근관내 주요 혐기성 병인균에 대한 수종 항생제와 근관충전용 세멘트의 항균효과에 관한 연구 (ANTIMICROBIAL EFFECT OF ANTIBIOTICS AND ROOT CANAL CEMENTS ON THE PREDOMINANT PATHOGENIC ANAEROBIC MICROFLORA IN ROOT CANALS)

  • The purpose of this study was to evaluate the susceptibility of anaerobic microorganisms to certain antibiotics and root canal cements. Prevotella intermedia(Bacteroides intermedius) ATCC 25611(serotype A), Fusobacterium nucleatum ATCC 25586, Actinomyces viscosus ATCC 15987 which are the predominant pathogenic anaerobes in dental root canals were cultured in BHI for 48 hours(Fig.1). After each $200{\mu}l$ of those broths with microorganisms was streaked on each surface of blood agar plate, 2 to 5 antibiotic discs which are impregnated with Tetrncycline, Erythromycin, Ampicillin, Clindamycin, or Vancomycin were applied on each surface of blood agar plate and cultured for 5 days anaerobically in the anaerobic chamber (Fig.2). 15 antibiotic discs for each kind of antibiotics and each species of microorganisms were tested. Also each kind of root canal cement tubes which include Zinc oxide eugenol cement, Zinc phosphate cement, Calcium hydroxide powder+DD.W., Calcium hydroxide paste(Pulpdent Tempcanal), or Vitapex(Table 1) were applied on the inoculated BAPs after $200{\mu}l$ of each experimental species of microorganisms was streaked on the surface of blood agar plates, and they were cultured for 5 days anaerobically in the anaerobic chamber(Fig.3). The sensitivity(antimicrobial effect) was determined by the diameter of the inhibition zone. The results are as follows: 1. The results of antibiotic susceptibility test(Table 2) 1) All of the tested antibiotics had antimicrobial activity with various degrees. 2) In Prevotella intermedia (old Bacteroides intermedius), the diameter of inhibition zone to Erythromycin($37.87mm{\pm}2.20$) was largest, those to Tetracycline($26.20mm{\pm}2.96$), Vancomycin($21.53mm{\pm}1.96$), Clindamycin($18.73mm{\pm}0.96$) was smaller than former orderly, and That to Ampicillin ($7.87mm{\pm}0.83$) was smallest. 3) In Actinomyces viscosus, the diameter of inhibition zone to Erythromycin($28.73mm{\pm}1.22$) was largest, those to Ampicillin($21.73mm{\pm}1.03$), Clindamycin($21.33mm{\pm}1.59$) was similarly next order, that to Vancomycin($19.00mm{\pm}1.96$) was smaller than Clindamycin, and that to Tetracycline($11.93mm{\pm}0.70$) was smallest. 4) In Fusobacterium nucleatum, the diameter of inhibition zone to Ampicillin($31.07mm{\pm}1.91$) was largest, that to Erythromycin($28.87mm{\pm}0.92$), Clindamycin($20.47mm{\pm}1.51$), Vancomycin ($16.73mm{\pm}0.96$), Tetracycline ($12.13mm{\pm}1.06$) are smaller than former orderly. 2. The results of root canal cements and pastes(Table 3) 1) The external diameter of tube is 4mm, so 4mm of the inhibition zone diameter means non-susceptable. Prevotella intermedia (old Bacteroides intermedius) was non-susceptable to Calcium hydroxide powder+D.D.W., Calcium hydroxide paste(pulpdent Tempcanal), and Actinomyces viscosus was non-susceptable to Zinc phosphate cement, Calcium hydroxide powder + D.D.W., Calcium hydroxide paste(pulpdent Tempcanal). 2) In Prevotella intermedia (old Bacteroides intermedius), the diameter of inhibition zone to Zinc oxide eugenol cement($13.67mm{\pm}3.30$) was largest, that to Vitapex($9.20mm{\pm}2.96$), Zinc phosphate cement($6.13mm{\pm}2.07$) was smaller than former. 3) In Actinomyces viscosus, the diameter of inhibition zone to Zinc oxide eugenol cement($17.40mm{\pm}5.20$) was largest and that to Vitapex($8.80mm{\pm}1.70$) was next order. 4) In Fusobacterium nucleatum, the diameter of inhibition zone to Vitapex($42.33mm{\pm}17.2$) was largest and those to Calcium hydroxide paste(Pulpdent Tempcanal)($14.47mm{\pm}3.72$) and Zinc oxide eugenol cement($8.93mm{\pm}2.71$), Zinc phosphate cement($8.20mm{\pm}2.27$), Calcium hydroxide powder+D.D.W.($5.53mm{\pm}2.10$)was next orderly. And then In Zinc oxide eugenol cement and Zinc phosphate cement group, two of fifteen samples showed no inhibition zone, in Calcium hydroxide powder + D.D.W. group, 8 of 15 samples showed no inhibition zone.

