  • 가축산업분야에서 항생제의 사용이 금지됨에 따라, 질병 예방을 통한 축산농가의 생산성 향상을 위해 사료첨가제인 미생물제재의 개발과 같은 예방적 수단이 필요하게 되었다. 본 연구는 가축의 생산성을 높이기 위해 사료 분해 능력이 좋고 항균활성이 뛰어난 포자 형성 Bacillus 균주 3종인 B. sutilis LCB7, B. licheniformis SHS14, B. amyloliquefaciens LCB10을 우수 균주로 선발하였다. 최종적으로 선발한 Bacillus 3종을 1:1:1 비율로 혼합하여 혼합 종균을 제조하여 항균시험(in vitro) 결과, 단일 3종 및 lincomycin과 비교하여 유사한 활성을 보여주었으며, 송아지를 이용하여 항균활성 시험(in vivo)을 실시한 결과에서도 lincomycin 투여 대비 90% 수준의 높은 활성을 보여주었다. 개발한 혼합 종균의 안정적 보존을 위해 혼합 종균을 제조하여 다시 증균을 통하여 미생물군집 분석을 통하여 확인한 결과, 초기 군집비율과 증균 후 군집비율이 매우 유사하게 유지되었다. 이로서 본 연구에서는 선발된 Bacillus 균주 3종을 이용하여 제조한 혼합 종균이 사료첨가제용 미생물제재로서 이용 가능함을 최종 확인하였다.

버섯 배지를 이용한 tyrosinase 저해제 발효

  • Tyrosinase is an enzyme which catalyzes an enzymatic browning of some foods and in vivo synthesis of melanin. In order to produce natural and edible inhibitor of the enzyme which is expected to have whitening effect on melanogenesis, a microorganism was selected from fermented foods. It was named as NU-7, and cultured in mushroom (Lentinus edodes, Shiitake) media. Optimal media to produce tyrosinase inhibitor was formulated by varing nitrogen or carbon content. If glucose content was in a range of 3-20% and ammonium sulfate was in a range of 0-0.25%, production of inhibitor was independent of cell mass. Addition of ammonium sulfate as a nitrogen source had little effect on inhibitor production. Production of inhibitor (Y) was proportionally related to shiitake content (X) with a regression equation of Y= -0.96X$^{2}$ + 13.07X + 14.43 (R = 0.96). These results indicate that shiitake and glucose are necessary for the production of tyrosinase inhibitor. In the analysis of mycotoxin in culture broth, aflatoxin was not detected, suggesting that it would be probably edible.

유색미 미강발효물의 면역활성 효과 (In Vivo Immunological Activity in Fermentation with Black Rice Bran)

