 항암활성 효과)

  • The antioxidant and anticancer activities of Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus according to extraction methods were measured. The contents of polyphenolic compounds were higher in fermented extracts than those in water extracts. The contents were 192.4mg/100 $m{\ell}$ for Inonotus obliquus vinegar, 186.9 mg/100 $m{\ell}$ for Phellinus linteus vinegar, and 156.5 mg/100 $m{\ell}$ for Pleurotus ostreatus vinegar, respectively. EDA(electron donating ability) in Phellinus linteus that was highest among mushrooms was 83.9% in the water extract and 96.8% in the fermented extract. The growth of stomach cancer cells, SNU-719 was inhibited 62.4%, 65.5%, and 53.0% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, respectively and also the growth of liver cancer cells, Hep3B was inhibited 67.1%, 68.3%, and 57.6% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, while the growth of normal cell, DC2.4 was not affected. Even though Phellinus linteus and Inonotus obliquus showed higher inhibition in cancer cell growth, Pleurotus ostreatus can be efficiently used for antioxidant and anticancer activities due to their cheap price in the market.

Analysis of Biological Activities of Medicinal Mushrooms (약용버섯의 생리활성 분석)

  • This study was carried out to observe the antioxidative activity, fibrinolytic activity, nitrite savenging ability and adenosine content of three medicinal mushrooms. Among water and ethanol extract of each mushroom, water extract of Inonotus obliquus (Czech) showed the highest antioxidative activity $(57.1{\mu}g/ml)$, nitrite scavenging ability (52.04%), fibrinolytic activity (86.8%) and adenosin content $(94.3{\mu}g/g)$, whereas nitrite scavenging ability of ethanol extract of Phellinus linteus showed higher than that of water extract. Apart from the above statements, water was effective than ethanol as extracting solvent in general. These results suggest that Inonotus obliquus (Czech) showed higher activities such as antioxidative activity, nitrite scavenging ability, fibrinolytic activity and adenosin content and water was effective as solvent except for nitrite scavenging ability.

Phylogeny of Phellinus and Related Genera Inferred from Combined Data of ITS and Mitochondrial SSU rDNA Sequences

  • To elucidate phylogenetic relationships of Phellinus and its related genera, nuclear internal transcribed spacer and mitochondrial small subunit ribosomal DNA sequences from 65 strains were determined and compared. The combined dataset of two sequences increased informative characters and led to the production of trees with higher levels of resolution. Phylogenetic analysis of the combined dataset revealed thirteen evolutionary lineages and several unresolved species that were together subdivided into two large clusters consisting of oligonucleate species and binucleate species. These results coincided with previous cytological, morphological, and molecular studies. It is newly recognized that the Phellinus linteus complex forms a sister clade to Inonotus, and that Fulvifomes is somehow related to Inocutis. The Phellinus linteus complex of dimitic perennial taxa made an independent clade from Inonotus and suggested that hyphal miticity and fruitbody permanence had enough phylogenetic significance to keep the complex within the traditional genus Phellinus. Taxa lacking setae were clustered into Fulvifomes, Phylloporia, Inocutis, and Fomitiporia, and the first three were closely related sister groups, but Fomitiporia was a genus distantly related to them. Several taxa with branched setae were shown among distantly related genera. Molecular evidence indicated that the ancestral nuclear type could be a binucleate feature, and that there might be parallel gains of branched setae and parallel losses of setae in the Hymenochaetales.

Molecular Identification of Two Strains of Phellinus sp. by Internal Transcribed Spacer Sequence Analysis

  • Two species of cultivated Phellinus sp. were identified as P. baumii by internal transcribed spacer (ITS) sequence analysis. The fruit bodies of the examined strains were similar to those of naturally occurring strains, having a bracket-like form, yellow-to-orange color, and poroid hymenial surfaces. The DNA sequences of ITS region of both strains showed a homology of 99% with ITS1 to ITS2 sequences of P. (Inonotus) baumii strain PB0806.

