Outbreak of contagious diseases to livestock animals is becoming prevalent worldwide and consequently, tremendous numbers of the infected or culled stocks are buried on the ground as the most common disposal method. The buried animals can generate a wide range of detrimental components such as leachate, nutrient salts, and pathogenic bacteria, consequently contaminating the surround environment. This implies that regular investigations are required to monitor any possible detrimental environmental aspect occurred around burial sites. Therefore, the current study was conducted to investigate whether the soil and groundwater nearby the burial sites had been contaminated by the substances originated from the burial sites, which can be applied for the establishment of the ideal burial site construction design and post management scheme. For this, two different burial sites located in Cheonan and Pyeongtaek were selected. Cheonan and Pyeongtaek sites were constructed in 2004 and 2008, respectively and both contained dead poultry infected by avian influenza (AI). Soil and groundwater samples were collected around the sites followed by determination of the nutrient concentrations and bacteria (Salmonella, Camphylobacter, and Bacillus) existence in both soil and groundwater. Some of the soil samples showed higher EC, $NH_4$-N, $NO_3$-N concentration compared to those of the background (control) soils. Also the concentration of $NH_4$-N in some of the groundwater samples appeared to exceed the USEPA guideline value for drinking water (10 mg $L^{-1}$). These results indicated that the soil and groundwater were influenced by the burial site originated nutrients. In the soil, Bacillus was isolated in most soil samples while there were no detections of Salmonella and Camplylobacter. Due to the Bacillus existing mainly as a spore in the soils, it was considered that the frequent detection of Bacillus in the soil samples was attributed to the nutrients originated from the burial sites.
In this study, we investigated the effect of bioprocessed polysaccharides (BPPs) from liquid culture of Lentinus edodes fungal mycelia containing rice bran (BPP-RB) on a chicken-derived macrophage cell line, HD-11, when infected with Salmonella Gallinarum, an etiological agent of fowl typhoid. Experimental results demonstrated water extract of BPP-RB did not show growth inhibitory effects on S. Gallinarum 277. Protein expression profiles were also not altered by its treatment. Nonetheless, it could (i) enhance phagocytic activity of HD-11 cells, (ii) activate transcriptional expression of Th1-type cytokines such as tumor necrosis factor-${\alpha}$ and interleukin $(IL)-1{\beta}$, iNOS, as well as an immunosuppressive cytokine IL-10, and (iii) negatively regulate Th2-type cytokines such as IL-4 and IL-6. Together results suggest that BPP-RB may be applicable for preventing fowl typhoid or other Salmonella infections in poultry farms as a potential feed additive.

Objective: The highly pathogenic avian influenza virus (HPAIV) is a threat to the poultry industry as well as the economy and remains a potential source of pandemic infection in humans. Antiviral genes are considered a potential factor for HPAIV resistance. Therefore, in this study, we investigated gene expression related to cytokine-cytokine receptor interactions by comparing resistant and susceptible Ri chicken lines for avian influenza virus infection. Methods: Ri chickens of resistant (Mx/A; BF2/B21) and susceptible (Mx/G; BF2/B13) lines were selected by genotyping the Mx dynamin like GTPase (Mx) and major histocompatibility complex class I antigen BF2 genes. These chickens were then infected with influenza A virus subtype H5N1, and their lung tissues were collected for RNA sequencing. Results: In total, 972 differentially expressed genes (DEGs) were observed between resistant and susceptible Ri chickens, according to the gene ontology and Kyoto encyclopedia of genes and genomes pathways. In particular, DEGs associated with cytokine-cytokine receptor interactions were most abundant. The expression levels of cytokines (interleukin-1β [IL-1β], IL-6, IL-8, and IL-18), chemokines (C-C Motif chemokine ligand 4 [CCL4] and CCL17), interferons (IFN-γ), and IFN-stimulated genes (Mx1, CCL19, 2'-5'-oligoadenylate synthase-like, and protein kinase R) were higher in H5N1-resistant chickens than in H5N1-susceptible chickens. Conclusion: Resistant chickens show stronger immune responses and antiviral activity (cytokines, chemokines, and IFN-stimulated genes) than those of susceptible chickens against HPAIV infection.