황금추출물이 함유된 Cosmeceuticals의 제조: 유화안정성 및 항균특성 최적화 (Preparation of Cosmeceuticals Containing Scutellaria baicalensis Extracts: Optimization of Emulsion Stability and Antibacterial Property)

  • 천연물인 황금추출물과 천연유화제인 올리브 유화왁스를 사용하여 제조된 cosmeceuticals의 유화안정성 및 대장균에 대한 항균능력을 최적화하였다. 독립변수로는 황금추출물 첨가량과 올리브 유화왁스의 첨가량으로 설정하였으며, 반응치로는 제조한 cosmeceuticals의 유화안정도지수(ESI)와 대장균 억제직경을 설정하였다. CCD-RSM을 통하여 95% 신뢰구간 내에서 통계학적으로 합리적이며 신뢰성이 우수한 회귀방정식을 얻었다. 다중반응의 최적화를 통하여 유화안정도지수와 대장균 억제직경을 동시에 만족하는 최적 유화조건으로 황금추출물 첨가량은 3.7 wt%, 올리브 유화왁스의 첨가량은 2.7 wt%이었으며, 산출된 유화안정도지수와 대장균 억제직경의 예측값은 각각 97.9%와 9.7 mm로 나타났다. 최적 조건에서 실제 실험을 진행한 결과 유화안정도지수 및 대장균 억제직경은 각각 95.0%와 9.4 mm로 측정되어 다중반응에 의한 예측값과의 평균오차율은 3.2 ± 0.4%로 나타났다.

한국산 마늘에 의한 Aspergillus parasiticus의 성장 억제 효과 (Inhibitory Effect of Garlic on the Growth of Aspergillus parasiticus)

  • This study was performed to investigate the possible effect of garlic produced in Korea on the inhibition/reduction of growth of A. parasiticus, a toxigenic strain. The effect was studied using different concentrations of freeze-dried garlic in potato-dextrose agar (PDA) and/or in yeast-extract sucrose (YES) broth at $25^{\circ}C$ for 15 days. While inhibition of the fungal growth due to increasing the concentration of garlic was observed, the more remarkable effect was observed on the ninth day. Reduction of fungal diameter as a result of addition of garlic on PDA was observed to range between 3.4% to 20.1 % while reduction of mycelial weight in YES broth ranged from 9.9% to 30.5%. The 0.5% and 1.0% concentrations of garlic significantly reduced fungal diameter in PDA on the 9th day, while 0.1 %, 0.5%, and 1.0% concentrations of garlic significantly reduced the mycelial weight in YES broth (p<0.05). Dose-response relationships were observed between the concentration of garlic and inhibition of growth both in solid culture and in liquid culture. This study indicates that garlic could be an effective inhibitor at a human consumption level of the growth of A. parasiticus. More research is needed to study the inhibitory effects of the main active component of garlic.

Antimicrobial Persistence of Silver Diamine Fluoride and Silver Fluoride against Streptococcus mutans

  • Purpose: To evaluate the antimicrobial persistence of silver diamine fluoride (SDF) and silver fluoride (AgF) on Streptococcus mutans. Materials and Methods: An in vitro experiment was conducted to observe changes in the diameter of the inhibition zone of various materials, including AgF (Riva Star AquaTM step 1; SDI), potassium iodine (Riva star aquaTM step 2; SDI), Fluor protector® (FP, Ivoclar Vivadent), SDF (Riva starTM step 1; SDI), Ampicillin (Sigma-Aldrich), Amphotericin B (Nexstar) and negative control on S. mutans. Result: SDF, AgF and FP exhibited significant antimicrobial persistence over the 4 weeks period (P<0.05). At day 28, the diameter of inhibition zone was larger in SDF than in AgF. Conclusion: SDF and AgF have significant antibacterial durability against bacteria commonly associated with dental caries, with the antimicrobial effect lasting for at least 4 weeks. Further clinical studies are needed to validate these findings in vivo.

