Early Disease Development and Stem and Leaf Water Content in the Seedlings of Pinus koraiensis Inoculated with Pinewood Nematodes in a Greenhouse

  • Korean pine (Pinus koraiensis Sieb. et Zucc.), a five-needle pine, has recently been suffering pine wilt disease caused by non-native pinewood nematode, Bursaphelenchus xylophilus. Three-year-old Korean pine seedlings were inoculated with 10,000 pathogenic nematodes in a greenhouse to investigate disease development, water content and the density of nematodes in stems. Needle dehydration, xylem drying and pith browning started 20 days after inoculation (DAI). There were significant differences between seedlings inoculated with nematodes and control seedlings in the relative water content of stems and leaves at 20 and 30 DAI. At 60 DAI, all remaining seedlings inoculated with nematodes had died, but control seedlings all remained alive. The average number of nematodes recovered from stems of Korean pine dramatically increased from 10 to 20 DAI, and then decreased at the end of the experiment at 60 days. This study suggests that the relative water content of stems and leaves in current-year branches could be used as a useful physiological indicator for early diagnosis of pine wilt disease.

Studies on the Pear Abnormal Leaf Spot Disease - 3. Graft Transmissibility of the Causal Agent - (배나무잎 이상반점증상에 관한 연구 - 3. 병원의 접목전염 -)

  • Nature of graft transmissibility of pear abnormal leaf spot disease was examined by various grafting methods in the greenhouse and field. When the diseased and symptomless twigs were collected in winter and grafted in the next spring to the seed-originated healthy root stock, the abnormal leaf spot was developed only in the case of the diseased twigs. Double grafting on a seed-originated healthy root stock, where the diseased and the symptomless twigs were used as 1st and 2nd scions, respectively, developed abnormal leaf spot lesions without exception on the 2nd scions. Tongue-graft with the diseased and the symptomless trees also incited abnormal leaf spots on the both trees. Abnormal leaf spots of were also developed on HN-39, an indicator pear tree, used as a 2nd scion in a double graft test, where the diseased twig and a seed-originated healthy tree were used as the 1st scion and the root stock, respectively. When the diseased twig was top-grafted to the healthy root stock, lesion development of abnormal spot was limited to the grafted twig itself in the 1st year, but expanded to the main branches in the 2nd year, and spread over the whole tree in the 3rd year. This result indicates that the causal agent of abnormal leaf spot disease is transmitted by graft.

Atmospheric Polycyclic and Nitropolycyclic Aromatic Hydrocarbons in an Iron-manufacturing City

  • Total suspended particulates (TSP) in the atmosphere were collected for 2 weeks during winter in Muroran, Hokkaido, Japan, a typical iron-manufacturing city. The concentrations of polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs) in TSP were determined by high-performance liquid chromatography (HPLC) using fluorescence and chemiluminescence detectors, respectively. No relationship was observed between the atmospheric PAH and NPAH concentration, or the atmospheric PAH and TSP concentration. However, there was a tendency that the atmospheric PAH concentration was higher when the wind blew from the coke-oven plant. Furthermore, the concentration ratios of 1-nitropyrene to pyrene, which is a suitable indicator of the contribution made by automobiles and coal combustion systems to urban air particulates, were smaller in Muroran and the values were close to those observed in particulates from coal combustion systems. Therefore, these results show that the PAH and NPAH compositions for Muroran are characteristic of an iron-manufacturing city.

1-Aminocyclopropane-1-Carboxylate Deaminase from Pseudomonas fluorescens Promoting the Growth of Chinese Cabbage and Its Polyclonal Antibody

  • Bacterial 1-aminocyclopropane-1-carboxlyate (ACC) deaminase (AcdS) is an enzyme that cleaves ACC, a precursor of the plant hormone ethylene, into ${\alpha}$-ketobutyrate and ammonia. The acdS gene was cloned from Pseudomonas fluorescens, which was capable of improving the seedling of Chinese cabbage under salinity condition. The recombinant AcdS (rAcdS) exhibited optimal activity at pH 8.5 and $30^{\circ}C$. Strong activity was sustained at up to 100 mM NaCl. The polyclonal anti-P. fluorescens AcdS antibody was produced in a rabbit that had been immunized with the purified rAcdS. This antibody successfully recognized the homologous antigens derived from the total proteins of isolated plant growth-promoting microorganisms. A statistically significant correlation was observed between the intensity of hybridization signal and AcdS activity measured by a biochemical method, suggesting its application as a useful indicator for active deaminases.