  • The Aphyllophorales is a large order containing about 2,000 known species. Many of these are the bracket and coral fungi. The vast majority of these fungi are saprophytic on the plant debris. Many species are significant in decomposing plant remains, as they are able to digest cellulose or lignin that occurs in plant cell walls. Many of these fungi have been involved in everyday human affairs. A few were used medicinally by the Greeks and Romans as a remedy for many complaints, including colic, fractured limbs and bruises. Other bracket fungi have been used as curry combs for horses, as snuff, as razor strops and as a source of dye for clothing. The texture of the basidiocarp may be similar to that of cork, wood, leather, paper, or cartilage. Unlike the basidiocarps of the Order Agaricales, the basidiocarps of the Aphyllophorales are not fleshly and moist. Division of the members of the Aphyllophorales into genera was originally made on the basis of gross morphology of the basidiocarp and hymenium and Donk(1964) recognizes 22 families in this order. The species and genus whose typical in Aphylloporales were listed in Table. with related information. The ITS region sequence of some genus were found by BLAST search. Sequences retrieved from GenBank were visually aligned by the program CLUSTAL G. As a result, the medicinal mushroom was separated in four groups. In this multiple alignment, the sequence analysis among Fomes group, Inonotus group and Phellinus group showed high genetic similarity except Hericium group and Sparassis group.

Artificial Cultivation Characteristics and Bioactive Effects of Novel Tropicoporus linteus (Syn. Phellinus linteus) Strains HN00K9 and HN6036 in Korea

  • Phellinus strains were collected from different areas in Korea. Of them, the fast mycelial growing strains were artificially cultivated on the oak logs to produce fruiting body. The varieties, Phellinus linteus ASI26099 (Korea Sanghwang) and P. baumii PBJS (Jangsoo Sanghwang) were grown under the same conditions as controls. Their cultivating characteristics including mycelial colonization, pinhead formation, and fruiting body formation rate were investigated on the logs. Basidiocarps of Phellinus strains HN00K9, HN6036, and ASI26099 were concentrically zonate and shallowly sulcate, and dark chestnut showing typical characteristics of Tropicoporus linteus (synonyum: P. linteus, Inonotus linteus, polyporus linteus), which is distinguishably different to PBJS. HN00K9 showed the highest yield of fruiting body among the mushroom strains. The β-glucan content in fruiting bodies of HN00K9 was 20% higher than those of other strains. Bioactive effects of polysaccharide samples from fruiting bodies of Phellinus strains, HN00K9, HN6036, ASI26099, and PBJS were assessed on cell viability and cytokine (IL-6 and TNF-α) inhibition and finally on anticancer to different human cancer cells.

The Effects of Extracts Mixture Drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on Hematopoietic Stem Cells and Lymphocyte Subset of Blood in Human (차가버섯, 상황버섯 및 영지버섯 복합추출물 복용이 인체의 혈중 조혈모세포와 면역세포에 미치는 영향)

  • This study was performed to investigate the effect of extract mixture(IPGE) drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on hematopoietic stem cells and lymphocyte subset[lymphocyte, $CD4^+T$ cell, $CD8^+T$ cell, Natural Killer(NK) cells] of blood in 37 participants who were healthy and about $40{\sim}70$ years old. They were divided into two groups; extract mixture drink administration group(n=27) and placebo administration group(n=12). They were given the test drink daily for 4 weeks. Blood was obtained from the subjects every two week in the beginning of administration day to evaluate the $CD34^+$ hematopoietic stem cells and immune cells. As results, $CD34^+$ hematopoietic stem cells were significantly increased after taking IPGE drink for 4 weeks compared to that before taking the drink (p<0.001). There was no significant changes in number of lymphocytes, $CD4^+T$ cells, $CD8^+T$ cells, NK cells and in the ratio of $CD4^+/CD8^+$ cell after taking the test drink. From these results, it was suggested that IPGE have a good health effect by promoting the proliferation of the hematopoietic stem cells.
