감마선 조사가 식용색소의 색도에 미치는 영향 (Effects of Irradiation on Color Values of Food Colorants)

  • 가공식품과 화장품 산업에 사용되는 식용색소의 감마선 조사로 인한 이화학적 특성 변화를 연구할 목적으로 기계적 색도 변화를 측정하였다. 적색 2호(Amaranth), 녹색 3호(Fast Green PE), 청색 2호(Indigo Carmine)용액을 0.01 및 $0.10\%$의 농도로 준비하였으며, 0.25, 0.5, 1 및 3 kGy로 조사를 실시하여 기계적 색도 (L, a, b, ${\Delta}E$)를 측정하였다. 감마선 조사는 $0.01\%$농도 시료의 색소에 영향을 미쳤으며, 전반적인 색차$({\Delta}E)$에서도 변화를 나타내었다. 적색 2호와 청색 2호의 명도(L)는 조사선량에 의해 증가되었으며, 적색도(a)는 증가되었다. 녹색 3호의 경우에는 감마선 조사에 의해 적색도(a)와 황색도(b)가 모두 증가되었다. 이에 반해 $0.10\%$ 농도의 식용색소에서는 유의적인 변화를 일으키지 않는 것으로 나타났다.

Decolorization of synthetic dyes by Rhodopseudomonas palustris P4

  • A newly isolated Rhodopseudomonas palustris P4 could decolorize various synthetic dyes containing different chromogenic groups such as azo linkage (Crocein Orange G, New Coccine, Chromotrope FB, Congo Red, Remazol Black B), anthraquinone Reactive blue 2, or indigo Indigo Carmine. Among them, the degradation rate of Black B was studied in detial. Degradation of Black B followed the Arrhenius equation in 25 - $40^{\circ}C$ with an activation energy of 7.79 kcal/mol. Optimum pH was 8. Glucose in the range of 5 - 50g/l did not affect the Black B decolorization. When Black B increased from 25 mg/l to 2000 mg/l, decolorization activity increased almost linearly but the extent of decolorization was constant at about 86% irrespective of dye concentration. Analyses by HPLC revealed that the Black B molecules were partially degraded and some chromogenic intermediates were produced. These results indicate that Rps. palustris P4 has an outstanding capability to degrade various dyes.

Intercalation of Functional Organic Molecules with Pharmaceutical, Cosmeceutical and Nutraceutical Functions into Layered Double Hydroxides and Zinc Basic Salts

  • Negatively charged functional organic molecules such as retinoic acid, ascorbic acid, indole acetic acid, citric acid, salicylic acid, acidic dye (indigo carmine, Food Blue 1) are intercalatively encapsulated by zinc basic salt (hydrozincite) and layered double hydroxide. Such functional organic-inorganic nanohybrids are realized via coprecipitation reaction involving simultaneous formation of layered inorganic lattice and intercalation of anionic species. The heterostructural nature of these nanohybrids, their particle morphology and textural characterizations are mainly discussed on the basis of Powder X-ray Diffraction and Field Emission Scanning Electron Microscopy results.

  • Functional cosmetic ingredients such as L-ascorbic acid, retinoic acid, indole-3-acetic acid, salicylic acid, acidic dye(indigo carmine) are intercalatively encapsulated by skin-friendly metal hydroxides and oxides matrices. Such functional organic-inorganic nanohybrids are realized via chemical coprecipitation and surface coating reactions. The hetero-structural nature of these nanohybrids, their particle morphology and textural characterizations are mainly discussed on the basis of powder X-ray diffraction, electron microscopies, and high performance liquid chromatographic analyses. The cosmetic ingredients encapsulated in inorganics show greatly improved storage stability, sustained releasing property as well as higher transdermal transfer efficiency.

  • [ $SnO_2$ ]-doped $TiO_2$ thin films were prepared from tin (IV) bis (acetylacetonate) dichloride and titanium diisopropoxide bis (acetylacetonate) with pluronic P123 or degussa P25 as a structural-directing agent. These hydrolyzed sol were spin coated onto Si(100) wafer substrate. The microstructure, morphology and bonding states of thin films were studied by field-emission scanning electron microscopy (FE-SEM), X-ray diffractometry (XRD), and X-ray photoelectron spectroscopy (XPS). The photocatalytic activity of these films was investigated by using indigo carmine solution.

  • From dye-assisted conventional chromoendoscopy to novel virtual chromoendoscopy, image-enhanced endoscopy (IEE) is continuously evolving to meet clinical needs and improve the quality of colonoscopy. Dye-assisted chromoendoscopy using indigo carmine or crystal violet, although slightly old-fashioned, is still useful to emphasize the pit patterns of the colonic mucosa and predict the histological structures of relevant lesions. Equipment-based virtual chromoendoscopy has the advantage of being relatively easy to use. There are several types of virtual chromoendoscopy that vary depending on the manufacturer and operating principle. IEE plays distinctive roles with respect to histologic characterization of colorectal polyps and prediction of the invasion depth of colorectal cancers. In addition, the newest models of IEE have the potential to increase adenoma and polyp detection rates in screening colonoscopy.

