옥수수 가공방법 및 두께가 in situ 건물 분해율과 in vitro 메탄 발생에 미치는 영향 (Effects of Processing Methods of Corn and their Thickness on in situ Dry Matter Degradability and in vitro Methane Production)

  • 본 연구에서는 옥수수의 가공방법 및 옥수수의 두께의 차이가 반추위 in situ 소화율 및 in vitro 반추위 발효와 메탄 발생에 미치는 영향에 대하여 알아보고자 실시하였다. Micronized corn(2.5 mm thickness) 처리구는 다른 처리구들에 비하여 in situ 48시간 건물 분해율이 모든 배양시간대에서 높게(P<0.05) 나타났다. 반면, steam flaked corn(3.3 mm thickness) 처리구는 모든 배양시간대에서 가장 낮은(P<0.05) in situ 건물 분해율을 보였다. 반추위 내에서 미생물에 의해 분해되는 b fraction은 이와 상반되는 결과인 steam flaked corn(3.1 mm thickness)과 steam flaked corn(3.3 mm thickness) 처리구에서 높게(P<0.05) 나타났다. b fraction이 반추위를 통과하는 속도인 건물 분해상수 k값은 micronized corn(2.5 mm thickness) 처리구에서 가장 높게 (P<0.05) 나타났고 steam flaked corn(3.3 mm thickness) 처리구에서 가장 낮게 (P<0.05) 나타났다. 반추위내 시간당 통과속도를 0.05로 적용한 유효 건물 분해도는 건물 분해상수와 유사하게 micronized corn(2.5 mm thickness) 처리구에서 가장 높게 (P<0.05), steam flaked corn(3.3 mm thickness) 처리구에서 가장 낮게(P<0.05) 나타났다. In vitro 반추위 48시간 건물 소실율은 micronized corn(2.5 mm thickness) 처리구에서 가장 높은 경향을(P=0.088) 보인 반면 steam flaked corn 처리구들 사이에서는 두께에 따른 건물 소실율의 상관관계는 나타나지 않았다. 총 가스발생량 및 메탄 발생량에 있어서도 건물 소실율과 같은 결과인 micronized corn(2.5 mm thickness) 처리구에서 가장 높게 나타났으며(P=0.001), 총 휘발성지방산은 steam flaked corn(2.9 mm thickness)의 처리구에서 가장 높게 나타났으나(P=0.015) 나머지 처리구들에서는 유의적인 차이를 보이지 않았다. aectate: propionate의 비율 또한 steam flaked corn(2.9 mm thickness)의 처리구에서 가장 높게, micronized corn(2.5 mm thickness) 처리구에서 가장 낮게 나타났다(P=0.008). 본 연구의 결과를 종합하면 steam flaked corn 처리구들에서 in vitro 휘발성지방산 농도에 있어 in situ 반추위내 건물 분해상수와 유효 건물 분해도의 결과와 비교하여 상관관계가 나타나지 않은 결과로 유추해 보면 옥수수 가공방법에 의한 두께의 차이는 반추위 메탄 발생량에 영향을 미치지 않은 것으로 판단된다.

Rumen Degradability and Post-ruminal Digestion of Dry Matter, Nitrogen and Amino Acids of Three Protein Supplements

  • This study evaluated the in situ ruminal degradability, and subsequent small intestinal digestibility (SID) of dry matter, crude protein (CP), and amino acids (AA) of cottonseed meal (CSM), sunflower seed meal (SFSM) and distillers dried grains with solubles (DDGS) by using the modified three-step in vitro procedure. The ruminal degradability and subsequent SID of AA in rumen-undegradable protein (RUP-AA) varied among three protein supplements. The result show that the effective degradability of DM for SFSM, CSM, and DDGS was 60.8%, 56.4%, and 41.0% and their ruminal fermentable organic matter was 60.0%, 55.9%, and 39.9%, respectively. The ruminal degradable protein (RDP) content in CP for SFSM, CSM, and DDGS was 68.3%, 39.0%, and 32.9%, respectively, at the ruminal solid passage rate of 1.84%/h. The SFSM is a good source of RDP for rumen micro-organisms; however, the SID of RUP of SFSM was lower. The DDGS and CSM are good sources of RUP for lambs to digest in the small intestine to complement ruminal microbial AA of growing lambs. Individual RUP-AA from each protein source was selectively removed by the rumen microorganisms, especially for Trp, Arg, His, and Lys (p<0.01). The SID of individual RUP-AA was different within specific RUP origin (p<0.01). Limiting amino acid was Leu for RUP of CSM and Lys for both RUP of SFSM and DDGS, respectively. Therefore, different protein supplements with specific limitations should be selected and combined carefully in growing lambs ration to optimize AA balance.

