Effects of Mito-TEMPO on the survival of vitrified bovine blastocysts in vitro

  • Vitrification methods are commonly used for mammalian reproduction through the long-term storage of blastocyst produced in vitro. However, the survival and quality of embryos following vitrification are significantly low compared with blastocyst from in vitro production (IVP). This study evaluates that the survival of frozen-thawed bovine embryos was relevant to mitochondrial superoxide derived mitochondrial activity. Here we present supplementation of the cryopreservation medium with Mito-TEMPO (0.1 µM) induced a significant (p < 0.001; non-treated group: 56.8 ± 8.7%, reexpanded at 24 h vs Mito-TEMPO treated group: 77.5 ± 8.9%, re-expanded at 24 h) improvement in survival rate of cryopreserved-thawed bovine blastocyst. To confirm the quality of vitrified blastocyst after thawing, DNA fragmentation of survived embryos was examined by TUNEL assay. As a result, TUNEL positive cells rates of frozen-thawed embryos were lower in the Mito-TEMPO treated group (4.2 ± 1.4%) than the non-treated group (7.1 ± 3.5%). In addition, we investigated the intracellular ROS and mitochondrial specific superoxide production using DCF-DA and Mito-SOX staining in survived bovine embryos following vitrification depending on Mito-TEMPO treatment. As expected, intracellular ROS levels and superoxide production of vitrified blastocysts after cryopreservation were significantly reduced (p < 0.05) according to Mito-TEMPO supplement in freezing medium. Also, mitochondrial activity measured by MitoTracker Orange staining increased in the frozen-thawed embryos with Mito-TEMPO compared with non-treated group. These results indicate that the treatment of Mito-TEMPO during cryopreservation might induce reduction in DNA fragmentation and apoptosis-related ROS production, consequently increasing mitochondrial activation for developmental capacity of frozen-thawed embryos.

한우에 있어서 초음파기기를 이용한 생체내 개체별 난자 채취 빈도 및 수정란 생산효율에 관한 연구 (Effect of Ovum Pick-up Frequency on In Vitro Production of Embryos in Hanwoo Cattle)

  • The ultrasound-guided oocytes cllection (ovum pick-up ; OPU) has become a substitution for superovlation in cattle. The objective of this study was to examine the effect of OPU frequency on the in vitro production of embryos in Hanwoo cattle. Six cycling Hanwoo cows were distributed into two groups for either once or twice weekly OPU sessions. Oocytes were collected by ultrasound-guided follicle aspiration(SA600) using a 6.5HMz transducer and attached with 18 gauge needle, with vacuum pressure of 40 mmHg. The cumulus-oocyte complexes (COCs) collected from each donor were matured in TCM 199 supplemented with 10% fetal bovine serum at 5% CO2 in air at 38.5$^{\circ}C$ for 22h and in vitro matured oocytes were co-incubated with sperm(separated by Percoll gradient) for 6h. The zygotes were co-cultured on cumulus cell monolayer in 10ul droplets in the same culture medium and conditions used for IVM for 7 days. On Day 7 of culture, development to blastocysts was examined. Although the number of oocytes collected was variable depending on individuals, overall embryo production in the twice per week OPU sessions was better that in the once per week sessions(6~21 vs 2~7 blastocysts produced, respectively). Two cows(E, A) were good oocyte donors and embryo production was superior in cow C ; however, cow F was a poor donor as compared to the others. In conclusion, these results suggest that for embryo production, twice weekly OPU sessions were better than once per week for producing embryos in vitro from Hanwoo cattle.

Effect of a short-term in vitro exposure time on the production of in vitro produced piglets

  • Although piglets have been delivered by embryo transfer (ET) with in vitro produced (IVP) embryos and blastocysts, a success rate has still remained lower level. Unlike mouse, human, and bovine, it is difficult to a production of piglets by in vitro fertilization (IVF) because of an inappropriate in vitro culture (IVC) system in pig. Therefore, the present study was conducted to investigate whether minimized exposure time in IVC can improve the pregnancy and delivery rates of piglets. Immediately after IVM, the oocytes were denuded and co-incubated with freshly ejaculated boar semen for 3.5 to 4 hours at $38.5^{\circ}C$ under 5% $CO_2$ in air. To avoid long-term exposure to in vitro state, we emitted IVC step after IVF. After that the presumptive zygotes were transferred into both oviducts of the surrogate on the same day or 1 day after the onset of estrus. Pregnancy was diagnosed on day 28 after ET and then was checked regularly every month by ultrasound examination. The 3 out of 4 surrogates were determined as pregnant (75%) and a total of 5 piglets (2 females and 3 males) were delivered at $118.3{\pm}2.5$ days of pregnancy period. In conclusion, a short-term exposure time may be an important factor in the production of IVP-derived piglets. It can be apply to the in vitro production system of transgenic pig by IVF, cloning, and pronuclear microinjection methods.

