Cytotoxicity of Listeriolysin O Produced by Membrane-Encapsulated Bacillus subtilis on Leukemia Cells

  • Encapsulation of biological material in the permiselective membrane allows to construct a system separating cells from their products, which may find biotechnological as well as biomedical applications in biological processes regulation. Application of a permiselective membrane allows avoiding an attack of the implanted microorganisms on the host. Our aim was to evaluate the performance of Bacillus subtilis encapsulated in an elaborate membrane system producing listeriolysin O, a cytolysin from Listeria monocytogenes, with chosen eukaryotic cells for future application in anticancer treatment. The system of encapsulating in membrane live Bacillus subtilis BR1-S secreting listeriolysin O was proven to exert the effective cytotoxic activity on eukaryotic cells. Interestingly, listeriolysin O showed selective cytotoxic activity on eukaryotic cells: more human leukemia Jurkat T cells were killed than human chronic lymphocytic B cells leukemia at similar conditions in vitro. This system of encapsulated B. subtilis, continuously releasing bacterial products, may affect selectively different types of cells and may have future application in local anticancer treatment.

Analgesic and Anti-inflammatory Activity of Resina Pini

  • In this study, we investigated the potential of Resina Pini (RP) for anti-inflammatory and analgesic agents to treat inflammatory diseases such as gingivitis and periodontitis. Crude RP (RP1), recrystallized RP (RP2), and Ramus Mori Albae-treated RP (RP3), plus their respective water extracts (RP1-WE, RP2-WE and RP3-WE) were prepared for in vitro and in vivo tests. We couldn't find any signs of heavy metals pollution in all the RP samples. RP2-WE exhibited the highest viability of human gingival fibroblasts (HGF) and the strongest scavenging activity on superoxide anion. RP1, RP2 and RP3, RP2 showed potent scavenging activity on DPPH free radical. RP2-WE displayed a stronger inhibition on hyaluronidase (HAase) activity and RP3 also displayed potent HAase inhibition. RP2-WE, RP3-WE, RP3 and RP2 were reduced admirably the production of $PGE_2$ in HGF. In addition, RP2-WE and RP3-WE exhibited potent inhibitory activities on arachidonic acid-induced ear edema in mouse. Moreover, RP-2 prevented completely acetic acid-induced writhing by 100.0% and RP1, RP3, RP1-WE and RP2-WE also exhibited excellent protective activities against writhing. While aminopyrine, the positive control, showed 76.9% analgesic effect at the same dose. Taken together, these results suggest that recrystallized aqueous extract of Resina Pini could be a promising drug for the treatment of periodontal diseases.

The Inhibitory Effects of Houttuynia cordata $T_{HUNB}$ against Cadmium induced Cytotoxicity (II) (어성초의 카드뮴에 대한 독성억제효과 (II))

  • This study was condo더ed to investigate the antitoxic component in aqueous extract of Houttuynia cordata $T_{HUNB}$. The results were as follows: Generally, detoxification effects by Houttuynia cordata $T_{HUNB}$ extract increased in proportion to the extract concentrations in rats. When 40 mBtg dosage of Houttuynia cordata $T_{HUNB}$ extract was administrated, Houttynia cordata $T_{HUNB}$ extract showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights increased in proportion to the extract concentrations. However, after 3 weeks, the body weight decreased insignificantly. From the above results, Houttuynia cordate $T_{HUNB}$ extract increased metallothionein concentration and decreased the toxicity of cadmium in rats. In vitro the antitoxic activity of aqueous extract of Houttuynia cordata $T_{HUNB}$ on NIH 373 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetra-zoliumbromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows; The concentration of 10$^{-2}$ mg/ml of Houttuynia cordata $T_{HUNB}$ extract was shown significant antitoxic activity The number of NIH 373 fibroblasts were increased and tend to regenerate. These results suggest that Houttuynia cordata THUNB extract retains a potential antitoxic activity.oxic activity.

  • Gekko has been used for several diseases including cancer in Oriental medicine and fork herbalogy. Nevertheless, its origin as herbal medicine and its efficacy and mechanism as anti-tumor drug have not yet been thoroughly reported in Korea. This study aimed to investigate anti-tumor effect of Gekko through selected articles from cqvip database in China. In vitro and In vivo, Gekko could obviously inhibit tumor growth, induce tumor cells apoptosis, reduce micro-vessel density in tumor tissue through down regulating VEGF & bFGF protein expression, promote cytotoxicity of lymphocyte. Gekko could improve survival rate, relive clinical symptoms, improve quality of life, and relieve anti-tumor treatment reaction, suggesting that Gekko might be a effective anti-tumor drug.