식품 중 합성첨가물 사용실태 조사 연구 -타르색소 중심으로- (Evaluation of Tar Dyes Used in Commercial Foods)

  • 타르색소는 식품의 색상을 아름답게 하여 제품의 가치를 높이기 위한 수단으로 가공단계에서 다양하게 사용되고 있다. 따라서 식품 중에 첨가된 타르색소에 대한 정확한 분석으로 위생관리가 요구되고 있으나 현행 식품공전에는 정성시험법만이 수재되어 있으며 지금까지 연구 보고된 분석법들도 시료전처리가 복잡하고 색소별로 분석조건이 달라 동시 정량이 어려운 실정이다. 이에 본 연구에서는 시중 유통되고 있는 사탕, 청량음료, 빙과 및 옥춘 등의 식품에 첨가된 타르색소에 대하여 새로운 전처리방법과 HPLC에 의한 동시정량으로 분석시간을 단축하고 정확한 사용실태를 파악하고자 하였다. 황색4호, 황색5호, 적색2호, 적색3호, 적색40호, 청색 1호, 청색2호 등 7종의 타르색소에 대하여 시료 전처리법을 검토한 결과 aminopropyl amine cartridge를 이용한 색소 추출법으로 기존의 방법에 비해 전처리과정을 크게 단축할 수 있었으며 대상식품별 회수율은 사탕 및 옥춘 66.1∼95.6%, 청량음료 69.2∼99.6%, 빙과 65.8∼96.8%의 결과를 얻었다. 대상 식품 4종 100개 품목에 대한 식품별 색소사용농도는 사탕류 N.D.∼50.l mg/kg, 청량음료 N.D.∼49.9 mg/kg, 빙과류 N.D.∼56.0 mg/kg로 비슷하였고, 옥춘의 경우 N.D.∼867.3 mg/kg로 다른 식품보다 15배 이상 진한 농도로 사용되었음을 알 수 있었다. 색소별 사용빈도는 사탕류의 경우 황색4호, 청량음료는 청색1호, 빙과류는 적색2호가 가장 많이 사용되었고 모든 대상식품에서 가장 많이 사용된 색소는 황색4호, 전혀 사용되지 않는 색소는 청색2호로 나타났으며 2개 이상의 혼합색소 사용율이 많아 종합적인 검토의 필요성이 제기되고 있다. 대상식품의 대부분은 타르색소가 FAO/WHO에서 권장한 일일허용섭취량보다 안전한 수준으로 사용되고 있었으나 적색3호와 적색2호가 867.3 mg/kg, 191.2 mg/kg이 검출된 옥춘과 적색2호가 44.8 mg/kg이 검출된 빙과류 등은 안전성이 우려되었다 또한 발암성의 이유로 미국에서는 사용이 금지된 적색 2호가 우리 나라에서는 사탕 등 어린이 선호식품에 빈번하게 사용되고 있었다.

Decolorization of Acid Green 25 by Surface Display of CotA laccase on Bacillus subtilis Spores

  • In this study, we expressed cotA laccase from Bacillus subtilis on the surface of B. subtilis spores for efficient decolorization of synthetic dyes. The cotE, cotG, and cotY genes were used as anchoring motifs for efficient spore surface display of cotA laccase. Moreover, a $His_6$ tag was inserted at the C-terminal end of cotA for the immunological detection of the expressed fusion protein. Appropriate expression of the CotE-CotA (74 kDa), CotG-CotA (76 kDa), and CotY-CotA (73 kDa) fusion proteins was confirmed by western blot. We verified the surface expression of each fusion protein on B. subtilis spore by flow cytometry. The decoloration rates of Acid Green 25 (anthraquinone dye) for the recombinant DB104 (pSDJH-EA), DB104 (pSDJH-GA), DB104 (pSDJH-YA), and the control DB104 spores were 48.75%, 16.12%, 21.10%, and 9.96%, respectively. DB104 (pSDJH-EA) showed the highest decolorization of Acid Green 25 and was subsequently tested on other synthetic dyes with different structures. The decolorization rates of the DB104 (pSDJH-EA) spore for Acid Red 18 (azo dye) and indigo carmine (indigo dye) were 18.58% and 43.20%, respectively. The optimum temperature for the decolorization of Acid Green 25 by the DB104 (pSDJH-EA) spore was found to be $50^{\circ}C$. Upon treatment with known laccase inhibitors, including EDTA, SDS, and $NaN_3$, the decolorization rate of Acid Green 25 by the DB104 (pSDJH-EA) spore decreased by 23%, 80%, and 36%, respectively.

Sonochemical Synthesis of $PbMoO_4$ Nanoparticles and Evaluation of its Photocatalytic Activity

  • $PbMoO_4$ nanoparticles were successfully obtained in the presence of ethylene glycol (EG) with the assistance of a prolonged sonication process. The nanoparticles were characterized by X-ray powder diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), diffuse reflectance spectroscopy (DRS) and adsorption-desorption $N_2$ isotherms (BET). The catalyst prepared sonochemically showed higher photocatalytic activity than $PbMoO_4$ prepared by solid-state reaction in the degradation reactions of rhodamine B (rhB), indigo carmine (IC), orange G (OG), and methyl orange (MO) under UV-Vis light radiation. In order to elucidate aspects of the degradation mechanism of the organic dyes, some experimental variables were modified such as pH, $O_2$ level in solution, and radiation source. In general, the photocatalytic activity for the degradation of organic dyes followed the sequence IC>OG>rhB>MO.