  • 본 연구는 유색미 미강을 표고버섯 균사체로 발효한 1차 발효물(CFP)과 1차 발효물을 효모와 유산균으로 2차 발효시킨 2차 발효물(CFP-S, CFP-L)을 이용하여 in vivo 상에서 면역 활성을 측정하였다. 4주간 각 물질을 250 mg/kg BW/day로 경구 투여시킨 후 마우스 복강 대식세포와 비장세포를 분리하여 각각의 세포 증식도을 평가하였으며, 각 세포에서 분비하는 사이토카인의 분비량을 측정하였다. 실험 결과, 마우스 대식세포의 증식은 CFP 경구 투여군과 CFP-S 경구 투여군이 높게 나타났다. 또한 비장세포의 증식에 있어서는 CFP 경구투여군이 대조군에 비해 높게 나타났다. 마우스 대식세포에서 분비하는 사이토카인의 분비량을 측정한 결과, CFP-S군은 IFN-${\gamma}$, TNF-${\alpha}$, IL-6의 분비량을 모두 증가시켰으며, CFP 경구 투여군은 LPS를 처리하여 대식세포를 활성화시킨 세포에서 IFN-${\gamma}$, IL-6의 분비량이 증가하였다. CFP-L 경구 투여군에서는 LPS를 처리한 세포에서 TNF-${\alpha}$의 분비량이 증가됨이 관찰되었지만 미비한 수준이었다. 비장세포에서 분비하는 사이토카인의 농도 측정 결과는 CFP 경구 투여군에서 모든 사이토카인(IFN-${\gamma}$, TNF-${\alpha}$, IL-6) 분비량을 증가시켰으며, CFP-S는 LPS를 처리한 비장세포에서 분비하는 사이토카인 중 TNF-${\alpha}$, IL-6의 분비량이 증가됨을 관찰하였다. CFP-L 경구 투여 군에서는 사이토카인 분비량이 대조군에 비해 모두 감소되었다. 이상의 연구 결과를 바탕으로 유색미 미강 1차 발효물(CFP)와 2차 발효물 중 효모로 발효시킨 CFP-S가 마우스에 경구 투여 시 면역과 관련된 세포의 활성을 증가시키고, 이 세포에서 분비되는 면역인자인 사이토카인의 분비량을 증가시키는 것으로 나타났다. 반면, 2차 발효물 중 유산균으로 2차 발효 시킨 CFP-L은 면역 활성에 효과를 나타내지 않았다. 여러 선행연구에서는 미강 표고버섯 균사체 발효물이 면역 증진작용 가진다고 보고하고 있다. 이는 발효물에 생리활성을 나타내는 기능성 다당체가 포함되어 있기 때문이다. 따라서 본 연구에서는 일반 미강에서 유색미 미강을 이용하고, 더 나아가 1차 발효물을 효모와 유산균으로 2차 발효를 함으로 좀 더 저분자화된 다당체를 생성함으로 면역 증진 기능의 상승효과를 기대하였다. 특히, 2차 발효에 사용한 효모는 자체 내세포벽에 베타-글루칸을 함유한 것으로 현재 기능식품소재로 사용되고 있으며, 여러 연구 결과, 면역 증강 작용이 확인되었다(Kim 등 2008). 또한 유산균종은 BRM(biological response modifier)이라 불리는 면역조절물질로 알려져 있다(Hong 2005). 그러나 연구 결과, 유산균을 통한 2차 발효물은 면역기능 평가 시 효과적이지 못하였다. 또한 본 연구에서는 유색미 미강발효물의 면역효과는 기능성 다당체와 더불어 유색미에 포함되어 있는 안토시아닌이라는 항산화 성분의 효과라고 생각된다. 그러나 본 연구에서는 유색미 미강 발효물과 일반미의 미강발효물의 면역활성 효과를 비교하지 않았고, 유색미 미강의 어떤 물질이 면역활성에 기여했을 지는 판단하기 어렵다. 따라서 앞으로 1차 발효물과 2차 발효물의 기능성 다당체의 성분과 또한 항산화 효과를 기대한 안토시아닌 함량 분석하고, 2차 발효시 미강에 존재하는 기능성 다당체들의 수율과 활성의 변화를 평가할 필요가 있다고 판단된다. 이 같은 연구는 유색미 미강의 이용가치를 높이고, 식품 소재 개발에 있어서 중요한 기초자료가 될 것으로 보인다.

Monascus pilosus M-15에 의한 콜레스테롤 합성저해제 Monacolin의 생산 (Production of Cholesterol Inhibitor, Monacolin Produced from Monasces pilosus M-15)

  • The monacolin retard in vivo synthesis of cholestrol. This study was conducted to select the best strain of Monascus sp. for monacolin production among 21 strains. In addition, research on suitable condition for mass production was conducted. Monascus pilosus M-15 was selected as the best strain for production of monacolin. The appropriate substrate for the monacolin production seem to be as follow : glycerin 5%, glucose 2%, corn steep liquor 0.5%, sodium nitrate 0.5% and length of fermentation was at $25^{\circ}C$ for 10~12days, respectively. The monacolin obtained from this organism was compared with standard monacolin using UV, IR, and GC/MS system and it was very close to the standard monacolin.