좌금환과 수련환의 항균, 항산화 및 미백 효과 (Antibiotic, Antioxidant and Whitening effects of Jwa Kum-Whan and Soo Ryeon-Whan)

  • Objectives : The purpose of this study was to research the antibiotic, antioxidant and whitening effects of Jwa Kum-Whan(JKW) and Soo Ryeon-Whan(SRW). Both Jwa Kum-Whan and Soo Ryeon-Whan are composed of Coptidis rhizoma and Evodiae fructus, but the ratios of the two species are different. JKW is composed of Coptidis rhizoma and Evodiae fructus by ratio of 6:1 and SRW's ratio is 1:1. Methods : Antibiotic activities of JKW and SRW's water extracts were studied by paper disc diffusion method. Four kinds of bacteria were applied in paper disc, and each extract was dropped, individually. The diameter of inhibition zone was measured. DPPH assay was used for studying free radical scavenging activity. DPPH is quantitatively decolorized from purple color by antioxidant material. Change of color was measured by spectrophotometer. Whitening activity was analyzed by tyrosinase inhibition assay. Tyrosinase is major enzyme for control process of making eumelanin. Optical Density was measured by spectrophotometer. Results : Candida albicans and Staphylococcus aureus's inhibition zone were made large available by Both prescriptions. Inhibition zone's diameter of JKW was preferable to SRW's. Radical scavenging activity was better at SRW, but JKW's activity was available, too. Tyrosinase inhibition activity was found out available only for JKW, not for SRW. Conclusions : JKW had antibiotic effects for Candida albicans, Staphylococcus aureus. And had antioxidant, whitening effects. SRW had antibiotic, antioxidant activities but not whitening effect.

상온에서 보관한 반코마이신 함유 항생제 시멘트의 메티실린 내성 포도상구균에 대한 항균력 조사 (In Vitro Study Evaluating the Antimicrobial Activity of Vancomycin-Impregnated Cement Stored at Room Temperature in Methicillin-Resistant Staphylococcus aureus)

  • Purpose: Treatment of diabetic foot infection due to methicillin-resistant Staphylococcus aureus (MRSA) remains challenging. Applying vancomycin-impregnated cement is one of the best methods of treatment. Vancomycin-impregnated cement has been used worldwide; however, to date, there is a limited number of studies regarding its use. We evaluated the duration of antimicrobial activity of vancomycin-impregnated cement stored at room temperature after manufacturing. Materials and Methods: The vancomycin-impregnated cement was manufactured by mixing 1 g of vancomycin with 40 g of polymer and adding 17.90 g of liquid monomer. The cement dough was shaped into flat cylinders with diameter and height of 6 mm and 2 mm, respectively. Another cement of the same shape without mixing vancomycin was prepared as the negative control. All manufactured cements were sterilized with ethylene oxide gas and stored at room temperature. Each cement was placed on Mueller Hinton agar plate lawned with standard MRSA strain. Standard vancomycin disk and gentamicin disk were placed together. After 24 hours, the diameter of inhibition zone was measured, and if the diameter was less than 15 mm, vancomycin-impregnated cement was regarded as a loss of antimicrobial activity. The study was repeated every 2 weeks until vancomycin-impregnated cements lost their antimicrobial activity. Results: Vancomycin-impregnated cement stored for a duration of 16 weeks created a 14 mm inhibition zone, while vancomycin disk created a 15 mm inhibition zone. Vancomycin-impregnated cement stored for a duration of 17 weeks created 7 mm and 9 mm inhibition zones, while vancomycin disk created 16 mm and 15 mm inhibition zones, respectively. Conclusion: We found a decrease of antimicrobial activity in vancomycin-impregnated cements after 16 weeks. After 17 weeks, they showed definite loss of antimicrobial activity. Therefore, we recommend not using vancomycin-impregnated cement spacers that has been stored for more than 16 weeks at room temperature.

Inhibition of Quorum Sensing Regulated Virulence Factors and Biofilm Formation by Eucalyptus globulus against Multidrug-Resistant Pseudomonas aeruginosa

  • Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.

