麻黃辛芎散의 卽時型 알레르기 反應 抑制 效果에 關한 硏究 (Studies on Inhibitory Effect of Immediate-Type Allergic Reaction of Mahwang-Shin-Gung-San)

  • Mahwang-Shin-Gung-San has been found inhibiting the mast cell-mediated anaphylactic reaction. This report describes an inhibitory effect of Mahwang-Shin-Gung-San (MSGS) on the immediate-type cutaneous allergic reactions. MSGS has concentration -dependently inhibited the ear swelling response induced by compound 48/80 in mouse by intradermal injection. The mast cells in mouse ear tissue undergone ear-swelling response by compound 48/80 were stained by alcian blue/nuclear fast red. MSGS significantly inhibited the compound 48/80-induced degranulation from mast cells in ear tissue. MSGS concentration-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. We also studied the effect of MSGS on mast cell-dependent passive cutaneous anaphylaxis (PCA) activated by dinitrophenyl IgE antibody. MSGS showed potent inhibition of PCA by oral administration. These results indicate that MSGS inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

The Activity of Apo-transferrin on the Growth of Staphylococcus pseudintermedius

  • Apo-transferrin is an iron-binding protein that has been reported to have an antimicrobial effect. It is considered a major component of the host defense mechanism as it limits microbial access to iron. This study was performed to investigate whether bovine apo-transferrin would have an inhibitory effect on the growth of S. pseudintermedius, which is one of the most isolated bacteria from dogs, and to compare the antimicrobial efficacy with bovine holo-transferrin. S. pseudintermedius were grown at $37^{\circ}C$ in 96-well culture plates using Muller Hinton broth containing bovine apo-transferrin or bovine holo-transferrin at concentrations ranging from 0.5 or 2.5 to 5.0 mg/ml. The optical densities of the wells were then measured at 570 nm. In this study, the apo-transferrin showed dose-dependent antimicrobial effect against S. pseudintermedius while holo-transferrin did not inhibit the growth of S. pseudintermedius effectively. The results suggest that iron deprivation is an important pathway for inhibiting bacterial growth and bovine apo-transferrin has great antimicrobial effects against S. pseudintermedius.

Erythrinae Cortex inhibits Synthesis of Inflammatory Cytokines induced by IL-1$\beta$ and TNF-$\alpha$ in Cultured Human Synovial Cells

  • Our study shows that EC extract has inhibitory effect on pro-inflammatory cytokines such as TNF-α, IL-1, IL-6, iNOS and COX2 in hFLSs. IL-1β, IL-6, iNOS and COX2 mRNA expression is suppressed at a low dasage (1㎍/ml) of EC extract. TNF-α was also suppressed at higher dosages (10 ㎍/ml, 100㎍/ml). EC extract also inhibited TNF-α, IL-1β and IL-6 production in pro-inflammatory cytokine stimulated-hFLSs. Expecially IL-1β(p<0.05) production are suppressed significantly. On the other hand, EC extract did not show any cytotoxicity. Thses data suggest that EC extract has anti-inflammatory effect mostly by inhibiting IL-1β production, and thus could be used to prevent or treat some inflammatory disease such as RA. It remains to be known what are the major components responsible for anti-inflammatory effect and what is the main mechanism.

Genistein이 유방암예방 활성에 미치는 영향 (Effect of Genistein on Chemopreventive Activity of Human Brest Cancer)

  • Genistein was tested for chemopreventive potential against breast cancer by measuring the effect on proliferation of human breast cancer cells, human placental aromatase activity and cyclooxygenases-2 (COX-2) expression and activity, Genistein inhibited the growth of estrogen-independent MDA-MB-231 human breast cancer cell. However, there is no inhibitory effect of genistein on human placental aromatase activity. The expression of COX-2 was inhibited by genistein in Western blot analysis. Genistein significantly inhibited COX-2 activity at the concentrations of 10 (p<0.05), 25 (p<0.05) and 50 ${\mu}M$ (p<0.01). These results suggest that genistein may have breast cancer chemopreventive potential by inhibiting the growth of human breast cancer cell and expression and activity of COX-2.

Effect of Packaging Methods on Colour, Lipid Quality and Microbial Growth of Beef Patties Enhanced with Flaxseed Flour

  • In this study, the effect of packaging methods [aerobic packaging (AP), vacuum packaging (VP) and modified atmosphere packaging (MAP: 75% $N_2$, 25% $CO_2$)] on colour, lipid quality and microbial growth of beef patties enhanced with flaxseed flour was investigated during storage at $2{\pm}1^{\circ}C$ for 10 d. L and a values of beef patties packaged in MAP and VP were higher (p<0.05) than that of the samples packaged in AP. Packaging in MAP and VP retarded the lipid oxidation (TBA value) and inhibited the bacterial growth of beef patties enhanced with flaxseed flour. Furthermore, TBA values in beef patties were correlated with a values (r = -0.340; p<0.05). Packaging in MAP was more effective than packaging in VP for inhibiting microbial growth. The samples packaged in VP lost their shape due to the compression by external atmosphere. Packaging treatment had no significant effect on saturated fatty acid (SFA), monounsaturated fatty acid (MUFA) and polyunsaturated fatty acid (PUFA) levels in beef patties. The a and b, TBA and MUFA values changed during storage time. TBA values for beef patties increased during storage time, but did not reach to the limit value (1 mg/kg) until the end of the storage time. The results suggest that the shelf life of beef patties enhanced with flaxseed flour can be extended by packaging in MAP.

