This study was carried out to investigate the effect of sucrose, boric acid, and calcium nitrate in optimum pH and relative humidity on pollen germination and pollen tube growth of boxthorn (Lycium chinense M.). Medium containing 100mg/L of $H_3BO_3$ showed maximum pollen germination and pollen tube elongation, 64.8% and $920{\mu}m$. Additionally supplied calcium nitrate to the medium lowered pollen germination rate and pollen tube elongation. Here we report the optimum conditions for the pollen germination and pollen tube development was observed at 10% sucrose, 1.0% agar, pH 6.0, and 100% relative humidity.
This paper deals with the study on comparative efficacy and in vitro activity of cow urine and cow dung for controlling root rot disease of cucumber caused by Fusarium solani f. sp. cucurbitae Snyder & Hansen following slide germination and mycelial growth inhibition tests. Results showed that both germination of conidia and the percentage inhibition of mycelial growth decreased or suppressed and varied greatly with respect to different hour and days of incubation and kind of bio-matters. In between two bio-matters cow urine was found more effective than that of cow dung in conidial germination. No germination of conidia was recorded after one hour of incubation in any medium whereas in cow urine germination of conidia was not also observed even after 2 hours of incubation. After 7 hours of incubation out of 200 conidia of F. solani f. sp. cucurbitae, 28 in cow urine and 64 in cow dung were germinated while in control a total germinated conidia was 185. In case of percentage inhibition of conidial germination the highest percentage(100%) was recorded in cow urine after 2 hours of incubation followed by 3 hours(96.0%), 4 hours(91.0%) and 6 hours(89.4%). During the test on inhibition of mycelial growth, the highest percentage(62.8%) was recorded in cow urine potato dextrose agar(CUPDA) medium tested after 4 days of incubation, followed by 3 days(60.5%), 5 days(56.5%) and 2 days(55.0%). In this test cow dung potato dextrose agar(CDPDA) had less efficacy in suppression of the percentage inhibition of mycelial growth.
Seed of Codonopsis lanceolata exhibits low germination due to impermeable seed coat. Prolonged seed dormancy can be overcome by the application of gibberellins, as it promotes growth of the embryo and weakens the structures surrounding of embryo. The effects of photoperiod, sugar and gibberellin concentration were investigated at constant temperature for 12 days and 22 days in vitro and invivo conditions respectively. The rate of germination of seeds of Codonopsis lanceolata in wet filter paper in both complete dark and light treatments was significantly lower than that of seed treated with $GA_3$. It clearly indicates that there is significant coat imposed dormancy in the seed of Codonopsis lanceolata. The rate of germination in vivo condition was lower than that of the in vitro condition supplemented with $GA_3$ Thus, the physical dormancy due to impermeable seed coat and low level of endogenous gibberellins in the seed was the cause of poor germination rate in Codonopsis lanceolata.

Background: This study aimed to investigate the effect of cold plasma treatment on the improvement of seed germination and surface sterilization of ginseng seeds. Methods: Dehisced ginseng (Panax ginseng) seeds were exposed to dielectric barrier discharge (DBD) plasma operated in argon (Ar) or an argon/oxygen mixture (Ar/O2), and the resulting germination and surface sterilization were compared with those of an untreated control group. Bacterial and fungal detection assays were performed for plasma-treated ginseng seeds after serial dilution of surface-washed suspensions. The microbial colonies (fungi and bacteria) were classified according to their phenotypical morphologies and identified by molecular analysis. Furthermore, the effect of cold plasma treatment on the in vitro antifungal activity and suppression of Cylindrocarpon destructans in 4-year-old ginseng root discs was investigated. Results: Seeds treated with plasma in Ar or Ar/O2 exhibited a higher germination rate (%) compared with the untreated controls. Furthermore, the plasma treatment exhibited bactericidal and fungicidal effects on the seed surface, and the latter effect was stronger than the former. In addition, plasma treatment exhibited in vitro antifungal activity against C. destructans and reduced the disease severity (%) of root rot in 4-year-old ginseng root discs. The results demonstrate the stimulatory effect of plasma treatment on seed germination, surface sterilization, and root rot disease suppression in ginseng. Conclusion: The results of this study indicate that the cold plasma treatment can suppress the microbial community on the seed surface root rot in ginseng.
