Study on the Anti-Cancer Effect of Acanthopanax Sessiliflorus (오가피(五加皮)의 항암효과에 관한 연구)

  • Objectives: Hepatoma is a very serious disease in Korea and vvorldwiclc. Hepatitis B vims (HBV) has proved the most significant cause of hepatoma. We canied out this study to investigate the effect of Acanthopanax sessilifloms on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell line and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT/3T3-HBx ceIl. Methods: To investigate the anti-cancer effect of Acanthopanax sessiliflorus, we did the CellTiter 96 Aqueous Non-radioactive Cell Proliferation assay (Promega); MTS/PMS assay, [$^3H$]-thymicline incorporation assay, and we measured the gene expression through westem blotting. Results: Acanthopanax sessiliflorus showed an inhibiting effect on the increase of HepG2.2.15 in the NTS/PMS assay. It also showed an inhibiting effect on DNA synthesis of HepG2.2.15 in the [$^3H$]-thymidine incorporation assay. Acanthopanax sessiliflorus showed an inhibiting effect of phosphorilation of MAP kinase in HBV - X genes. too. Conclusions: The results suggested that this herb had an anti cancer effect. We may discover an effective anti-cancer herb medicine through further studies on this herb medicine.

  • Objectives: Hepatoma is a very serious disease in Korea and worldwide. Hepatitis B virus (HBV) has proved the most significant cause of hepatoma. We carried out this study to investigate the effect of Injinchunggan-tang (Yinchenqinggan -tang) on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell lines and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT /3T3-HEx cells. Methods: First we confinned the Hepatitis B virus producing ability of HepG2.2.15 cells. To investigate the anti-cancer effect of Injinchunggan-tang (Yinchenqinggan-tang), we did the NTS/PMS assay, [3H]-thymidine incorporation assay and transfection of pcDNA-X. We also measured the gene expression through western blotting. Results: Injinchunggan-tang (Yinchenqing gan tang) showed the suppressing effect of HepG2.2.l5 increase in the MTS/PMS assay and the inhibiting effect of DNA synthesis of HepG2.2.15 in the [3H] thymidine incorporation assay. Injinchunggan-tang (Yinchenqinggan-tang) also showed the inhibiting phosphorilation effect of MAP kinase in HBV -X genes. Conclusions: From the above Injinchunggan-tang (Yinchenqinggan-tang) is thought to have an anti-cancer effect on the hepatoma from HBV. It is suggested that further studies on this prescription would give us a better medicine with an anti-cancer effect.

Effects of Serum Fractions Separated by Molecular Weight on the Development of Mouse Embryos Fertilized In Vitro (분자량에 따라 분획화된 혈청성분이 생쥐 체외수정란의 발생에 미치는 영향)

  • This study was carried out to investigate the inhibiting or promoting effect of fetal bovine serum fractionated by the molecular weight and to examine the effect of reconstruction of serum fractions on the development of 1- and 2-cell mouse embryos fertilized in vitro (IVEE) . The serum was separated by ultrafiltration or gel filtration methods and added in m-KRB medium for culture of IVFE. The developemental ability(cavitation and hatching) of embryos following culture of day 4 and 6 was compared among fractions. Small molecular weight fraction( <3 kDa) significantly inhibited the development of 1-and 2-cell IVFE to the blastocyst stages, compared with other fractions. One-cell IVFE were more sensitively damaged than 2-cell embryos by that fraction and arrested mainly at 2~4 cell stages. Moreover, small amount(<3%,v /v) of the inhibiting fraction acted even with protein rich fraction(100~30 kDa) and arrested the embryonic development. On the other hand, 100~30 kDa fraction promoted the embryonic development and no inhibiting effect was observed at the level of 50%(v /v) in culture medium In the experiment of gel filtraton, =30 kDa fraction showed the highest promoting effect on the embryonic development, but <4 kDa fraction inhibited significantly the development. These results suggest that serum contains not only small molecular weight inhibitory component(s) but also promoting one rather than albumin on embryonic development. And serum can be more effectively used in the IVF program after removal of inhibitory component(s) by one of above separation methods.

