Evaluation of Clubroot Resistance in Chinese Cabbage and Its Inheritance in the European Turnip Line 'IT033820', a New Genetic Resource

  • Clubroot caused by the protist Plasmodiophora brassicae is one of the most destructive diseases of Brassica crops. Developing Chinese cabbage cultivars with durable clubroot resistance (CR) is an important goal of breeding programs, which will require new genetic resources to be identified and introduced. In this study, we evaluated resistance to P. brassicae race 4 using 26 Chinese cabbage (B. rapa ssp. pekinensis ) cultivars compared to the clubroot-susceptible Chinese cabbage inbred line 'BP079' and the clubroot-resistant European turnip (B. rapa ssp. rapifera ) inbred line 'IT033820'. No symptoms of clubroot disease were found in 'IT033820' infected with P. brassicae race 4, whereas the Chinese cabbage cultivars exhibited disease symptoms to various degrees. The Chinese cabbage cultivars that were reported to be clubroot-susceptible were susceptible to P. brassicae race 4; however, seven of the 20 cultivars reported to be clubroot-resistant were susceptible to this race of P. brassicae to varying degrees. Resting spores of P. brassicae were abundant within the infected root tissues of 'BP079', as revealed by light microscopy and scanning electron microscopy (SEM), but they were not detected in root tissues of 'IT033820'. Although resting spores were not detected by light microscopy in root tissues of the clubroot-resistant Chinese cabbage cultivar 'Kigokoro 75', a few spores were observed by SEM. The $F_1$ hybrids from a cross between 'IT033820' and 'BP079' showed no disease symptoms, and all $BC_1P_1$ progenies from a cross between the $F_1$ hybrid and 'IT033820' exhibited a resistance phenotype. In the $BC_1P_2$ population from a cross between the $F_1$ hybrid and 'BP079', this trait segregated at a ratio of 3(R):1(S) (${\chi}^2=1.333$, p = 0.248) at a 5% significance level. Inoculated $BC_1P_2$ plants were either highly resistant or highly susceptible to the pathogen, indicating that the CR to race 4 of P. brassicae carried by 'IT033820' is dominant. In the $F_2$ population, this trait segregated at a ratio of 15(R):1(S) (${\chi}^2=0.152$, p = 0.696) at a 5% significance level, suggesting that CR in 'IT033820' is mainly controlled by two dominant genes. Therefore, 'IT033820' represents a promising genetic resource for developing durable CR breeding lines in Chinese cabbage.

배유 특이 프로모터에 의해 유도된 GUS 유전자의 형질전환 담배 내에서의 발현 및 유전 양상 (Expression and Inheritance Patterns of Gus Gene Driven by an Endosperm-Specific Promoter in Transgenic Tobacco)

  • 본 실험은 형질전환 담배내에서의 배유 특이 promoter에 의한 gus 유전자의 조직 특이적 발현 여부와 전이유전자의 후대로의 유전 양상을 확인하고자 수행하였다. 배유 특이 promoter에 의해 유도되는 gus 유전자(Z4pro-gus)와 kanamycin 저항성유전자를 운반하는 BV3 construct를 A. tumerfaciens을 이용하여 담배 형질전환체를 유기시켰다. 형질전환체 중에서 8개체를 선발하여 nptII primer를 이용하여 PCR을 실시한 결과 8개체 모두에서 700bp의 PCR 산물을 얻을 수 있었다. Promoter에 따른 유전자의 발현양상을 알아보기 위하여 Z4pro-gus가 전이된 형질전환체의 잎과 CaMV35S와 gus 유전자(35Spro-gus)로 구성된 pBI121 construct를 전이시킨 형질전환체의 잎으로부터의 발색정도를 비교하였다. 그 결과 Z4pro-gus가 전이된 형질전환체의 경우 잎에서 매우 부분적으로 극소량 발색되었으나 35Spro-gus가 전이된 형질전환체의 잎에서는 상대적으로 많은 양의 발색정도를 보여 promoter에 따른 발현정도의 차이를 보였다. 보다 명확한 Z4 promoter의 조직 특이 발현 양상을 확인하기 위하여 Z4pro-gus로 형질전환시킨 $T_0$ 식물체를 자가수분하여 얻은 $R_1$ 종자와 35Spro-gus를 형질전환시켜 같은 방법으로 얻은 $R_1$ 종자를 histochemical assay하였다. 그 결과 35Spro-gus로 형질전환된 담배 종자는 절단면 전체에서 gus 유전자가 발현되어 배유뿐만 아니라 종자 내 다른 조직에서도 발색되는 양상을 나타내었으나 Z4pro-gus를 형질전환 시켜 얻은 종자의 경우는 배유 부분만이 조직 특이적으로 파랗게 발색되었고 배 또는 그 이외의 조직에서는 gus 발색이 전혀 관찰되지 않았다. Kanamycin 저항성검정을 실시한 결과 모든 계통에서 전이유전자가 후대로 안정적으로 전이됨을 확인할 수 있었다.

