Comparative Genomic and Genetic Functional Analysis of Industrial L-Leucine- and L-Valine-Producing Corynebacterium glutamicum Strains

  • Corynebacterium glutamicum is an excellent platform for the production of amino acids, and is widely used in the fermentation industry. Most industrial strains are traditionally obtained by repeated processes of random mutation and selection, but the genotype of these strains is often unclear owing to the absence of genomic information. As such, it is difficult to improve the growth and amino acid production of these strains via metabolic engineering. In this study, we generated a complete genome map of an industrial L-valine-producing strain, C. glutamicum XV. In order to establish the relationship between genotypes and physiological characteristics, a comparative genomic analysis was performed to explore the core genome, structural variations, and gene mutations referring to an industrial L-leucine-producing strain, C. glutamicum CP, and the widely used C. glutamicum ATCC 13032. The results indicate that a 36,349 bp repeat sequence in the CP genome contained an additional copy each of lrp and brnFE genes, which benefited the export of L-leucine. However, in XV, the kgd and panB genes were disrupted by nucleotide insertion, which increase the availability of precursors to synthesize L-valine. Moreover, the specific amino acid substitutions in key enzymes increased their activities. Additionally, a novel strategy is proposed to remodel central carbon metabolism and reduce pyruvate consumption without having a negative impact on cell growth by introducing the CP-derived mutant $H^+$/citrate symporter. These results further our understanding regarding the metabolic networks in these strains and help to elucidate the influence of different genotypes on these processes.

Edge Detection System for Noisy Video Sequences Using Partial Reconfiguration (부분 재구성을 이용한 노이즈 영상의 경계선 검출 시스템)

  • In this paper, the Zynq system-on-chip (SoC) platform is used to design an adaptive noise reduction and edge-detection system using partial reconfiguration. Filters are implemented in a partially reconfigurable (PR) region to provide high computational complexity in real-time, 1080p video processing. In addition, partial reconfiguration enables better utilization of hardware resources in the embedded system from autonomous replacement of filters in the same PR region. The proposed edge-detection system performs adaptive noise reduction if the noise density level in the incoming video sequences exceeds a given threshold value. Results of implementation show that the proposed system improves the accuracy of edge-detection results (14~20 times in Pratt's Figure of Merit) through self-reconfiguration of filter bitstreams triggered by noise density level in the video sequences. In addition, the ZyCAP controller implemented in this paper enables about 2.1 times faster reconfiguration when compared to a PCAP controller.

A Mobile Payment System Based-on an Automatic Random-Number Generation in the Virtual Machine (VM의 자동 변수 생성 방식 기반 모바일 지급결제 시스템)

  • A mobile phone has became as a payment tool in e-commerce and on-line banking areas. This trend of a payment system using various types of mobile devices is rapidly growing, especially in the Internet transaction and small-money payment. Hence, there will be a need to define its standard for secure and safe payment technology. In this thesis, we consider the service types of the current mobile payments and the authentication method, investigate the disadvantages, problems and their solutions for smart and secure payment. Also, we propose a novel authentication method which is easily adopted without modification and addition of the existed mobile hardware platform. Also, we present a simple implementation as a demonstration version. Based on virtual machine (VM) approach, the proposed model is to use a pseudo-random number which is confirmed by the VM in a user's mobile phone and then is sent to the authentication site. This is more secure and safe rather than use of a random number received by the previous SMS. For this payment operation, a user should register the serial number at the first step after downloading the VM software, by which can prevent the illegal payment use by a mobile copy-phone. Compared with the previous SMS approach, the proposed method can reduce the amount of packet size to 30% as well as the time. Therefore, the VM-based method is superior to the previous approaches in the viewpoint of security, packet size and transaction time.

A Remote Applications Monitoring System using JINI (JINI 기반 원격 응용 모니터링 시스템)

  • In general, remote monitoring systems monitor the status of distributed hosts and/or applications in real-time for diverse managerial purposes. However, most of the extant systems have a few undesirable problems. First of all, they are platform-dependent and are not resilient to network and/or host failures. Moreover, they normally focus on the resource usage trends in monitored hosts, rather than on the status change of the applications running on them. We strongly believe that the latter has more direct and profound effect on the resource usage patterns on each host. In this paper, we present the design and implementation of the Remote Applications Monitoring System (RAMS) that enables us to effectively manage distributed applications through a real-time monitoring of their respective resource usages. The RAMS is a centralized system that consists of many distributed agents and a single centralized manager. An agent on each host is in charge of collecting and reporting the status of local applications. The manager handles agent registration and provides a central access point to the selection and monitoring of distributed applications. The salient features of the system include robustness and portability The adoption of JINI greatly facilitates an automatic recovery from partial network failure and host failure.

  • Recently, the research of GPS receiver which uses the Software-Defined Radio(SDR) technique is being actively proceeded instead of traditional hardware-based receiver. The software-based GPS receiver indicates that the signal acquisition and tracking treated by the hardware-based platform are processed as the software technique through a microprocessor. In this paper, GPS software receiver is designed by using SDR technique and then the signal acquisition, tracking, and the navigation message decoding parts are verified through the PC-based simulation. Moreover, the efficient algorithms are developed about the signal acquisition and tracking parts in order to obtain the accurate pseudorange. Finally, the pseudorange is calculated through the relative channel delay received through the different satellite of L1 frequency band. GPS software receiver proposed in this paper will be included in the element of GPS/Galileo complex system of development target and will provide not only the method that verifies the performance for Galileo Sensor Station standard but also usability by providing various debugging environments.

