Characterization of Prunus necrotic ringspot virus Isolate from Peach in Korea (국내 복숭아에서 분리한 Prunus necrotic ringspot virus의 특성)

  • In this paper, we report a characterization of Prunus necrotic ringspot virus (PNRSV) isolate. The virus was identified from 'Yumyeong' peach showing mild mosaic on leaves in commercial orchard of 'Umsung', Chungbuk province in Korea. The virus isolate produced ringspot symptom on the inoculated cotyledons and systemic mosaic and malformation on the upper leaves of Cucumis sativus. Systemic mottles were appeared in Chenopodium quinoa. When the buds of the virus infected stem were grafted on the healthy young Prunus persica GF305 seedlings, line pattern with mosaic appeared within 3 months. Isometric virus-like particles were found in parenchyma cells and plasmodesmata of C. sativus leaves inoculated mechanically with the virus. The cDNA fragments of PNRSV coat protein (CP) region, approximately 675bp, were synthesized from genomic RNA extracted from virus-infected leaves by RT-PCR using specific primer pairs. Partial nucleotide sequences of the CP regions were determined and analyzed with the known PNRSV. The CP gene of PNRSVKorea isolates showed 93.9~94.7% similarity to the 4 known PNRSV isolates.

Occurrence and Symptoms of Tomato Spotted Wilt Virus on Vegetables in Korea (I) (채소류의 토마토 반점 위조 바이러스 발생과 병징 (I))

  • Tomato spotted wilt virus (TSWV) occurred abruptly with a high incidence rate in 14 vegetable crops in Anyang area, Gyunggido in 2004. TSWV was identified by the symptoms on the indicator plants, immunocaptured reverse transcription-polymerase chain reaction (IC/RT-PCR), virion captured (VC)RT-PCR and RT-PCR using total RNA from the infected plants. Vegetable crops infected with TSWV included table tomato, cherry tomato, red pepper, lettuce, chicory, red leaf chicory, red mustard, dragon tongue, treviso, potato, perilla, sesame, pumpkin, and ssamchoo (hybrid of oriental cabbage and cabbage). The incidence of TSWV in fields ranged from 30 to $100\%$. TSWV usually produced necrosis, wilt and/or severe mosaic with typical single or double ring spots on the leaves. TSWV could be detected in Frankliniella occidentalis collected from the crops in the fields with $90\%$ rate by IC/RT-PCR.

Comparison of nutrient balance and nutrient loading index for cultivated land nutrient management (농경지 양분관리를 위한 양분수지 지표와 양분부하 지표간의 비교)

  • Recently, concerns regarding the environmental impact due to nutrient input in croplands have increased. Therefore, the government is promoting the introduction of a nutrient management system in croplands to solve the problem of excessive nutrient input. This study was carried out to establish nutrient indicators in regional croplands to facilitate the introduction of the national nutrient management system in Korea. The nutrient load and balance indicators for nitrogen and phosphorus were analyzed for nine provinces (Gang-won, Gyeong-gi, Chung-buk, Chung-nam, Jeon-buk, Jeon-nam, Gyeong-buk, Gyeong-nam, and Jeju). In the correlation analysis between the nutrient load and nutrient balance, the correlation coefficient (r) for nitrogen was 0.2504, which was not statistically significant at the 5% significance level. However, the correlation coefficient for phosphorus was 0.7375, which was statistically significant at the 5% significance level. In the nutrient management index, phosphorus showed mutual compatibility between the nutrient load and the nutrient balance indicators, but nitrogen showed no mutual compatibility between the nutrient load and the nutrient balance indicators. Therefore, utilization of the nutrient balance indicator, reflecting the characteristics of the agricultural environment, was more reasonable as a nutrient management index for regional nutrient management.

Changes of Lactulose Content during Heat Treatment of Milk (우유의 열처리 및 저장 조건에 따른 Lactulose의 함량 변화)

