Effect of Mild Heat and Organic Acid Treatments on the Quality of Fresh-Cut Lotus Roots (열처리 및 유기산처리가 신선편이 연근의 품질에 미치는 영향)

  • To extend the shelf-life of freshly cut lotus roots, the effect of their heat treatment in water at $50^{\circ}C$ was investigated and compared with that of their treatment using 1 percent ascorbic acid and citric acid during their storage. In addition, consecutive heat and acid treatment was applied to freshly cut lotus roots to determine its synergy effect. The level of molds in the sample that was treated with $50^{\circ}C$ water for 60 minutes was the lowest among the treatments and 3.89 log CFU/g after 12-day storage, whereas the population of the control was 6.81 log CFU/g for the same number of days. During the storage, the surface color of the samples showed higher ${\Delta}E$ values and lower L values than that of the initial sample. The color with the heat treatment hardly changed, unlike in the control. The consecutive treatments with hot water and acid showed less quality loss than the simple heat treatment. The marketable quality was maintained for three days without any treatment, for nine days with the heat treatment, and for 12 days with the consecutive treatments of hot water and acid. The consecutive treatments with hot water and acid, especially with 1 percent citric acid, extended the shelf-life of the freshly cut lotus roots by inhibiting the growth of microorganisms and the browning.

  • To anticipate and respond to harmful algae produced in a big artificial lake like Daecheong reservoir, development of a regional analysis computer system using GIS or RS technique is needed in addition to biological and chemical research. The purpose of this study is to develop a cyanobacterial blooming prediction model to prevent harmful algae produced in Daecheong reservoir and construct an early forecasting system based on GIS. For this purpose this paper examines previous studies related to the relationship between cyanobacteria and environmental factors in Daecheong reservoir and selects precipitation and air temperature as two important environmental factors for the development of cyanobacterial blooming prediction model. Data used in this study are water quality and weather data for three water regions in Daecheong reservoir between 2000 and 2004. Based on qualitative correlation analysis between cyanobacteria and environmental factors, this paper presents a Rump model which enables us to predict cyanobacteria in water regions of Daecheong reservoir. Under this model the prediction of initial occurrence time and growth period of cyanobacteria are possible. The model is also applied to the GIS-based early forecasting system for cyanobacteria, and finally a GIS which can predict cyanobacteria produced in Daecheong reservoir and can manage the related data is developed.

  • Recently, the transport quantity of goods between South and North Korea is rising rapidly by increasing of economical exchange between South and North Korea and growth of Gaeseoung industrial complex. In addition to a land transportation route, it is necessary to secure various logistics and transportation route for freight transportation between Incheon and Gaesung. In this paper, we investigated a construction of barge transportaion system between Incheon and Gaesung. The barge transportation system which has many merits such as a little initial investment and convenience of harbor loading/ unloading has been also used to transport freight widely in the EU and USA. Firstly, we investigated barge transport route, marine freight and barge fleet which consists of barge and pusher between Incheon and Gaesung. And next, we designed hull form and general arrangement of a barge and pusher based on shipping service condition, barge fleet, etc. Finally, a construction plans of barge transport system such as a operation of shipping service, berthing facilities, logistics center, intermodal transport are investigated.

Culture Conditions for Hydrogen Production of Enterobacter cloacae YJ-1 (수소생산을 위한 Enterobacter cloacae YJ-1의 배양조건)

  • We investigated the effective culture conditions of anaerobic bacteria, Enterobacter cloacae YJ-1 on hydrogen production. It was cultured with 60 mL of working volume at $35^{\circ}C$, 120 rpm for 40 h. With culture time, hydrogen production and cell growth increased, but residual glucose and pH decreased. When the $2\%$ of glucose was used as single carbon source, hydrogen production was 975.1 mL/L. To enhance hydrogen productivity, mixed carbon sources of glucose and sucrose were added. The maximum hydrogen production was earned at the mixing ratio of 25:75, and it was 1319.5 mL/L. When we added 50 mM of phosphate to protect the pH drop in culture broth, hydrogen production increased 1.3 times more than that of initial concentration. The organic nitrogen sources were more effective than inorganic nitrogen for hydrogen production. Among organic nitrogen, yeast extract was the most effective and its hydrogen production was 1691.3 mL/L. Among 9 of mineral sources, Ferric citrate and $NaMoO_4$ were especially effective, and their productions were 1782.3 mL/L and 1784.8 mL/L, respectively.