콩 우수 계통 '천알'에서 발견한 역병 저항성 유전자좌 (Identification of a Locus Associated with Resistance to Phytophthora sojae in the Soybean Elite Line 'CheonAl')

  • 콩 역병(Phytophthora root rot, PRR)은 난균(oomycete)인 Phytophthora sojae에 의해 발생하는 콩의 주요 병 중 하나로, 배수가 잘 안 되는 밭이나 습한 토양에서 심하게 발생한다. 역병의 피해를 효과적으로 줄일 수 있는 방법은 주로 역병 저항성 품종을 재배하는 것으로, 이는 저항성 유전자 Rps (resistance to P. sojae)에 대한 연구를 중심으로 이루어진다. 본 연구는 대풍 과 천알(계통명 SS0404-T5-76)을 교배하여 구축한 RIL (recombinant inbred line) 집단을 이용하여 콩 역병 균주40468과 연관된 저항성 유전자좌를 탐색하기 위해 수행되었다. 역병 균주40468에 대한 저항성 평가는 하배축 접종(hypocotyl inoculation) 방법으로 이루어졌다. 저항성 검정 결과, 천알은 저항성,대풍은 감수성을 보였고 집단 내에서는 계통들의 표현형이 분리되는 양상을 보였다. 집단 내에서 표현형 분포는 1:1 (R:S) (χ2 = 0.57, p = 0.75) 분리비와 일치하였으며, 이는 저항성 반응이 단일 유전자에 의해 조절됨을 나타낸다. 대풍, 천알과 각 RIL 계통들은 고밀도 SNP 유전자형 분석을 통해 데이터를 얻었고, 이를 바탕으로 유전자 지도를 작성하였다. 일원분산 분석(Single-marker ANOVA) 및 linkage analysis 결과, 18번 염색체의 55.9~56.4 Mbp에서 높은 통계적 유의성을 보였으며, 이 지역의 표현형 분산은 ~98%로 나타났다. 탐색된 영역은 다수의 선행연구에서 Rps의 위치로 보고된 지역과 겹치며, 콩 표준 유전체 정보를 기반으로 0.5 Mbp 범위 내에서 leucine-rich repeat (LRR) 또는 serine/threonine kinase(STK)을 합성하는 유전자 9개를 포함하고 있다. 천알은 역병 균주40468에 대한 저항성 유전자좌가 밝혀진 첫 국내 콩 품종으로, 본 연구에서 밝힌 천알의 저항성 유전자좌는 향후 역병 저항성 육종 및 연구에서 유용한 재료가 될 것이다.

Identification of a Causal Pathogen of Watermelon Powdery Mildew in Korea and Development of a Genetic Linkage Marker for Resistance in Watermelon (Citrullus lanatus)

