본 연구에서는 환경친화적인 무공해생물농약 재발을 위하여 곤충병원성 선충의 in vitro 배양방법을 개별하였다. 곤충병원선 층연 Steinernem$\alpha$ glasen 종으로부터 공생박테리아를 분리하여 동정한 결과 Xenorhabdus nem$\alpha$tophllus 종임을 확인하였으며, X enorhabdus nematophilus는 감염단계 선충의 장내에서와 in vitro 배양 동안에 그 생화학적 특성이에서 차이를 보이는 동 질형화 헨상을 나타냄을 확인하였다. 균주의 최적바지 조성은 5% yeast extract, 0.5% NaCl, $K_2HPO_4$, $0.02% MgSO_4$.$7H_2O$이였으며, $28^{\circ}C$가 최적배양 온도였다, 초기 pH 6-7에 관계 없이 성장이 진행됨에 따라서 약 90까지 층가하였다. Flask 배양에 비해서 fennentortor양에서 균주의 성장속도가 1.4배 빠르게 나타났으나, 그에 따른 상변회도 빠르게 진행되었다. 한편 Steinem$\xi$ma glaseri익 in vitro 배양을 위 힌 인공배지원으로 chIcken offal, dog food, peanut 퉁이 사용될 수 있으며 최적의 배지는 농축펀 bovine liver로 조사되었으며, 그 농도논 80%일 때 가장 높은 증식을 보였다. 또한 곤충병원선충으로부터 분리된 공생박테리아를 이용한 혼합배양방법은 공생박테리아를 사용하지 않는 경우보다 선충의 in vitro 증식속도가 2 배 가량 빨랐으며, 15일만에 약 $5.5\times10^4$/mL 의 선충이 수확되였다.
자동차 폐촉매로부터 백금족 금속(PGMs)의 회수를 위한 어트리션 스크러빙의 효과를 조사하였다. 자동차 폐촉매를 2mm이하로 분쇄한 후 60분 범위내에서 어트리션 스크러빙하였으며 이를 체가름하였다. 스크러빙을 통해 촉매층은 촉매 지지체 표면으로부터 탈리되어 $45{\mu}m$이하의 미립자로 분리되었다. $45{\mu}m$ 이하 미립자의 양은 스크러빙 시간이 증가함에 따라 증가하였으며, 스크러빙 시간 40분에서 미립자의 유가물 함량은 $CeO_2$ 19.3%, $ZrO_2$ 1.9%, PGMs 419ppm을 나타내었다. 미립자에서 감마 알루미나의 회수율은 스크러빙 시간이 증가함에 따라 증가하였으며, 동시에 $CeO_2$, $ZrO_2$ 및 PGMs의 회수율도 $CeO_2$ 82.9%, $ZrO_2$ 78.7%, PGMs 78.9%로 증가하였다. 시료의 고액비 및 초기 투입 입자 크기가 증가할 때 미립자의 양 및 감마알루미나의 회수율은 증가하였다. 어트리션 스크러빙은 분쇄 및 분리 기술로서 스크러빙 용기내에서 입자들 사이의 충격 및 전단 운동에 의해 코디어라이트 촉매 지지체로부터 감마알루미나의 분리에 효과적임을 알 수 있었다.
Nd-Fe-B 폐 영구자석으로부터 회수한 염화희토류용액을 PC88A에 의한 경희토류(Pr, Nd)/중희토류(Tb, Dy) 분리정제에 대한 용매추출 연구를 진행하였다. 회분식 실험 단계에서 추출제(PC88A) 농도 0.5 M, 추출 후 평형 pH는 0.8~1.0(feed의 초기 pH 1.3), 세정액(HCl)의 농도는 0.1 M, 탈거액(HCl)의 농도는 2 M 이상일 때 분리 효율이 좋은 것을 확인하였다. 이 결과값을 혼합침강조에 적용하여 초기 조건에서 운용 조건을 변화시키면서 180시간 운용하여 3N급의 경희토류(Pr, Nd)/중희토류(Tb, Dy)금속 이온으로 분리정제 후 회수하였다.
