Gamigunggui-tang (GMGGT) is one of the important prescriptions that has been used in oriental medicine. We investigated the inhibitory effect of an oral administration of hot water extract of GMGGT on the development of atopic dermatitis (AD) by using Balb/c mice. The induction of atopic dermatitis-like lesion was conducted by the removal of the back hairs and topical application of 2,4-Dinitrochlorobenzene (DNCB) on to the back skin repeatedly. GMGGT was orally administered at a different dose (10.0 mg/kg, 50.0 mg/kg). Skin reactions, consisting of increased ear thickness and the presence of ear inflammation, were observed in mice over three weeks. Oral administration of GMGGT significantly suppressed the skin lesions, ear swelling, spleen weight, total serum IgE in a concentration dependent manner, and also inhibited the infiltration of mast cells in the dorsal skin. In the present study, these results suggested that GMGGT extract inhibits inflammatory response atopic dermatitis. Therefore, GMGGT may be effective substances for the management of AD in human.
홍역이환개의 중추신경계에서 발생한 수초탈락을 수반하는 뇌척수염을 병리조직학적으로 관찰하고 조직반응과 수축탈락의 과정을 알아보기 위하여 중추신경계 조직을 파라핀 포매, 절편을 만들고 병소가 빈발하는 소뇌와 시신경로는 수초의 주요 구성단백인 MBP와 MAG 그리고 별아교세포의 Marker인 GFAP의 항혈청을 반응시켜 면역세포학적으로 관찰하였다. 조직학적으로는 대뇌에서 신경세포의 일부괴사, 신경아교세포의 결절형성, 소뇌에서는 4뇌실에 인접하여 백질부에서 수초의 공포변성, 수초탈락과 염증세포의 침윤이 현저하였고 피질부의 과립층에 인접한 부위와 시신경로에서도 수초탈락이 인정되었다. MBP와 MAG를 면역반응시킨 결과 수초의 공포화와 수초탈락이 현저한 부위에서는 MBP와 MAG가 동시에 소실되었고 부위에 따라서는 MBP또는 MAG가 먼저 소실되기도 하였다. 동시에 별아교세포의 반응은 수초탈락 초기에는 GFAP양성의 섬유와 세포가 크게 증가한 반면 수초탈락이 심하게 진행된 부위에서는 오히려 소실되는 경향이었다. 따라서 홍역이환 개에서 발생되는 수초탈락은 일차적으로 수초가 공격을 받아 파괴됨을 알 수 있었고 동시에 부위에 따라 다른 소견을 요인별로 비교하였다.

The augmentation of soft tissue defects is one of the critical problems in the oral and maxillofacial surgery. Various types of graft materials, both autologous and non-autologous, have been used for the augmentation of soft tissue in the facial region. However, it is not easy to choose an ideal material for soft tissue augmentation because each has its advantages and disadvantages. An ideal graft material should meet the following criteria : it should not leave a scar at the area from which it was taken; should have less likelihood of causing infection; should feel natural after implanted; and should be not absorbed. Among the materials meeting these criteria, human dermis and artificial dermis are commonly used for clinical purposes. The present study was aimed to investigate and compare the resorption rate and the histological change following the use of the autologous dermis, the human homogenous dermis $Alloderm^{(R)}$, and the artificial dermis $Terudermis^{(R)}$ to reconstruct the soft tissue defect. Twenty mature rabbits of either sex, weighing about 2 ㎏, were used. Each rabbit was transplanted with the autologous dermis, $Alloderm^{(R)}$, and $Terudermis^{(R)}$ size $1{\times}1-cm$ at the space between the external abdominal oblique muscle and the external abdominal oblique fascia. They were then divided into 4 groups (n=5 each) according to the time elapsed after the surgery: 1, 2, 4, and 8 weeks. The resorption rate was calculated by measuring the volume change before and after the transplantation, and H-E stain was preformed to observe the histological changes. The resorption rate after 8 weeks was 21.5% for the autologous dermis, 16.0% $Alloderm^{(R)}$, and 36.4% $Terudermis^{(R)}$, suggesting that $Alloderm^{(R)}$ is the most stable while $Terudermis^{(R)}$ is the most unstable. In microscopic examinations, the autologous dermis graft was surrounded by inflammatory cells and showed foreign body reactions. The epidermal inclusion cyst was observed in the autologous dermis graft. $Terudermis^{(R)}$ and $Alloderm^{(R)}$ demonstrated neovascularization and the progressive growth of new fibroblast. The results suggest that $Terudermis^{(R)}$ and $Alloderm^{(R)}$ can be availably for substituting the autologous dermis.