  • Bacillus subtilis KMU-13 was isolated from the Lillehammer forest soils at Norway and shown a strong antifungal activity on cucumber scab, Cladosporium cucumerinum KACC 40576. B. subtilis KMU-13 produced a maximum level of antifungal substance under incubation aerobically at $30^{\circ}C$, 180 rpm for 48 hours in LB broth containing 0.5% maltose and 0.5% bactopeptone and initial pH adjusted to 6.0. Butanol extract of cultured broth was confirmed inhibitory zone by plate assay and Rf 0.64 value substance by thin layer chromatography (TLC) represented high antifungal activity against C. cucumerinum KACC 40576 and also shown fungal growth inhibitory activity against Botytis cinerea KACC 40573, C. gloeosporioides KACC 40804, D. byoniae KACC 40669, F. oxysporum KACC 40037, F. oxysporum KACC 40052, F. oxysporum f. sp. radicis-lycopersici KACC 40537, F. oxysporum KACC 40902, M. cannonballus KACC 40940, P. cambivora KACC 40160, R. soiani AG-1 KACC 40101, R. solani AG-4 KACC 40142, and S. scleotiorum KACC by agar diffusion method.

Initial Development of Forest Structure and Understory Vegetation after Clear-cut in Pinus densiflora Forest in Southern Gangwon-do Province (강원도 남부 지역에서 소나무림 개벌 후 초기 임분 구조 및 하층식생 발달)

  • Open- to closed canopy stage and it's ecological characteristics in vegetation succession are commonly described, but poorly understood in Korea. Vegetation development on structure, environment and understory abundance were studied for 16 yr in post-clearcut Pinus densiflora forests in the southern Gangwon-do province by applying space-for-time approach. We sampled 210 plots (10 for structure and 200 for understory) for four seral stages (1yr, 3yr, 10yr and 16yr). After clear-cut, mean stem density increased gradually to $5,714{\pm}645$ stems/ha after 16 years and mean basal area was also from $5.5{\pm}0.7m^2/ha$ after 10 years and doubled at $10.0{\pm}1.6m^2/ha$ in 16 years. Woody debris and bared soil on the forest floor peaked at 11--- after 10 years and at 10.3--- after 3 years, respectively. In understory mean cover declined with all growth form groups following succession, but in richness, forb specie increased with structural development during 16 years. Our study suggested that overstory development did not suppressed whole understory properties especially in richness, thus appeared to act as a filter selectively constraining the understory characteristics. However only long-term studies are essential for elucidating patterns and processes that cannot be inferred form short-term or space-for-time researches. Strong negative relationship between overstory and understory characteristics in conventional models surely reexamined.

Quality Changes of Egg Products during Storage (계란가공품의 보관중 품질 변화)

  • To know the probable storage conditions for commercially prepared ‘baked egg’ and ‘rolled egg’ products, microbes and quality changes for samples immediately after the production and stored under the several conditions were tested. Sealed samples were stored at 5, 10, 15, $20^{\circ}C$, respectively and tested at 1 or 2-days intervals, opened samples were stored at 15, 20, 30, $37^{\circ}C$, respectively and tested at 6-hr intervals. Coliform bacteria, E. coli, Salmonella, Staphylococci were not detected, and viable cell counts level was under $10^2\;CFU/g$ for samples immediately after the production. The higher the storage temperature and baked eggs rether than rolled eggs showed more rapid microbial growth for both samples. For sealed condition, 14-days at $5^{\circ}C$ for both samples, 7-days for rolled eggs and 5-days for baked eggs at $10^{\circ}C$ were kept their initial microbial level and favorable flavor. For opened condition, it showed remarkable microbial increase after $18{\sim}24\;hr$ for $15{\sim}20^{\circ}C$ and 12 hr for $30{\sim}37^{\circ}C$. From these results, used egg products samples are able to be stored more than 1-week at below $5^{\circ}C$ and opened samples need to be stored at cold storage. At room temperature, it egg products.