To investigate the contribution of macroalgae to biogeochemical nutrients and carbon cycles, we measured the uptake rates of nutrients and $CO_2$ by Undaria pinnatifida using an incubation method in an acrylic chamber. From January to March 2010, U. pinnatifida was sampled at Ilkwang, a well-known area of macroalgae culture in Korea. The initial and final concentrations of nutrients, dissolved oxygen, total alkalinity, and pH of the chamber water were measured, and production/uptake rates were calculated using concentration changes, chamber volume, and incubation time. The production rate of dissolved oxygen by U. pinnatifida (n = 32) was about $5.4{\pm}4.0\;{\mu}mol\;g_{fw}^{-1\;}h^{-1}$. The uptake rate of total dissolved inorganic carbon (TDIC), calculated by total alkalinity and pH, was $7.9{\pm}6.5\;{\mu}mol\;g_{fw}^{-1}\;h^{-1}$. Nutrients uptake averaged $141.7{\pm}119.2$ nmol N $g_{fw}^{-1}\;h^{-1}$ and $15.0{\pm}9.1$ nmol P $g_{fw}^{-1}\;h^{-1}$. A positive linear correlation ($r^2$ = 9.6) existed between the production rate of dissolved oxygen and the uptake rate of total dissolved inorganic carbon, suggesting that these two factors serve as good indicators of U. pinnatifida photosynthesis. The relationships between fresh weight and uptake rates of nutrients and $CO_2$ suggested that younger specimens (<~50 g fresh weight) are much more efficient at nutrients and $CO_2$ uptake than are specimens >50 g. The amount of carbon uptake by the total biomass of U. pinnatifida in Korea during the year of 2008 was about 0.001-0.002% of global ocean carbon uptake. Thus, more research should be focused on macroalgae-based biogeochemical cycles to evaluate the roles and contributions of macroalgae to the global carbon cycle.
Two experiments were conducted to evaluate the effects of a complex Lactobacilli preparation on performance, resistance to E. coli infection and gut microbial flora of weaning pigs. In exp. 1, twelve pigs (7.65$\pm$1.10 kg BW), weaned at 28 d, were randomly allotted into 2 groups and placed in individual metabolic cages. During the first 7 d, one group of pigs was provided ad libitum access to water containing $10^5$ colony forming units (CFU) Lactobacilli per ml and the control group was provided tap water. The Lactobacilli preparation included Lactobacillus gasseri, L. reuteri, L. acidophilus and L. fermentum, which were isolated from the gastrointestinal (GI) tract mucosa of weaning pigs. On d 8, 20 ml of $10^8$ CFU/ml E. coli solution (serovars K99, K88 and 987P at the ratio of 1:1:1) was orally administered to each pig. Diarrhea scores and diarrhea incidence were recorded from d 7 to 14. On d 14, pigs were euthanized and digesta and mucosa from the stomach, duodenum, jejunum, ileum, cecum and colon were sampled using aseptic technique to determine microflora by culturing bacteria in selective medium. The results showed that Lactobacilli treatment significantly decreased E. coli and aerobe counts (p<0.01) but increased Lactobacilli and anaerobe counts (p<0.01) in digesta and mucosa of most sections of the GI tract. A 66 and 69.1% decrease in diarrhea index and diarrhea incidence, respectively, was observed in the Lactobacilli treated group. In exp. 2, Thirty-six crossbred Duroc$\times$Landrace$\times$Yorkshire piglets, weaned at 28$\pm$2 days, were selected and randomly allocated into 2 groups. There were 18 piglets in each group, 3 piglets in one pen and 6 replicates in each treatment with 3 pens of barrow and 3 pens of female piglet in each treatment. Piglets had ad libitum access to feed and water. The initial body weight of piglet was 7.65$\pm$1.09 kg. Dietary treatments included a non-medicated basal diet with Lactobacilli ($10^5$ CFU/g diet) or carbadox (60 mg/kg) as control. On d 21, six pigs per group (one pig per pen) were euthanized. Ileal digesta was collected to determine apparent amino acid digestibility. Microflora content was determined similarly to exp.1. The results showed that Lactobacilli treatment significantly improved average daily feed intake (ADFI) of pigs compared to carbadox (p<0.05) during the first 2 wks after weaning and average daily gain (ADG) and ADFI increased significantly (p<0.05) from d 8 to 14. Nitrogen and total phosphorus digestibility also increased (p<0.05). Bacterial counts were similar to exp. 1. The results indicate that the complex Lactobacilli preparation improved performance for 2 wks after weaning, enhanced resistance to E. coli infection, and improved microbial balance in the GI tract.