Longevity in medicine can be defined as a long life without mental or physical deficits. This can be prevented by Alzheimer's disease (AD). Current conventional AD treatments only alleviate the symptoms without reversing AD progression. Recent studies demonstrated that Panax ginseng extract improves AD symptoms in patients with AD, and the two main components of ginseng might contribute to AD amelioration. Ginsenosides show various AD-related neuroprotective effects. Gintonin is a newly identified ginseng constituent that contains lysophosphatidic acids and attenuates AD-related brain neuropathies. Ginsenosides decrease amyloid ${\beta}$-protein ($A{\beta}$) formation by inhibiting ${\beta}$- and ${\gamma}$-secretase activity or by activating the nonamyloidogenic pathway, inhibit acetylcholinesterase activity and $A{\beta}$-induced neurotoxicity, and decrease $A{\beta}$-induced production of reactive oxygen species and neuro-inflammatory reactions. Oral administration of ginsenosides increases the expression levels of enzymes involved in acetylcholine synthesis in the brain and alleviates $A{\beta}$-induced cholinergic deficits in AD models. Similarly, gintonin inhibits $A{\beta}$-induced neurotoxicity and activates the nonamyloidogenic pathway to reduce $A{\beta}$ formation and to increase acetylcholine and choline acetyltransferase expression in the brain through lysophosphatidic acid receptors. Oral administration of gintonin attenuates brain amyloid plaque deposits, boosting hippocampal cholinergic systems and neurogenesis, thereby ameliorating learning and memory impairments. It also improves cognitive functions in patients with AD. Ginsenosides and gintonin attenuate AD-related neuropathology through multiple routes. This review focuses research demonstrating that ginseng constituents could be a candidate as an adjuvant for AD treatment. However, clinical investigations including efficacy and tolerability analyses may be necessary for the clinical acceptance of ginseng components in combination with conventional AD drugs.
Endothelial cells are a vital constituent of most mammalian organs and are required to maintain the integrity of these tissues. These cells also play a major role in angiogenesis, inflammatory reactions, and in the regulation of thrombosis. Angiogenesis facilitates pulp formation and produces the vessels which are essential for the maintenance of tooth homeostasis. These vessels can also be used in bone and tissue regeneration, and in surgical procedures to place implants or to remove cancerous tissue. Furthermore, endothelial cell regeneration is the most critical component of the tooth generation process. The aim of the present study was to stimulate endothelial regeneration at a site of acute cyclophosphamide (CP)-induced endothelial injury by treatment with human umbilical cord-derived endothelial/mesenchymal stem cells (hEPCs). We randomly assigned 16 to 20-week-old female NOD/SCID mice into three separate groups, a hEPC ($1{\times}10^5$ cells) transplanted, 300mg/kg CP treated and saline (control) group. The mice were sacrificed on days 5 and 10 and blood was collected via the abdominal aorta for analysis. The alanine transaminase (ALT), aspartate aminotransferase (AST), serum alkaline phosphatase (s-ALP), and albumin (ALB) levels were then evaluated. Tissue sections from the livers and kidneys were stained with hematoxylin and eosin (HE) for microscopic analysis and were subjected to immunohistochemistry to evaluate any changes in the endothelial layer. CP treatment caused a weight reduction after one day. The kidney/body weight ratio increased in the hEPC treated animals compared with the CP only group at 10 days. Moreover, hEPC treatment resulted in reduced s-ALP, AST, ALT levels compared with the CP only group at 10 days. The CP only animals further showed endothelial injuries at five days which were recovered by hEPC treatment at 10 days. The number of CD31-positive cells was increased by hEPC treatment at both 5 and 10 days. In conclusion, the CP-induced disruption of endothelial cells is recovered by hEPC treatment, indicating that hEPC transplantation has potential benefits in the treatment of endothelial damage.