Effects of Tween 80 on In Vitro Fermentation of Silages and Interactive Effects of Tween 80, Monensin and Exogenous Fibrolytic Enzymes on Growth Performance by Feedlot Cattle

  • The effects of monensin, Tween 80 and exogenous fibrolytic enzymes on ruminal fermentation and animal performance were studied in vitro and in vivo. In Expt 1, the effects of the surfactant Tween 80 (0.2% wt/wt, DM basis) on ruminal fermentation of alfalfa, corn and orchardgrass silages were investigated using in vitro gas production techniques. Tween 80 did not affect (p>0.05) cumulative gas production at 24 h, but it reduced (p<0.05) the lag in fermentation of all three silages. With corn silage and orchardgrass silage, gas production rates and concentrations of total volatile fatty acids (VFA) were increased (p<0.05) by Tween 80; with alfalfa silage, they were reduced (p<0.05). Tween 80 increased (p<0.05) the proportion of propionate in total VFA, and reduced (p<0.05) acetate to propionate ratios (A:P) with all three silages. In Expt 2, exogenous fibrolytic enzymes (E; at 0, 37.5 or 75 g/tonne DM), monensin (M; at 0 or 25 ppm and Tween 80 (T; at 0 or 2 L/tonne DM) were added alone or in combination to backgrounding and finishing diets fed to 320 crossbred steers in a feeding trial with a $3{\times}2{\times}$2 factorial arrangement of treatments. The backgrounding and finishing diets contained barley grain and barley silage in ratios of 57.8:42.2 and 93.5:6.5 (DM basis), respectively. Added alone, none of the additives affected DM intake (p>0.1) in the backgrounding or in the finishing period, but interactive $M{\times}T$ effects were observed in the finishing period (p=0.02) and overall (p=0.04). In the finishing period, T without M tended to reduce DM intake (p=0.11), but T with M increased (p=0.05) DM intake. Monensin increased average daily gain (ADG) during backgrounding (p=0.07) and finishing (p=0.01), and this ionophore also improved overall feed efficiency (p=0.02). Warm carcass weight was increased (p<0.001) by M, but dressing percentage was reduced (p=0.07). In the backgrounding period, T increased ADG by 7% (p=0.06). Enzymes increased (p=0.07) ADG by 5 and 6% (low and high application rates, respectively) during backgrounding, but did not affect (p>0.10) ADG during finishing, or overall feed efficiency. Whereas T enhanced the positive effects of M on ADG during backgrounding (p=0.04) and overall (p=0.05), it had no impact (p>0.1) on the effects of E. Interactions between M and T suggest that the surfactant may have potential for enhancing the positive effects of monensin on beef production, but this requires further research.

Effects of L-glutamine supplementation on degradation rate and rumen fermentation characteristics in vitro

  • Objective: Two follow-up studies (exp. 1 and 2) were conducted to determine the effects of L-glutamine (L-Gln) supplementation on degradation and rumen fermentation characteristics in vitro. Methods: First, rumen liquor from three cannulated cows was used to test L-Gln (50 mM) degradation rate and ammonia-N production at 6, 12, 24, 36, and 48 h after incubation (exp. 1). Second, rumen liquor from two cannulated steers was used to assess the effects of five levels of L-Gln including 0% (control), 0.5%, 1%, 2%, and 3% at 0, 3, 6, 12, 24, 36, and 48 h after incubation on fermentation characteristics, gas production, and degradability of nutrients (exp. 2). Results: In exp. 1, L-Gln degradation rate and ammonia-N concentrations increased over time (p<0.001). In exp. 2, pH was reduced significantly as incubation time elapsed (p<0.001). Total gas production tended to increase in all groups as incubation time increased. Acetate and propionate tended to increase by increasing glutamine (Gln) levels, whereas levels of total volatile fatty acids (VFAs) were the highest in 0.5% and 3% Gln groups (p<0.001). The branched-chain VFA showed both linear and quadratic effects showing the lowest values in the 1% Gln group particularly after 6 h incubation (p<0.001). L-Gln increased crude protein degradability (p<0.001), showing the highest degradability in the 0.5% Gln group regardless of incubation time (p<0.05). Degradability of acid detergent fiber and neutral detergent fiber showed a similar pattern showing the highest values in 0.5% Gln group (p<0.10). Conclusion: Although L-Gln showed no toxicity when it was supplemented at high dosages (2% to 3% of DM), 0.5% L-Gln demonstrated the positive effects on main factors including VFAs production in-vitro. The results of this study need to be verified in further in-vivo study.