  • Crude extract of young dangyuja (Citrus grandis Osbeck) fruit was investigated for its antioxidant activity as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. Among the samples, including 4 Citrus species and various solvent-extracted-fractions of young dangyuja fruit, the water-extracted fraction (WF) and butanol-extracted fraction (BF) showed the greatest DPPH free radical scavenging activity. WF and BF were further examined for their antioxidant activities by three different in vitro assays. The cell viability tests using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay showed that both fractions significantly reduced $H_2O_2$-induced cytotoxicity in HepG2 cells dose-dependently. Generation of the reactive oxygen species (ROS) was also reduced in cells pretreated with both fractions. In addition, BF showed a higher level of lipid peroxidation inhibitory capacity than WF in $H_2O_2$-treated HepG2 cells. Taken together, these results indicate that young dangyuja fruit can be used as an easily accessible source of natural antioxidants.

Suppression of Human Fibrosarcoma Cell Metastasis by Phyllanthus emblica Extract in Vitro

  • Phyllanthus emblica (PE) is known to exhibit various pharmacological properties. This study aimed to evaluate the antimetastatic potential of a PE aqueous extract. Cytotoxicity to human fibrosarcoma cells, HT1080, was determined by viability assay using the 3-(4,5-dimethylthiazol,2-yl)-2,5-diphenyltetrazolium bromide (MTT) reagent. Cell migration and invasion were investigated using chemotaxis chambers containing membranes precoated with collagen IV and Matrigel, respectively. Cell attachment onto normal surfaces of cell culture plates was tested to determine the cell-adhesion capability. The molecular mechanism of antimetastatic activity was assessed by measuring the gene expression of matrix metalloproteinases, MMP2, and MMP9, using reverse transcription-polymerase chain reaction (RT-PCR) assay. The mRNA levels of both genes were significantly down-regulated after pretreatment with PE extract for 5 days. Our findings show the antimetastatic function of PE extract in reducing cell proliferation, migration, invasion, and adhesion in both dose- and time-dependent manners, especially growth arrest with low $IC_{50}$ value. A decrease in the expression of both MMP2 and MMP9 seems to be the cellular mechanism for antimetastasis in this case. There is a high potential to use PE extracts clinically as an optional adjuvant therapeutic drug for therapeutic intervention strategies in cancer therapy or chemoprevention.

Immunostimulant and Anti-Tumor Activity of Crude Extracts of Galium aparine L. (팔선초 물 추출물의 면역자극 및 항종양 활성)

  • We here demonstrate the evidence of increased anti-tumor and immunostimulating activities of crude extracts (GAL) from Galium. aparine L. In experimental lung metastasis of colon26-M3.1 carcinoma or B16-BL6 melanoma cells, prophylactically intravenous (i.v) administration of GAL significantly inhibited lung metastasis in a dose-dependant manner. In an in vitro cytotoxicity analysis, GAL at the concentration up to $500\;{\mu}g/ml$ did not affect the growth of B16-BL6 melanoma cells. In contrast, GAL showed the enhancement of splenocyte proliferating activity in a dose-dependent manner. Peritoneal macrophages stimulated with GAL produced various cytokines such as $1L-1{\beta},\;TNF-{\alpha},\;IFN-{\gamma}$ and IL-12. These data suggest that GAL has an antitumor activity to inhibit tumor metastasis, and its antitumor effects is associated with activation of nonspecific immnune related cells.

  • Porous Ti compacts were fabricated by spark plasma sintering (SPS) method and their in vitro and in vivo biocompatibilities were investigated. Alkaline phosphatase (ALP) activity representing the activity of osteoblast was increased when osteoblast-like MG-63 cells were cultured on the Ti powder surface. Some genes related to cell growth were over-expressed through microarray analysis. The porous Ti compact with 32.2% of porosity was implanted in the subcutaneous tissue of rats to confirm in vivo cytotoxicity. 12 weeks post-operation, outer surface and inside the porous body was fully filled with fibrous tissue and the formation of new blood vessels were observed. No inflammatory response was confirmed. To investigate the osteoinduction, porous Ti compact was implanted in the femur of NZW rabbits for 4 months. Active in-growth of new bone from the surrounded compact bone was observed around the porous body. From the results, The porous Ti compacts fabricated by spark plasma sintering might be available for the application of the stem part of artificial hip joint.

  • The discovery and development of the taxane class of antitumor compounds represent significant advances in the treatment of patients with a variety of malignancies. These drugs are effectively used in the treatment of breast cancer. In this study we evaluated the efficacy of fractionated usage of both paclitaxel and docetaxel as a single agent in the breast cancer cell line MCF-7. It has been shown that the cytotoxic effect of paclitaxel was increased when the divided $IC_{50}$ concentrations were used sequentially and in contrast to paclitaxel, cytotoxic effect of docetaxel was decreased with the same schema and the single dose of $IC_{50}$ concentration was optimal. The cause of the difference between the cytotoxic effects of two agents with this schedule is obscure. Demonstrating mechanisms, which are responsible for these differences, will be important for more rational use of taxoids and to provide basis for the following clinical trials.
