Production of pediocin by Chemical Synthesis and Bactericidal Mode of Action

  • To investigate the mode of bactericidal action for antimicrobial peptide, pediocin, synthetic and mutant pediocins were prepared by direct chemical synthesis. Native pediocin was purified from Pedio-coccus acidilactici M and its conformational structure and bactericidal functions were analyzed and compared to synthetic pediocin. Schematic mode of pediocin actions, how pediocin binds on the target cell membrane, penetrates and makes tunnel are proposed. For these purposes, primary and secondary structures of pediocin was analyzed and disulfide bond assignment was also done. The pediocin purified from P. acidilactici M had high effective bactericidal ability against gram positive bacteria, especially Listeria monocytogenes and was very stable at extreme pHs and even at high temperatures such as autoclaving temperature (121$^{\circ}C$). Pediocin was consisted of 44 amino acids with four cysteines. Novel synthetic peptides were achieved by solid phase peptide synthesis(SPPS) method. To explain the function of cysteine in C-terminal region, mutant pediocin, Ped[C24A+C44A], was synthesized and their structural and biological functions were analyzed. Second mutant pediocin, Ped[KllE], was prepared to explain the function of lysine at 11 of N-terminal part of pediocin, especially loop of $\beta$-sheet, and to predict the initial binding site of pediocin. The native and synthetic pediocins was showed random coil conformation by spectropolarimetry in moderate conditions. This conformation was observed in extreme conditions such as high temperature and low and high pHs, also. Circular dichroism(CD) data also showed the existence of $\beta$-turn structure in N-terminal part both native and synthetic pediocins. A structural model for pediocin predicts that 18 amino acids in the N-terminal part of the peptide assume a three-strand $\beta$-sheet conformation. This random coil in C-terminal part of pediocin was converted to folding structure, helix structure, in nonpolar solvents such as alcohol and TFE. The disulfide bond between $^{9}$ Cys and $^{14}$ Cys was concrete and inevitable, however, evidences of disulfide bond between $^{24}$ Cys and $^{44}$ Cys was not. Data of Ped[C24A+C44A], pediocin mutant showed that $^{44}$ Cys was required during killing the target cells but not inevitable, since Ped[C24A+C44A] still have bactericidal activity but much less than native pediocin. Another pediocin mutant, Ped[KllE], had still bactericidal activity, was controversial to propose that positive charge like as $^{11}$ Lys in loop or hinge in bacteriocin bound or helped to binding to microorganism with electrostatic interaction between cell membrane especially teichoic acid and positive amino acid nonspecifically. The conformation of pediocin among native, synthetic and mutant pediocins did not show big difference. The conformations between oxidized and reduced pediocin were almost similar regardless of native or synthetic.

  • SqueeSAR is a new technique to combine Persistent Scatterer (PS) and Distributed Scatterer (DS) for deformation monitoring. Although many PSs are available in urban areas, SqueeSAR analysis can be beneficial to increase the PS density in not only natural targets but also smooth surfaces in urban environment. The height of each targets is generally required to remove topographic phase in interferometric SAR processing. The result of PSInSAR analysis to use PS only is not affected by DEM resolution because the height error of initial input DEM at each PSs is precisely compensated in PS processing chain. On the contrary, SqueeSAR can be affected by DEM resolution and precision since it includes spatial average filtering for DS targets to increase a signal-to-noise ratio (SNR). In this study we observe the effect of DEM resolution on deformation measurement by PSInSAR and SqueeSAR. With ALOS-1 PALSAR L-band data, acquired over Daejeon city, Korea, two different DEM data are used in InSAR processing for comparison: 1 m LIDAR DEM and SRTM 1-arc (~30 m) DEM. As expected the results of PSInSAR analysis show almost same results independently of the kind of DEM, while the results of SqueeSAR analysis show the improvement in quality of the time-series in case of 1-m LIDAR DSM. The density of InSAR measurement points was also improved about five times more than the PSInSAR analysis.