The Effect of Yerba Mate (Ilex Paraguariensis) Supplementation on Nutrient Degradability in Dairy Cows: An In sacco and In vitro Study

  • This study was carried out to investigate the effects of Yerba Mate (YM) supplementation on nutrients' degradation, in vitro dry matter disappearance, gas production and rumen ammonia concentration. Three rumen-fistulated Holstein Friesian cows were used for the in situ incubations and provided rumen liquor for in vitro incubations. The inclusion of YM in a control diet (pasture+pellets) affected some in sacco degradation parameters. YM supplementation decreased the effective degradability and degradation rate of pasture crude protein (CP), and it seems to slow down the degradation of pasture neutral detergent fiber. A significant increase of degradation of pasture acid detergent fiber (ADF) was detected after YM inclusion in the control diet. YM supplementation reduced in vitro gas production of pasture and ammonia concentration of pellets. The addition of YM in ruminant diet could decrease ammonia production and increase protein availability for productive purposes. The moderate presence of tannins in YM could have affected the degradation kinetics of pasture CP and ADF and the ammonia production of pellets.

Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

  • Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.

Effects of Protein Supplement Sources on Digestibility of Nutrients, Balance of Nitrogen and Energy in Goats and Their In Situ Degradability in Cattle

  • The experiment was conducted to determine in situ rumen degradability of soybean meal (SM), fish meal (FM), sesame cake (SC) and Italian ryegrass hay (IRGH) and the effect of supplementing the above protein meals to IRGH on digestibility and balance of nutrients in three Saanen goats. For measuring the degradability, nylon bags containing each meal were incubated in the rumen of one fistulated dry cow for 3, 6, 12, 24, 36, and 48 hrs. Degradability revealed that SM protein was highly degradable in the rumen (99.1%), while FM protein was less degradable (76.8%) with SC protein being intermediate (91.2%) at 48 hrs of incubation (p < 0.01). Degradation rate of the potentially degradable fraction was estimated to be 12.12, 5.88 and 5.88%/hr for SM, FM and SC, respectively. In the metabolism trial, all goats were offered daily 900 g IRGH and one of the supplements, SM (100 g), FM (75 g) or SC (100 g). Intake of DM, OM, CP and GE was similar among diets. However, digestibility of OM for SC diet was significantly lower than that for diets supplemented with FM and SM (p < 0.10). Nitrogen (N) excreted in faeces (p < 0.05) and in urine (p < 0.10) was, respectively, higher and lower for SC diet than that for the other two diets. The same tendency was observed in energy losses in faeces (p < 0.10) and in urine (p < 0.05). There was no difference in energy loss in methane or in heat production among diets. Consequently, no significant difference was observed in N retention (2.13, 0.42 and -0.11 g/day for FM, SC and SM diet, respectively) or in energy retention (-1.49, -2.14 and -2.70 MJ/day for FM, SM and SC diet, respectively). These results showed that protein supplements affected the digestion of diets based on grass hay with 7.45% CP in DM in goats, although there was no significant influence on N or energy retention.

느타리, 새송이 및 팽이버섯 폐배지의 반추동물 조사료원으로서의 사료 영양적 가치평가 (Evaluation on Feed-Nutritional Value of Spent Mushroom(Pleurotus osteratus, Pleurotus eryngii, Flammulina velutupes) Substrates as a Roughage Source for Ruminants)

  • 본 연구에서는 느타리, 새송이 및 팽이버섯 폐배지를 버섯 종류별, 배지에 이용된 주원료별로 나누어 반추가축 사료로서의 영양적 가치를 구명하기 위해서 화학성분, in vitro 소실율 실험 및 in situ darcon bag 실험을 수행하였다. 버섯 폐배지는 고섬유소(NDF 64~78%), 저단백질(CP 7~11%) 조사료원에 속하였다. 주원료별 폐배지는 섬유소 성분이 가장 높은 톱밥 주원료 폐배지와 비교해서 폐면 주원료 폐배지는 NDF 함량이 낮고(P<0.05), NFC와 ash 함량이 높았다(P<0.05). 버섯 폐배지의 화학적 성분은 전반적으로 버섯 종류보다는 주원료 종류에 의해 주로 영향을 받았다. In vitro DM, NDF 소실율은 분석된 배양시간(24, 48, 72시간) 모두에서 옥공이, 폐면, 톱밥 주원료 폐배지 순으로 높게 나타났다. In situ DM, NDF, ADF 소실율은 배양 24시간에서 옥공이, 폐면, 톱밥 주원료 폐배지 순으로 높았다(P<0.05). 톱밥 주원료 폐배지와 비교해서 폐면 주원료 폐배지는 가소화성 분획(NDF, ADF)이 뚜렷이 높고(P<0.05) 비소화성 분획은 현저히 낮았으며(P<0.05), 그 결과 배양 72시간에서의 in situ DM, NDF 소실율(disappearance)과 예상 반추위 분해율(degradability)도 현저히 높았다(P<0.05). 결과적으로 폐면 주원료 폐배지는 톱밥 주원료 폐배지 보다 반추가축 조사료원으로서의 사료영양적 가치가 높아 우선적 이용이 권장된다.