Anti-Apoptosis Engineering Using a Gene of Bombyx mori

  • We have previously shown that the addition of silkworm hemolymph to a culture medium increases the longevity of insect and mammalian cells by inhibiting apoptosis. This indicates that the component which inhibits apoptosis is contained in the silkworm hemolymph, The apoptosis-inhibiting component was isolated from silkwonn hemolymph and characterized in our previous study. A database search using the N-terminal amino acid sequence of this component as a template resulted in a 95% homology with a low molecular weight lipoprotein, the so called ’30K protein' of unknown function. In this study, the 30K protein gene was expressed in mammalian and insect cells to confirm the apoptosis-inhibiting effect. The overexpression of 30K protein in mammalian cell inhibited the staurosporin-induced apoptosis by the prevention of the activation of caspase 3. Using an Autographa californicanuclear polyhedrosis virus (AcNPV) system, the 30K protein was overexpressed also in insect cells. The expression of the 30K protein increased the longevity of baculovirus-infected insect cells by inhibiting apoptosis. These results suggest that the 30K protein is a novel anti-apoptotic protein.

Effect on Promoting Gastrointestinal Function and Inhibiting of Decreasing Body Temperature of Ginger Extracts(Zingiber Officinale) (생강(生薑) 추출물의 위장관 기능개선 및 체온저하 억제효과에 대한 실험적 연구)

  • This study was performed to investigate the effect of promoting gastrointestinal function and inhibiting of decreasing body temperature of ginger extract(Zingiber officinale) in rats. In order to elucidate the gastrointestinal function and inhibiting effect of body temperature of native ginger and improved ginger, water extracts of ginger were orally administrated into rats. The results are as follows: The gastrointestinal transit time was significantly decreased in native ginger(7.66hrs) and improved ginger(7.72hrs) extract administrated groups compare to control group(8.44hrs). The mean red faecal weight was increased in native ginger(30.6%) and improved ginger(31.1%) extract administrated groups compare to control group(24.9%) for 24hrs. Inhibiting effect of decreasing body temperature induced by serotonin was increased in native ginger($1.116^{\circ}C$) and improved ginger($1.416^{\circ}C$) extract administrated groups compare to positive control group($0.384^{\circ}C$) during 40 minutes. Gastrin and CGRP immunoreactive density was more strongly expressed in native ginger and improved ginger extract administrated groups compare to control group. Serotonin immunoreactive density was more weakly expressed in native ginger and improved ginger extract administrated groups compare to control group. These results suggest that ginger extracts may enhance physiological activity such as gastrointestinal motility, protection of mucosa and gastric acid secretion in gastrointestinal tracts, and inhibits decreasing body temperature

  • Whitening agents are eagerly demanded especially by oriental women who often suffers from the pigmentary disorders such as melasma and solar lentigines. As these pigmentary disorders are exacerbated by ultraviolet (UV), the whitening agents could exert its effect not only by inhibiting melanin synthesis but also by inhibiting UV activated signals. Eumelanin protects UV-induced DNA damages so that the chemicals which could reduce UV-induced DNA damages might be the ideal lightening agents. The effect of newly synthesized antioxidants, a-tocopheryl ferulate, on protective effect for UV-induced DNA damages as well as inhibiting melanin synthesis are briefly shown. For clinical evaluation, our results of the efficacy of lightening agents on treating pigment macules in combination with chemical peeling are shown. Furthermore, newly developed facial image analyzers to quantitatively evaluate the improvement of pigment macules are introduced.