체외성숙, 수정 및 배양에 있어서 각기 다른 배양조건들이 소 체외수정란의 생산에 미치는 효과 (Effects of In Vitro Maturation, In Vitro Fertilization and In Vitro Culture Conditions on Bovine Embryo Production)

  • This study was conducted to establish the optimal conditions for in vitro embryo production using oocytes derived from follicles of slaughter-house ovaries. The ovaries of Hanwoo were obtained from a local slaughter-house. The oocytes were aspirated from visible follicles of 2~7mm in diameter. The recovered oocytes which were completely surrounded by at least 2 layers of cumulus cells and a homogeneous cytoplasmic pigmentation were used. The selected oocytes were washed 3 or 4 times with D-PBS containing 10% bovine calf serum (BCS) and matured in vitor (IVM) in Ham's F-10 supplemented with 10% BCS or 0.01 $\mu\textrm{g}$/ml epidermal growth factor(EGF) at 39$^{\circ}C$ under 5% CO2 in air for 24 hours. They were fertilizqed in vitro (IVF) with fresh sperm separated by Percoll density gradient or swim-up in TALP media. The zygotes were cultrued with or without bovine oviductal epitherial cells(BOEC) in media(HECM-6 supplemented with 11 amino acid and / or TCM-199 supplemented with 10% BCS) for 7 to 10 days. The results obtained were as follow: The cleavage rate and developmental rate to blastocyst after maturation and IVF were not significantly different between Ham's F-10 with EGF(76.0% vs. 44.0%) and BCS(75.9% vs. 43.6%)(P<0.05). The cleavage rate and development rate to blastocyst after fertilizing by swim-up or Percoll method were not signifciantly(P<0.05) different between swim-up (80.2% vs. 29.2%) and Percoll(81.9% vs. 26.5%) (P<0.05). The cleavage rate in TCM 199(80.5) was signficiantly higher than that in HECM-6 (72.0%) (P<0.05). However, developmental rate to blastocyst using TCM 199 following HECM-6 for 3 or 4 days (42.2%) was significantly higher than that in TCM-199 alone(26.7%)(P<0.05). The cleavage rate and development rate of embryos produced in vitro by exchange timing for HECM-6 media were not significantly different between in day 3(78.6% vs. 45.5%) and day 4(75.0% vs. 43.2%)(P<0.05). The cleavage rate and developmental rate to blastocysts according to co-culture system were not significantly different between with (74.2% vs. 41.4%) and without BOEC(73.95 vs. 43.5%) (P<0.05). The number of blastomere in blastocyst stage after co-culture with or without BOEC was not significantly different (106.7$\pm$5.1 and 96.6$\pm$4.0). In conclusion, the most transferable IVP embryos could be produced from Ham's F-10 medium for IVM, Percoll density gradient method for IVF sperm separation and in vitro culture in HECM-6 until day 3 or day 4, and then transferred into TCM-199 until day 9 within adequate embryo density in culture droplets after insemination.

Effect of Serum and Serum Free Media on the Developmental Competence of OPU Derived Bovine IVP Embryo

  • Embryos produced with serum show the alterations in their ultrastructure, impaired compaction, abnormal blastulation, aberrant mRNA expression profiles and large calf syndrome with greater incidences of stillbirths and deaths after birth. The aim of the present study was to describe in vitro embryo production by analyzing embryo production, fetal production and pregnancy rate in free-serum medium. The OPU-IVP data used in this study from 2016. Approximately, sixteen cows (Hanwoo), which belonged to the Institute of Gyeongsang National University, were used. Two experimental group is used in this study. Serum groups were conducted in March to July and free-serum group was conducted in September to December. The recovered cumulus-oocyte complexes were morphologically classified to four grades based on the compaction of cumulus cells layers and homogeneity of the cytoplasm. The number of oocyte was significantly greater in serum groups than that in free-serum groups (29.61 ± 0.63 vs. 15.6 ± 0.62; p < 0.05). Between serum and free-serum groups indicate that average of 1st and 2nd grade oocytes were no difference (2.38 ± 1.67 vs. 2.38 ± 1.48; p > 0.05), but number of 3rd and 4th grade oocytes were greater in serum groups than that in free-serum groups (7.31 ± 7.64 vs. 5.60 ± 6.29; p < 0.05). Embryo cleaved competence was higher in rate in free-serum groups than that in serum groups (62.1% vs. 58.3; p < 0.05). However, blastocyst developmental rate was no difference between serum and free-serum groups (33.1% vs. 43.5%; p < 0.05). 986 recipients were used for embryo transfer. Pregnancy rate was indicated that between serum and free-serum group was no difference (54.6% vs. 56.3%; p < 0.05). In conclusion, we developed the free-serum system for production of in vitro bovine embryos in order to meet the developmental and qualitative requirements for large scale commercial use.

