Induction of DNA Breakage by the Hot-water Extracts of Fructus Chaenomelis (Chaenomeles sinensis Koehne)

  • The possible mechanism of the DNA strand breaking activity of the hot-water extract of Fructus Chaenomelis (dried fruit of Chaenomeles sinensis) in a closed circular duplex replica form DNA (RFI DNA) was studied through agarose gel electrophresis under various conditions. Induction of DNA strand scission by the hot-water extract of C. sinensis occurred in dose and time-dependent manners. $Cu^{2+}$ was indispensable for the induction of DNA strand breakage. Exogeneous chelating agents inhibited the DNA breaking activity, conforming the catalytic action of $Cu^{2+}$ on generation of free radicals responsible for oxidative damage. Antioxidant enzymes and some radical scavengers were used to investigate the major radical species triggering the DNA strand scission, demonstrating that a highest inhibitory activity was found in the presence of catalase, while less in the presence of tiron (a scavenger for superoxide radical), 2-aminoethyl-isothiuroniumbromide-HBr, cysteamine (scavengers for hydroxyl radical), and 1,4-diazabicyclo [2,2,2] octane (a scavenger for singlet oxygen) in decreasing order. The findings implied that oxygen radical species generated in presence of transition divalent cation during the oxidation of some compounds contained in the hot-water extract of C. sinensis is mainly responsible for inducing genotoxicity.

Cloning, Expression and Hormonal Regulation of Steroidogenic Acute Regulatory Protein Gene in Buffalo Ovary

  • In mammalian ovary, steroidogenic acute regulatory (StAR) protein mediates the true rate-limiting step of transport of cholesterol from outer to inner mitochondrial membrane. Appropriate expression of StAR gene represents an indispensable component of steroidogenesis and its regulation has been found to be species specific. However, limited information is available regarding StAR gene expression during estrous cycle in buffalo ovary. In the present study, expression, localization and hormonal regulation of StAR mRNA were analyzed by semi-quantitative RT-PCR in buffalo ovary and partial cDNA was cloned. Total RNA was isolated from whole follicles of different sizes, granulosa cells from different size follicles and postovulatory structures like corpus luteum and Corpus albicans. Semi-quantitative RT-PCR analyses showed StAR mRNA expression in the postovulatory structure, corpus luteum. No StAR mRNA was detected in total RNA isolated from whole follicles of different size including the preovulatory follicle (>9 mm in diameter). However, granulosa cells isolated from preovulatory follicles showed the moderate expression of StAR mRNA. To assess the hormonal regulation of StAR mRNA, primary culture of buffalo granulosa cells were treated with FSH (100 ng/ml) alone or along with IGF-I (100 ng/ml) for 12 to 18 h. The abundance of StAR mRNA increased in cells treated with FSH alone or FSH with IGF-I. However, effect of FSH with IGF-I on mRNA expression was found highly significant (p<0.01). In conclusion, differential expression of StAR messages was observed during estrous cycle in buffalo ovary. Also, there was a synergistic action of IGF-I on FSH stimulation of StAR gene.

Identification and Characterization of an Antifungal Protein, AfAFPR9, Produced by Marine-Derived Aspergillus fumigatus R9

  • A fungal strain, R9, was isolated from the South Atlantic sediment sample and identified as Aspergillus fumigatus. An antifungal protein, AfAFPR9, was purified from the culture supernatant of Aspergillus fumigatus R9. AfAFPR9 was identified to be restrictocin, which is a member of the ribosome-inactivating proteins (RIPs), by MALDI-TOF-TOF-MS. AfAFPR9 displayed antifungal activity against plant pathogenic Fusarium oxysporum, Alternaria longipes, Colletotrichum gloeosporioides, Paecilomyces variotii, and Trichoderma viride at minimum inhibitory concentrations of 0.6, 0.6, 1.2, 1.2, and 2.4 μg/disc, respectively. Moreover, AfAFPR9 exhibited a certain extent of thermostability, and metal ion and denaturant tolerance. The iodoacetamide assay showed that the disulfide bridge in AfAFPR9 was indispensable for its antifungal action. The cDNA encoding for AfAFPR9 was cloned from A. fumigatus R9 by RT-PCR and heterologously expressed in E. coli. The recombinant AfAFPR9 protein exhibited obvious antifungal activity against C. gloeosporioides, T. viride, and A. longipes. These results reveal the antifungal properties of a RIP member (AfAFPR9) from marine-derived Aspergillus fumigatus and indicated its potential application in controlling plant pathogenic fungi.

Formulation of mBHT (Modified BoyangHwanoTang) Orally Disintegrating Tablet for Therapeutics of Stroke (뇌졸중 예방 및 치료를 위한 가미보양환오탕 구강붕해정 제형 연구)

  • According to the statistical data, stroke is about 13.9% of leading causes of death. Some herbal medicines including Paeonia lactiflora, Angelica gigas nakai and Prunus persica, etc., had been reported to be effective in preventing stroke and mBHT (Modified BoyangHwanoTang) was an advanced prescription used in Korean clinics. Orally disintegrating tablets (ODT) is useful for patients suffering from dysphagia, motion sickness, repeated emesis and mental disorders. Further, drugs exhibiting satisfactory absorption through the mucosa intended for immediate pharmacological action could be advantageously formulated in ODT. The aim of this study was to develop the most efficient ODT formulation of mBHT. Corresponding herbal medicines comprising mBHT were extracted with water for 3 hr at 95~$100{^{\circ}C}$ and then dried. mBHT extract was obtained with about 30% of yield. Subsequently, some pharmaceutical excipients such as spray-dried lactose, crospovidone, glyceryl behenate and/or cogrinded-treated arabia gum were used to achieve an immediate disintegration of mBHT ODT in oral cavity. The requirements of ODT with mechanical strength sufficient to stand the rigors of handling and capability of disintegrating within a few seconds in contact with saliva are indispensable. mBHT ODT prepared by the wet granulation method showed a disintegration time of below 30 sec.