A Comparative Analysis of Kinematics and Kinetics on Forehand Drive in Squash (스쿼시 Forehand 드라이브 동작 시 운동역학적 비교연구)

  • The purpose of the study is to give basic data for the improvement of the skill and to show an exemplary position for squash club members or trainers thru a comparative analysis on the kinematics and kinetics variables on the forehand drive motion in playing squash. The objects of the research are divided into two sections, skilled group(n=8) and unskilled group(n=8). The skilled group is composed of professional players currently working and unskilled group is career of six month, both of lives in B city. In this research, to gather the data 3D motion analysis and test result analysis using force platform was used. The variables are duration, position, segment velocity, segment acceleration and etc. in using force platform. The results are as follows: 1. The duration per phase of the skilled is 0.18sec P1(DS) while that of unskilled is 0.32sec. in P2(FT), the duration of the skilled is 0.29sec, that of unskilled is 0.34sec. Average of the duration of the skilled is 0.48sec, while the unskilled, 0.66sec. 2. Regarding positional movements per event, the unskilled has a relatively higher position in center of gravity, shoulder joint, elbow joint compared with that of the skilled. Generally speaking, positions of the unskilled is higher than the skilled. 3. In segment velocity per event, R-shank, R-upper arm, R-forearm and racket. The skilled is faster than the unskilled. we found a big dig difference in shank. 4. In acceleration per event, there was a big difference in upper-arm and fore-arm of the impact. 5. The skilled group on the force platform shows relatively stable and regular changes while the unskilled shows unstable from the touch down to initial 20% the force value of central support period after the impact moment decreases rapidly and the center of gravity is not moved well. 6. The maximum force value of the skilled is 1019.7N. it is found 19.86% of the total duration. That of the unskilled is 639.2N, it is found 20.67% of total duration.

Comparison of Intraperitoneal and Retroperitoneal/Pelvic Contrast Extravasation: The Characteristics and Prognosis of the Each Patient Group with Arterial Embolization according to the Abdominal Computed Tomography Scanning after Blunt Trauma (둔상 후 복부 전산화단층촬영에서 조영제 유출로 동맥색전술을 시행받은 환자의 복강내와 후복막강/골반강내 출혈 비교)

  • Purpose: This study compared the characteristics of and the prognosis for intraperitoneal and retroperitoneal/pelvic contrast extravasation, which had been confirmed by enhanced abdominal CT scan, after blunt trauma in patients who had undergone angiographic embolization. Methods: From January 2001 to March 2009, data were retrospectively collected regarding patients who had undergone contrast extravasation (CE) on CT scanning and arterial embolization after blunt trauma. The study patient group was divided into the intraperitoneal and the retroperitoneal/pelvic groups according to the area of contrast extravasation. We reviewed the initial demographic data, the location of injury, the solid organ injury, the embolized vessel, and the clinical outcome. Results: The mean age of the study subjects was $40.2{\pm}2.6$ years old, and there were 24 male patients. The intraperitoneal group included 10 patients, and retroperitoneal/pelvic group was comprised of 17 patients. The amount of transfusion from presentation to intervention and during the first 24 hours was greater in the retroperitoneal/pelvic group than in the intraperitoneal group. The intraperitoneal group showed a higher frequency and severity of liver injury than the retroperitoneal/pelvic group. Angiography revealed that the hepatic artery (n=4) was the most frequently embolized vessel in the intraperitoneal group, while the internal iliac artery (n=6), followed by the renal artery (n=4), internal pudendal artery (n=3), and the gluteal artery (n=2), were the most frequently injured vessels in the retroperitoneal/pelvic group. Conclusion: In patients with intra-abdominal contrast extravasation found on CT scanning and arterial embolization after blunt trauma, the need for transfusion was less in the intra-abdominal group than in the retroperitoneal/pelvic group. Liver injury was also more frequent and severe in the intraperitoneal group than in the retroperitoneal/pelvic group.

An Empirical Study on Comparative Analysis of Freight Demand Estimation Methods - Unimodal O/D Based Method and P/C Based Method : Focus on Korean Import/Export Container Freight (수단O/D기반 및 P/C기반 화물수요추정방식의 실증적 비교: 우리나라 컨테이너 화물을 중심으로)

  • This study deals with the comparative analysis between two freight demand estimation methods : Unimodal O/D based method and P/C based method. The data of access/egress truck trips has been omitted from the Korean freight unimodal O/D of KTDB. This is because KTDB's unimodal O/D has not marked the series of unlinked trips down as the whole freight intermodal transport and surveyed only the main-haul trips of them. For these reasons, freight intermodal transport mechanism has not been analysed perfectly with Korean unimodal O/D data. This study tries to estimate P/C table of Korean Import/Export container freight and develop the MCC(Multimodal Channel Choice) model. Then, comparing unimodal O/D based method and P/C based method in terms of the switch commodities between production point(the initial point of freight transport) and consumption point(the terminal point of freight transport), unimodal commodities, and commodities on links is conducted. The results show that the P/C based method is able to simulate the freight intermodal transport.