OPU 유래 한우 수정란 생산 및 이식 (Transplantation and Production of OPU Derived Hanwoo IVP Embryos)

  • This study was carried out to establish the system of OPU derived embryo production, management of recipients as well as offspring production. OPU derived embryo production system was carried out of aspiration of immature oocytes 2 times per week, total 24 times for 3 months by an ultrasonographic guided follicular aspiration system and then produced in vitro-produced blastocysts by in vitro maturation, fertilization and culture system. This work was collected total 13,866 oocytes, average $8.2{\pm}4.5$ oocytes per session and 8,170 G1 + G2 grade oocytes, average 4.8 oocytes per session by 1,692 times session of total 71 donors for 4 years from 2010 to 2013. The rate of cleavage and blastocyst developmental competence were obtained 11,825 (85.3%) and 5,032 (36.3%) that was $7.0{\pm}3.8$ cleaved embryos and $3.0{\pm}2.5$ blastocysts per session. OPU derived embryo transfer were taken place in 2, 4, 6 and 7 local governments at 2010, 2011, 2012 and 2013 for 4 years and pregnancy rate were obtained 41.2, 43.9, 46.5 and 49.7% in each years. It means that pregnancy rate was continuously improved according of every year for 4 years. Pregnancy rate was significantly different according to individual local government in which was 62.7% in B, but 24.2% in F at 2012. Paternity identification was carried out total 26 offspring in C local government of 2012 and then confirmed 100% agreement of its analysis. In conclusion, the results obtained the possibility of mass production of elite cow embryos as well as offspring by OPU derived embryo production system, of which could be decreased the required time of genetic improvement.

수정란의 밀도가 소 체외수정란의 체외발달에 미치는 효과 (Effects of Embryo Density on Development of In Vitro Produced Bovine Embryos)

  • 수정란이식에 필요한 다량의 수정란을 생산하는 수단인 배양액과 적정 수정란의 수는 수정란의 체외발달에 많은 영향을 미치므로 이들 관계를 조사하여 수정란의 체외배양체계를 확립하고자 본 연구를 실시하였다. 도축장에서 채취한 난소에서 미성숙 난자를 채란하여 10% FBS가 첨가된 TCM -199 에 LH(10 $\mu\textrm{g}$/$m\ell$), FSH(35 $\mu\textrm{g}$/$m\ell$), estradiol-17 $\beta$(1 $\mu\textrm{g}$/$m\ell$)가 첨가된 체외성숙배양액에서 24시간 동안 배양 후, 동결정액은 Percoll-density gradients(45 vs. 90%)을 이용하여 700 g 에서 30분 동안 처리한 다음, 체외수정배양액 (IVF-Fert)에서 체외수정을 유도하였다. 수정이 확인된 수정란은 50 ${\mu}\ell$ 배양액에 1, 25 또는 50개의 수정란을 난구세포와 공배양을 하여 9일 동안 배양하였다. 일정한 배양액에서 수정란의 수가 체외수정란의 발달에 미치는 요인을 분석하고, 개별배양 또는 그룹배양 시 난구세포와의 공동배양 효과를 조사한 결과는 다음과 같다. 1. 50 ${\mu}\ell$ 배양액에 1개, 25 또는 50개의 수정란을 수정 후 9일 동안 배양한 결과, 25와 50개의 수정을 배양했을 경우에는 발달율이 36.5 와 26.5%를 보여 1개의 수정란을 배양했을 경우에서의 발달율 6.2% 보다 유의적으로 높은 발달율을 얻었다(P<0.05). 2. 1, 25, 50개의 수정란을 배양시 수정 후 6일째 발달율은 1.0~3.5%였으나, 수정 후 7, 8, 9 일째의 발달율은 1개의 수정란을 배양하는 것보다 25, 50개의 수정란을 배양한 처리구에서 유의적으로 높은 발달율을 얻었다 (P<0.05). 3. 일정한 배양액에서 1, 25 및 50개의 수정란을 배양 시 수정 후 8일째의 배반포 수정란의 세포수를 조사한 결과, 1개의 수정란을 배양시 배반포 수정란의 수는 93.0개였으나, 25, 50개의 수정란을 배양시는 각각 112개의 세포수를 얻어 유의적으로 높은 세포수를 나타내었다 (p<0.01). 4. 일정한 배양액에 1개 또는 25개의 수정란을 난구세포와 공배양을 하거나, 하지 않았을 때의 발달율은 각각 15.0와 3.7% 또는 34.5와 9.0%로서 난구세포와 공배양을 하는 것이 유의적으로 높은 발달율을 나타내었다 (p<0.01 수정 후 8 일째의 배반포 수정란의 세포수도 각각 96.1와 82.0개 또는 116.4 와 96.5개로서 난구세포와 공배양을 한 처리구에서 유의적으로 높은 세포수를 나타내었다 (p<0.01). 이상의 결과를 요약하면, 수정란의 수가 체외수정란의 체외발달에 중요한 영향을 미친다는 것을 알 수 있었으며, 다량의 체외수정란을 생산하여 수정란 이식에 이용하기 위해서는 체외성숙 / 수정된 체외수정란을 일정한 배양액 (50${\mu}\ell$) 에 25, 50개의 수정란을 난구세포와 공배양하는 것이 높은 배 발달율과 세포 수를 얻 을 수 있을 것으로 사료된다.

