Neuroprotective Effects of Berberine in Neurodegeneration Model Rats Induced by Ibotenic Acid

  • Berberine, an isoquinoline alkaloid found in Coptidis Rhizoma(goldenthread) extract, has multiple pharmacological effects such as anti-inflammatory, antimicrobial and anti-ischemic effects. In the present study, we examined the effects of berberine on neuronal survival and differentiation in a hippocampal precursor cell line and in the memory deficient rat model. Berberine increased in a dose dependent manner the survival of hippocampal precursor cells as well as differentiated cells. In addition, berberine promoted neuronal differentiation of hippocampal precursor cells. In the memory deficient rat model induced by stereotaxic injection of ibotenic acid into entorhinal cortex(Ibo model), hippocampal cells were increased about 2.7 fold in the pyramidal layer of CA1 region and about 2 fold in the dentate gyrus by administration of berberine after 2 weeks of ibotenic acid injection. Furthermore, neuronal cells immunoreactive to calbindin were increased in the hippocampus and entorhinal cortex area by administration of berberine. Taken together, these results suggest that berberine has neuroprotective effect in the Ibo model rat brain by promoting the neuronal survival and differentiation.

The Development of Tolerance to and Dependence on Morphine are Reduced by Co-administration of Nalbuphine in Rat (Nalbuphine의 병용투여에 의한 morphine의 내성 및 의존성 형성 저하효과)

  • Morphine has been used widely on the treatment of many types of chronic pain. However the development of tolerance to morphine by repeat application is a major problem in pain therapy. The purpose of the present study was to investigate whether combined administration of nalbuphine with morphine affects the development of tolerance to and dependence on morphine. We hypothesize that the use of nalbuphine, k-agonist may prove to be useful adjunct therapy to prevent morphine-induced undesirable effects in the management of some forms of chronic pain. Morphine (10 mg/kg) was injected to rats intraperitoneally for 5 days. The variable dose of nalbuphine (0.1, 1.0 and 5.0 mg/kg) was administered (i.p.) in combination with morphine injection. The development of tolerance to morphine was assessed by measuring the antinociceptive effect with the Randall-Selitto apparatus. The development of dependence on morphine was determined by the scoring the precipitated withdrawal signs for 20 min after injection of naloxone (10 mg/kg, i.p.). Nalbuphine did not attenuate antinociceptive effect of morphine in rats. Interestingly, combined administration of morphine with nalbuphine (100:1) significantly attenuated the development of morphine tolerance and dependence. These results suggest that the co-administration of nalbuphine with morphine in chronic morphine treatment can be one of therapies to reduce the development of dependence on morphine.

Risperdal Sachet and Oral Lorazepam versus Intramuscular Haloperidol and Lorazepam Injection for Acute Psychotic Symptom in the Elderly Patients with Organic Mental Disorder (노인 기질성 정신장애 환자의 급성 정신증상에 대한 리스페달 사셰와 할로페리돌 근주의 효과 비교)

  • Objectives : The purpose of present study was to investigate the effect, safety and tolerability of risperdal sachet(oral solution) with lorazepam tablet versus intramuscular haloperidol and lorazepam injection for management of acute psychotic symptom in the elderly with organic mental disorder. Methods : Total 37 patients who have dementia, medical or physical diseases, associated with acute psychotic symptom were randomly assigned to oral treatment with 1mg of risperdal sachet(oral solution) plus 1mg of lorazepam(N=17) or to intramuscular treatment with 2.5mg of haloperidol plus 2mg of lorazepam (N=20). The change of CGI scores was used for the evaluation of efficacy. Results : Mean score improvements at 15, 30, 60, and 120 minutes after treatment were statistically significant at each time point in both groups(p<0.001) and were similar in both groups(p=0.189). Conclusion : A single oral dose of risperdal sachet(oral solution) plus lorazepam was as effective and tolerable as parenterally administered haloperidol plus lorazepam for the rapid control of acute psychotic symptom in the elderly with organic mental disorder.

