Spawning and Larval Development of the Jicon Scallop, Chlamys farreri (비단가리비, Chlamys farreri의 산란과 유생사육)

  • This study focused on spawning season, induce spawning, spawning and larval development of the Jicon scallop in Daehuksan Island of southwestern waters in Korea. The condition index and gonadosomatic index were used to investigate the reproductive pattern of the Jicon scallop. The major spawning season was from July to August, showing an unimodal gametogenic cycle per year. Several different tests were carried out to induce spawning of the mature male and female C. farreri. For females, the injection of serotonin, temperature induction technique and the combination of the both treatments produced significantly faster gamete release. Unlike females, males spawned only in response to the UV rays irradiation stimulation. Mean size of fertilized eggs was 69.5 $\mu$m in diameter. After fertilization, the zygote could be divided into 2 cells as early as 2 hours. It took about 8 hours to develop the 8-cell stage, about 20 hours to hatch trochophore larvae, and about 40 hours to be D-shaped larvae.

  • The study carried out a 15-day feeding trial of the fleshy shrimp broodstock that have just finished their first spawn using five different maturation feed types (three types of manufactured feed, fresh feed, and commercial feed) to induce multiple spawning in the shrimp and then analyzed and compared hemolymph levels of total protein, total lipid, hemocyanin, glucose, and cholesterol as well as ovary histology between the treatments to identify the potential of the experimental diets to be utilized as maturation feed. The study found that the manufactured diets of polychaetes + clam and polychaetes + clam + squid resulted in significantly higher levels of lipid and protein in shrimp hemolymph suggesting an indicator of multiple spawning in the fleshy shrimp. The other manufactured diet of polychaets + squid led to the highest 2nd spawning with significantly lower levels of lipid, protein, and cholesterol in shrimp hemolymph. The fresh feed treatment showed similar hematological as well as histological responses to the manufactured diet of polychaetes + squid, but with a significantly lower spawning volume compared to the other treatments. The study demonstrated that the manufactured diets could replace fresh feed as maturation feed and attain a higher shrimp maturation than the commercial feed.

  • A survey was conducted in 2011 to investigate the reproductive ecology and spawning behavior of the natural monumental and endangered species, Cobitis choii. C. choii exhibits sexual dimorphism with the development of lamina circularis on the second pectoral fin ray in males at one year old, facilitating easy differentiation between males and females. A total of 1,288 females and 881 males were collected from January to December, resulting in a sex ratio of 1 : 0.68. The spawning season was estimated to be from late June to early July (spawning peak from late June, with water temperatures ranging from 22 to 25℃), coinciding with a sharp decrease in the condition index. Gonadal maturity was observed with maturity indices of 17.5±2.11% for females and 2.9±0.46% for males, with fecundity of 2,444±838 eggs and egg size of 0.75±0.05 mm. Spawning behavior was induced by hormone injection (Human Chorionic Gonadotropin, HCG) in laboratory conditions. Experimental results revealed that males and females paired in a 1 : 1 ratio before spawning. Males pressed the abdomen of females using their lamina circularis to induce spawning, followed by immediate fertilization. Spawning behavior was repeated 15 to 25 (average 20) times per pair of males and females, and 80 to 120 eggs were spawned per time, and the total amount of spawned eggs were 2,500±250 on average.

  • To develop techniques for the production of artificial seedling of Corbicula japonica, protocols for induction of spawning and larval development were developed. During the assumed spawning period of August to mid-September, attempts were first made to induce spawning by gonadal incision and UV-irradiation but all in vain. At the end of August, elevated thermal induction evoked 90 % positive response in animals maintained at $3\textperthousand$ salinity. Immersion in (1/1000~3/1000 N) ammonium hydroxide ($NH_4$OH) also induced spawning in 15~45%) of the treated animals at $3\textperthousand$ salinity. Fertilized eggs measured 86$\mum. At 23.0~24.5$^{\circ}C$, the fertilized egos developed into 4-cell stage embryos within 2 hours, trochophores 15 hours, D-shaped larvae 2 days, umbo 9 days and fully grown veligers, ready to infiltrate into the sediment, within 16 days.

Spawning Induction and Egg Development of Surf Clam, Spisula sachalinensis (북방대합, Spisula sachalinensis의 산란유발 및 난발생)

  • In order to obtain the basic information for seedling production of surf clam, Spisula sachalinensis, spawning induction and egg development were investigated. $NH_4OH$ addition and serotonin injection could induce the spawning in surf clam. Water temperature rising, sperm suspension immersion, UV-ray irradiated seawater and $H_2O_2$ addition less affected on induction of spawning than $NH_4OH$ and serotonin did. On the other hand, males were more sensitive to the treatments than females. The response time to initial spawning in the case of $NH_4OH$ addition was $3\~4$ hours. However in the case of serotonin injection, it was within 5 minutes. The number of eggs released by $NH_4OH$ addition were significantly more than those released by serotonin injection. The serotonin injection induced higher rates of germinal vesicle breakdown than the $NH_4OH$ addition. Fertilizing and hatching rates of the eggs also were the similar results. Eggs of surf clam were demersal isolated eggs and averaging $65.2{\pm}±1.8\;{\mu}m$ in diameter after spawning. Optimum range of water temperature for the development of egg was $15\~20^{\circ}C$, The required time for development of D-shaped larvae was 42 hours at $15^{\circ}C$ and 27 hours at $20^{\circ}C$, respectively.