Populus. caspica L. is an Iranian indigenous poplar species which naturally distributed in the northern part of country. Unfortunately, overuse has removed many of the stems of better form, so that natural stands now usually appear small and crook. Therefore genetic variation for selection of new superior clone of this species is needed. Conventional hybridization system is currently used to induce genetic variation in poplar species but incompatibility barriers have been observed between them. In vitro ovule embryo culture was used to overcome incompatibility obstacle for interspecific hybridization between Populus caspica L. with Populus deltoids L.62/75. Female flowers of Populus caspica L. have artificially been pollinated with pollen grain of P. deltoides 62/75 in one direction using twig and pot crossing system. Ovaries at different ages (7, 14 and 21 days after pollination) were disinfected through 70% ethanol for 1 minute, 5% of sodium-hypochlorite solution for fifteen min followed by three time rising with sterile distil-water. Isolated ovaries were then transferred to MS hormone free medium containing 30 and 60 g/L sucrose for embryo development and germination. Collected data have been analyzed by two factorial experimental designs. The results indicated that there were significant differences between age of embryos for development and germination at ${\alpha}=0.01%$. Highest embryo germination (45%) was observed from 21 days old ovaries. No significant differences were observed between MS culture media containing 30 and 60 g/L for percentages of ovary-embryo germination and number of germinated embryo per ovary at ${\alpha}=0.05%$. Fourteen percentage of embryo germination obtained in MS medium supplemented with 60 g/L sucrose, while only 35% of isolated ovaries were able to germinate in MS containing 30 g/L sucrose. Induced plantlets in 4 cm height were transferred into pots containing soilless (1:1:1 peat, per lit and vermiculite) medium for acclimatization. After successful acclimatization, plants were delivered to nursery.

This study was conducted to evaluate the performance of Italian ryegrass cultivars for salt tolerance under in vitro condition. Italian ryegrass cultivars such as Greenfarm, Florida80, Kowinearly, and Hwasan101 were tested for their tolerance to various sodium chloride levels (0, 50, 150, 250, and 350 mM). The seed germination, growth, and activities of antioxidant enzymes were investigated under salt treatment. Physiological traits such as seed germination percentage, germination period, shoot and root length, and dry weight were suppressed under entire salt stress conditions. The results indicated that the highest germination percentage and shoot and root length were recorded at normal conditions. Increased sodium chloride levels caused a significant reduction in the seed germination and growth rate. Among the four tested cultivars, Italian ryegrass 'Hwasan101' could be considered as salt tolerant owing to its higher germination percentage, better seedling growth and antioxidant activities under salinity stress, whereas Greenfarm cultivar was more sensitive. The selection of Italian ryegrass cultivars for greater tolerance to saline environment would allow greater productivity from large saline lands.
Eleutherococcus senticosus (also called Acanthopanax senticosus), belonging to Araliaceae family, has been used as an important medicinal woody plant. Mature seeds of Eleutherococcus senticosus have rudimentary (extremely immature) zygotic embryos and require a long-term stratification for about 18 months to induce germination. Here, through the methods of endosperm removal and other exogenous treatments, we investigated the factors for inducing rudimentary embryos by in vitro culture, Rudimentary zygotic embryos in seeds were at globular to heart-shaped stage at about $250{\mu}m$ in length just after harvest of fruits. When the seeds without testa were cultured on 1/2 MS (Murashige and Skoog 1962) medium, they did not germinate regardless of medium and sucrose concentrations but the removal of endosperm tissue markedly stimulated the growth of rudimentary zygotic embryos. The embryo reached ear-lier maturation, once when the endosperm surrounding the rudimentary embryos was removed. Rudimentary zygotic embryos developed cotyledons within 3 weeks of culture after endosperm emoval. However, post-mature zygotic embryos failed to germinate though they were morphologically normal, indicating another dormancy of embryos. $GA_3\;(2.0\;\cal{mg/L})$ and/or charcoal ($0.2\%$) treatment rapidly enhanced the germination of zygotic embryos. These results suggest that E. senticosus seeds have double dormancy; i. e. morphological rudimentary dormancy influenced by surrounding endosperm and physiological dormancy after post-maturation of zygotic embryos. Based on the above findings, we established the rapid germination of rudimentary zygotic embryos by in vitro culture of excised seeds with endosperm removal and $GA_3$ treatment.