옥수수의 steam flake 처리가 in situ 건물소화율 및 저장기간별 aflatoxin과 ochratoxin 함량에 미치는 영향 (Effects of Steam Flaking on In situ DM Digestibility and Aflatoxin and Ochratoxin Contents during Storage of Corns)

  • 본 연구는 옥수수 수입원과 steam flake 처리에 따른 in situ 건물 분해율 및 보관기간별 mycotoxins 함량을 조사하기 위하여 수행되었다. 미국과 인도산 옥수수(yellow dent corn)를 공시재료로 하여, steam flake 처리 효과에 대해 조사하였다. 시험구는 총 4개의 처리구(USCW, 미국산 무처리 옥수수; USCF, 미국산 steam flaking 옥수수; IDCW, 인도산무처리 옥수수; IDCF, 인도산 steam flaking 옥수수)를 두고, 각 처리구당 보관기간(0, 1, 2, 3, 4, 5, 6, 7, 8주)별 4반복으로 수행되었다. In situ 시험을 위해서 반추위 누관이 장착된 Holstein 수소 2두가 이용되었고, 공장환경분야 표준상태 (STP, standard temperature and pressure, $25^{\circ}C$ 1 atm.)에서 보관기간 동안 매주, 곰팡이 독소인 aflatoxin과 ochratoxin의 농도를 측정하였다. 옥수수의 반추위 내 발효시간별 in situ 건물 소실율은 무처리구에서는 미국산이 인도산보다 약 $3.0{\sim}44.1%$ 높았으나, steam flaking 처리를 함으로써 건물소실율의 차이가 없어졌다. 미국산 옥수수는 무처리 알곡의 건물 분해율이 높은 편이었으나, flake처리에 의한 영향이 적은 반면, 인도산 옥수수는 무처리 알곡의 건물 분해율은 낮은 편이지만 flake 처리를 하였을 때, 분해율이 크게 증가되었다. 미국산 옥수수와 flake 처리 옥수수의 aflatoxin 함량은 보관기간 동안 매우 낮은 수준이었으나, 인도산 옥수수 알곡을 6주 이상 보관할 경우, 허용기준치를 초과하였다. 따라서 인도산 옥수수 알곡을 장기보관 해야 할 경우, steam flake 처리를 하는 것이 aflatoxin 오염방지를 위해서 필수적이며, 알곡으로 보관할 경우는 6주 이상을 넘기지 않는 것이 중요할 것으로 사료된다.

쌀, 보리, 옥수수의 반추위내 In situ 및 In vitro 분해율, 미생물 성장과 Gas 발생량에 대한 연구 (Studies on In situ and In vitro Degadabilities, Microbial Growth and Gas Production of Rice, Barley and Corn)

  • 본 시험은 반추위 누관이 시술된 한우 수소 3두를 이용하여 쌀, 옥수수 및 보리의 in situ 분해율과 in vitro 시험으로서 영양소 분해율, 미생물 성장량, pH 변화 및 gas 발생량을 비교분석함으로써 쌀의 사료적 이용성을 평가해 보고자 수행되었다.In situ 건물 및 유기물 소실율은 반추위내 24시간 발효 시, 보리와 쌀은 거의 대부분이 소실된 반면, 옥수수는 약 67%만 소실되어 분해 속도가 매우 느린 것으로 나타났다. 수용성 물질인 ‘a’ 부분은 옥수수, 쌀, 보리 순으로 많았으며, 천천히 분해되는 ‘b’ 부분은 ‘a’ 부분과 정 반대의 경향으로서 ‘b’ 부분의 분해 상수(‘c’ 부분)로부터 시간당 건물과 유기물의 분해속도는 보리의 경우 각각 38.3%, 37.2%로서 쌀(7.7%, 5.6%)이나 옥수수(4.1%, 1.3%)에 비해 매우 빠르게 일어났다. In vitro 건물 및 유기물 분해율은 in situ 분해율에 비해 수준은 현저히 낮았으나, 보리, 쌀, 옥수수 순으로 높아 곡류 간 순서는 일치하였다. In vitro 미생물 성장은 쌀이 옥수수나 보리보다 상대적으로 낮았으나, 가스 발생량은 높게 나타났으며, pH는 발효시간에 따른 저하속도가 쌀에서 가장 느린 것으로 나타났다. 발효시간에 따른 미생물 성장량은 발효 12시간까지 높아졌다가 이후로 감소하였고, 배양액의 pH는 발효초기에는 높았다가 이후로 점차 감소하였으며, gas 발생량은 발효 9시간까지는 빠르게 증가하였다가 이후로 증가폭이 둔화되었다.이상의 결과에서 반추위내 건물과 유기물의 분해율과 발효시간의 경과에 따른 pH의 저하속도 등을 고려하면, 쌀은 옥수수와 함께 반추위내 발효 안정을 위한 에너지원으로서 사용 할 수 있을 것으로 사료된다.