Antiplatelet activity of esculetin through the down-regulation of PI3K/MAPK pathway

  • Among the different cardiovascular disorders (CVDs), the activation of platelets is a necessary step. Based on this knowledge, therapeutic treatments for CVDs that target the disruption of platelet activation are proving to be worthwhile. One such substance, a bioactive 6,7-dihydroxy derived from coumarin, is 6,7-Dihydroxy-2H-1-benzopyran-2-one (esculetin). This compound has demonstrated several pharmacological effects on CVDS as well as various other disorders including diabetes, obesity, and renal failure. In various reports, esculetin and its effect has been explored in experimental mouse models, human platelet activation, esculetin-inhibited collagen, and washed human platelets exhibiting aggregation via arachidonic acid. Yet, esculetin affected aggregation with agonists like U46619 or thrombin in no way. This study investigated esculetin and how it affected human platelet aggregation activated through U46619. Ultimately, we confirmed that esculetin had an effect on the aggregation of human platelets when induced from U46619 and clarified the mechanism. Esculetin interacts with the downregulation of both phosphoinositide 3-kinase/Akt and mitogen-activated protein kinases, important phosphoproteins that are involved in activating platelets and their signaling process. The effects of esculetin reduced TXA2 production, phospholipase A2 activation, and platelet secretion of intracellular granules (ATP/serotonin), ultimately causing inhibition of overall platelet aggregation. These results clearly define the effect of esculetin in inhibiting platelet activity and thrombus formation in humans.

  • Platelet activation plays a major role in cardiovascular disorders (CVDs). Thus, disrupting platelet activation represents an attractive therapeutic target on CVDs. Esculetin, a bioactive 6,7-dihydroxy derivative of coumarin, possesses pharmacological activities against obesity, diabetes, renal failure, and CVDs. In other report, the effect of esculetin has been examined in human platelet activation and experimental mouse models, and esculetin inhibited collagen- and arachidonic acid-induced platelet aggregation in washed human platelets. However, it had no effects on other agonists such as thrombin and U46619, and its mechanism is not also clearly known. This study investigated the effect of esculetin on collagen-induced human platelet aggregation, and we clarified the mechanism. Esuletin has effects on the down regulation of PI3K/Akt and MAPK, phosphoproteins that act in the signaling process in platelet aggregation. The effects of esculetin reduced of TXA2 production and phospholipase A2 activation, and intracellular granule secretion including ATP and serotonin, leading to inhibit platelet aggregation. These results clearly clarified the effect of esculetin in inhibiting platelet activity and thrombus formation in humans.

Sinapic Acid Ameliorates REV-ERB α Modulated Mitochondrial Fission against MPTP-Induced Parkinson's Disease Model

  • Parkinson's disease (PD) is the second most common neurodegenerative disease worldwide, and accumulating evidence indicates that mitochondrial dysfunction is associated with progressive deterioration in PD patients. Previous studies have shown that sinapic acid has a neuroprotective effect, but its mechanisms of action remain unclear. The neuroprotective effect of sinapic acid was assayed in a PD mouse model generated by the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) as well as in SH-SY5Y cells. Target protein expression was detected by western blotting. Sinapic acid treatment attenuated the behavioral defects and loss of dopaminergic neurons in the PD models. Sinapic acid also improved mitochondrial function in the PD models. MPTP treatment increased the abundance of mitochondrial fission proteins such as dynamin-related protein 1 (Drp1) and phospho-Drp1 Ser616. In addition, MPTP decreased the expression of the REV-ERB α protein. These changes were attenuated by sinapic acid treatment. We used the pharmacological REV-ERB α inhibitor SR8278 to confirmation of protective effect of sinapic acid. Treatment of SR8278 with sinapic acid reversed the protein expression of phospho-Drp1 Ser616 and REV-ERB α on MPTP-treated mice. Our findings demonstrated that sinapic acid protects against MPTP-induced PD and these effects might be related to the inhibiting abnormal mitochondrial fission through REV-ERB α.

Streptococcus mutans에 대한 홍삼박 n-hexane 추출물과 Arginine의 병용 효과 (Combinatorial effects of arginine and n-hexane extract from Korean red ginseng marc against Streptococcus mutans)

  • 본 연구에서는 항충치 활성을 보이는 지용성의 홍삼박 n-hexane 추출물과 수용성의 arginine의 병용이 S. mutans의 생육에 미치는 영향을 조사하였다. Checkerboard assay로 분석한 결과, 홍삼박 n-hexane 추출물과 arginine은 fractional inhibitory concentration index 0.396으로 S. mutans의 생육 저해에 시너지 효과를 나타내었다. 홍삼박 n-hexane 추출물과 arginine의 병용은 핵산 성분의 유출과 생균수의 감소를 초래하였으며, 이는 모두 홍삼박 n-hexane 추출물의 농도에 비례하였다. 결론적으로 S. mutans의 생육 저해에 대한 홍삼박 n-hexane 추출물과 arginine의 시너지 효과는 주로 홍삼박 n-hexane 추출물이 기여하는 것으로 판단된다.

고수 에탄올 추출물의 고지방식이 비만 동물모델에서의 항비만효과 (Ant-Obesity Effect of Coriandrum sativum L. Ethanol Extract in High Fat-Induced Obesity Animal Model)

  • This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.