  • Watermelon production is often limited by powdery mildew in areas with a large daily temperature range. Development of resistant watermelon cultivars can protect against powdery mildew; however, little is known about the characteristics of its causal agents. Here, we identified the genus and race of a causal pathogen of powdery mildew in Ansung province of South Korea, and developed molecular markers for the generation of resistant watermelon cultivars. The causal pathogen was determined to be Podosphaera xanthii based on multiple sequence alignments of internal transcribed spacers (ITS) of rDNA. The physiological race was identified as 1W, and the Ansung isolate was named P. xanthii 1W-AN. Following inoculation with the identified P. xanthii 1W-AN, we found inheritance of the resistant gene fitting a single dominant Mendelian model in a segregated population ('SBA' ${\times}$ PI 254744). To develop molecular markers linked to fungus-resistant loci, random amplified polymorphic DNA (RAPD) was accomplished between DNA pooled from eight near-isogenic lines (NILs; $BC_4F_6$), originated from PI 254744 and susceptible 'SBB' watermelon. After sequencing bands from RAPD were identified in all eight NILs and PI254744, 42 sequence-characterized amplifiedregion (SCAR) markers were developed. Overall, 107 $F_2$ plants derived from $BC_4F_6$ NIL-1 ${\times}$ 'SBB' were tested, and one SCAR marker was selected. Sequence comparison between the SCAR marker and the reference watermelon genome identified three Nco I restriction enzyme sites harboring a single nucleotide polymorphism, and codominant cleavage-amplified polymorphic site markers were subsequently developed. A CAPS marker was converted to a high-resolution melt (HRM) marker, which can discriminate C/T SNP (254PMR-HRM3). The 254PMR-HRM3 marker was evaluated in 138 $F_{2:3}$ plants of a segregating population ('SBA' ${\times}$ PI254744) and was presumed to be 4.3 cM from the resistance locus. These results could ensure P. xanthii 1W-AN resistance in watermelon germplasm and aid watermelon cultivar development in marker-assist breeding programs.

수도품종의 도열병 저항성 유전분석 제1보 특정 도열병 균계에 대한 저항성 품종들의 저항성 유전분리와 II, VIII, XI 및 XII번 연관과의 관계 (Studies on Genetice of Blast Resistance in Rice L Inheritance of Resistance to Specific Races of Blast Fungus and Relationship between Their Resistance and II, VIII, XI and XII Linkage Groups in Some Rice Varieties)

  • 통일형품종을 침해하는 도열병 균계에 대한 저항성 품종의 저항성 유전현상과 그들 저항성 유전자와 표식유전자와의 연관관계를 구명하기 위하여 7개 저항성 품종과 유묘식별이 용이한 4개 이병성 표식유전자간 조합 F_2집단의 유묘 4~5 엽기에 T-2$^{+t}$ , N-2$^{+t}$ 및 C-8$^{+t}$ 균계를 각각 주사접종하여 저항성의 분리를 조사하고 표식유전자들과의 연관관계를 검토하였다. 그 결과를 요약하면 다음과 같다. 1. Semi-dwarf 검정친들은 Indica 저항성 품종들과 교배되어 평균 74% 정도의 임성을 나타내었다. Carreon 조합은 예외였다. 2 도열병 저항성은 품종이 따라, 균계에 따라 분리양상이 달랐는데 어느 경우에도 저항성이 우성으로 표현되었으며 육성계통보다는 모본품종들에서 더 복잡한 분리를 보였다. 3. Tadukan, Tetep 및 IR 747 품종에서는 균계에 따라 3:1, 9:7, 13:3 및 37:27 등 4가지의 분리비를, Carreon 품종에서는 3:1, 13:3 및 15:1 등 3가지의 분리비를 상정할 수 있었고, Suweon287, Suweon 208 및 Iri 34002 품종에서는 3:1과13:3 두 가지의 분리비를 상정할 수 있었다. 4. 균계에 따른 저항성의 분리는 T-2$^{2t}$ 에 대하여는 조합에 따라 3:1, 13:3 및 15:1의 분리비를, N-2$^{2t}$ 와 C-8$^{+t}$ 에 대하여는 각각 3:1, 13:3, 9:7 및 37:27 의 분리비를 상정할 수 있었다. 5. Suweon287, Suweon288 및 Iri 34002 품종들은 1개의 단순우성 유전자를 가지고 있으며 조합에 따라 억제유전자가 관여하고 있는 것으로 나타났고, Tadukan, Tetep 및 IR 747품종들은 1~3개의 저항성 유전자를 가지고 있으며 조합에 따라 단순우성, 억제, 중복 및 보족작용을 나라내었다. 6 본실험에 공시된 T-2$^{+t}$ 및 C-8$^{+t}$ 균계에 대하여 Tadukan, Tetep, Carreon , Suweon 287 Suweon 288 및 Iri 342 품종들이 가지는 도열병 저항성 유전자는 II(1g), VII(la), XI(be) 및 XII(gl) 연관군과는 상호착립적으로 나타났다.