  • Recently, message based data transmission plays an important role in modem computing systems. Especially JMS(Java Message Service) is one of the most popular messaging platform. However, because of its characteristics for maintaining reliability, if we want to use it for transmission of large scale messages in a distributed Internet environment by using a WAN connection which may not be robust enough, we need to employ a different method to minimize total transmission time of messages. We found the fact that the total time of message transmission heavily depends on size of a message. In order to achieve the ideal size of a message, we develope a novel mechanism and a system which finds the ideal size of a message and automatical1y control JMS applications for minimizing transmission time. Finally, we test the proposed mechanism and system using real-data in order to prove advantages and compared with the naive mechanism. As a conclusion, we showed that our proposed mechanism and system provide an effective way to reduce transmission time of large scale messages in distributed environment.

A SOA-based Dynamic Service Composition Framework using Web Services and OpenAPIs (웹 서비스와 OpenAPI를 사용한 SOA 기반 동적 서비스 합성 프레임워크)

  • With the advent of Web 2.0, OpenAPIs are becoming an increasing trend to emphasize Web as platform recently. OpenAPIs are used to combine services and generate new services by mashup. However because the standard documents for OpenAPIs do not exist, it may restrict the use of OpenAPIs. Previous studies of OpenAPIs mashup have been limited to tool design or language definition for service combination rather than dynamic composition. On the other hand, Web services that are a software technology implementing SOA provide standard documents such as WSDL to explain each service, UDDI to register it, and SOAP to transfer messages. Thus Web applications can interpret and execute services by using these technologies. Recent works have also been performed to provide semantic features and dynamic composition for SOA. If a dynamic and systematic approach is provided to combine Web services and OpenAPIs, Web applications can provide users with diverse services. In this study, we present a SOA based framework for mashup of OpenAPIs and Web services. The framework supports dynamic composition of OpenAPIs and Web services, where the process of composite services is described in OWL-S. A prototype is provided to validate our framework. The framework is expected to add diversity to typical Web services.

Storm-Based Dynamic Tag Cloud for Real-Time SNS Data (실시간 SNS 데이터를 위한 Storm 기반 동적 태그 클라우드)

  • In general, there are many difficulties in collecting, storing, and analyzing SNS (social network service) data, since those data have big data characteristics, which occurs very fast with the mixture form of structured and unstructured data. In this paper, we propose a new data visualization framework that works on Apache Storm, and it can be useful for real-time and dynamic analysis of SNS data. Apache Storm is a representative big data software platform that processes and analyzes real-time streaming data in the distributed environment. Using Storm, in this paper we collect and aggregate the real-time Twitter data and dynamically visualize the aggregated results through the tag cloud. In addition to Storm-based collection and aggregation functionalities, we also design and implement a Web interface that a user gives his/her interesting keywords and confirms the visualization result of tag cloud related to the given keywords. We finally empirically show that this study makes users be able to intuitively figure out the change of the interested subject on SNS data and the visualized results be applied to many other services such as thematic trend analysis, product recommendation, and customer needs identification.

Design and Implementation of Multiple Filter Distributed Deduplication System Applying Cuckoo Filter Similarity (쿠쿠 필터 유사도를 적용한 다중 필터 분산 중복 제거 시스템 설계 및 구현)

  • The need for storage, management, and retrieval techniques for alternative data has emerged as technologies based on data generated from business activities conducted by enterprises have emerged as the key to business success in recent years. Existing big data platform systems must load a large amount of data generated in real time without delay to process unstructured data, which is an alternative data, and efficiently manage storage space by utilizing a deduplication system of different storages when redundant data occurs. In this paper, we propose a multi-layer distributed data deduplication process system using the similarity of the Cuckoo hashing filter technique considering the characteristics of big data. Similarity between virtual machines is applied as Cuckoo hash, individual storage nodes can improve performance with deduplication efficiency, and multi-layer Cuckoo filter is applied to reduce processing time. Experimental results show that the proposed method shortens the processing time by 8.9% and increases the deduplication rate by 10.3%.

A Study on the Promotion of Mobile Easy Payment Services in the Fintech Era (핀테크 모바일 간편결제 서비스 활성화 방안)

  • With the growing interest being reflected in the FinTech industry, much attention has been paid to mobile easy payment services as well. In the era of mobile commerce, the core advantage of using mobile easy payment services(simplifying complex payment procedures and thus facilitating user convenience and reducing the chance of giving up payment) are being more emphasized. Mobile easy payment service market not only includes mobile easy payment service providers but also users, non-users, affiliates, banks, and credit card companies as main stakeholders. Exploring those stakeholders is thus important to thoroughly understand such market. However, extant literature on mobile easy payment services mostly focuses on examining adoption intention of users or non-users. This study, an exploratory research based on interviews, thus aims to extract driving as well as inhibiting factors of mobile easy payment service use from six different perspectives (i.e., social platform, bank, credit card company, affiliate, user, and non-user) and analyze a sequence of cause and effect for each factor. For this, the causal loop diagram was developed to deduce key issues and propose an alternative. Theoretical and practical implications of this study will also be discussed.