고지방식이로 유도된 비만 C57BL/6 쥐에서 발효 무궁화 추출물의 항산화 활성 (The Anti-oxidative Activities of Fermented Hibiscus syriacus Extract in High Fat Diet-induced obese C57BL/6 Mice)

  • 본 연구의 목적은 비만 쥐에서의 발효 무궁화 추출물의 항산화 효과를 평가를 위해 수행되었다. 항산화 실험은 in vivo와 in vitro 둘 다 진행하였다. In vitro 실험은 산화질소 자유 라디칼 소거활성을 사용하여 측정하였다. 발효 무궁화 추출물에서의 산화질소 자유 라디칼 소거활성은 비발효 무궁화 추출물보다 높게 나타났다. In vivo 실험은 정상식이군(NOR), 고지방식이군(CON), 고지방식이+발효 무궁화 추출물군(FH)과 고지방식이+비발효 무궁화 추출물군(NFH)로 나눴고, 4군은 4주 동안 복강투여하였다. 그리고 간 균질액의 SOD, CAT, MDA 수준을 측정하였다. 그 결과 FH군의 SOD와 CAT 수준은 CON군보다 높게 나타났으며, FH군의 MDA 수준은 CON군에 비해 현저하게 감소하였다. 결론적으로 발효 무궁화 추출물은 비만 쥐에서의 산화적 스트레스 억제 효과와 항산화 효소 활성 증가 효과를 가지며, 유용한 천연 항산화제의 원료로 사용할 수 있음을 제안한다.

Effects of Lactic Acid Bacteria, Storage Temperature and Period on Fermentation Characteristics, and in vitro Ruminal Digestibility of a Total Mixed Ration

  • This study evaluated the effect of lactic acid bacteria (LAB, a mixture of Enterococcus faecium and Lactobacillus plantarum) supplementation, the storage temperature, and storage period on the fermentation characteristics and in vitro ruminal digestibility of a total mixed ration (TMR). The TMR was prepared into two groups, namely, CON (control TMR without the LAB) and ML (supplementing a mixture of E. faecium and L. plantarum in the ratio of 1% and 2% (v/w), respectively). Both groups were divided and stored at 4℃ or 25℃ for 3, 7, and 14 d fermentation periods. Supplementing LAB to the TMR did not affect the chemical composition of TMR except for the lactate and acetate concentration. Storage temperatures affected (p<0.05) the chemical composition of the TMR, including pH, lactate, and acetate contents. The chemical composition of TMR was also affected (p<0.05) by the storage period. During in vitro rumen fermentation study, the ML treatment showed lower (p<0.05) dry matter digestibility at 24 h incubation with a higher pH compared to the CON. There was no difference in the in vitro dry matter digestibility (IVDMD) of TMR between the CON and ML treatment however, at 24 h, ML treatment showed lower (p<0.05) IVDMD with a higher pH compared to the CON. The effects of storage temperature and period on IVDMD were not apparent at 24 h incubation. In an in vivo study using Holstein steers, supplementing LAB to the basal TMR for 60 d did not differ in the final body weight and average daily gain. Likewise, the fecal microbiota did not differ between CON and ML. However, the TMR used for the present study did include a commercial yeast in CON, whereas ML did not; therefore, results were, to some extent, compromised in examining the effect of LAB. In conclusion, storage temperature and period significantly affected the TMR quality, increasing acetate and lactate concentration. However, the actual effects of LAB supplementation were equivocal.