Regeneration of the periodontal tissue destroyed by periodontal disease is one of the final goals of periodontal therapy. In the past few years, periodontists have used various alloplastic grafting materials in an attempt to regenerate bone lost from periodontal disease. These materials have used widely because they have shown to be nontoxic, biologically compatible with surrounding host tissue and chemically similar to bone. The purpose of this study was to investigate the effect of Porous Resorbable Calcium Carbonate and Porous Replamineform Hydroxyapatite on the regeneration of the alveolar bone and the healing of roots transplanted into the periodontally diseased extraction sockets of dogs. The experimental chronic periodontitis was induced by elastic ligatures on the 2nd and 3rd mandibular premolars of 2 adult dogs for 8weeks after surgically creating periodontal defect. The extracted root were split in half along the long-axis, and the extend of plaque exposure was marked on the root surfaces with burs. The roots were inserted in extraction sockets with Porous Resorbable Calcium Carbonate(PRCC) in left side and with Porous Replaminefrom Hydroxyapatite(PRH) in right side. The flaps were sutured to cover the sockets completely. The animals were sacrificed after 12 weeks of healing, and the specimens were examined histologically. The results were as follows: 1. No inflammatory reactions were observed in either groups. 2. Hoot resorption was observed in both groups while the general outline of the roots were maintained. 3. PRCC was almost completely resorbed and replaced with new bone, while R.H.A. was not resorbed & remained encased in newly-formed C-T and alveolar bone. 4. PRH was encapsulated with alveolar bone which has been deposited from apical & lateral area of the sockets, while the coronal portion of the sockets were filled with C-T. 5. In both groups, the resorbed portions of the roots were replaced with new bone. These results suggest that either PRCC or PRH may not interfere with bone formation or healing in extraction sockets, and in some degree, retard the root resorption. Because the roots maintained in anatomy, we think that graft materials prevent the root resorption.
주방세제로 유도한 건성피부에 대한 Palmarosa, Neroli essential oil의 유효성 연구결과는 다음과 같다. 단백질 분석결과 Neroli essential oil 처리 군이 대조군과 가장 유사하게 나타났고 그 다음으로 Palmarosa + Neroli essential oil 처리 군이 대조군과 유사하게 복구되어 가고 있는 것으로 나타났고 그 다음으로 Palmarosa의 순이다. 표피층의 형태적 관찰결과로는 Palmarosa essential oil에 의한 각질층의 이탈과 Neroli essential oil에 의한 각질층의 복구에 의한 표피층의 비후 현상을 볼 수 있었으나 각각의 오일 처리군에서 계면활성제 처리 후 피부 염증에 대한 재생효과가 있음을 알 수 있었다. 염증 반응을 알아보기 위한 Mast Cell 실험에서 보면 대조군에 비해 계면활성제 연속처리군에서는 mast cell의 크기와 수가 증가되었음을 확인 할 수 있다. Neroli essential oil이 처리된 각각의 군에서는 mast cell의 수가 확연히 감소된 것을 볼 수 있다.
문제 진술: 임플랜트 시스템은 임플랜트와 지대주 사이에서 빈 공간이 발생하여 세균의 저장소로서 작용하게 되어 임플랜트 주위 연조직에 염증반응을 야기할 수 있다. 목 적: 이 연구의 목적은 임플랜트-지대주 간극의 미세 누출에 관여하는 치주질환원인균으로 추정되는 Porpyromonas gingivalis, Prevotella intermedia, Tannerella forsythia, Treponema denticola, Aggregatibacter actinomycetemcomitans의 검출률을 조사하는 것이다. 연구재료 및 방법: 시료채취는 27명의 환자들에서 행하였고 소독된 페이퍼포인트를 이용하여 $1{\times}PBS$로 옮겼다. 치주질환원인균의 검출은 16 rDNA에 근거한 종특이적인 프라이머를 지닌 중합효소체인반응을 이용하였다. 결과: 임플랜트 고정체내에서의 Porphyromonas gingivalis와 Prevotella intermedia의 검출률은 59%와 82%였다. 환자의 임플랜트 연구에서는 Porphyromonas gingivalis와 Prevotella intermedia의 검출률은 44%와 82%였다. 환자혀에서의 Porphyromonas gingivalis와 Prevotella intermedia의 검출률은 82%와 82%였다. 결 론: 현재의 임플랜트 시스템은 임플랜트 내부의 세균 군집화와 미세누출을 안전하게 예방할 수 없다.