Rhodamine B(RhB) 폐수의 색도와 COD 제거를 위해 $O_3$, $UV/TiO_2$, $O_3/UV$ 및 $O_3/UV/TiO_2$ 공정과 같은 고급산화법의 영향을 연구하였다. 오존 공정의 경우 오존 농도가 높을수록 탈색속도가 높아졌으며, $UV/TiO_2$와 $O_3/UV/TiO_2$ 공정의 최적 $TiO_2$ 농도는 0.4 g/L 였다. $O_3/UV$ 공정은 오존 공정에 비해 초기 탈색속도상수가 높고 탈색종결시간도 줄어드는 것으로 나타났다. 실험에 적용한 공정의 초기 탈색속도상수의 순서는 $O_3/UV/TiO_2>O_3/UV>O_3>UV/TiO_2$로 나타났다. 모든 공정의 탈색속도가 COD로 표시한 무기화 속도보다 빠르고, COD는 처리에 시간이 더 소요되는 것으로 나타났다. COD 제거속도는 $O_3/UV/TiO_2>O_3/UV>UV/TiO_2{\geqq}O_3$의 순서로 나타났다. 네 공정 중에서 광촉매와 오존 공정을 조합한 $O_3/UV/TiO_2$ 공정이 염료 폐수와 같은 폐수의 색도와 COD 제거에 적절한 것으로 나타났다.
최근 미국과 일본을 중심으로 높은 공극과 압축성이라는 장점을 지닌 압축성 섬유여재 여과에 대한 다양한 기술이 개발되면서 이를 이용한 집약적 하 폐수 처리에 대한 관심이 높아지고 있다. 이에 따라 본 연구에서는 다양한 유입수질의 하수에 대하여 압축성 섬유여재 여과의 적용성을 검토하고자 하며, 해당 유입수에 대한 최적의 운전조건을 도출하고자 한다. 이를 위하여 예비 실험으로서 추적자 시험을 진행하였으며, 이를 통하여 다양한 압축률 및 여과율에서의 여과 양상을 검토하였다. 그 결과 높은 압축률에서 높이에 따라 다른 공극률이 적용되어 이론적인 결과값과의 뚜렷한 차이를 관찰할 수 있었다. 하수처리장의 침사지 유출수를 대상으로 다양한 압축률 조건에서 압축성 섬유여재 여과의 TSS 제거 및 제거 입자크기분포를 살펴본 결과, 압축률 30%가 최적의 운전조건으로서 제거효율 70% 이상을 나타냈으며, 작은 크기의 입자 제거율 증대를 위해 최소 15% 이상의 압축률이 요구되었다. 또한 여과 유출수 농도의 주요 영향인자인 $10{\mu}m$ 이하 입자의 제거 효율을 증대시키기 위해서는 응집과 같은 추가 공정이 필요하다고 판단되었다. 모델링 결과를 통하여 압축률이 증가할수록 여과 초기에 입자 크기에 따른 TSS 제거효율이 두드러지게 나타남을 관찰하였고, 유입수의 입자크기분포에서 가장 큰 비율을 차지하는 $10{\mu}m$ 크기 입자를 대상으로 높이에 따른 모델링 결과 높이 150-300 mm 여재층이 해당 입자 크기에 대하여 가장 활발한 여과 작용을 나타내었다.