  • The aim of this work to determine the formation of lactulose during heat treatment process as a contribution to the estabilishment of limits of chemical indicators for different types of heat processed milk and analyze of lactulose for the reconstituted milk added samples. The HTST(75$\^{C}$/15s) and UHT(130$\^{C}$/2∼3s) treatment realized with a pilot plant and heat-treated samples were stroed at 4, 10, 30$\^{C}$ for 4 weeks. Changes in lactulose was evaluated at 7 days intervals. The other heat treatment was sealed in glass tube and heated at 75$\^{C}$ for 10 to 120s and heated at 130$\^{C}$ for 2 to 60s in a thermostatically controlled constant temperature bath of glycerol. The reconstituted milk was made with full fat dry milk that reconstituted with deionized water to 10% total solid, and was added to milk at 10, 20, 30% respectively. The samples processed with a HTST pilot plant showed that lactulose was contained at 1.47∼1.52mg/10()ml and 8.19 ∼8.32mg/100ml for UHT-treated samples. Changes in the lactulose content of heat-treated samples during storage at 4 and 10$\^{C}$ for 4 weeks caused a slight increase, however a noticeable increase was observed at 30$\^{C}$ for 4 week. The glass tube samples showed that high correlations between relative increase in content of lactulose and increasing processing times(75$\^{C}$ : r = 0.986, 130$\^{C}$ : r = 0.987, respectively). Added with reconstituted milk would cause a increase of the lactulose content linear with increasing addition amount(r = 0.982). This results observed for lactulose in commercial milk samples would applied to the detection of chemical changes during heat treatment and illegal use of reconstituted milk.

The Vascular Plants in Mt. Gaji (Gyeongsang nam-do) (가지산(경남)의 관속식물상)

  • This study was carried out to investigate the flora of Mt. Gaji(Gyeongsang nam-do). The vascular plants which are collected in 12 times(from May to Oct. 2006 and Mar. to Oct. 2009) were identified as 573 taxa in total, including 106 families, 314 genera, 503 species, 6 subspecies, 56 varieties, 8 forms. There were many useful plants such as 20 taxa of Korean endemics and 10 taxa of specially designated plants which were determined by the Korea Forest Service. The designated in endangered plants by the Ministry of Environment, which should be protected by the wildlife protection law, were identified as 3 taxa and 64 taxa of specially designated floristic regional indicator plants by the Ministry of Environment. Based on the list of approved for delivering overseas of plants, 23 taxa were recorded in the investigated area. The naturalized plants were identified as 22 taxa, and their naturalization ratio were found to be 3.8% respectively. The 573 taxa listed consists of 217 taxa(37.8%) of edible plants, 181 taxa(31.5%) of medicinal plants, 91 taxa(15.8%) of pasture plants, 86 taxa(15%) of ornamental plants, 19 taxa(3.3%) of timber plants, 11 taxa(1.9%) of fiber plants and 7 taxa(1.2%) of industrial plants.

Distribution of Medicinal Plants and Vascular Plants in Mt. Nochu (Jeongseon-gun, Gangneung-si), Korea (노추산(정선군, 강릉시)의 관속식물상 및 약용식물 분포 특성)

  • This study was carried out to investigate the vascular plants of Mt. Nochu (1,322m) from April, 2014 to October, 2015. The vascular plants were summarized as 515 taxa, including 89 families, 304 genera, 448 species, 4 subspecies, 57 varieties and 6 forms. Among the investigated 515 taxa, 11 Korean endemic plants and 14 rare and endangered plants were included. The floristic regional indicator plants found in this area were 11 taxa of grade IV, 30 taxa of grade III. Naturalized plants consisted of 24 taxa that made up 4.6% of the total vascular plants in this area. Medicinal plants of the Korean Pharmacopoeia and Korean Herbal Pharmacopoeia distributed in this area consisted of 67 taxa.

Floristic study of Sobaeksan National Park in Korea (소백산 국립공원의 관속식물상)

  • The flora in the region of Mt. Sobaeksan National Park ($N36^{\circ}50^{\prime}-37^{\circ}50^{\prime}$, $E128^{\circ}20^{\prime}-128^{\circ}43^{\prime}$) was surveyed from April of 2005 to October of 2011. The vascular plants from 27 field surveys were revealed to total 869 taxa, comprising 112 families, 429 genera, 756 species, four subspecies, 91 varieties and 18 forms. Sixty-eight taxa were the first records from this region. This field study discovered significant plants in various categories. Korean endemic plants numbered 25 taxa, and 18 taxa designated by the Korean Forest Service as rare plants were investigated in this region. The taxa in category II of rare and endangered plants and higher than the third degree among the floristic regional indicator plants designated by the Korean Ministry of the Environment were three taxa and 75 taxa, respectively. From a geographical perspective, limited distribution of Saxifraga octopetala, Corydalis grandicalyx, Aegopodium alpestre, and Polygonatum acuminatifolium at a latitude higher than that of Sobaeksan National Park suggest that Sobaeksan National Park might be the limit line of the distribution of the taxa. Stewartia koreana demonstrate a reversed pattern from the taxa above, occurring at a latitude lower than that of Sobaeksan National Park. In addition, naturalized plants of 53 taxa were recorded.

