New winter wheat winter barley hybrids were produced (Mv9 kr1 Igri, Mv9 kr1 Osnova, Asakazekomugi Manas). The wheat-barley hybrids showed entire male sterility and were multiplied in tissue culture. Chromosome configurations were studied with GISH in meiosis in the Mv9 krl x Igri hybrid and in its progenies multiplied in vitro. Chromosome pairing between wheat and barley has been observed in some cells in the hybrids multiplied in vitro. Backcross plants with 43 and 44 chromosomes were selected with the aim of developing new wheat-barley addition lines. Wheat-barley translocations were demonstrated with GISH in backcross progenies originating from in vitro regenerated wheat (Triticum aestivum L. cv. Chinese Spring) x barley (Hordeum vulgare L. cv. Betzes) hybrids. Five different translocations were observed. Sequential N-banding and GISH analyses were performed to further identify the translocations. The N-banding pattern of the Robertsonian translocation suggests that this chromosome consists of the short arm of barley chromosome 4H translocated to the long arm of 2B of wheat. Plants with four different homozygous translocations were selected from the following BC2F3 generation.
Plants regenerated from in vitro cultures carry chromosomal variations, especially in long-term culture. Reducing the duration of plant tissue culture is one of the ways to reduce genetic and epigenetic changes. In this study, we reduced the duration of long-term culture and repeat subculture using small bulblets derived from bulb scales in two lily cultivars. The adventitious bulblets derived from bulb-scale tissue were cultured on three different media containing Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. About seven weeks later, the number of newly propagated multiple shoots in the two media, A and B media, showed little differentiation. Compared to both media, the number of propagated multiple shoots increased 5-fold in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal (C medium). The number of propagated multiple shoots ranged from 5 to 6 and 4 to 6 with an average of 5 in TropicalPink and GreenStar cultivars, respectively. The flow cytometric measurements indicated no variation in the ploidy level between control and in vitro propagated plants.
The in vitro regeneration was established, and the genetic stability among the mother plants (control) and the micropropagated green plants was evaluated using ISSR markers in Imperata cylindrica 'Rubra', Poaceae which containing important bioenergy plants. Green shoots were multiply induced from growing point culture via callus on MS medium supplemented with 0.01 mg/L NAA and 2 mg/L BA, and the shoots were proliferated on the MS medium with rooting. Rooted plantlets were transplanted to the pot with 100% survival rate. Using ISSR markers, somaclonal variation was analyzed in eight mother plants (control), ten green-regenerant cultivated at culture room (ReR) and ten green-regenerant cultivated at field condition (ReF). All ISSRs produced a total of 97 bands, and the scorable bands varied from one to seven with an average of 4.4 bands per primer. The polymorphism rate of ReRs and ReFs was 4.1% and 3.1% respectively, showing higher rate than that of control (0%). The genetic similarity matrix (GSM) among all accessions ranged from 0.919 to 1.0 with a mean of 0.972. According to the clustering analysis, ReFs and mother plants were divided into two independent groups. The results indicate that no clear genetic diversity was detected among regenerated plants, and ISSR markers were useful tool for identification of somaclonal variation of regenerants.

During the past two decades, China has seen her great progress in plant biotechnology. Since the Chinese market of herb medicine is huge, while the plant resources are shrinking, particular emphasis has been placed in plant tissue and cell cultures of medicinal plants, this includes fast propagation, protoplast isolation and regeneration, cell suspension cultures and large scale fermentation. To optimize culture conditions for producing secondary compounds in vitro, various media, additives and elicitors have been tested. Successful examples of large scale culture for the secondary metabolite biosynthesis are quite limited : Lithospermum ery throrhizon and Arnebia euchroma for shikonin derivatives, Panax ginseng, P. notoginseng, P. quinquefolium for saponins, and a few other medicinal plants. Recent development of genetic transformation systems of plant cells offered a new approach to in vitro production of secondary compounds. Hairy root induction and cultures, by using Ri-plasmid, have been reported from a number of medicinal plant species, such as Artemisia annua that produces little artemisinin in normal cultured cells, and from Glycyrrhiza uralensis. In the coming five years, Chinese scientists will continue their work on large scale cell cultures of a few of selected plant species, including Taxus spp. and A. annua, for the production of secondary metabolites with medicinal interests, one or two groups of scientists will be engaged in molecular cloning of the key enzymes in plant secondary metabolism.
Endophytic bacterial communities of tidal flat plants antagonistic to oomycete plant pathogens were studied by the isolation of 256 root colonizing endophytic bacteria from surface-disinfected root tissues of six plants ($Rosa$$rugosa$, $Suaeda$$maritima$, $Vitex$$rotundifolia$, $Carex$$scabrifolia$, $Glehnia$$littoralis$ and $Elymus$$mollis$) growing in a tidal flat area of Namhae Island, Korea. To understand the antagonistic potential, an $in$$vitro$ antagonistic assay was performed to characterize and identify strains that were antagonistic to the oomycete plant pathogens $Phytophthora$$capsici$ and $Pythium$$ultimum$ from the total population. Nine percent of the total number of isolated bacteria exhibited in vitro inhibitory activity against target plant pathogenic oomycetes. Taxonomic and phylogenetic placement of the antagonistic bacteria was investigated by analysis of the 16S rRNA gene sequences. The sequence analysis classified the antagonistic strains into four major classes of the domain bacteria ($Firmicutes$, ${\alpha}-Proteobacteria$, ${\gamma}-Proteobacteria$ and $Actinomycetes$) and 10 different genera. Further production of secondary metabolites, hydrolytic enzymes and plant growth promoting traits were determined for the putative new species of antagonistic endophytic bacteria. These new strains could not be identified as known species of ${\alpha}-Proteobacteria$, and so may represent novel bacterial taxa. The unexpected high antagonistic bacterial diversity associated with the tidal flat plants may be indicative of their importance in tidal flat plants as a promising source of novel antimicrobial compounds and biocontrol agents.