Chemical Composition, In vitro Gas Production, Ruminal Fermentation and Degradation Patterns of Diets by Grazing Steers in Native Range of North Mexico

  • The objective of the study was to quantify annual and seasonal differences in the chemical composition, in vitro gas production, in situ degradability and ruminal fermentation of grazing steers… diets. Diet samples were collected with four esophageal cannulated steers ($350{\pm}3$ kg BW); and four ruminally cannulated heifers ($342{\pm}1.5$ kg BW) were used to study the dry matter degradation and fermentation in rumen. Data were analyzed with repeated measurements split plot design. The crude protein, in vitro dry matter digestibility and metabolizable energy were higher during the first year of trial and in the summer (p<0.01). The values of calcium, phosphorus, magnesium, zinc and copper were higher in summer (p<0.05). The gas produced by the soluble and insoluble fractions, as well as the constant rate of gas production were greater in summer and fall (p<0.01). The ammonia nitrogen ($NH_3N$) and total volatile fatty acids concentrations in rumen, the soluble and degradable fractions, the constant rate of degradation and the effective degradability of DM and NDF were affected by year (p<0.05) and season (p<0.01). Our study provides new and useful knowledge for the formulation of protein, energetic and mineral supplements that grazing cattle need to improve their productive and reproductive performance.

Estimation of Nutritive Value of Whole Crop Rice Silage and Its Effect on Milk Production Performance by Dairy Cows

  • The nutritive value and utilization of whole crop rice silage (WCRS), Hamasari, at yellow mature stage was determined by three studies. In first study, chemical composition, in vivo digestibility and metabolizable energy (ME) content of WCRS was determined by Holstein steers. WCRS contains 6.23% CP, its digestibility is 48.4% and estimated TDN is 56.4%. Its ME content was 1.91 Mcal/kg DM. Gross energy (GE) retention (% of GE intake) in steers is only 22.7% most of which was lost through feces (44.7% of GE intake). It takes 81 minutes to chew a kg of WCRS by steers. In another study, the effect of Hamasari at yellow mature stage at three stages of lactation (early, mid and late lactation) and two levels of concentrate (40 or 60%) on voluntary intake, ME content and ME intake, milk yield and composition using lactating Holstein dairy cows were investigated. Total intake increased with the concentrate level in early and mid lactation, but was similar irrespective of concentrate level in late lactation. WCRS intake was higher with 40% concentrate level than with 60% concentrate. ME intake by cows increased with the concentrate level and WCRS in early lactating cows with 40% concentrate can support only 90% of the ME requirement. Milk production in accordance with ME intake increased with the increase in concentrate level in early and mid lactating cows but was similar in late lactating cows irrespective of concentrate level. Fat and protein percent of milk in mid and late lactating cows were higher with for 60% concentrate than 40%, but reverse was in early lactating cows. Solids-not-fat was higher with for 60% concentrate than 40% concentrate. Finally in situ degradability of botanical fractions such as leaf, stem, head and whole WCRS, Hamasari at yellow mature stage was incubated from 0 to 96 h in Holstein steers to determine DM and N degradability characteristics of botanical fractions and whole WCRS. Both DM and N solubility, rate of degradation and effective degradability of leaf of silage was lower, but slowly degradable fraction was higher compared to stem and head. Solubility of DM and N of stem was higher than other fractions. The 48 h degradability, effective degradability and rate of degradation of leaf were always lower than stem or head. In conclusion, voluntary intake of silage ranged from 5 to 12 kg/d and was higher with low levels of concentrate, but milk yield was higher with high levels of concentrate. Fat corrected milk yield ranged from 19 to 37 kg per day. For consistency of milk, early lactating cows should not be allowed more than 40% whole crop rice silage in the diet, but late lactating cows may be allowed 60% whole crop rice silage.
