Anti-inflammatory and Radical Scavenging Effects of Spirodela polyrrhiza

  • Anti-inflammatory and anti-oxidant effect of Spirodela polyrrhiza (Lemnaceae), a widely used traditional medicinal plant were investigated. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. 85% MeOH extracts of S. polyrrhiza (0.01, 0.1, 1 mg/mL) suppressed nitric oxide production in interferone-$\gamma$ (IFN-$\gamma$) and lipopoloysaccharide(LPS)-stimulated macrophages. It also attenuated the expression of inflammatory enzymes like inducible NOS (iNOS) and cyclooxygenase-2(COX-2) as assessed by immunoblotting with specific antibodies. Moreover, the values obtained with DPPH radical, superoxide anion and NO radical scavenging assay showed that S. polyrrhiza has potent antioxidant properties as a natural ROS scavenger. The results of the present study suggest the potential use of S. polyrrhiza in the treatment of ROS-mediated chronic inflammatory diseases such as atherosclerosis and rheumatoid arthritis.

  • The interphalangeal joint (IPJ) of the hallux has received little attention compared with the first metatarsophalangeal joint. But, the hallucal IPJ has several disorders such intra-articular fractures, dorsal dislocation, alignment disorder, and inflammatory or degenerative arthritis. Among these disorders septic arthritis of the IPJ of the hallux is rare. We report a case of sepsis of the hallucal IPJ and adjacent underlying osteomyelitis without neuropathic problem and was performed through infected soft tissue and osseous debridement, temporary antibiotic-impregnated cement spacer, and delayed intercalary allogenic fibular bone graft with K-wire fixation.

  • Spontaneous rupture of the extensor tendon has been reported in association with predisposing inflammatory conditions including rheumatoid arthritis, diabetes, trauma, tophaceous gout, and steroid injection. The authors experienced a case of spontaneous rupture of the extensor digitorum longus tendons caused by an osteophyte of the tarsometatarsal joint in a patient with rheumatoid arthritis. The authors stress that aggressive treatment including surgery could be considered for prevention of spontaneous tendon rupture in a patient with predisposing conditions despite an asymptomatic spur.

  • Lonicera japonica Thunb. is a twining shrub that has been used as an antidote and to treat urinary disorders, fever and headache. It has been known as an anti-inflammatory agent in Korea from ancient times and is used widely for treating upper-respiratory tract infections, diabetes mellitus and rheumatoid arthritis.   • Joins (SKI 306X) is now clinically used for the treatment of osteoarthritis (OA). In previous reports, Joins a natural herbal product extracted from three herbs Clematis Radix. Trichosanthes Radix and Prunella Flos, was shown to have good analgesic and anti-inflammatory effects in several in vivo models. e.g., acetic acid-induced pain, carrageenan-induced paw edema and adjuvant-induced arthritis.   • Daidzein and its methyl derivatives were synthesized and their anti-inflammatory activities were examined. Daidzein suppressed the carrageenin-induced edema, but its methyl derivatives showed decrease or abolition of the anti-inflammatory effect. Daidzein did not significantly suppress the complete Freund's adjuvant-induced arthritis, and the mixed phlogistics (histamine+serotonin)-induced edema. Daidzein inhibited the leukocyte emigration and protein exudation when it was administered into the CMC pouch at low doses (5,25mg/pouch). Daidzein significantly suppressed the cotton-pellet granuloma formation.

  • A various ${\alpha}$-arylmethylpropionic acids(profen) have been widely used as non-steroidal anti-inflammatory drugs for the relief of acute and chronic rheumatoid arthritis and osteoarthritis, as well as for other connective tissue disorders and pains. Example is fenoprofen, ibuprofen, ketoprofen, and naproxen. All are chiral and, except for naproxen and ibuprofen, are marketed in racemic form. 전갈약침이 adjuvant 유발 흰쥐의 관절활액막내 cytokine과 prostaglandin E2에 미치는 영향 (The Effect of Buthus martensi Karsch Herbal-acupuncture on Immune Responses to Adjuvant Induced Arthritis in Rats)

  • Objective : The purpose of this study is to investigate the immunohistological effect of Buthus martensi Karsch Herbal-acupuncture in treating the arthritis, performed several experimental items : those are paw edema, $IL-1{\beta}$, IL-6, IL-8, $TNF-{\alpha}$ and $PGE_2$. Methods : All the male Sprague Dawley rats used in this study were bred and maintaned in our pathogen-free rat colony and were 8 weeks of age at the start of to experiment. The experimental model of arthritis was induced by injection of $50{\mu}g/{\mu}{\ell}$ adjuvant(mineral oil mixed Mycobacterium butyricum). Buthus martensi Karsch Herbal-acupuncture was injected into ST36(足三里) of rats daily for 21 days. Immunohistological analysis was carried out to assess paw edema, $IL-1{\beta}$, IL-6, IL-8, $TNF-{\alpha}$ and $PGE_2$ expression in synovial membrane and sera Buthus martensi Karsch Herbal-acupuncture injected. Results : Buthus martensi Karsch Herbal-acupuncture group showed a decrease with statistical significance, in paw edema, $IL-1{\beta}$, IL-6, IL-8, $TNF-{\alpha}$ and $PGE_2$ in synovial membrane and sera compared with control group. Conclusion : Buthus martensi Karsch Herbal-acupuncture stimulation inhibited the development of immunity to adjuvant induced arthritis in rats. Thus, Buthus martensi Karsch Herbal-acupuncture may have preventive effects on autoimmune inflammatory joint diseases as arthritis. The effect of Buthus martensi Karsch Herbal-acupuncture on the immune function and the disease activity in patients with arthritis warrants further investigation.

상심자추출복합물의 퇴행성관절염 개선 효능에 대한 동물실험 및 임상연구 (Animal and Clinical Study of the Efficacy of Mulberry Extract Complex on Degenerative Arthritis)

  • Objectives : This study was performed to investigate the efficacy and safety of the Mulberry Extract Complex in a placebo-controlled randomized clinical trial and animal study on degenerative arthritis. Methods : Animal study: Mulberry Extract Complex is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To evaluate the serum level of interleukin-2, interferon-$\gamma$, and prostaglandin E2, an animal model of degenerative arthritis induced by monosodium iodoacetate was employed. Clinical study: The efficacy index (VAS, K-WOMAC) was compared among patients with symptoms of degenerative arthritis before and after Mulberry Extract Complex ingestion as well as the one in groups. Evaluations of the improvement by the subjects and by doctor assessment were also performed. Results : Animal study: Mulberry Extract Complex reduced the serum level of interferon-$\gamma$ and prostaglandin E2 in an animal model with degenerative arthritis. Clinical study: The VAS change showed statistical significance in the experimental groups after 4 weeks (PP set) and 8 weeks (ITT set) of ingestion. When the K-WOMAC was analyzed using a modified ITT set to determine the effectiveness, statistically significant results were obtained in the fields of pain & symptom within the Mulberry Extract Complex group as well as between the Mulberry Extract Complex and placebo groups after 8 weeks of ingestion. Results from the improvement evaluation by subjects and the assessment of improvement by doctors showed statistical significance in the experimental groups (PP set) after 8 weeks. Conclusions : Mulberry Extract Complex could be useful for the improvement of various symptoms of degenerative arthritis based on its anti-inflammatory activity and its reduction of VAS and K-WOMAC pain scores.