High Glucose Induces Connective Tissue Growth Factor Expression and Extracellular Matrix Accumulation in Rat Aorta Vascular Smooth Muscle Cells Via Extracellular Signal-Regulated Kinase 1/2

  • Connective tissue growth factor (CTGF) is a potent pro-fibrotic factor, which is implicated in fibrosis through extracellular matrix (ECM) induction in diabetic cardiovascular complications. It is an important downstream mediator in the fibrotic action of transforming growth factor ${\beta}$ ($TGF{\beta}$) and is potentially induced by hyperglycemia in human vascular smooth muscle cells (VSMCs). Therefore, the goal of this study is to identify the signaling pathways of CTGF effects on ECM accumulation and cell proliferation in VSMCs under hyperglycemia. We found that high glucose stimulated the levels of CTGF mRNA and protein and followed by VSMC proliferation and ECM components accumulation such as collagen type 1, collagen type 3 and fibronectin. By depleting endogenous CTGF we showed that CTGF is indispensable for the cell proliferation and ECM components accumulation in high glucose-stimulated VSMCs. In addition, pretreatment with the MEK1/2 specific inhibitors, PD98059 or U0126 potently inhibited the CTGF production and ECM components accumulation in high glucose-stimulated VSMCs. Furthermore, knockdown with ERK1/2 MAPK siRNA resulted in significantly down regulated of CTGF production, ECM components accumulation and cell proliferation in high glucose-stimulated VSMCs. Finally, ERK1/2 signaling regulated Egr-1 protein expression and treatment with recombinant CTGF reversed the Egr-1 expression in high glucose-induced VSMCs. It is conceivable that ERK1/2 MAPK signaling pathway plays an important role in regulating CTGF expression and suggests that blockade of CTGF through ERK1/2 MAPK signaling may be beneficial for therapeutic target of diabetic cardiovascular complication such as atherosclerosis.

However, robot applications are transformed from industrial field to human life in recent tendency Nowadays, final goal of robot is to make a intelligent robot that can understand what human say and learn by itself and have internal emotion. For example Home service robots are able to provice functions such as security, housework, entertainment, education and secretary To provide various functions, home robots need to recognize human`s requirement and environment, and it is indispensable to use artificial intelligence technology for implementation of home robots. In this paper, implemented robot system takes data from several sensors and fuses the data to recognize environment information. Also, it can select a proper behavior for environment using soft computing method. Each behavior is composed with intuitive motion and sound in order to let human realize robot behavior well.

Interaction of 2-Hydroxyquinoxaline (2-HQ) on Soil Enzymes and Its Degradation: A Review

  • The United Nations project the world population to reach 10 billion by the year 2057. To increase the food of the ever-increasing world population, agrochemicals are indispensable tools to the boon in agriculture production. These agrochemicals are a serious threat to the health of humans, plants, and animals. Agrochemicals are ultimately reached to the main reservoir/sink such as soil and contaminating the groundwater, disturb the soil health and in turn a serious threat to biogeochemical cycling and the entire biosphere. Among agrochemicals, quinalphosis one of the most repeatedly and widely used insecticides in the control of a wide range of pests that attack various crops. Quinalphos is shown to be primarily toxic in organisms by acetylcholinesterase enzyme action. Hydrolysis of quinalphos produces amajor metabolite 2-hydroxyquinoxaline (2-HQ), which has shown secondary toxicity in organisms. 2-HQ is reported to be mutagenic, carcinogenic, growth inhibition and induce oxidative stress in organisms. Quinoline is a heterocyclic compound and structural resemblance of 2-HQ with minor changes, but its degradation studies are enormous compared to the 2-HQ compound. Biotic factors in fate and behavior of 2-HQ in the environment are least studied. 2-HQ interactions with soil enzymes are vary from soil to soil. Based on the toxicity of 2-HQ in our stockpile we need to isolate a handful of microorganisms to treat this persistent metabolite and also other metabolites/compounds.This brief review will be significant from the point of biological and environmental safety.

  • In recent years, with the rapid development of machine learning technology, research utilizing machine learning has been actively conducted in fields such as cognition, reasoning and judgment, and action among various technologies constituting intelligent systems. In order to utilize this machine learning, it is indispensable to collect data for learning. However, the types of data generated vary according to the environment in which the data is generated, and the types and forms of data required are different depending on the learning model to be used for machine learning. Due to this, there is a problem that the existing data collection method cannot be reused in a new environment, and a specialized data collection module must be developed each time. In this paper, we propose a specification-based methology for data collection in artificial intelligence system to solve the above problems, ensure the reusability of the data collection method according to the data collection environment, and automate the implementation of the data collection function.

Proprioception and the Sense of Ownership (고유수용성 감각과 신체 소유감)

  • This paper aims to examine how proprioception provides a sense of ownership over one's body and physical actions. Proprioception is a sense that relies on somatic interoceptors rather than exteroceptors that receive external stimuli and has epistemic importance of knowing the presence, location, and movement of bodily parts. In this paper, I will argue that Shoemaker's principle, "Immunity to Error through Misidentification (IEM)" can be applied to proprioception by focusing on one of the intrinsic features of proprioception: First-Person Perspective. I will advocate the following two arguments by defeating each of Marcel's pathological counter-examples in turn. Proprioception is infallible in that it provides a sense of ownership over one's body and physical actions. Second, proprioception is indispensable for the sense of ownership of one's body and physical actions.