  • The obtained results are summarized as follows 1. New findings are reporting year by year as for the study related to Anti-inflammatory mechanism of Bee Venom therapy. 2. The Anti-inflammatory effect of Bee Venom therapy is achieved through counterirritation, stimulations to adrenal cortex, immuno-regulation, antioxidation, removal of free radicals, modulation of AGP gene induction. 3. The chief components of Bee Venom related to Anti-inflammatory effect are Melittin, MCD peptide, Apamin, Adolapin etc. 4. Melittin binds to secretory phospholipase A2 and inhibits its enzymatic activity. 5. Melittin blocks neutophil O2-production. 6. MCD peptide(Peptide 401) stimulates the mast cell secrets histamine, Anti-inflammatory effect caused by this is 'conterirritation'. 7. Melittin & Apamin have an anti-inflammatory effect by inducing cortisone secretion. 8. MCD peptide & Apamin increase immunologic fuction by stimulating hypophysis & adrenal cortex and have an anti-inflammatory effect by inhibiting synthesis of prostaglandin from arachidonic acid. 9. Adolapin have an anti-inflammatory effect by inhibiting COX. 10. Bee Venom have an anti-inflammatory effect by suppressing AGP($\alpha$-acid glycoprotein). 11. Bee Venom have an anti-inflammatory effect by inhibiting NO, iNOS, PLA2, COX-2, TNF-$\alpha$, IL-1, NF-${\kappa}B$, MAP kinase.

In Vitro Anti-Helicobacter pylori Activity of Panaxytriol Isolated from Ginseng

  • This study investigated the effect that some polyacetylenes and protopanaxatriol, which were isolated from heated ginseng (family Araliaceae), have on inhibiting Helicobacter pylori (HP) growth. Among the compounds tested, panaxytriol was quite effective in inhibiting HP growth with an MIC of 50 ${\mu}g/ml$. Cinsenoside Rhl and protopanaxatriol weakly inhibited $H^{+}/K^{+}$-ATPase from a rat stomach.

  • Up to now, many crack repair techniques have been developed for inhibiting crack growth in structural components. However, the simplest way for inhibiting crack growth is to apply a indentation at the crack tip or at some distance ahead of the expected crack growth path so as to produce residual compressive stresses that can reduce the effective stresses around the crack tip. In spite of its importance to the aerospace industry, little attention has been devoted to evaluation of the indentation residual stress effect on the fatigue crack initiation life quantitatively. Therefore, in the present work, the magnitude and distribution of the indentation residual stresses were investigated in order to estimate the beneficial effect on fatigue crack initiation by using finite element method. Furthermore, to examine the validity of finite element analysis results, residual stress distribution in the indented specimen was measured by using X-ray diffraction technique, and fatigue crack behavior at fastener hole in aluminum alloy 7075-T6 before and after indentation processes was investigated.

Antimicrobial activity and inhibition effect on the artificial dental plaque formation of herbal medicine extract (생약 추출물의 항균력 및 인공치태형성 억제효과)

  • This study was executed to evaluate the effects of natural herbal medicine extract on the antimicrobial activity and the artificial plaque formation. Schisandra chinensis, Sophora flavescens, Dryopteris crassirhizoma and Aristolochia contorta were extracted by 80% MeOH respectively and each concentration(5mg, 10mg, 20mg) of herbal medicine extract was tested to show the antimicrobial activity against S. mutans. To show the inhibition effect of herbal medicine extract on the artificial dental plaque formation each extract of 0.4% concentration was added into M 17 broth and inoculated with $5.0{\times}10^6$ of S. mutans. After 24 hour incubation each weight of artificial dental plaque was measured and compared with each other. Results obtained are as follows: 1. Dryopteris crassirhizoma showed very intense antimicrobial effect but Schisandra chinensis, Sophora flavescens and Aristolochia contorta showed a little difference against control. 2. Regarding artificial plaque formation inhibiting activity by each herbal extract at 0.4% concentration, Dryopteris crassirhizoma showed the perfect inhibition effect. Schisandra chinensis and Aristolochia contorta followed in order but Sophora flavescens showed the worst value. In conclusion, Dryopteris crassirhizoma showed strong antimicrobial effect against S. mutans and significant inhibiting effect on the artificial dental plaque formation.