생장조절체 처리가 원통형 종이포트묘의 도장 억제 및 근권부 발달에 미치는 영향 (Effects of Different Application Approaches with Diniconazole on the Inhibition of Stem Elongation and the Stimulation of Root Development of Cylindrical Paper Pot Seedling)

  • 본 연구는 원통형 종이포트 토마토 육묘시 Diniconazole의 처리방법이 도장억제 및 근권발달에 미치는 영향을 검토하기 위하여 수행되었다. 그 결과, 엽면적, LAR, 초장, 충실도, 생체중, RGR 및 R/S 에서 시험구간 유의한 차이를 보였다. 동일한 농도를 처리했을 경우, 근권부와 지상부의 흡수도 차이로 인해 저면관수가 엽면살포에 비해 도장억제에 효과적이었다. 저면관수는 엽면시비의 10분의 1의 농도만으로도, 20~30%정도의 동일한 도장억제 효과를 얻을 수 있었다. 디니코나졸 처리에의한 근권부 반응이 흥미로웠는데, 저면관수시 총근장, 근권부피, 평균 근경 및 근단수가 증가하였다. 특히, 0.3mm 이하의 초미세근이 감소하고 0.3~0.6mm의 세근이 증가하였다. 따라서 원통형 종이포트 육묘시 저면관수를 하는 것이 기존 엽면시비에 비해 사용량이 적으면서도 도장억제 및 근권부 활착률을 높힐 수 있을 것으로 판단된다.

전나무와 잣나무 잎 추출물의 대장균과 황색포도상구균에 대한 항균특성 (Antibacterial Properties of Extracts from Abies holophyllaand Pinus koraiensisNeedles Against Escherichia coli and Staphylococcus aureus)

  • 본 연구에서는 전나무와 잣나무 잎으로부터 항균특성을 갖는 기능성 물질을 추출한 후 반응표면분석법 중 중심합성 계획법(CCD-RSM)을 이용하여 최적화하였다. 최적 추출 조건은 전나무의 경우 추출온도(59.5 ℃)와 주정/초순수 부피비(69.5 vol.%)이었으며, 이때 반응치는 추출수율(13.5%), 대장균 억제직경(11.6 mm), 황색포도상구균 억제직경(9.3 mm)로 산출되었다. 또한 잣나무의 경우에는 최적 추출조건인 추출온도(59.2 ℃)와 주정/초순수 부피비(67.8 vol.%)에 서 추출수율(4.8%), 대장균 억제직경(7.9 mm), 황색포도상구균 억제직경(12.5 mm)로 산출되었다. 각 천연물의 최적조건에서 실제 실험 결과 전나무 잎의 추출수율은 13.0%, 대장균 억제직경은 11.7 mm, 황색포도상구균 억제직경은 9.2 mm로 약 ± 2.30% 오차율을 나타내며, 잣나무 잎의 추출수율은 5.1%, 대장균 억제직경은 7.5 mm, 황색포도상구균 억제직경은 12.3 mm로 ± 4.23% 오차율을 나타내었다.

시험 방법에 따른 벌꿀의 항균 특성 (Antibacterial Activities of Honey Measured With Different Test Methods )

  • 배경: 벌꿀의 항균 특성은 시험 검사법에 따라 영향을 받으나, 검사 방법에 따른 차이에 대해 알려진 바가 거의 없다. 목적: 시험 검사법에 따라 벌꿀의 항균 특성을 평가하기 위함이다. 방법: 천연 벌꿀 시료에 대해 3가지 정량적인 시험 검사법 (최소억제농도시험(Minimum Inhibition Concentration test: MIC test), 억제대 측정법(Zone Diameter of Inhibition test: ZDI test or Halo Test) 및 ASTM E2315 Time-Kill Assay)을 이용하여, 벌꿀의 희석 농도에 따른 항균력을 비교 평가했다. 결과: 최소억제농도시험에서는 대장균(E.coli) 및 황색포도상구균(S.aureus) 모두 25% 희석 벌꿀 농도에서 균의 성장이 억제되는 것을 확인했다. 억제대 측정법에서는 대장균 및 황색포도상구균 모두 원액에서부터 억제대가 확인되지 않았다. 대장균 균주에 대한 Time-Kill assay에서 50% 희석 벌꿀 농도에서 3 로그 감소(99.9% 감소) 이상의 항균 효과를 보였고 6.25% 희석 벌꿀 농도에서부터 < 0.3 이하의 로그 감소(약 50% 이하 감소)를 확인했다. 황색포도상구균 균주에 대한 Time-Kill assay에서 50% 희석 벌꿀 농도에서 0.5 로그감소(68.4% 감소)를 확인했고 25% 희석 벌꿀 농도에서부터 < 0.3 이하의 로그 감소(약 50% 이하 감소)를 확인했다. 결론: 벌꿀의 항균 특성은 시험 방법에 유의한 차이를 보이는 것으로 나타났다. 벌꿀의 항균 효과를 활용하기 위해서는 표준화된 정량적인 향균 시험 검사 기술 확립이 필요하다.
