In order to establish micropropagation system Anthurium andreanum 'Atlanta', dwarf type, shoots of A. andreanum were cultured on medium supplemented with cytokinin. Callus was formed from the base of shoots. high frequency callus induction was obtained on medium with 10.0mg/L BA or 10.0mg/L TDZ(thidiazuron) at more than 71.8%. The shoots were cultured on media with various combinations and concentrations of TDZ, BA and 2.4-D to enhance callus induction. Callus was induced at more than 72.6% and grew vigorously on media containing 10.0mg/L BA and 0.0∼0.5mg/L 2.4-D, or 1.0mg/L TDZ. Stimulation effects of cytokinin by 2.4-D did not occur in combined treatments of cytokinin and 2.4-D. Callus was cut into sections(7${\times}$10mm), and then cultured on media with BA alone or BA and 2.4-D to regenerate shoots and to stimulate the callus growth. Shoot regeneration and callus growth were effective on media with 10.0mg/L BA alone, or 10.0mg/L BA and 0.1mg/L 2.4-D. In combined treatments of BA and 2.4-D, stmulation effects of cytocinin by 2.4-D also did not occur. Callus growth was decreased, accordiong to increasing the concentration of 2.4-D. In cimbined treatments of TDZ and 2.4-D in shoot regeneration and callus proliferation, stimulated effects of cytokinin by 2.4-D did not occur entirely. Media with 0.5∼1.0mg/L TDZ ingibited the regeneration and rooting of shoots, and callus growth from callus sections. Addition of 2.4-D on medium with TDZ ingibited the regeneration and rooting of shoots, and callus growth. Rooted plantdts were acclimatized in greenhouse. The plantlets were survived more than 98% in soil of vermiculite alone or mixed perlite 1 and vermiculite 1.
Stem cell research arose from the need to explore new therapeutic possibilities for intractable and lethal diseases. Although musculoskeletal disorders are basically nonlethal, their high prevalence and the relative ease of performing clinical trials have facilitated the clinical application of stem cells in this field. On the other hand, despite the plethora of in vitro and preclinical studies in stem cell research for regenerative medicine in the musculoskeletal system, few reliable clinical studies have been published. Stem cell therapy can be applied locally for bone, cartilage, and tendon regeneration. The candidate disease modalities in bone regeneration include large bone defects, nonunion of fractures, and osteonecrosis. Focal osteochondral defect and osteoarthritis are the current targets for cartilage regeneration. For tendon regeneration, bone-tendon junction problems, such as rotator cuff tears are hot topics in clinical research. To date, the literature supporting stem cell-based therapies comprises mostly case reports or case series.
The beneficial paracrine roles of mesenchymal stem cells (MSCs) in tissue repair have potential in therapeutic strategies against various diseases. However, the key therapeutic factors secreted from MSCs and their exact molecular mechanisms of action remain unclear. In this study, the cell-free secretome of umbilical cord-derived MSCs showed significant anti-fibrotic activity in the mouse models of liver fibrosis. The involved action mechanism was the regulation of hepatic stellate cell activation by direct inhibition of the $TGF{\beta}$/Smad-signaling. Antagonizing the milk fat globule-EGF factor 8 (MFGE8) activity blocked the anti-fibrotic effects of the MSC secretome in vitro and in vivo. Moreover, MFGE8 was secreted by MSCs from the umbilical cord as well as other tissues, including teeth and bone marrow. Administration of recombinant MFGE8 protein alone had a significant anti-fibrotic effect in two different models of liver fibrosis. Additionally, MFGE8 downregulated $TGF{\beta}$ type I receptor expression by binding to ${\alpha}v{\beta}3$ integrin on HSCs. These findings revealed the potential role of MFGE8 in modulating $TGF{\beta}$-signaling. Thus, MFGE8 could serve as a novel therapeutic agent for liver fibrosis.
Objectives: Previous reports have shown that Bogijetong-Tang (BJT) is effective in peripheral neuropathy induced by taxol and crush injury. In this study, we researched the effects of BJT on diabetic neuropathy induced by STZ in the mouse. Methods: We performed both in vitro and in vivo experiments to verify the effects of BJT on diabetic neuropathy induced by STZ in mice. Changes in axonal recovery were observed with immunofluorescence staining using NF-200, Hoechst33258, $S100{\beta}$, caspase 3 and anti-cdc2. Proliferation and degeneration of Schwann cells were investigated by immunofluorescence staining and western blot analyses. Results: BJT showed considerable effects on neurite outgrowth and axonal regeneration in diabetic neuropathy. BJT contributed to the creation of NF-200, GAP-43, Cdc2, phospho-vimentin, ${\beta}1$, active ${\beta}1$, ${\beta}3$ integrin, phospho-Erk1/2 protein. Conclusions: Through this study, we found that BJT is effective for enhanced axonal regeneration via dynamic regulation of regeneration-associated proteins. Therefore, BJT had a pharmaceutical property enhancing recovery of peripheral nerves induced by diabetic neuropathy and could be a candidate for drug development after more research.

Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. O. japonicus was cultured in media containing various growth regulators and carbon sources. The highest regeneration rate was achieved in 1.0 and $3.0\;mg\;l\;^{-1}$ of 2,4-D concentration, while the lowest was obtained in $10.0\;mg\;l\;^{-1}$ 2,4-D concentration. When different carbone sources were added in the culture medium, plant growth was high in 3% sucrose treatment. The micropropagated shoots were successfully acclimatized in artificial soils and produced comparable amont of polysaccharide compred to parent cultivated plants.
Hibiscus syriacus L., the national flower of Korea, is a commonly grown ornamental species found in parks, gardens and along roadsides in Korea. This study sought to establish a plant regeneration system of H. syriacus 'Nanpa' using mature leaves as an explant. Sterilized mature leaf explants were cultured on modified MS medium with combinations of NAA and BAP at various concentrations for 6 weeks. Among the various media evaluated, modified MS media containing 0.46 mg/L NAA and 0.5 mg/L BAP (CI-1) or 0.92 mg/L NAA and 1 mg/L BAP (CI-4) were the most effective for callus formation. Whitish-yellow calluses were observed on CI-1 medium, while green calluses formed on CI-4 medium. The whitish-yellow callus derived from CI-1 medium showed a higher frequency of shoot induction than the green callus derived from CI-4 on modified MS medium containing 0.5 mg/L BAP. Among the various media evaluated in this study, modified MS medium containing 0.46 mg/L NAA and 0.5 mg/L BAP was the most optimal for shoot-forming callus production. Our findings show that mature leaves of H. syriacus 'Nanpa' can be used for in vitro plant regeneration, and the regeneration system described here may be a powerful tool for molecular breeding of H. syriacus 'Nanpa' in the future.
An efficient procedure was developed for adventitious shoot bud induction and plantlet regeneration from various explants of the ten genotypes of Pepper (Capsicum annuum L.) using Thidiazuron (TDZ). Among various treatments at 1.0-3.0 mg/L TDZ Induced maximum number of adventitious shoots depending upon the explant type and genotype compared to other treatments. Among the explants tested, leaf induced maximum number of adventitious shoots than the cotyledons. TDZ-mediated organo-genesis was possible in 10 pepper cultivars, the extent of the response being genotype-dependent. Of the ten genotypes tested, C. annuum cvs CA960, $G_4$ and X-235 were produced maximum number of adventitious shoots and Sell was the least, and all other genotypes gave moderate response. Elongation of multiple shoots was observed on medium supplemented with SA (0.05 mg/L) in combination of IAA (0.05 mg/L). Differences in ability for in vitro shoot regeneration and elongation depend upon the variety and explant type. The elongated shoots were success. Fully rooted on MS medium containing at 1.0 mG/L IAA. Plantlets regenerated from different explants of ten genotypes were found to be diploid (2n=24) and were devoid of any chromosomal aberrations. Regenerated plants were successfully established in soil where 85-90% of them developed into morphologically normal and fertile plants.
This study was carried out to address an efficient in vitro regeneration system from seed-derived callus of Phragmites communis, and to evaluate genetic variations of the regenerants using ISSR markers. Shoot regeneration via calli was greatly influenced by N6 medium compared with MS medium, and plant regeneration frequency was 90% in N6 supplemented with BA 0.25 mg/L and BA 0.5 mg/L. According to ISSR analysis of the thirty regenerants, out of 94 loci detected overall, 16 were identified to be polymorphic with a rate (PR) of 17.0%. The mean gene diversity (h) of different in vitro condition was 0.03 and ranged from 0.008 for N6 with BA 5 mg/L, to 0.040 for MS with IAA 0.1 mg/L+kinetin 2 mg/L. The results indicate that the regenerants have a low genetic variation, and ISSR analysis is effective to detect genetic variation of regenerants.

Pea (Pisum sativum) is one of important legume crops in the world. It is commonly used as a protein source for animal and human diet, and also used as a natural nitrogen source which is produced by a symbiotic bacterium in their root nodule and helpful for terrestrial ecosystem. The successful in vitro manipulation is depended on three main factors including physiology of plant donor, in vitro manipulation approach, and stress physiology during plant cultivation. Moreover, genotype is an important for plant manipulation; different genotype gives the different response to regeneration efficiency. An efficient condition of shoot induction for pea (Pisum sativum cv. 'Sparkle') was developed by using optimum explant, plant growth regulator concentrations, and pretreatment of BA onto explant. The average shoot number per explant showed the highest on two kinds of shoot induction media (MSB5 media containing 2 mg/L BA and a combination of 2 mg/L BA and 1 mg/L TDZ) with cotyledonary node explants culture. Moreover, the pretreatment of explant in 200 mg/L BA solution was found to be more effective in shoot induction than that of non-pretreatment. The analysis of genetic stability of regenerants by using 13 ISSR markers presented that in vitro regenerated plants showed polymorphism with 8.3% compared with their mother plants.

Callus and shoot formation of leaf explants from in vitro propagated shoots and field grown plants depending on the position of leaf, and four boxthron cultivars were investigated. Callus formation of explants from both in vitro shoot and field grown plants as easily achieved at the cut surfaces of explants but the callus formed from leaf of in vitro shoots was hardened as the duration of culture was proceed. Calli were effectively induced from leaves detached from the middle position of both in vitro and in vitro plants on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA, and the growth of calli were better in field grown leaves than in vitro grown leaves. Shoot formation were effectively induced from leaves detached from the upper position in vitro plants, and the middle parts of in vitro plants on MS medium containing 0.01 mg/L (NAA with 0.2 mg/ BA. There was difference on the frequency of shoot formation among four different cultivars; 'Jindojaerae' was the best for shoot formation followed by 'Cheonyang', 'Younghagukija' and 'Cheongyangjaerae'.