친환경 청보리, 이탈리안 라이그라스, 볏짚사일리지의 In vitro 반추위 발효성상 비교연구 (In vitro Rumen Fermentation Patterns of Environment Friendly Whole Crop Barley, Italian Ryegrass and Rice Straw Silages)

  • Rumen fermentative characteristic is useful indicators of the quality of ruminant feed stuffs and diets. An in vitro rumen fermentation experiment was therefore carried out to compare fermentation patterns among three forage sources. These were whole crop barley (WCBS), Italian ryegrass silage (IRGS) and rice straw silages (RSS). Rice straw (RS) was the control, making the treatments 4 in total. Forages were randomly allocated to serum bottles. The incubation times were arranged 0, 3, 6, 9, 12, 24, 48 and 72h at $39^{\circ}C$, respectively. Each forage source was replicated 3 times per incubation time. At each sampling time, total gas and pH were measured, whilst individual volatile fatty acids (VFAs), total volatile fatty acids (TVFAs) and ammonia nitrogen ($NH_3$-N) were determined later after storing samples at $-20^{\circ}C$. Acetate: Propionate ratio (A/P) was then calculated. Forage source had a significant effect (P<0.001) on pH and $NH_3$-N. RSS maintained higher pH values than the rest of the forage sources. A decreasing pH trend with increased time of incubation, in agreement with literature, was observed for all forage sources. WCBS recorded $NH_3$-N values higher than all the other treatments. Total gas, individual and total VFA and A/P ratio were not affected by forage source. However, there was a significant difference in all parameters (p<0.05) among forage sources at sampling periods at 3 to 72h. Therefore, the present results indicating that WCBS, IRGS, RS and RSS maintained in vitro rumen pH above the critical value. Also, WCBS produced the highest NH3-N and on this merit could be of better nutritive value, in vivo, in the ruminant.

Effects of fermented soybean meal with Bacillus velezensis, Lactobacillus spp. or their combination on broiler performance, gut antioxidant activity and microflora

  • Objective: A series of experiment were conducted to evaluate the effects of replacing a part of soybean meal (SBM) at 6% of broiler diets with fermented soybean meal (FSBM) obtained by single or two-stage fermentation by measuring growth performance, antioxidant activity in the jejunum and distal intestinal microflora. Methods: Soybean meal samples were prepared by single-stage fermentation using Bacillus velezensis (Bv) (FSBMB), or Lactobacillus spp. (as commercial control) (FSBML). Additional SBM sample was prepared by two-stage fermentation using Bv and subsequently using Lactobacillus brevis ATCC 367 (Lb) (FSBMB+L). Enzyme activity, chemical composition, trichloroethanoic acid-nitrogen solubility index (TCA-NSI) and antioxidant activity were measured. Then, in an in vivo study, 320 Ross308 broilers were divided into four groups with ad libitum supply of feed and water. Four groups were fed either a corn-soybean meal diet (SBM), or one of fermented SBM diets (FSBMB+L, FSBMB, and FSBML). Growth, serum characteristics, microflora, and the mRNA expression of selected genes were measured. Results: Compared to SBM, FSBMB+L contained lower galacto-oligosaccharide, allergic protein, and trypsin inhibitor, and higher TCA-NSI by about three times (p<0.05). Reducing power and 1,1-diphenyl-2-picrylhydrazyl free radical scavenging ability correlated positively with the TCA-NSI content in FSBM. Growth performances were not significantly different among four groups. In jejunum of 35-day-old broilers, partial replacement of SBM by FSBMB+L increased the activity of superoxide dismutase and catalase (CAT), and the FSBMB group had the highest catalase activity (p<0.05). Partial replacement of SBM by FSBM increased relative mRNA expressions of nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and peptide transporter 1 (PepT1) (p<0.05); however, FSBMB+L increased CAT mRNA level to 5 times of the control (p<0.05). Conclusion: Using Bv- and Lb-processed SBM through two-stage fermentation to partially replace 6% of diets will improve the gut's antioxidant activity under commercial breeding in broilers.