Life Cycle Assessment (LCA) for Calculation of the Carbon Emission Amount of Organic Farming Material -With Emphasis on Hardwood Charcoal, Grass Liquid and Microbial Agents- (유기농자재의 탄소배출량 산정을 위한 전과정평가(LCA) -참숯, 목초액, 미생물제재를 중심으로-)

  • Since 1997, Korean Ministry of Knowledge Economy and Ministry of Environment have established data on some 400 basic raw and subsidiary materials and process like energy, petro-chemical, steel, cement, glass, paper, construction materials, transportation, recycling and disposal etc by initiating establishment of LCI database. Regarding agriculture, Rural Development Administration has conducted establishment of LCI database for major farm products like rice, barley, beans, cabbage and radish etc from 2009, and released that they would establish LCI database for 50 items until 2020 later on. The domestic LCI database for seeds, seedling, agrochemical, inorganic fertilizer and organic fertilizer etc is only at initial stage of establishment, so overseas LCI databases are brought and being used. However, since the domestic and overseas natural environments differ, they fall behind in reliability. Therefore, this study has the purpose to select organic farming materials, survey the production process for various types of organic farming materials and establish LCI database for the effects of greenhouse gas emitted during the process in order to select carbon basic units for agricultural production system compliant in domestic situation instead of relying on overseas data and apply life cycle assessment of greenhouse gas emitted by each crop during the process. As for selecting methods, in this study organic farming materials were selected in the method of direct observation of material and bottom-up method a survey method with focus on the organic farming materials admitted into rice production. For the basic unit of carbon emission amount by the production of 1kg of organic farming material, the software PASS 4.1.1 developed by Korea Accreditation Board under Ministry of Knowledge Economy was used. The study had the goal to ultimately provide basic unit to calculate carbon emission amount in executing many institutions like goal management system and carbon performance display system etc in agricultural sector to be conducted later on. As a result, emission basic units per 1kg of production were calculated to be 0.0088kg-$CO_2$ for charcoal, 0.1319kg-$CO_2$ for grass liquid, and 0.2804kg-$CO_2$ for microbial agent.

Simulation Model for Estimating Soil Temperature under Mulched Condition (멀칭에 따른 지온변화 모델의 작성 및 토양온도의 추정)

  • A numerical model using soil surface energy balance and soil heat flow equations to estimate mulched soil temperature was developed. The required inputs data include weather data, such as global solar radiation, air temperature, wind speed, atmospheric water vapor pressure, the optical properties of mulching material, and soil physical properties. The observed average soil temperature at 50 cm depth was used as the initial value of soil temperature at each depth. Soil temperature was simulated starting at 0 hour at an interval of 10 minutes. The model reliably described the variation of soil temperature with time progress and soil depth. The correlation between the estimated and measured temperature yielded coefficient values of 0.961, 0.966 for 5cm and 10cm depth of the bare soil, respectively, 0.969, 0.965 for the paper mulched soil, and 0.915, 0.938 for the black polyethylene film mulched soil. The percentages of absolute differences less than 2$^{\circ}$C between soil temperatures measured and simulated at 10 minute interval were 97.4% and 98.5% for 5 cm and 10cm for the bare soil, respectively, and 95.8% and 97.4% for the paper mulched soil, and 70.1% and 92.5% for the polyethylene film mulched soil. The results indicated that the model was able to predict the soil temperature fairly well under mulched condition. However, in the night time, the model performance was a little poor as compared with day time due to the difficulty of accurate determination of the atmospheric long wave radiation.