본 연구에서는 다양한 pH 조건에서 활성탄에 Fe(III)를 첨착시킨 Fe-첨착활성탄(Fe-AC)을 제조하였으며, 제조한 Fe-AC가 여과재질로서의 안정성 유무를 파악하기 위하여 내산성 실험을 수행하였다. 또한 Fe-AC의 중금속 제거능을 파악하기 위하여 회분식 실험을 수행하였다. 흡착제의 안정성 실험결과 모든 pH 조건에서 시간이 지날수록 철의 용출량은 증가하였지만, 용액의 pH가 높아질수록 철의 용출량이 감소하는 경향을 보였다. 용액의 pH를 2로 고정했을 경우 시간경과에 따라 철의 용출량이 점차 증가하여 12시간 후에는 Fe-AC에 함유된 총 철 함량의 13%가 용출되었으나, pH 3 이상에서 철의 용출량은 급격히 줄어들어서 무시할 수 있는 것으로 나타났다. 회분식 실험결과 Fe-AC에 의한 Cu(II) 제거는 흡착질의 농도가 감소하고 용액의 pH가 증가할수록 Cu(II)의 제거율은 증가하는 경향을 보였다. 모델링에 의한 흡착결과 예측은 이중확산층 이론에 의한 inner-sphere type의 표면착물화를 고려하여 MINTEQA2 프로그램을 사용하여 실시하였다.
Background: In 2006, the European Union (EU) has decided to forbid use of antibiotics as growth promoters. Although many researches had been conducted about fiber source as alternatives of antibiotics, there are still lack of reports in the literature about the optimum level of sugar beet pulp supplementation, affecting growth performance and nutrient digestibility in weaning pigs. Therefore, different level of sugar beet pulp was added to diets to determine the effects of sugar beet pulp supplementation on growth performance, nutrient digestibility, fecal microflora, blood profile and incidence of diarrhea in weaning pigs. Methods: A total of 200 weaning pigs [$(Yorkshire{\times}Landrace){\times}Duroc$], averaging $9.01{\pm}1.389kg$ of initial body weight were, allotted to 5 treatments in a randomized complete block (RCB) design. Each treatment was composed of 4 replicates with 10 pigs per pen. The treatments were control treatment: Corn-SBM basal diet + ZnO (phase 1: 0.05%; phase 2; 0.03%) and four different levels of sugar beet pulp were supplemented in Corn-SBM basal diet (3, 6, 9 or 12%). Two phase feeding programs (phase 1: 1-2 weeks; phase 2: 3-5 weeks) were used for 5 week of growth trial. Results: In feeding trial, there were no significant differences in growth performance and incidence of diarrhea among treatments. The E.coli counts were not significantly different among dietary treatments but linear response was observed in Lactobacillus counts as sugar beet pulp supplementation increased (P < 0.05). In addition, IGF-1, IgA and IgG were not affected by dietary treatments. However, the BUN concentration was decreased when pigs were fed the treatments of diets with SBP compared to that of control treatment (P < 0.05). In nutrient digestibility, crude fiber and NDF digestibilities were improved as the sugar beet pulp increased (P < 0.05). However, digestibilities of crude ash, crude fat, crude fiber and nitrogen retention were not affected by dietary sugar beet pulp levels. Conclusion: This experiment demonstrated that sugar beet pulp can be supplemented in weaning pigs' diet instead of ZnO to prevent postweaning diarrhea without any detrimental effect on growth performance.
토양으로부터 생전분을 분해하는 세균을 분리하였으며, Bacillus sp.로 동정하였다. 분리한 세균으로부터 생전분 분해효소의 생성을 위한 최적조건을 검토하였다. 세균은 탄소원으로 wheat 및 soluble starch를, 질소원으로 polypeptone을 사용했을 때 최대 효소생성을 얻을 수 있었다. 그리고 탄소원을 0.5, 질소원을 0.5 수준으로 사용했을 때 효소 생성을 극대화 하였다. 그리고 배지의 초기 pH를 6.5, 배양온도를 $35^{\circ}C$로 유지했을 때 효소생성이 높았다. Seepharose CL-6B 젤 여과 및 DEAE-Sephacel 이온교환수지를 사용하여 부분정제한 효소활성의 최적조건은 pH6.5, 온도 $70^{\circ}C$ 였다.