Effect of Methylprednisolone Sodium Succinate on Innate Immune Function of Canine Peripheral Blood Phagocytes

  • Glucocorticoids (GCs) are the most widely used immunosuppressive agents, but animals treated with GCs may experience deleterious side effects which limit their use in many clinical conditions. In the present study, we examined whether methylprednisolone sodium succinate (MPSS), a glucocorticoid, modulates circulating leukocyte numbers, phagocytic capacity and oxidative burst activity (OBA) of canine peripheral blood phagocytes, and whether tumor necrosis factor-alpha (TNF-$\alpha$) release is affected by MPSS injection. Neutrophilia and monocytosis were induced by the administration of a high dose of MPSS, which is the recommended protocol for canine patients with acute spinal cord injury. The injection of MPSS decreased the phagocytic capacity of canine PMNs but not PBMCs, and recovered 12 hours (hr) after the completion of MPSS dosing. The OBA of both PMNs and PBMCs was suppressed by MPSS, and restored 24 hr after the completion of dosing. The lipopolysaccharide-induced TNF-α release by PBMCs but not PMNs exposed to MPSS was reduced 12 hr after the completion of dosing, and recovered 48 hr after the completion of dosing. These results suggest that the application of MPSS protocol inhibits the innate immune functions of canine peripheral blood phagocytes for short time relatively.

Inhibitory Effects of Citaowan on Metastatic Cancer Growth and Agiogenesis in an Orthotopic Model of Breast Cancer (생쥐의 동소이식 유방암에서 자도환(慈桃丸)의 in vivo 전이암 성장 억제 및 혈관신생 억제 효과)

  • To investigate the inhibitory effects of Citaowan (CTW) on the growth and angiogenesis of breast cancer in vivo. Orthotopic breast cancer model was established by injection of MDA-MB-231 cells into mammary fat pad of nude mice. Seven weeks after injection, CTW was orally administered at dose of 50, 100 mg/mouse every day for 40 days. Body weight, tumor volume, tumor apoptosis, microvessel density and tumor proliferation were evaluated, after the mice were sacrificed. The body weight and tumor volume were not significantly changed in CTW group compared with the control group. Tumor apoptosis, proliferation and microvessel density were significantly reduced in CTW group (100 mg/mouse) compared with the control group. These data indicate that CTW has anti-angiogenic and proapoptotic effects on breast cancer.

$^{86}Rb$ Distribution in the Lung of the Rabbit with Pneumothorax (가토 기흉에서 본 $^{86}Rb$의 분포)

  • $^{86}Rb$ uptake of some organs and tissues, ego both lungs, both renal cortices. small intestine, liver and skeletal muscle were studied in the control and the rabbit subjected to pneumothorax. $^{86}Rb$ in the form of chloride mixed with physiological saline was intravenously. injected. The doses were $100{\mu}c$ for a rabbit. The rabbits were sacrificed at intervals of 10, 20, 40, and 60 seconds after the injection of $^{86}Rb$, by the injection of saturated KCI solution. After sacrification, the organ and tissue sample were quickly removed. $^{86}Rb$ uptake in gm of the organs and tissues were measured. On the basis of uptake value, administered doses and body weight, % dose/gm tissues per 200 gm body weight was calculated. Followings were the results; 1. Pneumothorax resulted in a marked elevation in $^{86}Rb$ uptake value of collapsed lung and returned to normal level lately. 2. Contralateral lung of pnemothorax also showed marked elevation in $^{86}Rb$ uptake value and recovered to normal level. 3. Initial $^{86}Rb$ uptake value of liver, small intestine of the rabbit with pneumothorax showed some elevation as compared to control, but that of late stage were similar with control. 4. Local blood flow determination by means of $^{86}Rb$ uptake were inadequate in the collapsed lung of pneumothorax. 5. It was suggested that the mechanism for the initial elevation of $^{86}Rb$ uptake value in each organs and tissue were different from each other.