The purpose of this study was to evaluate the influence and safety of spraying Farm Clean-S (FC-S) containing 70% Chamaecyparis obtusa essential oil on the elimination of red mite in laying hens. In total, 300 laying hens (ISA brown) aged 72 weeks and infected with red mite were assigned to individual cages for five treatments with five replicates for 18 days. Treatments consisted of distilled water (negative control; NC), commercial spray product (positive control; PC), FC-S 0 (original concentration), FC-S 50 (50 times diluted) and FC-S 100 (100 times diluted). The number of red mites, egg production and weight of each group were evaluated daily throughout the whole experimental period. Egg quality and blood biochemical parameters were measured at the end of the experiment. Safety tests were confirmed by feed intake, respiration, lethargy, diarrhea and mortality symptoms after the feeding trial. As the result, more than 80% of red mites were eliminated by the FC-S 0 spray treatment, and FC-S 50 also showed a higher elimination percentage than did PC (P<0.01). However, the elimination effect of the FC-S 100 spray was lower than that of the PC, but higher than that of the NC (P<0.01). These results confirmed that the optimum FC-S dilution rate ranged from 0~50 times with distilled water to eliminate the red mite effectively. The performance, egg quality, blood parameter and safety tests were not significantly different among treatments. Thus, FC-S 0 and FC-S 50 spray treatments can effectively and sustainably control red mite infestations without detrimental effects on the performance, egg quality and plasma biochemical properties of laying hens.
This study was conducted to investigate the in vivo and in vivo antibiotic effect of crude fucoidan extracted from Hizikia fusiforme, and to investigate any possible structural changes of broiler chick's intestinal villi by the supplementation of fucoidan. Total 84 broiler chicks were randomly assigned to 7 treatments, control and Salmonella typhimurium infection groups. The broiler chicks was infected with Salmonella typhimurium at third days, and antibiotics, fucoidan, dried Hizikia fusiforme, dried Undaria pinnatifida and yeast cell debris was respectively supplemented for each group. Each treatment had 4 chicks with three replications. Extraction yield of crude fucoidan from Hizikia fusiforme was 5.453%. Antibiotic effect of fucoidan was not detected in vitro, inhibition zone and micoorganism growth test. Weight gains of broiler chicks were tend to higher in fucoidan treatment group and yeast cell significance was not found. In in vivo test, the number of viable Salmonella typhimurium was low in the antibiotics and fucoidan treatment groups. The intestinal villi were short in the fucoidan and marine algae treatment groups. The intestinal villi were densely distributed on the large intestinal wall, but the morphology was not different among treatments.
Field strains of Ornithobacterium rhinotracheale (OR) were tested on their virulence in specific pathogen free (SPF) embryonated chicken eggs and 3-week-old broilers. When infected with three different OR isolates (OR-161, OR-240 and OR-295) through yolk sac infection route, all strains appeared to be highly pathogenic with responsible mortality 66% and 100% within 12 days post infection (DPI). To test the virulence of OR in the commercial broilers, 3 week-old broilers were grouped depends on the inoculation route of OR isolate (OR-295) through five different infection routes; group 1 (IT: intratracheal), group 2 (IM: intramuscular), group 3 (IV: intravenous), group 4 (aerosol) and group 5 [Mixed: NDV (LaSota)+OR aerosol]. Within 5 to 7 days after inoculation, only broilers given NDV+OR were slightly depressed and coughing, and had mild facial redness. Grossly, foamy and yellow-white yogurt like exudate in the air sacs, predominantly in the abdominal air sacs was present. In histology, infiltration of the air sac epithelium and lamina propria by macrophage and polymorphonuclear granulocytes was seen with cell debris and inflammatory cells, correlated with the presence of OR antigen, as demonstrated by immunohistochemistry. Field strains of OR were able to induce high mortality in the embryonated chicken eggs, whereas broilers were less susceptible to OR infection. Interestingly, in the absence of NDV infection, the four groups of OR single infection only different route showed minimal and temporary microscopic air sac lesions. Thus, Newcastle disease virus (LaSota strain) showed triggering effects on the OR infection in chickens.