A study was conducted to evaluate the nutritive potential value of different aquatic plants: duckweed (Lemna trisulaca), duckweed (Lemna perpusila), azolla (Azolla pinnata) and water-hyacinth (Eichhornia crassipes) from Bangladesh. A wide variability in protein, mineral composition, gas production, microbial protein synthesis, rumen degradable nitrogen and in situ dry matter and crude protein degradability were recorded among species. Crude protein content ranged from 139 to 330 g/kg dry matter (DM). All species were relatively high in Ca, P, Na, content and very rich in K, Fe, Mg, Mn, Cu and Zn concentration. The rate of gas production was highest in azolla and lowest in water-hyacinth. A similar trend was observed with in situ DM degradability. Crude protein degradability was highest in duckweed. Microbial protein formation at 24 h incubation ranged from 38.6-47.2 mg and in vitro rumen degradable nitrogen between 31.5 and 48.4%. Based on the present findings it is concluded that aquatic species have potential as supplementary diet to livestock.
Fusarium wilt on strawberry plants caused by Fusarium oxysporum f. sp. fragariae (Fof) is a major disease in Korea. The prevalence of this disease is increasing, especially in hydroponic cultivation in strawberry field. This study assessed the effect of nutrition solution pH and electrical conductivity (EC) on Fusarium wilt in vitro and in field trials. pH levels of 5.0, 5.5, 6.0, 6.5, 7.0, and 7.5 were assayed in vitro and in field trials. EC levels at 0, 0.5, 0.8, 1.0, and $1.5dS{\cdot}m^{-1}$ were assayed in field trials. Mycelial growth of Fof increased with increasing pH and was highest at $25^{\circ}C$ pH 7 and lowest at $20^{\circ}C$, pH 5.0 in vitro. The incidence of Fusarium wilt was lowest in the pH 6.5 treatment and highest in the pH 5 treatment in field trials. At higher pH levels, the EC decreased in the drain solution and the potassium content of strawberry leaves increased. In the EC assay, the severity of Fusarium wilt and nitrogen content of leaves increased as the EC increased. These results indicate that Fusarium wilt is related to pH and EC in hydroponic culture of strawberry plants.

The objective of this study was to determine inhibitory activities of four volatile plant essential oils (cinnamon oil, fennel oil, origanum oil and thyme oil) on in vitro growth of Fusarium oxysporum f. sp. fragariae causing Fusarium wilt of strawberry plants. Results showed that these essential oils inhibited in vitro conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in a dose-dependent manner. Cinnamon oil was found to be most effective one in suppressing conidial germination while fennel oil, origanum oil and thyme oil showed moderate inhibition of conidial germination at similar levels. Cinnamon oil, origanum oil and thyme oil showed moderate antifungal activities against mycelial growth at similar levels while fennel oil had relatively lower antifungal activity against mycelial growth. Antifungal effects of these four plant essential oils in different combinations on in vitro fungal growth were also evaluated. These essential oils demonstrated synergistic antifungal activities against conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in vitro. Simultaneous application of origanum oil and thyme oil enhanced their antimicrobial activities against conidial germination and fungal mycelial growth. These results underpin that volatile plant essential oils could be used in eco-friendly integrated disease management of Fusarium wilt in strawberry fields.
An efficient shoot regeneration condition for pea cv. 'Sparkle' was developed by using optimum explant, plant growth regulator concentrations, and pretreatment of BA onto explant. The average shoot number per explant showed the highest on two kinds of shoot induction media (MSB5 media containing 2 mg/L BA and a combination of 2 mg/L BA and 1 mg/L TDZ) when cotyledonary node explants were cultured. Moreover, the pretreatment of explant in 200 mg/L BA solution was found to be more effective in shoot induction than that of non-pretreatment. By histological study, cell division and proto-meristem were formed near the surface of the sub-epidermal and epidermal cell layers of cotyledonary node in earlier than 3 days after culture. The analysis of genetic stability of regenerants by using thirteen ISSR markers showed that in vitro regenerated plants showed polymorphism with 8.3% compared with their mother plants.
Young haploid leaf derived from the anthers of tobacco plant was cultuerd and plantlets of various ploidies were obtained. When the leaf was put on the medium supplemented with kinetin as growth regulator, plantlets developed directly from the leaf, and the plants coming out in early stage of culture were all haploid. Plants developing in later stage were mostly haploids with some exception of diploid and aneuploid. Leaves were also cultured on the callus-inducing media supplemented with 2,4-D and kinetiion, and the calluses were sub-cultured for six months. Plants developed from these calluses were mostly aneuploids of various chromosome numbers. In view of the fact that the plants directly developed from the leaf were all haploid, the tissue of the original leaf explant was assumed to be uniform as far as chromosome number was concerned. On the other hand, it seemed that the occurrence of various ploidies in the plants derived from the calluses of same origin was the result of the influence of in vitro culture. Apical meristem tissues and various multicellular bodies were formed in the epidermal and inner mesophyll tissues as well as in the sub-epidermal cells.