  • A Series of trials for the spawning induction of the Israeli strain of common carp were carried out at the fish culture research station of National Fisheries University of Pusan from 1978 to 1981. 1, During 3 years up to the first periods in 1980 carp pituitaries and/or commercial gonadotropic hormone was used to induce the brood fish to spawn without success. In the trials in the later periods in 1980 and in 1981, water temperature increase method was used with success in 40 females of 16 trials leaving a single failure with 2 females. 2. Brood fish had been kept in a small circular tank ($7\;m^2$) continuously supplied with cool well water enough to keep the water temperature in the tank relatively cool, ranging from 16.9 to $20.2^{\circ}C$, and the water in the spawning tank ($15\;m^2$) was 3.0 to $6.5^{\circ}C$ higher than in the brood fish holding tank. 3. It could be said that the spawning of Israeli strain of common carp in Korea hardly occur without water temperature increase even though treated with hormone.

  • To induce an early spawning, the adult red sea bream, Pagrus major, were exposed to predesignated condition of photoperiod and water temperature during February 21 to June 30, 1988. Spawning began from April 20, a month earlier than that in the natural condition, when the photoperiod and water temperature were controlled at 14L: l0D and $17.0^{\circ}C$, respectively, and the spawning continued until June 15, 1988. There were no particular differences between fish under the controlled condition and those in natural condition in terms of the fertilization rate, surface floating rate, the number of oil glouble and the hatching rate.

  • The egg development of various fishes is studied by various methods including electron microscopy(Matsuyama et al, 1991) and hormone treatment(Schoonbee et al. 1980). In addition to attempt to induce final maturation and spawning, the present experiments were conducted to investigate the influence of hCG upon reproductive cycles in catfish(Silurus asotus) with developing gonads and to stimulate the reproductive system during the rapid phase of gonadal development. Quantification of Reproductive Effort and Microscopic Observation on the Larval Development of Manila clam Ruditapes philippinarum (Adams and Reeve, 1850) (바지락 (Ruditapes philippinarum)의 번식량 측정 및 유생발달에 관한 연구)

  • Larval development of the Manila clam Ruditapes philippinarum reared in an indoor tank system was examined in this study using light microscope and scanning electron microscope. To induce spawning and subsequent larval development, clams were collected from the intertidal zone at Gim-nyeong harbor in Jeju Island in August 2011. After 2 days of rearing in the tank, all Manila clams spawned in the midnight. Non-feeding trochophore larvae appeared 7hrsafter fertilization and the first D-shape larvae could be observed at 19 hrs. Twenty one days after fertilization the pediveliger larvae crawling on the bottom of the tank with well-developed foot were observed. Histology indicated that all the clams used in this study were in the ripe stage prior to spawning and the gonad-somatic index (GSI), a ratio of the egg mass to the tissue weight, of the ripe female measured by ELISA was 28.6%. The GSI of female clam declined to 17.3% after the massive spawning in the tank, suggesting that Manila clam discharged 40% of the total eggs during the first spawning event. In conclusion, spawning and subsequent larval development of Manila clam was successfully carried out in this study using an indoor tank system, and the information obtained in the present study could be useful in future Manila clam hatchery development.

Induced Ovulation in rabbitfish, siganus canaliculatus, with Human chorionic Gonadotropin (HCG) (태반성 성선자극호르몬(HCG) 처리에 의한 독가시치, Siganus canaliculatus의 배란유도)

  • Successful produciton of seedlings for marine fish species are highly influenced by the aquisiton of sufficient number of good-quality eggs on required time. human chorionic gonadotropin has been used to induce successful ovulation for the purpose. Rabbitfish, siganus canaliculatus, is inhabited along the coast of Cheju Province and treated valuably for raw fish and fish roasted with seasoning. Female wild-captured rabbitfish (314 to 279 g in body weigth) were injected into intraperitoneal space with human chorionic gonadotropin (HCG) of 5,00,1,000, and 1,500 IU/kg body weight during spawning season of July 14 (trial 1), and July 27 (trial 2). Test fish in trial 1 were injected two times, first on July 14 and second on July 21 with one weekinterval because of no ovulation symptoms, but just one time in trial 2. All females injected both 1,000 and 1,500 IU/kg body weight ovulated in all trials. Among 500 IU/kg injection treatments, just one female ovulated in trial 1 showing 25% of ovulation rate, but none ovulated in trial 2. Ovulation was not occurred in control goups during test period. Time to reach ovulation after injection was 172 to 270 hours in trial 1 and 77 to 132 hours in trial 2, showing shorter as spawning season approached. Generally, ovulation of smaller females were late than that of larger ones. Gonadosomatic index (GSI) and pseudo-gonadosomatic index (Pseudo-GSI) were ranged 22 to 33% and 19 to 28%, respectively. One female spawned 406,200 to 1,032,000 eggs, and obtainable eggs per 100g of body weight were calculated by 130,000 to 190,000. Spawning rates were comparatively high by 96.0 to 98.4%. Rates to reach embryo-formation and hatching were higher in 500 high by 96.0 to 98.4%. Rates to reach embryo-formation and hatching were higher in 500 and 1,000 IU/kg treatments than in 1,500 IU/kg treatment. The result of present study demontrates that HCG treatment during spawning season could become very useful for ovulation of rabbitfish and the suitable dosage was suggested as 1,000 IU/kg of body weight.