Effects of environmental factors on in vitro pine pollen performance were investigated. Pinus densiflora and P. rigida pollen grains collected at Mt. Kwanak, Korea were used. Three environmental factors, such as pollen storage temperature, pollen culture temperature and nutrient condition in medium, were tested. To determine the storage temperature effects on pollen viability, pine pollen was stored at $-70^{\circ}C$, $-12^{\circ}C$, $4^{\circ}C$ and $22^{\circ}C$. Pollen viability was substantially extended at the storage temperatures of $-12^{\circ}C$ and $4^{\circ}C$ for more than 300 days. To elucidate the culture temperature effects on pine pollen germination and tube growth, pollen grains were cultured at the temperatures from $5^{\circ}C$ to $40^{\circ}C$ at $5^{\circ}C$intervals. The germination rate and tube growth were highest at $25^{\circ}C$ and decreased above $30^{\circ}C$. To investigate boron and sucrose effects on pollen tube growth, the pollen was cultured at different sucrose and boric acid concentrations. Germination rate was optimal in germination medium containing 3 or 5$\%$ sucrose with 0.01 $\%$ boric acid. These results indicate that the pine pollen can be stored for considerable length of time without noticeable loss of viability at storage temperature below or near $0^{\circ}C$. Optimal germination medium conditions were established for pine pollen. Therefore, pine pollen can be used for many biological and environmental monitoring researches.
In vitro and in vivo activities of a biocontrol agent, Bacillus stearothermophilus strain YC4194 was evaluated for the control of Pythium damping-off of cucumber. B. stearothermophilus YC4194 inhibited germination of cystospores and formation of zoosporangia of Pythium aphanidermatum in vitro. Incorporation of a bentonite and talc based formulation(10$^{9}$ cfu/g) of B. stearothermophilus YC4194 to the nursery soils (10 g/ι soil) resulted In a significant (p=0.01) reduction in the disease severity of cucumber damping-off after inoculation with P. aphanidermatum. The control efficacy of B. stearothermophilus YC4194 formulation was not different from that of the fungicides, dimethomorph, metalaxyl, ethaboxam. When the cucumber plants were transplanted to the soil inoculated with P. aphanidermatum zoospores, the B. stearothermophilus YC4194 maintained the high population density in rhizosphere soil upto 10$^{7}$ cfu/g until 15 days after treatment.
This study is aimed to understand the effects of temperature, light and GA soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping hentgrass 'Penlinks', tall fescue 'Arid' and Kentucky hluegrass 'Nasaw'. These experiments are carried in vitro, and their results obtained are summarized as the followings ; The optimum temperature for seed germination of perennial ryegrass 'Dandy' was found to he ranged at 20~30$^{\circ}C$, and the optimum temperature for germination of creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw' seeds was found to be ranged at 20~25$^{\circ}C$, whereas the germination percentage under the below or above temperature of this range decreased. The germination of perennial ryegrass 'Dandy' and tall fescue 'Arid' seeds showed no effect on light, and while the germination percentage of creeping hentgrass 'Penlinks' increased more highly under the dark condition than under the light condition. The germination percentage decreased according to the higher temperature. The germination of Kentucky bluegrass 'Nasaw' seeds increased more highly under the dark condition than under the light condition at 20~25$^{\circ}C$, whereas the germination percentage under the dark condition at 15$^{\circ}C$ and 30$^{\circ}C$ decreased. In case of GA soaking treatment of seeds, all the cool season turfgrasses tested showed their own promoting effects on seed germination. The early germination of tall fescue 'Arid' was good without the soaking times. Creeping bentgrass 'Penlinks' and perennial ryegrass 'Dandy' had re-spectively the highest germination percentage at 10min and 1hr soaking treatment, and the longer the soaking time, the lesser the germination percentage. Kentucky bluegrass 'Nasaw' obtained the best results at 24has soaking treatment, and the longer the soaking time, the better the ger-mination percentage.