The outbreak of Newcastle disease occurred for the first time at a commercial chicken farm near Ulaanbaatar, Mongolia in August 2010. Newcastle disease virus (NDV) obtained from infected chickens in Mongolia was characterized by biological and molecular biological approches. Mongolian NDV isolate killed all of chicken embryos within 60 h in the mean death time assay, indicating virulent for chicken. A genomic region of 695 nts between nts 1055 of the M gene and 508 of the F gene was amplified by RT-PCR and sequenced. The deduced amino acid sequence of the F protein cleavage site was $^{112}RRQKRF^{117}$, which is a typical sequence of velogenic strains of NDV and is agreement with the result of the MDT assay. The sequence of the partial F gene (nts 47 to 435) was used for genotyping by phylogenetic analysis. The phylogenetic analysis showed that the Mongolian isolate was of genotype VII within class II of NDV. Further phylogenetic analysis on the genotype VII strains revealed that the isolates placed in a genetic sublineage of VIId and most closely related with velogenic strains of NDV circulating in Far-east Asian region especially China, suggesting the introduction of velogenic NDV into Mongolia from neighboring countries.
Newcastle disease virus (NDV) Kr005/V strain was generated through 55 serial passages of NDV Kr005 strain in Vero cells. The Kr005/V virus yielded high infective titers of $10^{7.8}$$TCID_{50}/mL$ in Vero cells and the infected cells showed cytopathic effects such as marked cell rounding, though less frequent syncytia. The Kr005/V virus was heat-stable and classified into the lentogenic type with a Mean Death Time (MDT) of 120h or greater while the Kr005 strain was heat-labile and velogenic (MDT of 49.6 h). Only the single amino acid substitution (T to S) was observed at position 433 of the HN protein of the Kr005/V strain, whereas no amino acid change was found in the F protein. The Kr005/V input virus correlated well (correlation coefficient $r^2$=0.97) with the Kr005 virus when ten field sera were tested by virus neutralization test. The biological properties and usefulness of Vero cell-adapted Kr005/V virus were discussed.

Three strains of infectious bronchitis viruses (IBVs), designated VNUA3, VNUA8 and VNUA11, were isolated from diseased/infected chickens in Hanoi, Thainguyen, and Haiphong provinces of Vietnam. These birds had received a live IBV vaccination but still suffered from infectious bronchitis. VNUA3, VNUA8 and VNUA11 harbor cleavage sites (RRTGR, HRRRR, and HRRKR, respectively) within the S protein. A BLASTN search revealed that the S gene of VNUA3, VNUA8, and VNUA11 showed the highest nucleotide identity with those of IBV strains CK/Italy/I2022/13, CK/CH/LHLJ/08-6, and GX-NN120084, respectively. Phylogenetic analyses based on the S gene nucleotide sequences revealed that VNUA3, VNUA8 and VNUA11 clustered with Q1-like, QX-like and TC07-2-like genotypes, respectively, and were closely related to reference IBV strains from China. However, the Vietnam IBVs showed high divergence from vaccine strains 4/91 and Ma5, which are used in the Vietnamese farms from which the isolates were obtained. Taken together, these results indicate that at least three genotypes of IBV are circulating among chickens in North Vietnam. This is the first report of the molecular epidemiology of IBV in Vietnam.
Humoral immune response of young chicks to Brucella abortus strain 1119-3 inoculation was monitored to verify the degree of immunosuppression caused by infection with Cryptosporidium baileyi. Young chicks (2-day-old) were orally inoculated each with $2{\times}10^6$ oocysts of C. baileyi, and then injected intramuscularly with 0.3 $m\ell$ B. abortus strain 1119-3 containing $1{\times}10^9$ living organisms on day 14 postinoculation (PI). Serum samples were tested by plate agglutination test on day 17 PI onwards at an interval of 3-6 days over a period of 36 days. Infected chicks with the coccidium showed significantly lower antibody titers than those of uninfected controls (P<0.05). These findings document that C. baileyi infection in early life stage may predispose chicks easily to other potential poultry diseases.