Equilibrium, Kinetic and Thermodynamic Parameter Studies on Adsorption of Allura Red from Aqueous Solution by Granular Activated Carbon (입상활성탄에 의한 수용액으로부터 오로라 레드의 흡착에 대한 평형, 동력학 및 열역학 파라미터에 관한 연구)

  • Allura Red (AR) is a water-soluble harmful tar-based food colorant (FD & C Red 40). Batch adsorption studies were performed for the removal of AR using bituminous coal based granular activated carbon as adsorbent by varying the operation parameters such as adsorbent dosage, initial concentration, contact time and temperature. Experimental equilibrium adsorption data were analyzed by Langmuir, Freundlich and Temkin isotherms. The equilibrium process was described well by Freundlich isotherm. From determined separation factor ($R_L$), adsorption of AR by granular activated carbon could be employed as effective treatment method. Temkin parameter, B was determined to 1.62~3.288 J/mol indicating a physical adsorption process. By estimation of adsorption rate experimental data, the value of intraparticle diffusion rate constant ($k_m$) increased with the increasing adsorption temperature. The adsorption process were found to confirm to the pseudo second order model with good correlation. Thermodynamic parameters like change of free energy, enthalpy, and entropy were also calculated to predict the nature adsorption in the temperature range of 298~318 K. The negative Gibbs free energy change (${\Delta}G$ = -2.16~-6.55 kJ/mol) and the positive enthalpy change (${\Delta}H$ = + 23.29 kJ/mol) indicated the spontaneous and endothermic nature of the adsorption process, respectively.

Characteristics of Isotherm, Kinetic, and Thermodynamic Parameters for Reactive Blue 4 Dye Adsorption by Activated Carbon (활성탄에 의한 Reactive Blue 4 염료의 흡착에 대한 등온선, 동력학 및 열역학적 특성)

  • The isotherm, kinetic, and thermodynamic parameters of reactive blue 4 adsorbed by activated carbon were investigated for activated carbon dose, pH, initial concentration, contact time, and temperature data. The adsorption of the RB 4 dye by activated carbon showed a concave shape in which the percentage of adsorption increased in both directions starting from pH 7. The isothermal adsorption data were applied to Langmuir, Freundlich, and Temkin isotherms. Both Freundlich and Langmuir isothermal adsorption models fit well. From determined Freundlich separation factor (1/n = 0.125 ~ 0.232) and Langmuir separation factor (RL = 1.53 ~ 1.59), adsorption of RB 4 by activated carbon could be employed as an effective treatment method. The constant related to the adsorption heat (BT = 2.147 ~ 2.562 J mol-1) of Temkin showed that this process was physical adsorption. From kinetic experiments, the adsorption process followed the pseudo second order model with good agreement. The results of the intraparticle diffusion model showed that the inclination of the first straight line representing the surface diffusion was smaller than that of the second straight line representing the intraparticle pore diffusion. Therefore, it was confirmed that intraparticle pore diffusion is the rate-controlling step. The negative Gibbs free energy change (ΔG = -3.262 ~ -7.581 kJ mol-1) and the positive enthalpy change (ΔH = 61.08 kJ mol-1) indicated the spontaneous and endothermic nature of the adsorption process, proving this process to be spontaneous and endothermic.

Drug-Induced Anaphylactic Shock at the Emergency Department (응급실 기반 약물에 의한 아나필락시스 쇼크 환자의 임상적 특성)

  • Purpose: This study was conducted to investigate the characteristics of drug induced anaphylactis and anaphylactic shock in patients who were admitted to the emergency department Methods: We retrospectively collected the data on patients with drug induced anaphylaxis and who were admitted to the emergency department from January 2001 to June 2009. The study group was divided into the non-shock and shock groups according to whether the systolic blood pressure more than 90mmHg. The initial demographic data, the causes of drug-induced anaphylaxis, the clinical manifestations, the treatment and the prognosis were reviewed for 72 patients. Results: The mean age of the study subjects was $47.9P{\pm}14.2$ years old and there were 40 male patients and 32 female patients. There were 26 patients in the non-shock group and 46 in the shock group. The mean age was older in the shock group than in the non-shock group ($51.5{\pm}15.1$ vs $42.5{\pm}10.6$, p-0.002). A history of drug allergy was more common in the shock group, but no difference was found for the comorbid chronic diseases between the two groups. Radio-contrast media was the most common cause, followed non-steroidal anti inflammatory drugs and antibiotics, but there is no difference in the causes between the two groups. The symptoms of cyanosis, syncope, sweating and dizziness were more frequently manifested in the shock group. The administration of intravenous fluid and injection of subcutaneous epinephrine at the emergency department were more frequent in the shock group than in the non-shock group. Conclusion: For the patients who were admitted to the emergency department with drug induced anaphylaxis, the mean age was older and the symptoms of cyanosis, syncope, sweating, dizziness were more frequent in the anaphylactic shock patients than in the non-shock group. More treatments were given at the emergency department to the anaphylactic shock patients.