Hypoglycemic Effect of Fractions of Cassia tora Extract in Streptozotocin-Induced Diabetic Rats (결명자 분획물이 당뇨 유발 흰쥐의 혈당에 미치는 영향)

  • The hypoglycemic effect of fractions of methanol extract of Cassia tora was investigated in diabetic rats. Sprague-Dawley male rats weighing 180∼230 g were divided into 6 groups. Diabetes was induced in the male rats by intravenous injection of streptozotocin at a dose of 45 mg/kg dissolved in citrate buffer (pH 4.5). The diabetic animals then had plasma glucose concentration of above 300 mg/㎗. Fractions of methanol extract of Cassia tora were administered orally into the diabetic rats for 14 days after streptozotocin injection. The food intake and body weight gain were monitored and plasma levels of glucose, triglyceride, free fatty acid, HDL-cholesterol, cholesterol and ALT activity were determined. Levels of glycogen, cholesterol and triglyceride in liver were analysed. Weights of liver and kidney were lighter in all the fraction-administered groups than streptozotocin-control group. Plasma glucose level was significantly decreased by the administration of butanol fraction at 14 days. Plasma triglyceride levels were lower in chloroform, butanol and H$_2$O fraction-administered groups compared to streptozotocin-control group. Plasma cholesterol and HDL-cholesterol levels were not significantly different in all groups. Administrations of each of the four fractions have decreased plasma free fatty acid level, ALT activity and liver triglyceride levels in the diabetic rats. Liver glycogen levels in chloroform, butanol and H$_2$O fraction-administered groups were higher than streptozotocin-control group. It is suggested from the results that butanol fraction of methanol extract of Cassia tora may contain the antihyperglycemic compounds.

  • The efficiency of manure treatment was investigated in terms of the pH, BOD, COD, SS, T-N and TP with a variation in the injection amount of PAC, polymer and ozone. The wastewater flow rate to the AOF is of $7.4m^3/hr$ with a reaction time of 30 minutes. The amount of PAC and polymer changed by 30, 35, 40 ml/min, and 30, 40, 50 ml/min, respectively. The amount of ozone injected varied from 110, 125, and $150kg-O_3/hr$. The optimum manure treatment performance was found for a PAC of 35 ml/min for the COD and SS, with polymer of 30 ml/min, and ozone injection of $150kg-O^3/hr$. A substantially optimum dose for each PAC, polymer, and ozone was also found to exist.

Effects of Swainsonine on the Humoral Immune Response of Lipopolysaccharide

  • Effects of swainsonine (SW;8${\alpha}$, ${\beta}$-indolizidine-${\alpha}$, $2{\alpha}$8-triol from Locoweed) on the humoral immune responses of lipopolysaccharide (LPS) wer studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7 mg/kg of swainsonine, by i.p. injection twice a week for 14 days at a dose of 2 mg/kg. Humoral immune responses were evaluated by hemagglutination (HA) titer and splenic plaque forming cells (PFC). The results of this study were summarized as follows: Mice administrated each of LPS and SW showed significant enhancement of the weight ratios of spleen to body, HA titer, 2-mercaptoethanol-resistant HA(MER-HA) titer and PFC compared with those in controls. However, the LPS plus SW treatment decreased HA titer, MER-HA titer and PFC corresponding to humoral immunity, as compared with those in the mice treated with LPS alone. These findings indicated that LPS significantly enhanced humoral immune responses, but their enhancement effects were lowered somewhat by SW.

Effects of Swainsonine on the Cell-mediated Immune Responses of Lipopolysaccharide (리포포리사카라이드의 세포성 면역반응에 미치는 스와인소닌의 영향)

  • Effects of swainsonine (SW: 8${\alpha}$, ${\beta}$-indolizidine-1alpha, 2${\alpha}$, 8${\beta}$-triol from Locoweed) on the cellular and nonspecific immune responses of lipopolysaccharide (LPS) wer e studied in ICR mice. Mice were divided into 4 groups (10mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7mg/kg of SW by i.p. injection twice a week for 14 days at a dose of 2mg/kg. Immune responses of the delayed-type hypersensitivity response (DTH) to sheep red blood cells (s-RBC), phagocytic activity and natural killer (NK) cell activity were evaluated. LPS treatment didn`t affect NK cell activity, phagocytic activity, DTH to s-RBC compared with those in controls, and phagocytic activity of sareoma 180 tumor bearing mice. However, circulating leukocytes were significantly decreased. Combinaton of LPS and SW increased circulating leukocytes significantly compared vath that in LPS alone, and DTH to s-RBC, NK cell activity and phagocytic activities of normal and sarcoma tumor bearing mice were not affected. These findings indicate that SW didn`t affected the cellular immune responses suppressed by LPS but significantly increased circulating leukocytes.