고추 탄저병 저항성 관련 양적형질 유전자좌 분석 (Identification of Quantitative Trait Loci Associated with Anthracnose Resistance in Chili Pepper (Capsicum spp.))

  • 고추 탄저병은 국내외에서 고추 생산에 있어 많은 피해를 주는 병중의 하나로서, $Colletotrichum$ spp.에 의해 발생하며 현재 가장 많은 빈도를 나타내는 종은 $C.$ $acutatum$으로 보고되고 있다. 살균제에 대한 내성이 있는 것으로 보고되어 저항성 품종의 육성이 요구되어지고 있다. 본 연구의 목적은 $Capsicum$ $baccatum$ 종내 교잡을 통해 고추 탄저병 저항성 육종을 위한 MAS(marker assisted selection) 체계를 개발하는 기초연구로서 저항성과 관련한 QTL분석을 수행하였다. 저항성 검정결과 고추 탄저병저항성은 85%의 높은 광의의 유전력을 보였다. 양적형질 유전자좌 분석 결과 신뢰수준 이상의 LOD 수준을 보인 2개의 QTL($An8.1$, $An9.1$)이 탐색되었고, $R^2$가 2.04%와 14.36%이었다. 유전적인 효과는 $An8.1$은 상가적 효과가 0.46으로, $An9.1$은 상가적 효과가 -4.52로 나타났다. $An8.1$$An9.1$은 상처접종에 의해 나타나는 저항성 반응과 관련이 있는 것으로 생각되었다. 포장검정(08NR)과 관련한 QTL들은 신뢰수준이상의 LOD수준을 보이는 QTL은 탐색할 수 없었으나, $R^2$가 19.9%인 $An3.1$와 30.8%인 $An3.1$을 포함한 5개 QTL의 전체 $R^2$가 60.73%로 나타나 고추 탄저병저항성 유전과 관련한 인자가 다수 존재하는 것을 확인하였다. 따라서 이들 QTL은 더욱 정밀한 집단과 종간교잡 후대에서 적용성을 검정하여 고추 탄저병저항성 품종육성을 위한 MAS체계를 확립할 것이다.

Improvement of Transformation Efficiencies using Agrobacterium-Mediated Transformation of Korean Rice

  • A reproducible transformation system via optimized regeneration media for Korean rice cultivars was established using Agrobacterium tumefeciens LBA4404 (pSBM-PPGN; gusA and bar). Although japonica rice genotypes were easier to produce transgenic plants compared to Tongil type cultivars, transformation efficiencies were not always correlated with regeneration efficiencies of non-transgenic callus on the control medium. Regeneration efficiencies of Donganbyeo, Ilmibyeo, and Manchubyeo were over 50% in non-transgenic control, however, transformation efficiencies were significantly low when only sucrose was added to the media as a carbon source. However, the medium, MSRK5SS-Pr (or MSRK5SM-Pr), that contains $5\textrm{mgL}^{-1}$ kinetin, $0.5\textrm{mgL}^{-1}$ NAA, 2 % sucrose (or maltose), 3% sorbitol, and $500\textrm{mgL}^{-1}$ proline, was the most efficient not only for regeneration of non-transgenic callus but also for regeneration of transgenic callus in the presence of L-phosphinotricin (PPT). Average transformation efficiencies of 16 Korean rice cultivars were significantly enhanced by using the optimized medium from 1.5% to 5.8% in independent callus lines and from 2.9% to 19.4% in tromsgenic plants obained. Approximately 98.9% (876 out of 885) transgenic plants obtained on optimized media showed basta resistance. Stable integration, inheritance and expression of gusA and bar genes were continued by GUS assay and PCR and Southern analysis of the bar gene. With Pst1 digestion of genomic DNA of transgenic plants, one to five copies of T-DNA segment were observed; however, 76% (19 out of 25 transgenic plants) has low copy number of T-DNA. The transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.