림프절은 이차성 면역기관중 하나이다. 림프절은 복잡한 3차원적 뼈대 구조물과 스트로마 세포로 구성되어 있다. Fibroblastic reticular cells (FRC)는 T세포와 상호작용을 위해 T zone에 주로 분포하고 있는 세포이다. FRC는 CCL21, CCL19같은 홍밍유도 케모카인을 분비하거나 감염에 대비하여 림프절 세포외기질 형성에 중요한 역할을 한다. 하지만, FRC가 직접 면역반응에 관여하는지에 대하여 많이 알려져 있지 않다. 본 연구는 면역반응에 대한 FRC의 특성 규명에 대한 것이다. 이를 위해 FRC와 대식세포의 공배양, lipopolysaccharide (LPS), TNFα 자극에 노출시켜 반응성을 조사하였다. FRC와 대식세포를 공배양 하였을 때 FRC의 형태적 변화가 유도 되었고 이로 인해 FRC가 빈 공간이 형성되는 것을 확인하였다. 용해성 ICAM-1 (sICAM-1)의 발현량이 대식세포와 ROCK 저해제, Y27632를 처리 했을 경우 증가하는 것을 단백질 수준에서 확인하였다. Matrix metalloproteinase (MMP) 활성이 LPS를 처리한 FRC에서 반응시간 의존적으로 증가하는 것을 확인하였다. 더욱이, 세포외기질에 대하여 염증물질인 TNFα를 처리 했을 경우 조절되는 것을 gene chip assay를 통해서 확인하였다. 이상의 결과는 FRC가 면역반응에 직접 관여하고 있다는 것을 의미하며 이는 림프절 스트로마도 면역반응에 관여하고 있는 것으로 사료된다.
프로폴리스는 꿀벌에 의해 생산된 수지 물질로 벌집을 보호하기 위해 사용되어 왔다. 꿀벌은 다양한 나무와 식물에서 배출된 수분을 침과 밀랍과 섞어서 프로폴리스를 생산한다. 프로폴리스는 기원전 300년경부터 상처를 치료하는 민간요법으로 사용되어 왔다. 프로폴리스에는 플라보노이드, 페놀계 화합물, 밀랍과 같은 많은 생리 활성 화합물이 포함되어 있다. 프로폴리스의 기능적 요소들 때문에, 프로폴리스에는 생물학적 응용을 위한 넓은 스펙트럼이 있다. 프로폴리스와 프로폴리스의 생물학적 활동의 화합물은 각각 과즙의 출처와 추출 방법에 따라 달라질 수 있다. 프로폴리스의 박테리아, 바이러스, 곰팡이를 포함한 반미생물 활동이 가장 잘 알려진 특징이다. 면역조절 메커니즘의 경우 아르테필린 C와 카페인산 페네틸에스테르가 주로 조절 화합물로 확인되었으며 각 화합물은 염증 반응을 감소시키고 T 림프구에 면역억제 반응을 일으킨다. 프로폴리스는 항염증 효과를 통해 암세포 증식 억제, 종양 신호 전달 캐스케이드 차단, 항혈관신생 등 항종양 활성을 보였다. 그러나 프로폴리스의 더 많은 응용을 위해서는 과즙의 원천 분석, 프로폴리스 화합물의 식별, 프로폴리스 분자 메커니즘, 화합물 시너지 효과의 조사가 더 필요할 것으로 보인다.