한우 체외 수정란의 배반포 단계, 체외 배양 일령 및 등급이 산자의 생산에 미치는 영향 (Effects of the In Vitro Produced Hanwoo Blastocyst Stage, In Vitro Culture Day and Blastocyst Grade on the Production of Claves)

  • 본 연구는 수정란이식의 현장 적용 측면을 중심으로, 한우 체외 수정란의 발생 단계, 일자 및 등급이 젖소 대리모에 이식 후 임신과 유산, 분만과 관련된 임신기간, 생시체중, 난산 및 쌍자율을 조사하였다. 또한 단자 및 쌍자 분만에 따른 임신기간, 생시체중, 성비 및 폐사율을 분석하였다. 발생 단계에 따른 임신율은 배반포 단계(64.4%), 유산율은 부화 배반포(HB) 단계(21.4%)가 유의하게 높았다(p<0.05). 발생 일령에 따른 임신율은 7일령 배가 49.0%로서 8일 및 9일령의 36.4 및 15.4%보다 유의하게 높았으나(p<0.05), 유산율은 0$\sim$10.7%로 비슷한 경향이었다. 등급에 따른 임신율은 41.4$\sim$42.5%, 유산율은 9.3$\sim$16.5%로서 차이가 없었다. 발생 단계에 따른 임신기간, 송아지의 체중 및 난산율은 비슷한 경향이었으나, 쌍자율은 HB 단계가 44%로서 배반포 단계의 15.8%보다 유의하게 높았다(p<0.05). 발생 일령에 따른 임신 기간, 난산율 및 쌍자율은 차이가 없었으나, 송아지의 체중은 9일령의 것이 평균 35.5 kg으로서 7일령 및 8일령의 평균 25.2 및 26.2kg보다 유의하게 무거웠다(p<0.05). 등급에 따른 임신 기간은 1등 및 2등급이 평균 278.5 및 276.1일로서 3등급의 평균 273.7일에 비하여 유의하게 길었으나(p<0.05), 체중, 난산율 및 쌍자율은 차이가 없었다. 분만한 송아지의 개체 수에 따른 임신기간은 단자가 평균 278.5일로서 쌍자의 평균 272.5일보다 유의하게 길었다(p<0.05). 생시 체중도 단자가 평균 29.6 kg으로서 쌍자의 평균 22.3kg에 비하여 유의하게 무거웠으나(p<0.05), 성비와 폐사율은 유사한 경향이었다.

체외생산된 소 배반포강 내의 아미노산 농도 (Amino Acid Concentrations in the Blastocoelic Fluid of In Vitro-Produced Bovine Blastocysts)

  • 체외생산된 소 배반포 및 탈출 배반포강 내의 유리 아미노산 농도를 측정하였다. 체외 배양은 소 혈청알부민이 함유된 합성난관배양액(synthetic oviduct fluid; SOF)에서 실시하였으며 수정 후 180시간의 배반포 및 216시간 후의 탈출 배반포를 실험에 공여하였다 아미노산 측정은 알부민 대신 Polyvinyl alcohol이 함유된 SOF 미소적에 수정란을 분주하고 미세조작기를 이용하여 배반포강 내의 액을 추출하여 20종류의 아미노산을 측정하였다. 탈출 배반포는 isoleucine, leucine 및 methionine 농도가 배반포보다 유의적(p〈0.05)으로 높게 나타났으며, glutamate, aspartate는 두 군간에 차이를 나타내지 알았다. 반면 alanine 및 threamine (p〈0.01)과 cystine을 제외한 나머지 12종류의 아미노산(p〈0.001)은 배반포가 탈출 배반포에 비해서 유의적으로 높은 측정치를 나타내었다. 비록 glutamine의 경우 배양액 내에 첨가되지 않았으나 두 군, 특히 배반포 내에서 높은 측정치를 보였다. 본 연구결과로 보아 체외생산된 배반포 및 탈출 배반포는 내강에 필수 및 비필수 아미노산을 각기 다른 농도로 함유하고 있는 것으로 사료된다.