Pseudohypoparathyroidism type 1b due to paternal uniparental disomy of chromosome 20q: A case report

  • Pseudohypoparathyroidism type 1b (PHP 1b) is the result of end organ resistance to parathyroid hormone (PTH) in the absence of any features of Albright's hereditary osteodystrophy. There are two subtypes of PHP 1b with different genetic mechanisms. One subtype is related to a maternally derived 3kb microdeletion involving STX 16 gene, and is inherited in an autosomal dominant mode. Familial autosomal dominant inheritance of PHP 1b is relatively rare. The other subtype is associated with more extensive loss of imprinting at the GNAS locus that affects at least one additional differential methylated (hypermethylation at neuroendocrine secretory protein and hypomethylation at antisense transcript and or extra-large stimulatory G protein region) without microdeletion of the STX 16 or AS gene. It can be sporadic due to an imprinting defect in the GNAS gene. In our case, an 8-year-old girl was referred for suspected PHP with no feature of Albright hereditary osteodystrophy. Blood test results revealed hypocalcemia and hyperphosphatemia. Elevated PTH was also checked. There was no family history of endocrine or developmental problem. Her intelligence was normal, but she had inferior sociability at that time. Based on above, we diagnosed a rare case of paternal uniparental disomy of the long arm of chromosome 20 as the cause of PHP 1b by microsatellite marker test of chromosome 20.

Contemporary Issues Surrounding Folic Acid Fortification Initiatives

  • The impact of folate on health and disease, particularly pregnancy complications and congenital malformations, has been extensively studied. Mandatory folic acid fortification therefore has been implemented in multiple countries, resulting in a reduction in the occurrence of neural tube defects. However, emerging evidence suggests increased folate intake may also be associated with unexpected adverse effects. This literature review focuses on contemporary issues of concern, and possible underlying mechanisms as well as giving consideration the future direction of mandatory folic acid fortification. Folate fortification has been associated with the presence of unmetabolized folic acid (PteGlu) in blood, masking of vitamin $B_{12}$ deficiency, increased dosage for anti-cancer medication, photo-catalysis of PteGlu leading to potential genotoxicity, and a role in the pathoaetiology of colorectal cancer. Increased folate intake has also been associated with twin birth and insulin resistance in offspring, and altered epigenetic mechanisms of inheritance. Although limited data exists to elucidate potential mechanisms underlying these issues, elevated blood folate level due to the excess use of PteGlu without consideration of an individual's specific phenotypic traits (e.g. genetic background and undiagnosed disease) may be relevant. Additionally, the accumulation of unmetabolized PteGlu may lead to inhibition of dihydrofolate reductase and other enzymes. Concerns notwithstanding, folic acid fortification has achieved enormous advances in public health. It therefore seems prudent to target and carefully monitor high risk groups, and to conduct well focused further research to better understand and to minimize any risk of mandatory folic acid fortification.

일본(日本)의 임목육종(林木育種) 및 삼림경영연구동향(森林經營硏究動向) (The Tendency of Scientific Research of Tree Improvement and Forest Management in Japan)

  • The direction of scientific researches on tree improvement and forest management in several universities and research institutes in Japan can be summarized as follows: They put a great emphasis on sugi, Cryptomeria japonica and cypress, Chamaecyparus oblusa which are two major conifer species largerly planted in the Japanese forestry. In the research of sugi, a great concern has been made in evaluating inheritance of forest tree, quantitative characters and genetic parameter of growth, and in breeding for resistance to diseases and insects and to all the natural calamities. Interaction between environmental conditions and genetic nature of tree can be concerned factors in relation with forest damage, together with silvicultural conditions and pest infestation. Selfing hybrids of $F_1$ made from crossing twisted-leaf sugi, defomity leaf type and midori sugi, normal leaf type segregated the normal needle, twisted needle, green leaf and albino leaf type. It seemed that separation of many defomity individuals can be governed by two dominant complementary genes and from the near loci of which it was detected lethal genes. 52% of Japanese forestry is occupied by the small forest landowners like Korean forestry. This made difficulty for forest improvement such as progressive afforestation and for capital accumulation form forestry. The Forest Corporation was established at first in 1959 to aming at productive forestry structure and forest management, and afforestation. For these purpose, 35 Forest Corporations are at moment operating throughout Japan. However, investment in forestry business becomes less attractive since the wage in forest production duction increased in higher trend. than timber price. Therefore, an artifical afforestation becomes yearly decreased. At present. the self-sufficient rate of timber production in Japan is about 35%, and so a great effort is being made to increase self-sufficient rate of timber production.