Floristic study of bryophytes in a subtropical forest of Nabeup-ri at Aewol Gotjawal, Jejudo Island

  • This study presents a survey of bryophytes in a subtropical forest of Nabeup-ri, known as Geumsan Park, located at Aewol Gotjawal in the northwestern part of Jejudo Island, Korea. A total of 63 taxa belonging to Bryophyta (22 families 37 genera 44 species), Marchantiophyta (7 families 11 genera 18 species), and Anthocerotophyta (1 family 1 genus 1 species) were determined, and the liverwort index was 30.2%. The predominant life form was the mat form. The rates of bryophytes dominating in mesic to hygric sites were higher than the bryophytes mainly observed in xeric habitats. These values indicate that such forests are widespread in this study area. Moreover, the rock was the substrate type, which plays a major role in providing micro-habitats for bryophytes. We suggest that more detailed studies of the bryophyte flora should be conducted on a regional scale to provide basic data for selecting indicator species of Gotjawal and evergreen broad-leaved forests on Jejudo Island.

Sequestration of Orthophosphate by D(+)-Mannose Feeding Increases Nonphotochemical Quenchings in Chinese Cabbage Leaves (Mannose 처리된 배추 잎의 무기인산 감소에 따른 비광화학성 소산의 증가)

  • Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.

On the Accumulation of Radioactive Materials in Marine Organisms Along the Coast of Korea 3. Cesium-137 Activities in Several Edible Marine Algae

  • Continued to the previous papers, the present investigation is carried out to clarify cesium-137 activities and potassium contents among 54 samples of edible marine algae collected along the coast of Korea during September, 1973 and April, 1974. The potassium contents are variable, 1.5-29.6%, and 11.36% on an average, while most of the members contain them about 10-16% generally. Among the algal phyla, they are 13.49% in green, 10.82% in brown and 16.46% in red algae, respectively. Cesium-137 activities are variable, 0.19-8.13 pCi$\^$137/ Cs/g K, and 2.35 pCi$\^$137/Cs/g K on an average in the samples investigated. Among the algal phyla they are 4.35 in green, 2.17 in brown and 0.89 pCi $\^$137/Cs/g K in red algae, respectively. Green algae contain especially higher activities compared with the other two groups. The brown alga, Myelophycus caespitosus shows the highest activities, 8.13 pCi$\^$137/Cs/g K, among the members investigated. Considering the concentration factor, this plant would be an indicator plant of cesium-137 among the marine algae along the coast of Korea.

Distribution of Fecal Sterols, Nonylphenol, and Polycyclic Aromatic Hydrocarbons in Surface Water from Masan Bay, Korea

  • Fecal sterols, nonylphenolic compounds (NPs), and polycyclic aromatic hydrocarbons (PAHs) were determined in surface water from Masan Bay and its adjacent rivers in February 2005. Concentrations of coporstanol (Cop), an indicator of fecal pollution, in surface water ranged from <10 to 13,853 ng/L, and concentrations of nonylphenol, the most toxic of the NPs, ranged from 10.2 to 481 ng/L, and concentrations of PAHs ranged from 8.61 to 223 ng/L. The concentrations of the compounds measured in this study were lower than or comparable to those at other locations in Korea and other countries. The contamination of Cop and PAHs in surface water was associated with the discharge from rivers passing through cities and/or industrial complexes. The NP contamination was associated with wastewater treatment plant (WWTP) effluents through outfalls as well as riverine discharge. Compared to ecotoxicological values, the concentrations of NPs from rivers, the mouths of rivers, and WWTP outfall areas exceeded guidelines, suggesting that hot spot areas may pose a potential risk to sensitive species.

Phyto-toxicological Effects of PAHs on the Germination and Growth of Alfalfa, Barley, and Tall Fescue (종자 발아율 지표 및 초기 묘조 성장에 의한 다환방향족탄화수소의 식물 독성 측정)

  • The test of germination has been used as a good indicator to assess the toxicity of chemicals to plant. This study was carried out to investigate the effect of PAHs, anthracene and phenanthrene, on germination ratio, germination index and germination time of barley, tall fescue and alfafa. Anthracene and phenathrene induced to decrease the seedling length and increase the germination time of the experimental plants. But they did not affect the maximum germination ratio. While it was not correlated between seedling length and final germination ratio, it was shown a close relationship between seedling length and mean germination time of the plants. These results suggested that the mean of germination time and seedling growth could be useful to exmine the phyto -toxicological effect of PAHs. The mixture of anthracene and phenanthrene was examined to investigate the combined effect on seedling length; the mixture induced the reduction of seedling. This was meant that the mixture toxicity of PAHs might be not shown in the sum of each chemical toxicity.

  • In the face of accelerating biodiversity loss and its significance in our coexistence with nature, biodiversity is becoming more crucial in sustainable development perspective. To estimate biodiversity in the future which provides valuable information for decision making system especially in the national level, a quantitative approach must be studied forehand as a baseline of the present status. In this study, we developed a large-scale map of Plant Species Richness (PSR, typical indicator of biodiversity) for Young-dong and Pyung-chang provinces. Due to the accessibility of appropriate data and advance of modelling techniques, reduction of variables without deteriorating the predictive power is considered by applying Genetic algorithm. In addition, a number of Correctly Classified Instances (CCI) with 10-fold cross validation which indicates the predictive power, was carried out for evaluation. This study, as a fundamental baseline, will be beneficial in future land work as well as ecosystem restoration business or other relevant decision making agenda.