Flora of the vascular plants of the Baekdudaegan conservation area: Deok-chi to Yuk-sim-nyeong (백두대간보호지역의 식물상: 덕치-육십령 구간)

  • Baekdudaegan, the largest mountain range in eastern Asia, is a biodiversity hotspot in Korea that may have served as a glacial refugium. This study presents the flora of vascular plants on Deok-chi upto the Yuk-sim-nyeong area of the Baekdudaegan conservation area. The survey area was divided into four subareas and fieldwork was conducted for a total of twelve days in 2015. Voucher specimens were collected during the survey and were deposited at Daejeon University. A list of vascular plants was prepared based on the voucher specimens. The results of the survey showed that a total of 441 taxa, consisting of 100 families, 265 genera, 398 species, 9 subspecies, 32 varieties, and 2 forms, were found in the survey area. There was one endangered species, Aconitum coreanum, in the Bonghwasan Mt. area. Sixteen endemic taxa, 74 floristic regional indicator plants, as designated by the Ministry of the Environment, and eleven naturalized plants were distributed. The results of this study can serve as basic information to establish conservation and management plans for the Baekdudaegan conservation area.

Ecological Studies on Several Forest Communities in Kwangnung. A Study of the Site Index and the ground vegetation of Larch (광릉삼림의 생태학적 연구 낙엽송의 Site Index와 임상식생에 관하여)

  • In order to determine the factors related to site quality, 13 areas of Larch growing in the Kwangung and its vicinity forest as sample plots, were examined. Sample plots included various site classes as well as age classes. Three were divided into two groups (major and minor trees). Average height of dominant trees was determined through messurement of 5 to 6 dominant tree in each sample plots. Average height of dominant 30 year-old trees was the basis for site index. A Standard Yield Table for the larch produced in Kwangnung forest was made by various data, which included age class 5, ranging from 10 to 45 years. The relationship of the height of the trees, the site conditions, and ground vegetation are investigated in this paper. The site indexes of 40 forest class age in 28-B and 28-G forest classes of the larch associations for ground vegetation had comparatively rarge differences due to the sampled areas. The relation of the direction of forest communities to the height and the diameter of the tree shwoed that its communiteis of northest and northwest parts appeared higher valueof the height and the diameter. The diameter and the height of trees were closely realted to each other. The samller the occupied area per tree and the smaller the average distance among trees, the more density was increased. The larger the density was the lower height of the trees. In the ground vegetation of the larch communities, there seems to be a definite correlation between the height of trees and the occupied area per tree or the average distance among the trees. The height of trees and site index of two larch communities were as follow: 28-B forest class site index 20.8, height 24.0m, 28-G forest class site index 18.4, height 20.9m. The ground layer was analyzed by the method of Quadrat(20/20sq. cm) with an interval of 1M. It set up 40 Quadrats of the larch communiteis. The community structure of the ground vegetation of two larch was analyzed, and important value was calculated and then evaluated. The ground vegetation under the larch had developed Burmannii Beauv stratal society below the 28-B and 28-G the forest class. Accordingly, the first important value of Burmannii Beauv was found in two ground vegetation below the larch. Therefore, this species could be quantitatively considered as the forest indicator species. Common species of each community appeared 18 species out of 34 species in the ground vegetation under two larch communities. The ground vegetation of the 28-B forest class showed more than that of the 28-G forest class. the similarity of the ground vegetation was measrued by the Frequency Index Community Coefficient. The differences between the associations were lcearly manifested by the ground vegetation tested by Gleason's Frequency Index of Community Coefficient for the analysis of each stratal society of all associations. According to F.I.C.C. the ground vegetation under two larch(28-B and 28-G) forest classes showed higher value. An investigation into the relationship of physical and chemical properties of soil and site was considered the next step to be taken in the study of the larch site classification.

Analysis of Combining Ability for Quality and Yielding Characteristics in Rapeseed (Brassica napus L.) (유채의 품질과 수량 특성에 대한 조합능력 분석)

  • Naehanyuchae and Youngsanyuchae were originated from the cross between "Erra(female)" and "Tower(male)". The cultivars were made at Bioenergy Crop Research Center, National Institute of crop science, Rural Development Administration, Korea in order to develop the high cold tolerant and yielding rapeseed variety with non-erucic acid and non-glucosinolate as good seed quality indicator. The superiorities of these varieties have been shown through the regional performance trials and demonstration farmers fields. Naehanyuchae and Youngsanyuchae had good agronomic characteristics related to yield potential such as many valuable branches and pods, and long pod length. The seeds of these new varieties composed of the 80% of oleic acid and linoleic acid without erucic acid, and glucosinolate was not extracted in the oil cake. The average grain yields increased by the from 16% to 22% according to the regional performance trials compared to check variety.