The Effect of Drinking Water Fluoride on the Fine Structure of the Ameloblast in the Fetal Rat (음용수 불소가 흰쥐태아 법랑모세포의 미세구조에 미치는 영향)

  • The response of ameloblast to long term (3 weeks) exposure to fluoride was examined in continuously erupting mandibular incisors of pregnancy rats as compared to control rats receiving a similar diet (Teklad L-356) but no sodium fluoride in there drinking water. Rats were started on water containing 0 ppm, 100 ppm, 200 ppm, and 300 ppm NaF at the beginning of pregnancy. To examine on the ultrastructural changes of the ameloblast, electron microscopy was used. The results indicated that rat incisors expressed two major changes in normal amelogenesis that could be attributed to chronic fluoride treatment. The fluoride produces marked alteration in the fine structure of ameloblast from teeth of young rats, such as large confluent distensions of the endoplasmic reticulum and swelling of isolated mitochondria, in particular on the morphology of the rough-surfaced endoplasmic reticulum. A graded series of alterations to these organelles were produced, and the severity of the changes would seem to be dependent on dose and time. This experimental data suggested that exposure prolonged of animal to high level of fluoride appears to induce morphological changes in the normal appositional growth and initial mineralization of enamel created during amelogenesis.

A Microscope Study of Fluoride Effects on the Rat Incisor Enamel Formation (흰쥐 절치의 법랑질 형성에 미치는 불소의 효과에 관한 현미경적 연구)

  • The aim of the present study was to examine in detail, both at light and electron microscopical levels, the morphological variations in ameloblast of the fetal rat incisor enamel organ. Rats were started on distilled water at the beginning of pregnancy. The pups were sacrificed 11 days after delivery and animals were perfused intravascularly with glutaraldehyde and the incisors were removed. To examine on the ultrastructure of the ameloblast, the study employed primary light microscopy but electron microscopy was used to clarify some of the light microscopic finding. Longitudinal sections through the incisors of the rat show a continuous layer of ameloblasts on the labial surface of the tooth. This layer contains the entire sequence of developmental stages in enamel production. The ameloblast layer was divided into three main zones: 1) Presecretory zone, region of ameloblasts facing pulp. 2) Secretory zone, region of inner and outer enamel secretion. 3) Maturation zone, region of reduced ameloblasts. In particularly, the present study has shown that two distinctively different types of ameloblasts appear in the enamel organ during enamel maturation in the rat incisor. These two types have been designated ruffle-ended ameloblasts (rAB) and smooth-ended ameloblasts (sAB). Effect of Barley Yellow Mosaic Virus (BaYMV)- Infested Soil on the Agronomic Characters of Three Different Barley Varieties

  • Barley Yellow Mosaic Virus (BaYMV) caused significant reduction in barley yield and is difficult to control due to alive parasitic soil-borne fungus, Palmyra gamines that transmits the, virus. Previous studies have indicated that a virus-free soil could be infested by using virus-contaminated farming machineneries and implements. For the further confirmation of this finding, different proportions of BaYMV-infested soil were mixed into virus free soil. Three barley varieties (Hordum vulgarae, cv "Olbori", "Baegdong" and "Sacheon 6") were sown in pots treated with different rate of P. graminis-infested soil ranging from 0% to 100% in October 20, 2001. Results showed that BaYMV infection increased as the rate of infested soil increased. Initial symptoms were observed in a pots treated with 10% infested soil in all the 3 varieties of barley. "Olbori" had about 5% infection in 20% infested soil and about 10% infection in 40% or 50% infested soil and about 20% infection in 60% infested soil. In "Baegdong", the trend of BYMV occurrence was similar with "Olbori" but the time of severe infection was earlier than "Olbori". BaYMV infection in "Sacheon 6" was even earlier than "Baegdong" with much more severe symptoms than "Baegdong". The growth rate of barley was affected by about 19-22% when grown in 20% infested soil. As the rate of BaYMV infested soil increased the heading date was delayed but the maturing date was early in "Olbori" and "Sacheon 6". Also, reduction rate of culm length in 3 varieties increased with increase of infested soil content. However, "Olbori" showed the highest reduction. "Sacheon 6", have been characterized with long spike length, however was significantly reduced as the infested soil increased. On the other hand, spike length of "Olbori" was not significantly affected despite of increased of infested soil. The reduction rate of 1000 kernel weight was higher in large kernel size cultivar "Sacheon 6" and "Olbori" than small kernel size "Baegdong" as increase of BaYMV-infested soil content.
